1Distinct immune signatures discriminate between asymptomatic and presymptomatic SARS-CoV-2^(pos)subjects显示文摘Increasing numbers of SARS-CoV-2-positive(SARS-CoV-2^(pos))subjects are detected at silent SARS-CoV-2 infection stage(SSIS).Yet,SSIS represents a poorly examined time-window wherein unknown immunity patterns may contribute to the fate determination towards persistently asymptomatic or overt disease.Here,we retrieved blood samples from 19 asymptomatic and 12 presymptomatic SARS-COV-2^(pos)subjects,47 age/gender-matched patients with mild or moderate COVID-19 and 27 normal subjects,and interrogated them with combined assays of 44-plex CyTOF,RNA-seq and Olink.Notably,both asymptomatic and presymptomatic subjects exhibited numerous readily detectable immunological alterations,while certain parameters including more severely decreased frequencies of CD107a^(low)classical monocytes,intermediate monocytes,non-classical monocytes and CD62L^(hi)CD8^(+)Tnaive cells,reduced plasma STC1 level but an increased frequency of CD4^(+)NKT cells combined to distinguish the latter.Intercorrelation analyses revealed a particular presymptomatic immunotype mainly manifesting as monocytic overactivation and differentiation blockage,a likely lymphocyte exhaustion and immunosuppression,yielding mechanistic insights into SSIS fate determination,which could potentially improve 5ARS-CoV-2 management.Shanhe Yu Caixia Di Shijun Chen Mingquan Guo Jiayang Yan Zhaoqin Zhu Li Liu Ruixue Feng Yinyin Xie Ruihong Zhang Juan Chen Mengxi Wang Dong Wei Hai Fang Tong Yin Jinyan Huang Saijuan Chen Hongzhou Lu Jiang Zhu Jieming Qu 2021Cell Research2021,31,11:4
2A Single-cell Transcriptome Atlas of Cashmere Goat Hair Follicle Morphogenesis显示文摘Cashmere,also known as soft gold,is produced from the secondary hair follicles(SHFs)of cashmere goats.The number of SHFs determines the yield and quality of cashmere;therefore,it is of interest to investigate the transcriptional profiles present during cashmere goat hair follicle development.However,mechanisms underlying this development process remain largely unexplored,and studies regarding hair follicle development mostly use a murine research model.In this study,to provide a comprehensive understanding of cellular heterogeneity and cell fate decisions,single-cell RNA sequencing was performed on 19,705 single cells of the dorsal skin from cashmere goat fetuses at induction(embryonic day 60;E60),organogenesis(E90),and cytodifferentiation(E120)stages.For the first time,unsupervised clustering analysis identified 16 cell clusters,and their corresponding cell types were also characterized.Based on lineage inference,a detailed molecular landscape was revealed along the dermal and epidermal cell lineage developmental pathways.Notably,our current data also confirmed the heterogeneity of dermal papillae from different hair follicle types,which was further validated by immunofluorescence analysis.The current study identifies different biomarkers during cashmere goat hair follicle development and has implications for cashmere goat breeding in the future.Wei Ge Weidong Zhang Yuelang Zhang Yujie Zheng Fang Li Shanhe Wang Jinwang Liu Shaojing Tan Zihui Yan Lu Wang Wei Shen Lei Qu Xin Wang 2021Genomics, Proteomics & Bioinformatics2021,19,3:3

4Optimal Weighting Method for Reducing Digital Satellite TV Differential Timing Error显示文摘We illustrate the principle of Digital satellite TV differential timing(DSTVDT) and propose an optimal weighting method that reduces the timing error introduced by satellite ephemeris error. A mathematical formula for the timing error introduced by satellite ephemeris error is derived and analyzed. On the basis of the derived formula, the proposed optimal weighting method based on multi-base stations is presented. 12Influence of Initial Water Content on Synthesis of Silicalite-1 Zeolite显示文摘The effect of initial water content on the synthesis of silicalite-1 zeolite and its reaction performance in methanol to propylene(MTP) reaction was comprehensively investigated. The as-synthesized samples were characterized by XRD,N_2 adsorption and desorption, SEM, and FTIR techniques, and studied for the methanol to propylene(MTP) reaction. The results showed that the initial water content played a crucial role on the particle size and porous structure of the silicalite-1 zeolites. It was found that adding initial water to reach a certain amount, the crystal size reduced obviously and the mesoporous structure appeared. Upon applying the silicalite-1 zeolite samples in the MTP reaction, they showed excellent catalytic performance to reach a highest propene mass yield of 26.81%(on the CH_2 basis) and a P/E ratio of 6.28 in 2 h. 19Integrated microfluidic single-cell immunoblotting chip enables high-throughput isolation,enrichment and direct protein analysis of circulating tumor cells显示文摘Effective capture and analysis of a single circulating tumor cell(CTC)is instrumental for early diagnosis and personalized therapy of tumors.However,due to their extremely low abundance and susceptibility to interference from other cells,high-throughput isolation,enrichment,and single-cell-level functional protein analysis of CTCs within one integrated system remains a major challenge.Herein,we present an integrated multifunctional microfluidic system for highly efficient and label-free CTC isolation,CTC enrichment,and single-cell immunoblotting(ieSCI).The ieSCI-chip is a multilayer microfluidic system that combines an inertia force-based cell sorter with a membrane filter for label-free CTC separation and enrichment and a thin layer of a photoactive polyacrylamide gel with microwell arrays at the bottom of the chamber for single-cell immunoblotting.The ieSCI-chip successfully identified a subgroup of apoptosis-negative(Bax-negative)cells,which traditional bulk analysis did not detect,from cisplatin-treated cells.Furthermore,we demonstrated the clinical application of the ieSCI-chip with blood samples from breast cancer patients for personalized CTC epithelial-to-mesenchymal transition(EMT)analysis.The expression level of a tumor cell marker(EpCAM)can be directly determined in isolated CTCs at the single-cell level,and the therapeutic response to anticancer drugs can be simultaneously monitored.Therefore,the ieSCI-chip provides a promising clinical translational tool for clinical drug response monitoring and personalized regimen development.Aynur Abdulla Ting Zhang Shanhe Li Wenke Guo Antony RWarden Yufang Xin Nokuzola Maboyi Jiatao Lou Haiyang Xie Xianting Ding 2022Microsystems & Nanoengineering2022,8,1:1
