1Allosteric inhibition reveals SHP2-mediated tumor immunosuppression in colon cancer by single-cell transcriptomics显示文摘Colorectal cancer(CRC), a malignant tumor worldwide consists of microsatellite instability(MSI) and stable(MSS) phenotypes. Although SHP2 is a hopeful target for cancer therapy, its relationship with innate immunosuppression remains elusive. To address that, single-cell RNA sequencing wasperformed to explore the role of SHP2 in all cell types of tumor microenvironment(TME) from murine MC38 xenografts. Intratumoral cells were found to be functionally heterogeneous and responded significantly to SHP099, a SHP2 allosteric inhibitor. The malignant evolution of tumor cells was remarkably arrested by SHP099. Mechanistically, STING-TBK1-IRF3-mediated type I interferon signaling was highly activated by SHP099 in infiltrated myeloid cells. Notably, CRC patients with MSS phenotype exhibited greater macrophage infiltration and more potent SHP2 phosphorylation in CD68;macrophages than MSI-high phenotypes, suggesting the potential role of macrophagic SHP2 in TME. 3Optimization of compression formulation and load of food-grade tracers for grain traceability using central composite design显示文摘Food-grade tracers have been developed as an identification technology for grain traceability from original harvest to final destination for transportation.The characteristics of food-grade tracers must be able to satisfy the environmental demands for grain traceability.To optimize the food-grade tracer production process,the effects of direct compression formulation and load on the mechanical characteristics were studied using response surface methodology(RSM)with central composite design(CCD).Among the four tested formulations,Formulations#2(consisting of 35.00%lactose 100 mesh,64.50%microcrystalline cellulose 102 and 0.50%magnesium stearate)and#4(consisting of 38.00%lactose 100 mesh,50.00%microcrystalline cellulose 102,11.00%pregelatinized starch and 1.00%magnesium stearate)were selected for tracer production based on their physical properties as powders.The value of Carr’s flowability index was 68 for both Formulations#2 and#4,which was the highest among all the formulations.Therefore,Formulations#2 and#4 also had the best powder flowability.The magnesium stearate ratio(1.00%-3.00%)and pressure(6.00-16.00 kgf)were used as independent variables to detect changes in the breaking rate,peak shear force and friction coefficient of tracers compressed by the selected formulations.The optimal production parameters could be achieved at a magnesium stearate ratio of 2.25%and pressure of 16.00 kgf for Formulation#2 and at a magnesium stearate ratio of 1.02%and pressure of 16.00 kgf for Formulation#4.Under these optimal conditions,the tracers had good impact characteristics(breaking rate),compression characteristics(peak shear force)and frictional characteristics(friction coefficient).Moreover,Formulation#2 was more suitable for production because compared to Formulation#4,its breaking rate and friction coefficient values were lower,and its peak shear force value was higher.Liang Kun Zhang Lingling Lu Wei Cedric Sean Okinda Shen Mingxia 2017International Journal of Agricultural and Biological Engineering2017,10,6:2
4Loss of hnRNP A1 in murine skeletal muscle exacerbates high-fat diet-induced onset of insulin resistance and hepatic steatosis显示文摘Impairment of glucose(Glu)uptake and storage by skeletal muscle is a prime risk factor for the development of metabolic diseases.Heterogeneous nuclear ribonucleoprotein A1(hnRNP Al)is a highly abundant RNA-binding protein that has been implicated in diverse cellular functions.The aim of this study was to investigate the function of hnRNP A1 on muscle tissue insulin sensitivity and systemic Glu homeostasis.Our results showed that conditional deletion of hnRNP Al in the muscle gave rise to a severe insulin resistance phenotype in mice fed a high-fat diet(HFD).Conditional knockout mice fed a HFD showed exacerbated obesity,insulin resistance,and hepatic steatosis.In vitro interference of hnRNP Al in C2C12 myotubes impaired insulin signal transduction and inhibited Glu uptake,whereas hnRNP Al overexpression in C2C12 myotubes protected against insulin resistance induced by supraphysiological concentrations of insulin.The expression and stability of glycogen synthase(gysl)mRNA were also decreased in the absence of hnRNP A l.Mechanistically,hnRNP Al interacted with gys l and stabilized its mRNA,thereby promoting glycogen synthesis and maintaining the insulin sensitivity in muscle tissue.Taken together,our findings are the first to show that reduced expression of hnRNP Al in skeletal muscle affects the metabolic properties and systemic insulin sensitivity by inhibiting glycogen synthesis.Mingxia Zhao Lihong Shen Zijun Ouyang Manru Li Guoliang Deng Chenxi Yang Wei Zheng Lingdong Kong Xuefeng Wu Xudong Wu Wenjie Guo Ye Yin Qiang Xu Yang Sun 2020Journal of Molecular Cell Biology2020,12,4:2

12Laboratory Diagnosis and Monitoring the Viral Shedding of SARS-CoV-2 Infection显示文摘The worldwide epidemic of coronavirus disease 2019(COVID-19)is ongoing.Rapid and accurate detection of the causative virus SARSCoV-2 is vital for the treatment and control of COVID-19.In this study,the comparative sensitivity of different respiratory specimen types were retrospectively analyzed using 3,552 clinical samples from 410 COVID-19 patients confirmed by Guangdong CDC(Center for Disease Control and Prevention).Except for bronchoalveolar lavage fluid(BALF),the sputum possessed the highest positive rate(73.4%–87.5%),followed by nasal swabs(53.1%–85.3%)for both severe and mild cases during the first 14 days after illness onset(d.a.o.).Viral RNA could be detected in all BALF samples collected from the severe group within 14 d.a.o.and lasted up to 46 d.a.o.Moreover,although viral RNA was negative in the upper respiratory samples,it was also positive in BALF samples in most cases from the severe group during treatment.Notably,no viral RNA was detected in BALF samples from the mild group.Despite typical ground-glass opacity observed via computed tomographic scans,no viral RNA was detected in the first three or all upper respiratory tract specimens from some COVID-19 patients.In conclusion,sputum is most sensitive for routine laboratory diagnosis of COVID-19,followed by nasal swabs.Detection of viral RNA in BALF improves diagnostic accuracy in severe COVID-19 patients.Yang Yang Minghui Yang Jing Yuan Fuxiang Wang Zhaoqin Wang Jinxiu Li Mingxia Zhang Jinli Wei Ling Peng Gary Wong Haixia Zheng Weibo Wu Chenguang Shen Mingfeng Liao Kai Feng Jianming Li Qianting Yang Juanjuan Zhao Lei Liu Yingxia Liu 2020The Innovation2020,1,3:1
13Optimization of tracer coating parameters and their effects on the mechanical properties and quality of food-grade tracers for grain traceability显示文摘The purpose of this study was to optimize the coating process of food-grade tracers to manufacture tracers with good physical,mechanical and practical properties and an excellent appearance.The effects of the coating weight gain(1.00%-5.00%),coating solution spray rate(1.50-7.50 g/min)and tablet bed temperature(30℃-40℃)on the coating appearance quality,moisture absorption rate,friction coefficient,peak shear force,breaking rate,barcode recognition rate,transport wear rate and transport recognition rate were analysed using a Box-Behnken design(BBD)of response surface methodology(RSM).The experimental data were fitted to quadratic polynomial models by multiple regression analysis.The mathematical models of the barcode recognition rate,transport wear rate and transport recognition rate exhibited no statistically significant difference in these data.The optimum coating parameters were as follows:a 5.00%coating weight gain,spray rate of 5.47 g/min and tablet bed temperature of 35.42℃.Under the optimized conditions,the tracers had a good appearance(coating appearance quality),moisture resistance(moisture absorption rate),and frictional(friction coefficient),compression(peak shear force),and impact characteristics(breaking rate).Kun Liang Lingling Zhang Xiaohe Chen Mingxia Shen 2019International Journal of Agricultural and Biological Engineering2019,12,2:0
14Examination of Huntington's disease in a Chinese family显示文摘We report brain imaging and genetic diagnosis in a family from Wuhan,China,with a history of Huntington's disease.Among 17 family members across three generations,four patients(II2,II6,III5,and III9) show typical Huntington's disease,involuntary dance-like movements.Magnetic resonance imaging found lateral ventricular atrophy in three members(II2,II6,and III5).Moreover,genetic analysis identified abnormally amplified CAG sequence repeats(> 40) in two members(III5 and III9).Among borderline cases,with clinical symptoms and brain imaging features of Huntington's disease,two cases were identified(II2 and II6),but shown by mutation analysis for CAG expansions in the important transcript 15 gene,to be non-Huntington's disease.Our findings suggest that clinical diagnosis of Huntington's disease requires a combination of clinical symptoms,radiological changes,and genetic diagnosis.Mingxia Yu Xiaogai Li Sanyun Wu Ji Shen Jiancheng Tu 2014Neural Regeneration Research2014,9,4:0
15MANF brakes TLR4 signaling by competitively binding S100A8 with S100A9 to regulate macrophage phenotypes in hepatic fibrosis显示文摘The mesencephalic astrocyte-derived neurotrophic factor(MANF)has been recently identified as a neurotrophic factor,but its role in hepatic fibrosis is unknown.Here,we found that MANF was upregulated in the fibrotic liver tissues of the patients with chronic liver diseases and of mice treated with CCl4.MANF deficiency in either hepatocytes or hepatic mono-macrophages,particularly in hepatic mono-macrophages,clearly exacerbated hepatic fibrosis.Myeloid-specific MANF knockout increased the population of hepatic Ly6C^(high)macrophages and promoted HSCs activation.Furthermore,MANF-sufficient macrophages(from WT mice)transfusion ameliorated CCl4-induced hepatic fibrosis in myeloid cells-specific MANF knockout(MKO)mice.Mechanistically,MANF interacted with S100A8 to competitively block S100A8/A9 heterodimer formation and inhibited S100A8/A9-mediated TLR4-NF-κB signal activation.Pharmacologically,systemic administration of recombinant human MANF significantly alleviated CCl_(4)-induced hepatic fibrosis in both WT and hepatocytes-specific MANF knockout(HKO)mice.This study reveals a mechanism by which MANF targets S100A8/A9-TLR4 as a“brake”on the upstream of NF-κB pathway,which exerts an impact on macrophage differentiation and shed light on hepatic fibrosis treatment.Chao Hou Dong Wang Mingxia Zhao Petek Ballar Xinru Zhang Qiong Mei Wei Wang Xiang Li Qiang Sheng Jun Liu Chuansheng Wei Yujun Shen Yi Yang Peng Wang Juntang Shao Sa Xu Fuyan Wang Yang Sun Yuxian Shen 2023Acta Pharmaceutica Sinica B2023,13,10:0
16Short-term feeding behaviour sound classification method for sheep using LSTM networks显示文摘A deep learning approach using long-short term memory(LSTM)networks was implemented in this study to classify the sound of short-term feeding behaviour of sheep,including biting,chewing,bolus regurgitation,and rumination chewing.The original acoustic signal was split into sound episodes using an endpoint detection method,where the thresholds of short-term energy and average zero-crossing rate were utilized.A discrete wavelet transform(DWT),Mel-frequency cepstral,and principal-component analysis(PCA)were integrated to extract the dimensionally reduced DWT based Mel-frequency cepstral coefficients(denoted by PW_MFCC)for each sound episode.Then,LSTM networks were employed to train classifiers for sound episode category classification.The performances of the LSTM classifiers with original Mel-frequency cepstral coefficients(MFCC),DWT based MFCC(denoted by W_MFCC),and PW_MFCC as the input feature coefficients were compared.Comparison results demonstrated that the introduction of DWT improved the classifier performance effectively,and PCA reduced the computational overhead without degrading classifier performance.The overall accuracy and comprehensive F1-score of the PW_MFCC based LSTM classifier were 94.97%and 97.41%,respectively.The classifier established in this study provided a foundation for an automatic identification system for sick sheep with abnormal feeding and rumination behaviour pattern.Guanghui Duan Shengfu Zhang Mingzhou Lu Cedric Okinda Mingxia Shen Tomas Norton 2021International Journal of Agricultural and Biological Engineering2021,14,2:0

18Fine-grained detection of caged-hen head states using adaptive Brightness Adjustment in combination with Convolutional Neural Networks显示文摘Timely identification and tracking of abnormal hens in stacked cages are of great significance for precision treatment and the elimination of sick individuals.The head features of the caged-hens are used to overcome observation difficulties caused by the cage and feathers blocking,but it is still hard to identify similar head states.To solve this problem,the fine-grained detection of caged-hens head states was developed using adaptive Brightness Adjustment in combination with Convolutional Neural Networks(FBA-CNN).Grid Region-based CNN(R-CNN),a convolution neural network(CNN),was optimized with the Squeeze-and-Excitation(SE)and Depthwise Over-parameterized Convolutional(DO-Conv)to detect layer heads from cages and to accurately cut them as single-head images.The brightness of each single-head image was adjusted adaptively and classified through the deep convolution neural network based on SE-Resnet50.Finally,we returned to the original image to realize multi-target detection with coordinate mapping.The results showed that the AP@0.5 of layer head detection using the optimized Grid R-CNN was 0.947,the accuracy of classification with SE-Resnet50 was 0.749,the F1 score was 0.637,and the mAP@0.5 of FBA-CNN was 0.846.In summary,this automated method can accurately identify different layer head states in layer cages to provide a basis for follow-up studies of abnormal behavior including dyspnea and cachexia.Jia Chen Qi’an Ding Wen Yao Mingxia Shen Longshen Liu 2023International Journal of Agricultural and Biological Engineering2023,16,3:0
19Prevalence and Severity of HBV-Associated Acute-on-Chronic Liver Failure Due to Irregular Medication of Nucleos(t)ide Analogs显示文摘Hepatitis B virus(HBV)represents the commonest etiologic agent of acute-on-chronic liver failure(ACLF)in most Asian countries.Nucleos(t)ide analogs(NAs)are effective in the treatment of chronic HBV infections,but may also exacerbate the disease and stimulate its development into HBV-associated ACLF if not used appropriately.The current study aimed to assess the prevalence and severity of HBV-associated ACLF as a result from irregular medication of NAs(IMNA).A total of 1134 individuals with HBV-associated ACLF in nine hospitals in Heilongjiang Province were enrolled in this study between 2005 and 2015.Among these,777 chronic hepatitis B(CHB)and 357 HBV-associated liver cirrhosis cases were classified based on various predisposing factors,including IMNA,HBV reactivation(HBVR),infections,treatment drugs,alcohol use and others(hepatitis C virus,hepatitis E virus,gastrointestinal bleeding and unknown reasons).The percentage and improvement rate were examined.Among individuals with HBV-associated ACLF and CHB,IMNA was found in 9.01%,HBVR in 46.20%,infections in 9.52%,treatment drugs in 14.67%,alcohol in 11.71%,and others in 24.58%as predisposing factors.Improvement rates in cases with IMNA,HBVR,infections,treatment drugs,alcohol and others were 41.43%,58.50%,58.11%,56.14%,53.85%,and 65.97%,respectively.Multivariable analysis showed that IMNA,others,infections,hepatic encephalopathy and hepatorenal syndrome were associated with prognosis.Only IMNA independently predicted HBV-associated ACLF prognosis.Overall,our study demonstrated that the percentage of IMNAinduced HBV-associated ACLF was 12.61%,and worse disease conditions resulted from IMNA compared with other factors.Thus,the suitability of treatment with NAs should be thoroughly evaluated.Ying Zheng Shu Chen Yanxin Huang Lisheng Jiang Yongguo Li Yinghua Lan Shuchen Li Yuqin Xu Xiaodong Li Hongwei Zhao Yanbo Wang Ying Shen Chao Wei Honglin Zhou Rongshan Fan Xiqiu Zeng Mingxia Jiang Shupeng Song Mingyan Xu 2021Infectious Microbes & Diseases2021,3,4:0
