| 1 | A telomere-to-telomere gap-free reference genome of watermelon and its mutation library provide important resources for gene discovery and breeding显示文摘Watermelon,Citrullus lanatus,is the world's third largest fruit crop.Reference genomes with gaps and a narrow genetic base hinder functional genomics and genetic improvement of watermelon.Here,we report the assembly of a telomere-to-telomere gap-free genome of the elite watermelon inbred line G42 by incorporating high-coverage and accurate long-read sequencing data with multiple assembly strategies.All 11 chromosomes have been assembled into single-contig pseudomolecules without gaps,representing the highest completeness and assembly quality to date.The G42 reference genome is 369321829 bp in length and contains 24205 predicted protein-coding genes,with all 22 telomeres and 11 centromeres characterized.Furthermore,we established a pollen-EMS mutagenesis protocol and obtained over 200000M1 seeds from G42.In a sampling pool,48 monogenic phenotypic mutations,selected from 223M1and 78 M2 mutants with morphological changes,were confirmed.The average mutation density was 1 SNP/1.69Mband1 indel/4.55 Mb per M1 plant and 1SNP/1.08Mb and 1 indel/6.25 Mb per M2 plant.Taking advantage of the gap-free G42 genome,8039 mutations from 32 plants sampled from M1 and M2 families were identified with 100%accuracy,whereas only 25% of the randomly selected mutations identified using the 97103v2 reference genome could be confirmed.Using this library and the gap-free genome,two genes responsible for elongated fruit shape and male sterility(CiMs1)were identified,both caused by a single basechange from G to A.The validated gap-free genome and its EMS mutation library provide invaluable resources for functional genomics and genetic improvement of watermelon. | Yun Deng Shoucheng Liu Yilin Zhang Jingsheng Tan Xiaopeng Li Xiao Chu Binghua Xu Yao Tian Yudong Sun Bosheng Li YunbiXu Xing Wang Deng Hang He Xingping Zhang | 2022 | Molecular Plant2022,15,8: | 9 |
| 2 | Isolation,Identification,and Characterization of an Aluminum-Tolerant Bacterium Burkholderia sp.SB1 from an Acidic Red Soil显示文摘Aluminum(Al) toxicity usually occurs in acidic soils worldwide, which is detrimental to the growth of organisms. An Al-tolerant bacterium, SB1, was isolated from an acidic red soil of Chingkang Mountain, located in Jiangxi Province of China. Polyphasic analysis,including a 16 S rDNA phylogenetic tree as well as morphological and physicochemical properties, revealed that the isolate was a gramnegative, rod-shaped bacterium, which was recognized as Burkholderia sp. SB1 and had extreme acidity tolerance(pH 2.2) and excellent Al resistance(270 mg L^(-1) Al^(3+)). It could remove Al by up to 97.7% at a concentration of 54 mg L^(-1) Al^(3+). SB1 behavior under different temperatures and antibiotics was also examined. SB1 preferred moderate temperature conditions, ranging from 25 to37?C, and exhibited notable resistance to multiple antibiotics(including ampicillin, streptomycin, and tetracycline), except for being sensitive to chloramphenicol. Therefore, as the first reported bacterium to possess favorable Al resistance and excellent Al removal,Burkholderia sp. It is discussed the relationship between the subtropical anticyclone and subtropical easterlies to aim at the problem of high temperature and drought in the globe,further more,the comparison analysis has been made to the earth nutation and the variation of rotation. | 4 | Discussion on Super High-temperature Weather and City Construction显示文摘With the method of structure analysis, this work analyzes the real data of the high-temperature weather of China in 2003 and those in history, and finds out that the structure character of the high-temperature weather process corresponds to the distribution of urban buildings. The result shows that excessive dense buildings could influence the atmosphere structure, which leads to the urban temperature increasing sharply. | 5 | Lithosphere architecture characterized by crust±mantle decoupling controls the formation of orogenic gold deposits显示文摘This study,via combined analysis of geophysical and geochemical data,reveals a lithospheric architecture characterized by crust-mantle decoupling and vertical heat-flow conduits that control orogenic gold mineralization in the Ailaoshan gold belt on the southeastern margin of Tibet.The mantle seismic tomography indicates that the crust-mantle decoupled deformation,defined from previous seismic anisotropy analysis,was formed by upwelling and lateral flow of the asthenosphere,driven by deep sub duction of the Indian continent,Our magnetotelluric and seismic images show both a vertical conductor across the Moho and high Vp/Vs anomalies both in the uppermost mantle and lowest crust,suggesting that crust-mantle decoupling promotes ponding of mantle-derived basic melts at the base of the crust via a heat-flow conduit.Noble gas isotope and halogen ratios of gold-related ore minerals indicate a mantle source of ore fluid.A rapid decrease in Cl/F ratios of lamprophyres under conditions of 1.2 GPa and1050℃suggests that the ore fluid was derived from degassing of the basic melts.Similar lithospheric architecture is recognized in other orogenic gold provinces,implying analogous formational controls. | Zengqian Hou Qingfei Wang Haijiang Zhang Bo Xu Nian Yu Rui Wang David IGroves Yuanchuan Zheng Shoucheng Han Lei Gao Lin Yang | 2023 | National Science Review2023,10,3: | 2 |

| 11 | Photoluminescence properties and thermal stability of Eu^(3+)-activated La_(7)Ta_(3)W_(4)O_(30) red-emitting phosphors for near-UV-excited w-LEDs显示文摘Novel trivalent europium(Eu^(3+))-activated La_(7)Ta_(3)W_(4)O_(30):xEu^(3+)(x=0.5 mol%-40 mol%) red-emitting phosphors were synthesized by means of a high-temperature solid-state reaction.The structure,morphology,photoluminescence,thermal-stability properties,lifetime,and color-rendering of the prepared phosphors were investigated in detail.The La_(7)Ta_(3)W_(4)O_(30):Eu^(3+) phosphors show five emission peaks under near-ultraviolet(n-UV) at 397 nm,and these peaks are ascribed to the transitions of ^(5)D0-^(7)Fj(j=0,1,2,3 and 4) by Eu^(3+) ions.The optimal doping concentration of Eu^(3+) is 20 mol%,and the critical distance of the energy transfer between the Eu^(3+)ions was calculated to be 1.768 nm.The quenching temperature(T_(0.5)) of La_(7)Ta_(3)W_(4)O_(30):20 mol%Eu^(3+) is about 440 K.The quantum yield(QY) was measured to be 85.85%.The fabricated white-light-emitting diodes(w-LEDs) possess high color-rendering index(R_(a)) of 90,and high correlated color temperature(CCT) of 5810 K,respectively.The Commission Internationale de L’Eclairage(CIE) coordinates are(0.311,0.322).Therefore,the prepared phosphor has a promising application for w-LEDs. | Yu Ren Bin Deng Ting Liang Shuang Shu Junlan Guo Shoucheng Zhao Ruijin Yu | 2021 | Journal of Rare Earths2021,39,8: | 1 |

| 19 | Identification and analysis of mouse non-coding RNA using transcriptome data显示文摘Transcripts are expressed spatially and temporally and they are very complicated, precise and specific; however, most studies are focused on protein-coding related genes. Recently, massively parallel c DNA sequencing(RNA-seq) has emerged to be a new and promising tool for transcriptome research, and numbers of non-coding RNAs, especially linc RNAs, have been widely identified and well characterized as important regulators of diverse biological processes. In this study, we used ultra-deep RNA-seq data from 15 mouse tissues to study the diversity and dynamic of non-coding RNAs in mouse. Using our own criteria, we identified totally 16,249 non-coding genes(21,569 non-coding RNAs) in mouse. We annotated these non-coding RNAs by diverse properties and found non-coding RNAs are generally shorter, have fewer exons, express in lower level and are more strikingly tissue-specific compared with protein-coding genes. Moreover, these non-coding RNAs show significant enrichment with transcriptional initiation and elongation signals including histone modifications(H3K4me3, H3K27me3 and H3K36me3), RNAPII binding sites and CAGE tags. The gene set enrichment analysis(GSEA) result revealed several sets of linc RNAs associated with diverse biological processes such as immune effector process, muscle development and sexual reproduction.
