2Quantitative analysis of ruminal methanogenic microbial populations in beef cattle divergent in phenotypic residual feed intake(RFI)offered contrasting diets显示文摘Background:Methane(CH_4) emissions in cattle are an undesirable end product of rumen methanogenic fermentative activity as they are associated not only with negative environmental impacts but also with reduced host feed efficiency.The aim of this study was to quantify total and specific rumen microbial methanogenic populations in beef cattle divergently selected for residual feed intake(RFI) while offered(i) a low energy high forage(HF) diet followed by(ii) a high energy low forage(LF) diet.Ruminal fluid was collected from 14 high(H)and 14 low(L) RFI animals across both dietary periods.Quantitative real time PCR(qRT-PCR) analysis was conducted to quantify the abundance of total and specific rumen methanogenic microbes.Spearman correlation analysis was used to investigate the association between the relative abundance of methanogens and animal performance,rumen fermentation variables and diet digestibility.Results:Abundance of methanogens,did not differ between RFI phenotypes.However,relative abundance of total and specific methanogen species was affected(P< 0.05) by diet type,with greater abundance observed while animals were offered the LF compared to the HF diet.Conclusions:These findings suggest that differences in abundance of specific rumen methanogen species may not contribute to variation in CH_4 emissions between efficient and inefficient animals,however dietary manipulation can influence the abundance of total and specific methanogen species.Ciara A Carberry David A Kenny Alan K Kelly Sinead M Waters 2014Journal of Animal Science and Biotechnology2014,5,4:6

Recently-published guidelines bring much needed structure to the diagnostic work-up of patients with suspected chronic pancreatitis. In addition, novel diagnostic modalities bring promise for the future. 4Assessment of serum angiogenic factors as a diagnostic aid for small bowel angiodysplasia in patients with obscure gastrointestinal bleeding and anaemia显示文摘AIM To assess the use of serum levels of angiopoietin-1(Ang1), Ang2 and tumor necrosis factor-α(TNFα) as predictive factors for small bowel angiodysplasia(SBA).METHODS Serum samples were collected from patients undergoing capsule endoscopy for any cause of obscure gastrointestinal bleeding(OGIB) or anaemia. Based on small bowel findings patients were divided into 3 groups:(1) SBA;(2) other bleeding causes; and(3) normal, according to diagnosis. Using ELISA technique we measured serum levels of Ang1, Ang2 and TNFα and compared mean and median levels between the groups based on small bowel diagnosis. Using receiver operator curve analysis we determined whether any of the factors were predictive of SBA.RESULTS Serum samples were collected from a total of 120 patients undergoing capsule endoscopy for OGIB or anaemia: 40 with SBA, 40 with other causes of small bowel bleeding, and 40 with normal small bowel findings. Mean and median serum levels were measured and compared between groups; patients with SBA had significantly higher median serum levels of Ang2(3759 pg/mL) compared to both other groups, with no significant differences in levels of Ang1 or TNFα based on diagnosis. There were no differences in Ang2 levels between the other bleeding causes(2261 pg/mL) and normal(2620pg/mL) groups. Using Receiver Operator Curve analysis, an Ang2 level of > 2600 pg/mL was found to be predictive of SBA, with an area under the curve of 0.7. Neither Ang1 or TNFα were useful as predictive markers.CONCLUSION Elevations in serum Ang2 are specific for SBA and not driven by other causes of bleeding and anaemia. 8Residual feed intake phenotype and gender affect the expression of key genes of the lipogenesis pathway in subcutaneous adipose tissue of beef cattle显示文摘Background: Feed accounts for up to 75% of costs in beef production systems,thus any improvement in feed efficiency(FE) will benefit the profitability of this enterprise.Residual feed intake(RFI) is a measure of FE that is independent of level of production.Adipose tissue(AT) is a major endocrine organ and the primary metabolic energy reservoir.It modulates a variety of processes related to FE such as lipid metabolism and glucose homeostasis and thus measures of inter-animal variation in adiposity are frequently included in the calculation of the RFI index.The aim of this study was to determine the effect of phenotypic RFI status and gender on the expression of key candidate genes related to processes involved in energy metabolism within AT.Dry matter intake(DMI) and average daily gain(ADG) were measured over a period of 70 d for 52 purebred Simmental heifers(n = 24) and bulls(n = 28) with an initial BW±SD of 372±39.6 kg and 387±50.6 kg,respectively.Residual feed intake was calculated and animals were ranked within gender by RFI into high(inefficient; n = 9 heifers and n = 8 bulls)and low(efficient; n = 9 heifers and n = 8 bulls) groups.Results: Average daily gain ±SD and daily DMI ±SD for heifers and bul s were 1.2±0.4 kg and 9.1±0.5 kg,and 1.8±0.3 kg and 9.5±1 kg respectively.High RFI heifers and bulls consumed 10% and 15% more(P < 0.05) than their low RFI counterparts,respectively.Heifers had a higher expression of all genes measured than bulls(P < 0.05).A gender × RFI interaction was detected for HMGCS2(P < 0.05) in which high RFI bulls tended to have lower expression of HMGCS2 than low RFI bulls(P < 0.1),whereas high RFI heifers had higher expression than low RFI heifers(P < 0.05) and high RFI bulls(P < 0.05).SLC2 A4 expression was consistently higher in subcutaneous AT of low RFI animals across gender.Conclusion: The findings of this study indicate that low RFI cattle exhibit upregulation of the molecular mechanisms governing glucose metabolism in adipose tissue,in particular,glucose clearance.The decreased expression of SLC2 A4 in the inefficient cattle may result in less efficient glucose metabolism in these animals.We conclude that SLC2 A4 may be a potential biomarker for RFI in cattle.Clare McKenna Richard K.Porter Kate A.Keogh Sinead M.Waters Mark McGee David A.Kenny 2019Journal of Animal Science and Biotechnology2019,10,1:2

10Three paralogous clusters of the miR-17-92 family of microRNAs restrain IL-12-mediated immune defense显示文摘MicroRNAs(miRNAs)have been widely implicated in immune regulation,but evidence for the coordinated function of paralogous miRNA clusters remains scarce.Here,by using genetically modified mice with individual or combined cluster deficiencies,we found that three paralogous clusters of the miR-17-92 family of miRNAs collectively suppressed IL-12 production in macrophages.Accordingly,miR-17-92 family miRNAs deficiencies resulted in heightened production of IL-12 and thus enhanced the host defense against intracellular pathogen Listeria monocytogenes in vivo.Mechanistically,different members of the miR-17-92 family of miRNAs acted on a common target,PTEN,to inhibit IL-12 expression by modulating the PI3K-Akt-GSK3 pathway.In addition,the expression of miR-17-92 family miRNAs was collectively inhibited by the transcription factor RBP-J,and RBP-J-associated macrophage functional defects were genetically rescued by deleting three clusters of miR-17-92 family miRNAs on a RBP-J null background.Thus,our results illustrated key roles of three clusters of miR-17-92 family miRNAs in cooperatively controlling IL-12-mediated immune responses and identified miR-17-92 family miRNAs as functional targets of RBP-J in macrophages.Xiang Zhang Sinead MSmith Xi Wang Baohong Zhao Li Wu Xiaoyu Hu 2021Cellular & Molecular Immunology2021,18,7:2
11Timing of surgical repair of bile duct injuries after laparoscopic cholecystectomy: A systematic review显示文摘BACKGROUND The surgical management of bile duct injuries(BDIs)after laparoscopic cholecystectomy(LC)is challenging and the optimal timing of surgery remains unclear.The primary aim of this study was to systematically evaluate the evidence behind the timing of BDI repair after LC in the literature.AIM To assess timing of surgical repair of BDI and postoperative complications.METHODS The MEDLINE,EMBASE,and The Cochrane Library databases were systematically screened up to August 2021.Risk of bias was assessed via the Newcastle Ottawa scale.The primary outcomes of this review included the timing of BDI repair and postoperative complications.RESULTS A total of 439 abstracts were screened,and 24 studies were included with 15609 patients included in this review.Of the 5229 BDIs reported,4934(94%)were classified as major injury.Timing of bile duct repair was immediate(14%,n=705),early(28%,n=1367),delayed(28%,n=1367),or late(26%,n=1286).Standardization of definition for timing of repair was remarkably poor among studies.Definitions for immediate repair ranged from<24 h to 6 wk after LC while early repair ranged from<24 h to 12 wk.Likewise,delayed(>24 h to>12 wk after LC)and late repair(>6 wk after LC)showed a broad overlap.CONCLUSION The lack of standardization among studies precludes any conclusive recommendation on optimal timing of BDI repair after LC.This finding indicates an urgent need for a standardized reporting system of BDI repair.Patryk Kambakamba Sinead Cremen Beat Möckli Michael Linecker 2022World Journal of Hepatology2022,14,2:2
