1Olfactory ecto-mesenchymal stem cell-derived exosomes ameliorate murine Sjögren’s syndrome by modulating the function of myeloid-derived suppressor cells显示文摘Sjögren’s syndrome(SS)is a systemic autoimmune disease characterized by progressive inflammation and tissue damage in salivary glands and lacrimal glands.Our previous studies showed that myeloid-derived suppressor cells(MDSCs)exhibited impaired immunosuppressive function during disease progression in patients with SS and mice with experimental Sjögren’s syndrome(ESS),but it remains unclear whether restoring the function of MDSCs can effectively ameliorate the development of ESS.In this study,we found that murine olfactory ecto-mesenchymal stem cell-derived exosomes(OE-MSC-Exos)significantly enhanced the suppressive function of MDSCs by upregulating arginase expression and increasing ROS and NO levels.Moreover,treatment with OE-MSC-Exos via intravenous injection markedly attenuated disease progression and restored MDSC function in ESS mice.Mechanistically,OE-MSC-Exo-secreted IL-6 activated the Jak2/Stat3 pathway in MDSCs.In addition,the abundant S100A4 in OE-MSC-Exos acted as a key factor in mediating the endogenous production of IL-6 by MDSCs via TLR4 signaling,indicating an autocrine pathway of MDSC functional modulation by IL-6.Taken together,our results demonstrated that OE-MSC-Exos possess therapeutic potential to attenuate ESS progression by enhancing the immunosuppressive function of MDSCs,possibly constituting a new strategy for the treatment of Sjögren’s syndrome and other autoimmune diseases.Ke Rui Yue Hong Qiugang Zhu Xiaofei Shi Fan Xiao Hailong Fu Qing Yin Yida Xing Xinfeng Wu Xiaodan Kong Huaxi Xu Jie Tian Shengjun Wang Liwei Lu 2021Cellular & Molecular Immunology2021,18,2:12
2Aerosol influence domain of Beijing and peripheral city agglomeration and its climatic effect显示文摘The aerosol distribution in Beijing and peripheral cities agglomeration (BPCA) and its re- gional climatic effect are investigated on the basis of the statistical analyses of satellite Total Ozone Map- ping Spectrometer (TOMS) retrieval aerosol optical depth (AOD) and the meteorological data of sunshine duration, fog days, and low cloud cover, observed at Beijing and its peripheral meteorological stations. The analysis on multi-samples variational correction of the satellite remote sensing Moderate Resolution Imaging Spectroradiometer (MODIS) AOD under the clear sky and stable weather condition in conjunction with surface observations reveal that there was a “triangle-like” distribution pattern of the high values of aerosols in the southern “valley” of the “U-shape” megarelief of Beijing and its peripheral areas. The distribution pattern suggests that the large-scale transfer and diffusion of city agglomeration pollutants might form a relatively persistent characteristic spatial distribution of city agglomeration pollutants much larger than city-scale. Under the background of the particular megarelief effect of Beijing and periph- eral areas, the high value area of TOMS AOD, as well as regional correlation distribution between clear sky sunshine duration and TOMS AOD are also similar to the composite image of MODIS AOD variational fields, that is to say, the effect of atmospheric aerosols was very distinctive in Beijing and peripheral areas. The high value area of the negative correlation between clear sky sunshine duration and TOMS AOD ap-proximately accorded with the significant negative value area of the sunshine duration deviations of the 1980s to the 1990s, and the daily variations of the AOD also showed an anti-phase relation with those of clear sky sunshine duration. The above high cor- relation area of the urban aerosol impact of Beijing- Tianjin region leant towards south peripheral area, with its “center of gravity” in the south of Beijing- Tianjin agglomeration, and the main body of the high correlation area prolongating southwards, forming an aerosol influence domain of “eccentric ellipse” shape, that is to say, there was an aerosol influence domain of some “radius” in Beijing City and its peripheral areas, within and outside which sunshine duration, low cloud cover, and fog days showed a remarkable difference in interannual variation trend. At the downstream of the city agglomeration there was a significant interdecadal increment area of fog or low cloud cover, which might be associated with the local climatic feature of the regional flow convergent field in the diffusion process of city agglomeration pollut- ants in the aerosol influence domain, and such a local dynamical convergence feature might lead to the regional exacerbation of aerosol impact downstream of the city agglomeration. 3Analysis to significant climate change in aerosol influence domain of Beijing and its peripheral areas by EOF mode显示文摘Using the total ozone mapping spectrometer (TOMS) aerosol optical depth (AOD)data and the sunshine duration, fog days, Iow cloud cover (LCC), etc. meteorological data in 1979-2000 in North China, as well as empirical orthogonal function (EOF) mode statistical analyses method, the winter aerosol distributive character of Beijing and peripheral city agglomeration and its influence effect on regional climate are investigated in this paper, especially the relation between aerosol influence effect and distinct change regions of eigenvectors of EOF mode. It is found from analyzing the regional distribution of the long-term averaged winter TOMS AOD that there is a large-scale relatively stable high value zone of aerosol concentration in the valley of the Beijing and peripheral U-shape megarelief. A high correlation area of AOD between Beijing and its southern peripheral exists in winter, and in this significant region of aerosol interaction, there is 'in-phase' character of the interannual variations of winter AOD, fog days, and LCCs. It indicates that the variations of aerosol in Beijing and its peripheral areas have impacts on interannual changes of fog days and LCCs in this area. The EOF analyses of the meteorological data further reveal the climate change regions and long-term trends of winter sunshine duration-reducing, and LCC- and fog days-increasing in North China. The areas of significant changes of the first EOF eigenvectors (FEE) of sunshine duration, fog days, LCCs almost superpose on corresponding marked regions of interdecadal differences between the 1990s and 1980s, and all accord with the S-N zonal high value pattern and high correlation region of winter AOD in Beijing and its peripheral areas. Interannual variations of their associated time coefficients (ATC) are in phase with that of regional mean AOD, and both of them have a secular rising trend. Results by EOF mode analyses depict the regional climatic change principal character of winter sunshine duration-reducing, and LCC- and fog days-increasing in peripheral areas to the south of Beijing, and reveal the regional influence effect of aerosol, i.e. the high value zone of long-term averaged winter AOD, significant change regions of FEE of sunshine duration, fog days, and LCC all lie in peripheral city agglomeration to the south of Beijing. 4The Tibetan Plateau Surface–Atmosphere Coupling System and Its Weather and Climate Effects: The Third Tibetan Plateau Atmospheric Science Experiment显示文摘The Tibetan Plateau(TP) is a key area affecting forecasts of weather and climate in China and occurrences of extreme weather and climate events over the world. The China Meteorological Administration, the National Natural Science Foundation of China, and the Chinese Academy of Sciences jointly initiated the Third Tibetan Plateau Atmospheric Science Experiment(TIPEX-Ⅲ) in 2013, with an 8–10-yr implementation plan. Since its preliminary field measurements conducted in 2013, routine automatic sounding systems have been deployed at Shiquanhe, Gaize, and Shenzha stations in western TP, where no routine sounding observations were available previously. The observational networks for soil temperature and soil moisture in the central and western TP have also been established. Meanwhile, the plateau-scale and regional-scale boundary layer observations, cloud–precipitation microphysical observations with multiple radars and aircraft campaigns, and tropospheric–stratospheric air composition observations at multiple sites, were performed. The results so far show that the turbulent heat exchange coefficient and sensible heat flux are remarkably lower than the earlier estimations at grassland, meadow, and bare soil surfaces of the central and western TP. Climatologically, cumulus clouds over the main body of the TP might develop locally instead of originating from the cumulus clouds that propagate northward from South Asia. The TIPEX-Ⅲ observations up to now also reveal diurnal variations, macro-and microphysical characteristics, and water-phase transition mechanisms, of cumulus clouds at Naqu station. Moreover, TIPEX-Ⅲ related studies have proposed a maintenance mechanism responsible for the Asian 'atmospheric water tower' and demonstrated the effects of the TP heating anomalies on African, Asian, and North American climates. Additionally, numerical modeling studies show that the Γ distribution of raindrop size is more suitable for depicting the TP raindrop characteristics compared to the M–P distribution, the overestimation of sensible heat flux can be reduced via modifying the heat transfer parameterization over the TP, and considering climatic signals in some key areas of the TP can improve the skill for rainfall forecast in the central and eastern parts of China. 7Red blood cell distribution width improves the prediction of 28-daymortality for patients with sepsis-induced acute kidney injury:A retrospective analysis from MIMIC-IV database usingpropensity score matching显示文摘Background:The predictive value of red blood cell distribution width(RDW)for mortality in patients withsepsis-induced acute kidney injury(SI-AKI)remains unclear.The present study aimed to investigate the potentialassociation between RDW at admission and outcomes in patients with SI-AKI.Methods:The Medical Information Mart for Intensive Care(MIMIC)-IV(version 2.0)database,released in Juneof 2022,provides medical data of SI-AKI patients to conduct our related research.Based on propensity scorematching(PSM)method,the main risk factors associated with mortality in SI-AKI were evaluated using Coxproportional hazards regression analysis to construct a predictive nomogram.The concordance index(C-index)and decision curve analysis were used to validate the predictive ability and clinical utility of this model.Patientswith SI-AKI were classified into the high-and low-RDW groups according to the best cut-off value obtained bycalculating the maximum value of the Youden index.Results:A total of 7574 patients with SI-AKI were identified according to the filter criteria.Compared withthe low-RDW group,the high-RDW group had higher 28-day(9.49%vs.31.40%,respectively,P<0.001)and7-day(3.96%vs.13.93%,respectively,P<0.001)mortality rates.Patients in the high-RDW group were moreprone to AKI progression than those in the low-RDW group(20.80%vs.13.60%,respectively,P<0.001).Basedon matched patients,we developed a nomogram model that included age,white blood cells,RDW,combinedhypertension and presence of a malignant tumor,treatment with vasopressor,dialysis,and invasive ventilation,sequential organ failure assessment,and AKI stages.The C-index for predicting the probability of 28-day survivalwas 0.799.Decision curve analysis revealed that the model with RDW offered greater net benefit than that withoutRDW.Conclusion:The present findings demonstrated the importance of RDW,which improved the predictive ability ofthe nomogram model for the probability of survival in patients with SI-AKI.Honghao Lai Guosheng Wu Yu Zhong Guangping Chen Wei Zhang Shengjun Shi Zhaofan Xia 2023Journal of Intensive Medicine2023,3,3:0
8Identification of twist-angle-dependent excitons in WS^(2)/WSe_(2) heterobilayers显示文摘Stacking atomically thin films enables artificial construction of van der Waals heterostructures with exotic functionalities such as superconductivity,the quantum Hall effect,and engineered light-matter interactions.In particular,heterobilayers composed of monolayer transition metal dichalcogenides have attracted significant interest due to their controllable interlayer coupling and trapped valley excitons in moirésuperlattices.However,the identification of twist-angle-modulated optical transitions in heterobilayers is sometimes controversial since both momentum-direct(K–K)and-indirect excitons reside on the low energy side of the bright exciton in the monolayer constituents.Here,we attribute the optical transition at~1.35 eV in the WS_(2)/WSe_(2) heterobilayer to an indirectΓ–K transition based on a systematic analysis and comparison of experimental photoluminescence spectra with theoretical calculations.The exciton wavefunction obtained by the state-of-the-art GW-Bethe-Salpeter equation approach indicates that both the electron and hole of the excitons are contributed by the WS_(2) layer.Polarization-resolved k-space imaging further confirms that the transition dipole moment of this optical transition is dominantly in-plane and is independent of the twist angle.The calculated absorption spectrum predicts that the so-called interlayer exciton peak coming from the K–K transition is located at 1.06 eV,but with a much weaker amplitude.Our work provides new insight into the steady-state and dynamic properties of twist-angle-dependent excitons in van der Waals heterostructures.Ke Wu Hongxia Zhong Quanbing Guo Jibo Tang Jing Zhang Lihua Qian Zhifeng Shi Chendong Zhang Shengjun Yuan Shunping Zhang Hongxing Xu 2022National Science Review2022,9,6:0
9An isoleucine-zipper motif enhances costimulation of human soluble trimeric GITR ligand显示文摘Glucocorticoid-induced tumor-necrosis factor receptor(GITR)and its ligand,GITRL,play significant roles in regulating immune responses.It is clear that human soluble GITRL(hsGITRL)transduces signal activity through multiple oligomerization states.To develop human soluble trimeric GITRL protein as a potential therapeutic target,we explored the link of the isoleucine-zipper(ILZ)motif to the N-terminus of the human soluble GITRL with two leucine sequences.hsGITRL,with the ILZ motif(ILZ-hsGITRL),was firstly expressed in Escherichia coli,which exhibited a predominant trimer when identified by Sephadex G-100 filtration and non-reducing SDS–polyacrylamide gel electrophoresis(SDS-PAGE).The significantly higher biological activity of the ILZ-hsGITRL compared with hsGITRL was confirmed by CD41 T proliferation,interferon-c(IFN-c)secretion and binding activity assay.To reveal and compare the underlying mechanisms,the level of extracellular signal-regulated kinase-1/2(ERK1/2)phosphorylation was examined,indicating that ILZ-hsGITRL induced more persistent and stronger ERK1/2 activation than hsGITRL.In conclusion,the incorporation of an ILZ motif could markedly improve the costimulation of hsGITRL.Dawei Cui Shengjun Wang Yu Chen Jia Tong Jie Ma Li Tang Xianzhi Yang Ye Shi Jie Tian Liwei Lu Huaxi Xu 2010Cellular & Molecular Immunology2010,7,4:0
