1MicroRNAs:a new ray of hope for diabetes mellitus显示文摘Diabetes mellitus has become one of the most common chronic diseases,thereby posing a major challenge to global health.Characterized by high levels of blood glucose(hyperglycemia),diabetes usually results from a loss of insulin-producing β-cells in the pancreas,leading to a deficiency of insulin(type 1 diabetes),or loss of insulin sensitivity(type 2 diabetes).Both types of diabetes have serious secondary complications,such as microvascular abnormalities,cardiovascular dysfunction,and kidney failure.Various complex factors,such as genetic and environmental factors,are associated with the pathophysiology of diabetes.Over the past two decades,the role of small,single-stranded noncoding microRNAs in various metabolic disorders,especially diabetes mellitus and its complications,has gained widespread attention in the scientific community.Discovered first as an endogenous regulator of development in the nematode Caenorhabditis elegans,these small RNAs post-transcriptionally suppress mRNA target expression.In this review,we discuss the potential roles of different microRNAs in diabetes and diabetes-related complications.Munish Kumar Sayantan Nath Himanshu K Prasad G D Sharma Yong Li 2012Protein & Cell2012,3,10:14
2Matrix metalloproteinases and gastrointestinal cancers: Impacts of dietary antioxidants显示文摘The process of carcinogenesis is tightly regulated by antioxidant enzymes and matrix degrading enzymes, namely, matrix metalloproteinases(MMPs). Degradation of extracellular matrix(ECM) proteins like collagen, proteoglycan, laminin, elastin and fibronectin is considered to be the prerequisite for tumor invasion and metastasis. MMPs can degrade essentially all of the ECM components and, most MMPs also substantially contribute to angiogenesis, differentiation, proliferation and apoptosis. Hence, MMPs are important regulators of tumor growth both at the primary site and in distant metastases; thus the enzymes are considered as important targets for cancer therapy. The implications of MMPs in cancers are no longer mysterious; however, the mechanism of action is yet to be explained. Herein, our major interest is to clarify how MMPs are tied up with gastrointestinal cancers. Gastrointestinal cancer is a variety of cancer types, including the cancers of gastrointestinal tract and organs, i.e., esophagus, stomach, biliary system, pancreas, small intestine, large intestine, rectum and anus. The activity of MMPs is regulated by its endogenous inhibitor tissue inhibitor of metallopro-teinase(TIMP) which bind MMPs with a 1:1 stoichiometry. In addition, RECK(reversion including cysteinerich protein with kazal motifs) is a membrane bound glycoprotein that inhibits MMP-2,-9 and-14. Moreover, α2-macroglobulin mediates the uptake of several MMPs thereby inhibit their activity. Cancerous conditions increase intrinsic reactive oxygen species(ROS) through mitochondrial dysfunction leading to altered protease/anti-protease balance. ROS, an index of oxidative stress is also involved in tumorigenesis by activation of different MAP kinase pathways including MMP induction. Oxidative stress is involved in cancer by changing the activity and expression of regulatory proteins especially MMPs. Epidemiological studies have shown that high intake of fruits that rich in antioxidants is associated with a lower cancer incidence. Evidence indicates that some antioxidants inhibit the growth of malignant cells by inducing apoptosis and inhibiting the activity of MMPs. 3槲皮素诱导肿瘤细胞P53非依赖的G2/M周期阻滞和凋亡显示文摘目的研究槲皮素对肿瘤细胞的增殖、周期和凋亡的影响。方法用槲皮素12.5,25,50和100μmol·L^(-1),分别作用于人肝癌HepG2和Hep3B细胞、人乳腺癌MDA-MA-231细胞、人结肠癌HCT116 p53野生型(HCT116 p53WT)和p53敲除型(HCT116 p53KO)细胞12,24和48 h。荧光显微镜观察细胞形态变化;MTT法测定细胞存活率;采用Hoechst33258染色观察细胞核染色质的变化;采用流式细胞术检测细胞周期的变化;Western印迹法检测胱天蛋白酶3和聚腺苷二磷酸核糖聚合酶(PARP)蛋白水平的变化。结果槲皮素对肿瘤细胞增殖的抑制作用显著且呈浓度(rHepG2=0.972,P<0.01;rHep3B=0.959,P<0.01;rMDA-MB-231=0.979,P<0.01;rHCT116p53WT=0.983,P<0.01;rHCT116p53KO=0.980,P<0.01)和时间(rHepG2=0.974,P<0.01;rHep3B=0.956,P<0.01;rMDA-MB-231=0.959,P<0.01;rHCT116p53WT=0.971,P<0.01;rHCT116p53KO=0.988,P<0.01)依赖性,与细胞对照组相比,槲皮素处理组细胞数量明显减少。槲皮素诱导G_2/M周期阻滞(P<0.01);镜下观察见核染色质浓缩和碎裂等典型的凋亡特性(P<0.01)。Western印迹法结果显示,胱天蛋白酶3蛋白表达水平明显降低(P<0.05,P<0.01),活化的胱天蛋白酶3表达水平无显著变化,PARP表达水平明显降低(P<0.01),活化的PARP明显升高(P<0.01);在加入泛胱天蛋白酶抑制剂后,与槲皮素单独作用相比,胱天蛋白酶3和PARP蛋白表达水平升高,活化的PARP表达水平明显降低(P<0.05,P<0.01,P<0.05)。结论槲皮素能通过诱导P53非依赖性的G_2/M细胞周期阻滞和细胞凋亡,从而实现对肿瘤细胞增殖的抑制作用。王梓萱 周静 唐玥 石欢 黄晓薇 邓敬桓 胡晓晓 陈攀红 李忠杰 戴惠淇 冯吉 Sayantan CHATTERJEE 卢国栋 2018中国药理学与毒理学杂志2018,32,10:9
4Critical review of bone health,fracture risk and management of bone fragility in diabetes mellitus显示文摘The risk of fracture is increased in both type 1 diabetes mellitus(T1DM)and type 2 diabetes mellitus(T2DM).However,in contrast to the former,patients with T2DM usually possess higher bone mineral density.Thus,there is a considerable difference in the pathophysiological basis of poor bone health between the two types of diabetes.Impaired bone strength due to poor bone microarchitecture and low bone turnover along with increased risk of fall are among the major factors behind elevated fracture risk.Moreover,some antidiabetic medications further enhance the fragility of the bone.On the other hand,antiosteoporosis medications can affect the glucose homeostasis in these patients.It is also difficult to predict the fracture risk in these patients because conventional tools such as bone mineral density and Fracture Risk Assessment Tool score assessment can underestimate the risk.Evidence-based recommendations for risk evaluation and management of poor bone health in diabetes are sparse in the literature.With the advancement in imaging technology,newer modalities are available to evaluate the bone quality and risk assessment in patients with diabetes.The purpose of this review is to explore the patho-physiology behind poor bone health in diabetic patients.Approach to the fracture risk evaluation in both T1DM and T2DM as well as the pragmatic use and efficacy of the available treatment options have been discussed in depth.Rajan Palui Subhodip Pramanik Sunetra Mondal Sayantan Ray 2021World Journal of Diabetes2021,12,6:4
5Liver abscess caused by Burkholderia pseudomallei in a young man: A case report and review of literature显示文摘Pyogenic liver abscess is a common entity in Indian subcontinent and is mostly caused by gram negative bacteria. Melioidosis is not commonly seen in India and only a few cases are reported. It can give rise to multiple abscesses at different sites including liver. We report a case of isolated liver abscess caused by Burkholderia pseudomallei(B. pseudomallei) in a 29-yearold recently diagnosed diabetic, immunocompetent male. Diagnosis was made by imaging and culture of pus aspirated from the abscess and he was treated with percutaneous pigtail catheter drainage followed by antibiotics(meropenem and trimethoprim-sulphmethoxazole). Melioidosis is an emerging infection in India and has high mortality rate, so early diagnosis and promptmanagement is warranted which requires clinical vigilance and an intensive microbiological workup. 6Inhibition of Cdk5 increases osteoblast differentiation and bone mass and improves fracture healing显示文摘Identification of regulators of osteoblastogenesis that can be pharmacologically targeted is a major goal in combating osteoporosis,a common disease of the elderly population. Here, unbiased kinome RNAi screening in primary murine osteoblasts identified cyclin-dependent kinase 5(Cdk5) as a suppressor of osteoblast differentiation in both murine and human preosteoblastic cells. Cdk5 knockdown by si RNA, genetic deletion using the Cre-lox P system, or inhibition with the small molecule roscovitine enhanced osteoblastogenesis in vitro. Roscovitine treatment significantly enhanced bone mass by increasing osteoblastogenesis and improved fracture healing in mice. Mechanistically, downregulation of Cdk5 expression increased Erk phosphorylation, resulting in enhanced osteoblast-specific gene expression. Notably, simultaneous Cdk5 and Erk depletion abrogated the osteoblastogenesis conferred by Cdk5 depletion alone, suggesting that Cdk5 regulates osteoblast differentiation through MAPK pathway modulation. 7Fine needle aspiration diagnosis of isolated pancreatic tuberculosis: A case report显示文摘Tuberculosis(TB) involving the pancreas are uncommon, especially when present in immunocompetent hosts. Pancreatic TB is more frequently associated with miliary TB or widely disseminated disease. Pancreatic TB may present as cystic or solid pancreatic masses, pancreatic abscess or acute or chronic pancreatitis. Majority of the cases are diagnosed after surgical exploration for presumed pancreatic malignancy and preoperative diagnosis is quite difficult. However, improvement in imaging techniques and the resulting imageguided interventions gradually can obviate the need for more invasive diagnostic surgical procedures and expedite the planning of therapy. Herein, we report a rare case of isolated pancreatic TB which presented with pancreatic mass lesion in an immunocompetent host. Diagnosis was made by contrast enhanced computed tomography and guided fine needle aspiration of the pancreatic mass which revealed acid-fast bacillion Ziehl-Neelsen stain. The case was treated successfully with antituberculous drugs. Pancreatic tuberculosis should be considered in the differential diagnosis of a pancreatic mass when the patient is young, residing in the endemic zone of tuberculosis. 12Unilateral neuropathic arthropathy of the shoulder secondary to syringomyelia: Diagnostic challenges显示文摘Neuropathic arthropathy of the shoulder is a rare disorder characterized by joint degeneration, and is associated with loss of sensory innervation. Syringomyelia is a disease in which fluid-containing cavities(syrinxes) form within the spinal cord. Here, we report a case of neuropathic arthropathy of the shoulder secondary to syringomyelia in a 40-year-old woman. X-rays of the left shoulder revealed damage to bone and joint architecture. Blood tests indicated vitamin D deficiency and secondary hyperparathyroidism. Magnetic resonance imaging of the cervical spine showed a large syrinx from the second cervical spine to the second dorsal spine. Although neuropathic arthropathy is uncommon, it should be considered in cases of unexplained pain, discomfort, or limited range of motion of the affected joint. Symptoms related to the affected joint may precede or overshadow neurological deficits. 20Induced defense response in red mango fruit against Colletotrichum gloeosporioides显示文摘Mango fruit exposed to sunlight develops red skin and are more resistant to biotic and abiotic stresses.Here we show that harvested red mango fruit that was exposed to sunlight at the orchard is more resistant than green fruit to Colletotrichum gloeosporioides.LCMS analysis showed high amounts of antifungal compounds,as glycosylated flavonols,glycosylated anthocyanins,and mangiferin in red vs.green mango skin,correlated with higher antioxidant and lower ROS.However,also the green side of red mango fruit that has low levels of flavonoids was resistant,indicated induced resistance.Transcriptomes of red and green fruit inoculated on their red and green sides with C.gloeosporioides were analyzed.Overall,in red fruit skin,2,187 genes were upregulated in response to C.gloeosporioides.On the green side of red mango,upregulation of 22 transcription factors and 33 signaling-related transcripts indicated induced resistance.The RNA-Seq analysis suggests that resistance of the whole red fruit involved upregulation of ethylene,brassinosteroid,and phenylpropanoid pathways.To conclude,red fruit resistance to fungal pathogen was related to both flavonoid toxicity and primed resistance of fruit that was exposed to light at the orchard.Pradeep Kumar Sudheeran Noa Sela Mira Carmeli-Weissberg Rinat Ovadia Sayantan Panda Oleg Feygenberg Dalia Maurer Michal Oren-Shamir Asaph Aharoni Noam Alkan 2021Horticulture Research2021,8,1:1
