1Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy.Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen 2021Cancer Communications2021,41,11:29
2Dynamics of total culturable bacteria and its relationship with methylmercury in the soils of the water level fluctuation zone of the Three Gorges Reservoir显示文摘Field investigations were conducted to study the temporal and spatial distribution characteristics of total culturable bacteria(TCB)and its relationship with methylmercury(MeHg)in the soils of the water level fluctuation zone of the Three Gorges Reservoir.Different altitudes(170–180,170–175,165–170 and 160–165 m)in Zhenxi(Site 1),Shibaozhai(Site 2)and Tujing(Site 3),Chongqing,China were chosen as sampling sites.Results indicated that TCB did not have significant difference in the top(0–10 cm)and sub(10–20 cm)soil of the non-inundated area(175–180 m),but showing a significant difference in the water level fluctuation zone(\175 m,suggesting that water level fluctuation had an important effect on TCB.Moreover,TCB in soils of various altitudes of Site 1 and 2had significant difference,while for Site 3,this difference was not significant.And the difference of TCB in Site 2was much greater than that in Site 1.These results suggested that there were significant differences for TCB in soils of mainstream and tributaries.In addition,TCB in soils of 10–20 cm had significant or highly significant positive correlations with MeHg level(r C 0.762,P B 0.048),thus we assumed that there may be some aerobic microorganisms playing dominant roles in mercury methylation.Yuping Xiang Hongxia Du Hong Shen Cheng Zhang Dingyong Wang 2014Chinese Science Bulletin2014,59,24:10
3A hybrid quantum-based PIO algorithm for global numerical optimization显示文摘A novel hybrid quantum-based pigeon-inspired optimization(PIO) algorithm for global numerical optimization is proposed to perceive deceptiveness and preserve diversity. In the proposed algorithm,the current best solution is regarded as a linear superposition of two probabilistic states, namely positive and deceptive. Through a quantum rotation gate, the positive probability is either enhanced or reset to balance exploration and exploitation. Simulation results reveal that the hybrid quantum-based PIO algorithm demonstrates an outstanding performance in global optimization owing to preserving diversity in the early evolution. As a result, the stability of the algorithm is enhanced so that the precision of optimization is improved statistically. 7Performance limitations in trajectory tracking control for air-breathing hypersonic vehicles显示文摘An integrated approach that considers the performance limitations of tracking control systems for air-breathing hypersonic vehicles is proposed. First, a set of ascent trajectories is obtained as candidates for tracking control through a trajectory design method that considers the available acceleration. Second, the basic theory of performance limitations, which is adopted to calculate the limits on control performance through the trajectory, is integrated. The openloop dynamics of air-breathing hypersonic vehicles is responsible for these limits on the control system. Comprehensive specifications on stability, tracking accuracy, and robustness are derived, and the flight envelope with constraints and control specifications is identified. 9Water level fluctuations influence microbial communities and mercury methylation in soils in the Three Gorges Reservoir,China显示文摘Reservoirs tend to have enhanced methylmercury(MeHg) concentrations compared to natural lakes and rivers, and water level fluctuations can promote MeHg production. Until now, little research has been conducted on the effects of microorganisms in soils for the formation of MeHg during different drying and flooding alternating conditions in the Three Gorges Reservoir(TGR). This study aimed to understand how water level fluctuations affect soil microbial composition and mercury concentrations, and if such microbial variations are related to Hg methylation. The results showed that MeHg concentrations and the ratios of MeHg to THg(MeHg%) in soils were higher in the seasonally drying and flooding alternating areas(DFAs, 175–155 m) than those in the non-inundated(NIAs, > 175 m) and inundated areas(IAs, < 145 m). However, MeHg% in all samples was less than 1%, indicating that the Hg methylation activity in the soils of the TGR was under a low level. 454 highthroughput sequencing of 16 S rRNA gene amplicons showed that soil bacterial abundance and diversity were relatively higher in DFA compared to those in NIA and IA, and microbial community composition varied in these three areas. At the family level, those groups in Deltaproteobacteria and Methanomicrobia that might have many Hg methylators were also showed a higher relative abundance in DFA, which might be the reason for the higher MeHg production in these areas. 10Effect of Zibushenjing Fang on skeletal development and brain tissue antioxidation in mice with kidney essence insufficiency: a mechanistic study显示文摘OBJECTIVE: To investigate the effects of Zibushenjing Fang (formula for tonifying the kidney essence) on skeletal development and brain tissue antioxidation in mice with kidney essence insufficiency. METHODS: Fifty male Kunming mice were randomly divided into five groups: normal group, model group, Jinkuishenqi Wan group, Zibushenjing Fang high dose group, and Zibushenjing Fang low dose group, with 10 mice in each group. The model of kidney essence insufficiency syndrome was established in all the mice except the normal group by using a cat to threaten the mice and by swimming until exhaustion daily which lasted about 21 days. Mice in the model group were administered 20 mL/ kg -1 · d -1 of normal saline intragastrically. The Jinkuishenqi Wan group was given 2.7 g/kg-1 · d-1 ofa solution of Jinkuishenqi Wan. The Zibushenjing Fang high dose group was given 20 g/kg -1 · d -1 and the Zibushenjing Fang low dose group was given 10 g/kg -1 · d -1 of a solution of Zibushenjing Fang.The general condition of all the groups was observed, including the quantity of food and water intake, swimming time, length of femur, and weight of the femur and musculus quadriceps femoris. The total superoxide dismutase (T-SOD), glutathione peroxi- dase (GSH-Px), and methane dicarboxylic aldehyde (MDA) activities in the brain tissues were detected. RESULTS: Zibushenjing Fang could improve the manifestation of kidney essence insufficiency, increase the quantity of food and water intake, swimming time, femur length, and femur and musculus quadriceps femoris weight. It could also regulate the activities of T-SOD, GSH-Px, and MDA in brain tissue. 14A photo-assisted electrochemical-based demonstrator for green ammonia synthesis显示文摘Bridging laboratory research and practical utilization is of crucial importance for the development of green ammonia synthetic technologies. A decentralized photo-assisted electrochemical-based demonstrator has been proposed for green ammonia synthesis from renewable electricity, air and water, where well-known defect-laden WO_(3) is used as the working electrode, and a commercially available PV panel supplies renewable electricity. In this demonstrator, defect-laden WO_(3) exhibits the optimum electrochemical NH_(3) formation rate(4.51 × 10^(-12)mol s^(-1)cm^(-2)) in 0.1 M K_(2)SO_(4)in a photovoltaic electrochemical(PV-EC) system. A system-level energy and cost analysis was conducted to investigate its economic viability and a general evaluation tool for system performance and cost estimation was proposed. 20Feature of thermohaline circulation in two-layer conceptual model based on energy constraint显示文摘The assertion that the thermohaline circulation(THC)is driven and sustained by mechanical energy has been increasingly accepted.The simplest conceptual model describing the THC is the Stommel two-box model.Given the vertical stratification in the real ocean,layered models were designed and used.In this research,using a two-layer conceptual model based on energy constraint,we studied basic features of thermal-mode and saline-mode circulations.We focused on the effects of freshwater flux and mixing energy on the intensity and multiple equilibrium states of the THC.The results show that more important than affecting the THC intensity,both the decrease of freshwater flux and increase of mixing energy can lead to an'abrupt transition'in the THC from a stable saline to a stable thermal mode,which further develops the THC energy theory.SHEN Yang GUAN YuPing 2015Science China Earth Sciences2015,58,8:1
