| 1 | DNA-based vaccination induces humoral and cellular immune responses against hepatitis B virus surface antigen in mice without activation of Cmyc显示文摘AIM To develop a safe and effective DNAvaccine for inducing humoral and cellularimmunological responses against hepatitis Bvirus surface antigen(HBsAg).METHODS BALB/c mice were inoculated withNV-HB/s,a recombinant plasmid that had beeninserted S gene of hepatitis B virus genome andcould express HBsAg in eukaryotes.HBsAgexpression was measured by ABC immunohis-tochemical assay,generation of anti-HBs byELISA and cytotoxic T lymphocyte(CTL),byMTT method,existence of vaccine DNA bySouthern blot hybridization and activation ofoncogene C-myc by in situ hybridization,RESULTS With NV-HB/s vaccination byintramuscular injection,anti-HBs was initiallypositive 2 weeks after inoculation while all micetested were HBsAg positive in the muscles.Thetiters and seroconversion rate of anti-HBs weresteadily increasing as time went on and weredose-dependent.All the mice inoculated with100 μg NV-HB/s were anti-HBs positive onemonth after inoculation,the titer was 1:1024 ormore.The humoral immune response was similarinduced by either intramuscular or intradermalinjection.CTL activities were much stronger(45.26%)in NV-HB/s DNA immunized mice as compared with those(only 6%)in plasma-derived HBsAg vaccine immunized mice.Twomonths after inoculation,all muscle sampleswere positive by Southern-blot hybridization forNV.HB/s DNA detection,but decreased to 25%and all were undetectable by in situhybridization after 6 months.No oncogene C-myc activation was found in the muscle ofinoculation site.CONCLUSION NV-HB/s could generatehumoral and cellular immunological responsesagainst HBsAg that had been safely expressed insitu by NV-HB/s vaccination. | Lian San Zhao Shan Qin Tao You Zhou Hong Tang Li Liu Bing Jun Lei | 2000 | World Journal of Gastroenterology2000,6,2: | 24 |
| 2 | Immunization with chlamydial plasmid protein pORF5 DNA vaccine induces protective immunity against genital chlamydial infection in mice显示文摘To validate the immune protective efficacy of pORF5 DNA vaccine and to analyze potential mechanisms related to this protection. In this study, pORF5 DNA vaccine was constructed and evaluated for its protective immunity in a mouse model of genital chlamydial infection. Groups of BALB/c mice were immunized intranasally with pORF5 DNA vaccine. Humoral and cell mediated immune responses were evaluated. The clearance ability of chlamydial challenge from the genital tract and the chlamy- dia-induced upper genital tract gross pathology and histopathological characterization were also de- tected. The results showed that the total and the IgG2a anti-pORF5 antibody levels in serum were sig- nificantly elevated after pcDNA3.1-pORF5 vaccination, as were the total antibody and IgA levels in vaginal fluids. pcDNA3.1-pORF5 induced a significantly high level of Th1 response as measured by robust gamma interferon (IFN-γ). Minimal IL-4 was produced by immune T cells in response to the re-stimulation with pORF5 protein or the inactive elementary body in vitro. pcDNA3.1-pORF5-vacci- nated mice displayed significantly reduced bacterial shedding upon a chlamydial challenge and an accelerated resolution of infection. 100% of pcDNA3.1-pORF5 vaccinated mice successfully resolved the infection by day 24. pcDNA3.1-pORF5-immunized mice also exhibited protection against patho- logical consequences of chlamydial infection. The stimulated index was significantly higher than that of mice immunized with pcDNA3.1 and PBS (P<0.05). | 3 | A novel mutation at the JK locus causing Jknull phenotype in a Chinese family显示文摘Urea transporters are a group of proteins that facilitate urea movement across biological membranes. Kidd blood group (JK antigen) and urea transporter of human erythrocytes are carried by the same protein UT-B. To investigate the molecular basis of the Jknull phenotype in the Chinese population, blood samples from Chinese individuals were screened using the 2 mol/L urea solution hemolysis test. Urea and water permeability of erythrocytes membrane was measured by stopped-flow light scattering. Genomic DNA was extracted from lymphocytes. UT-B gene of Jknull's family was analyzed using genomic PCR by primers designed to cover sequences of all exons and exon-intron boundaries in human UT-B gene. One Jknull subject was found from twenty thousand screened Chinese individuals, and it was confirmed that this individual did not express the erythrocyte urea transporter. Genomic sequence analysis of the Jknull individual showed that there were two point mutations, G→C, which is novel, and G→A, at the 3(-acceptor splice site (AG) of intron 5 of UT-B gene. Exon 6 is spliced out in the UT-B transcript due to either of these mutations. | 6 | Advancement toward Polymer Electrolyte Membrane Fuel Cells at Elevated Temperatures显示文摘Elevation of operational temperatures of polymer electrolyte membrane fuel cells(PEMFCs)has been demonstrated with phosphoric acid-doped polybenzimidazole(PA/PBI)membranes.The technical perspective of the technology is simplified construction and operation with possible integration with,e.g.,methanol reformers.Toward this target,significant efforts have been made to develop acid-base polymer membranes,inorganic proton conductors,and organic-inorganic composite materials.This report is devoted to updating the recent progress of the development particularly of acid-doped PBI,phosphate-based solid inorganic proton conductors,and their composite electrolytes.Long-term stability of PBI membranes has been well documented,however,at typical temperatures of 160℃.Inorganic proton-conducting materials,e.g.,alkali metal dihydrogen phosphates,heteropolyacids,tetravalent metal pyrophosphates,and phosphosilicates,exhibit significant proton conductivity at temperatures of up to 300℃ but have so far found limited applications in the form of thin films.Composite membranes of PBI and phosphates,particularly in situ formed phosphosilicates in the polymer matrix,showed exceptionally stable conductivity at temperatures well above 200℃.Fuel cell tests at up to 260℃ are reported operational with good tolerance of up to 16%CO in hydrogen,fast kinetics for direct methanol oxidation,and feasibility of nonprecious metal catalysts.The prospect and future exploration of new proton conductors based on phosphate immobilization and fuel cell technologies at temperatures above 200℃ are discussed. | Jin Zhang David Aili Shanfu Lu Qingfeng Li and San Ping Jiang | 2020 | Research2020,,1: | 6 |
| 7 | Analysis of PM2.5 concentrations in Heilongjiang Province associated with forest cover and other factors显示文摘Atmospheric particulate matter(PM2.5) seriously influences air quality. It is considered one of the main environmental triggers for lung and heart diseases. Air pollutants can be adsorbed by forest. In this study we investigated the effect of forest cover on urban PM2.5 concentrations in 12 cities in Heilongjiang Province,China. The forest cover in each city was constant throughout the study period. The average daily concentration of PM2.5 in 12 cities was below 75 lg/m^3 during the non-heating period but exceeded this level during heating period. Furthermore, there were more moderate pollution days in six cities. This indicated that forests had the ability to reduce the concentration of PM2.5 but the main cause of air pollution was excessive human interference and artificial heating in winter. We classified the 12 cities according to the average PM2.5 concentrations. The relationship between PM2.5 concentrations and forest cover was obtained by integrating forest cover, land area,heated areas and number of vehicles in cities. Finally,considering the complexity of PM2.5 formation and based on the theory of random forestry, we selected six cities and analyzed their meteorological and air pollutant data. | 10 | Human adenovirus(HAdV)infection in children with acute respiratory tract infections in Guangzhou,China,2010–2021:a molecular epidemiology study显示文摘Background Human adenovirus(HAdV)infection can cause a variety of diseases.It is a major pathogen of pediatric acute respiratory tract infections(ARIs)and can be life-threatening in younger children.We described the epidemiology and subtypes shifting of HAdV among children with ARI in Guangzhou,China.Methods We conducted a retrospective study of 161,079 children diagnosed with acute respiratory illness at the Guangzhou Women and Children’s Medical Center between 2010 and 2021.HAdV specimens were detected by real-time PCR and the hexon gene was used for phylogenetic analysis.Results Before the COVID-19 outbreak in Guangzhou,the annual frequency of adenovirus infection detected during this period ranged from 3.92%to 13.58%,with an epidemic peak every four to fve years.HAdV demonstrated a clear seasonal distribution,with the lowest positivity in March and peaking during summer(July or August)every year.A signifcant increase in HAdV cases was recorded for 2018 and 2019,which coincided with a shift in the dominant HAdV subtype from HAdV-3 to HAdV-7.The latter was associated with a more severe disease compared to HAdV-3.The average mortality proportion for children infected with HAdV from 2016 to 2019 was 0.38%but increased to 20%in severe cases.After COVID-19 emerged,HAdV cases dropped to 2.68%,suggesting that non-pharmaceutical interventions probably reduced the transmission of HAdV in the community.Conclusion Our study provides the foundation for the understanding of the epidemiology of HAdV and its associated risks in children in Southern China. | Yi Chen Tao Lin Chang‑Bing Wang Wan‑Li Liang Guang‑Wan Lian Mark Zanin Sook‑San Wong Xin‑Gui Tian Jia‑Yu Zhong Ying‑Ying Zhang Jia‑Hui Xie Ling‑Ling Zheng Fei‑Yan Chen Run Dang Ming‑Qi Zhao Yi‑Yu Yang Rong Zhou Bing Zhu | 2022 | World Journal of Pediatrics2022,18,8: | 5 |

| 16 | A review of the thermal stability of metastable austenite in steels:Martensite formation显示文摘Metastable austenite plays a critical role in achieving improved combinations of high strength and high ductility/toughness in the design of advanced high-strength steels(AHSS). The thermal stability of metastable austenite determines the transformation characteristics of AHSS and thus primarily determines the microstructure evolution during complex processes, e.g., the quenching and partitioning process, to achieve the desirable microstructure. This study provides a review of the thermal stability of austenite and its influence on martensitic transformation from both experimental and theoretical modeling perspectives. From the experimental perspective, factors affecting the thermal stability are analyzed,the relative sensitivities are compared, and their corresponding mechanisms are discussed. From the theoretical modeling perspective, the most representative kinetic models that describe athermal and isothermal martensitic transformation are reviewed. The advantages, shortcomings, and applicability of each model are discussed. | 19 | Dimethyl sulfide in the atmospheric surface layer of the Equatorial Pacific Ocean显示文摘This paper reports a case study of atmospheric stability effect on dimethyl sulfide(DMS) concentration in the air. Investigation includes model simulation and field measurements over the Pacific Ocean. DMS concentration in surface sea water and in the air were measured during a research cruise from Hawaii to Tahiti. The diurnal variation of air temperature over the sea surface differed from the diurnal cycle of sea surface temperature because of the high heat capacity of sea water. The diurnal cycle of average DMS concentration in the air was studied in relation to the atmospheric stability parameter and surface heat flux. All these parameters had minima at noon and maxima in the early morning. The correlation coefficient of the air DMS concentration with wind speed (at 15 m high) was 0. 64. The observed concentrations of DMS in the equatorial marine surface layer and their diurnal variability agree well with model simulations.
