| 1 | ACSL4 is a predictive biomarker of sorafenib sensitivity in hepatocellular carcinoma显示文摘Sorafenib is the first-line treatment of advanced hepatocellular carcinoma(HCC).However,there is a lack of validated biomarkers to predict sorafenib sensitivity.In this study we investigated the role of ACSL4,a positive-activating enzyme of ferroptosis,in sorafenib-induced cell death and HCC patient outcome.We showed that ACSL4 protein expression was negatively associated with IC50 values of sorafenib in a panel of HCC cell lines(R=−0.952,P<0.001).Knockdown of ACSL4 expression by specific siRNA/sgRNA significantly attenuated sorafenib-induced lipid peroxidation and ferroptosis in Huh7 cells,and also rescued sorafenib-induced inhibition of xenograft tumor growth in vivo.We selected 29 HCC patients with surgery as primary treatment and sorafenib as postoperative adjunct therapy from a hospital-based cohort.A high proportion(66.7%)of HCC patients who had complete or partial responses to sorafenib treatment(according to the revised RECIST guideline)had higher ACSL4 expression in the pretreated HCC tissues,compared with those who had stable or progressed tumor growth(23.5%,P=0.029).Since ACSL4 expression was independent of sorafenib treatment,it could serve as a useful predictive biomarker.Taken together,this study demonstrates that ACSL4 is essential for sorafenib-induced ferroptosis and useful for predicting sorafenib sensitivity in HCC.This study may have important translational impacts in precise treatment of HCC. | Ji Feng Pei-zhi Lu Guang-zhi Zhu Shing Chung Hooi Yong Wu Xiao-wei Huang Hui-qi Dai Pan-hong Chen Zhong-jie Li Wen-jing Su Chuang-ye Han Xin-ping Ye Tao Peng Jing Zhou Guo-dong Lu | 2021 | Acta Pharmacologica Sinica2021,42,1: | 11 |

| 6 | Revisiting an ancient inorganic aggregation-induced emission system:An enlightenment to clusteroluminescence显示文摘Organic and inorganic clusteroluminescence have attracted great attention while the underlying mechanisms is still not well understood.Here,we employed a series of ancient inorganic complexes platinocyanides with aggregation-induced emission property to elucidate the mechanism of clusteroluminescence including how does the chromophore form and how does the solid structures influence the luminescence behaviors.The results indicate that the isolated platinocyanide cannot work as a chromophore to emit visible light,while their clusterization at aggregate state can trigger the d-orbitals coupling of the platinum atoms to facilitate the electron exchange and delocalization to form a new chromophore to emit visible light.Furthermore,the counter ions and H2O ligands help to rigidify the three-dimensional network structure of the platinocyanides to suppress the excited state nonradiative decay,resulting in the high quantum yield of up to 96%.This work fundamentally helps understanding both the organic and inorganic clusteroluminescence phenomenon. | Zheng Zhao Zaiyu Wang Javad Tavakoli Guogang Shan Jianyu Zhang Chen Peng Yu Xiong Xuepeng Zhang Tsz Shing Cheung Youhong Tang Bolong Huang Zhaoxun Yu Jacky W.Y.Lam Ben Zhong Tang | 2021 | Aggregate2021,2,2: | 1 |

| 20 | High Microplastic Contamination in Juvenile Tri-Spine Horseshoe Crabs:A Baseline Study of Nursery Habitats in Northern Beibu Gulf,China显示文摘Microplastics(MPs)are ubiquitous in marine sedimentary environments.Their occurrence in horseshoe crabs and interactions with the sedimentary environment has not been determined.In this study,MPs,mostly microfibers,were found in all extracted gastrointestinal tract(GIT)samples of the juvenile Tachypleus tridentatus from the northern Beibu Gulf,China.The MP concentrations(4–53 items ind-1)were higher than those in most marine benthic invertebrates(<15 items ind-1)reported in Chinese waters,despite their generally low level in habitat sediments(9–1818 items kg-1).The correlation between the juveniles and sediments was not evident,but the abundance in juvenile horseshoe crab GITs decreased with ages.The findings were relevant to the life-history characteristics of the species,typically with limited dispersal capability and their spending nine years or longer time living in mangrove wetlands during low tides,with apparent ontogenetic changes in their diets.These baseline data enable a better understanding of MP availability in benthic macroinvertebrates,and the ecological risks present in the ecosystems. | WANG Xueping LO Hoi Shing FU Yijian WU Zhou QIN Danmei HUANG Xing ZHU Jingmin CHEUNG Siu Gin KWAN Kit Yue | 2022 | Journal of Ocean University of China2022,21,3: | 0 |
