1Polymorphisms of three gene-derived STS on pig chromosome 13q41 are associated with susceptibility to enterotoxigenic Escherichia coli F4ab/ac in pigs显示文摘Neonatal diarrhea caused by enterotoxigenic Escherichia coli(ETEC)F4 is a common and serious disease,resulting in significant economical loss in the pig industry.The locus encoding ETEC F4 receptor has been mapped to pig chromosome(SSC)13q41,and one of the most significantly linked markers is S0075.In this study,we selected three genes including SLC12A8,MYLK and KPNA1 from a chromosomal region flanking S0075 on SSC13 to develop pig specific sequence tagged sites(STS). Seven single nucleotide polymorphisms were identified in the three pig STS using DNA of four full-sib susceptible and resistant animals in a White Duroc×Erhualian intercross.All grandparents,parents and 755 offspring in the intercross were genotyped for three polymorphisms,including SLC12A8 g.159A>G,MYLK g.1673A>G and KPNA1 g.306A>G.Family-based transmission disequilibrium test(TDT) revealed that all polymorphisms and the corresponding haplotypes are significantly associated with ETEC F4ab/ac(especially F4ac)brush border adhesion phenotypes,indicating that these polymor- phism are in linkage disequlibrium with causal mutation(s)of the gene encoding ETEC F4ab/ac receptor. 2Effects of Salvia miltiorrhiza Bge.f.alba on neuronal regeneration following cerebral ischemia/reperfusion显示文摘BACKGROUND:Subsequent to cerebral ischemic injury, endogenous neural stem cells are activated, but ischemia-induced neuronal loss is not compensated by ischemic injury-induced neural regeneration.Salvia (S.) miltiorrhiza Bge.f.alba (Baihua Danshen, a Chinese herbal medicine) could enhance learning and memory functions, as well as promote neural regeneration.OBJECTIVE:To observe the effects of S.miltiorrhiza Bge.f.alba on recovery from cerebral ischemia-reperfusion injury, and the influence on neuronal regeneration and differentiation.DESIGN, TIME AND SETTING:Randomized, controlled, animal experiments were performed at the Experimental Animal Center and Neurobiology Laboratory of Taishan Medical College in September of 2006.MATERIALS:S.miltiorrhiza Bge.f.alba was provided by Taishan Medical College Botanic Garden, Taian, China; dl-3n-butylphthalide (NBP) soft capsule was purchased from NBP Pharmaceutical, Shijiazhuang, China; mouse anti-bromodeoxyuridine antibody, rabbit anti-NF200 antibody, and bromodeoxyuridine were purchased from Sigma, Louis, MO, USA; Annexin V-fluorescein isothiocyanate/PI apoptosis kit was purchased from Nanjing Comissariado Biological Technology Development, Nanjing, China.METHODS:Adult Sprague Dawley rats were randomly assigned to sham surgery, model (cerebral ischemia and reperfusion, without administration), S.miltiorrhiza Bge.f.alba, and NBP groups.Following establishment of the cerebral ischemia/reperfusion model, S.miltiorrhiza Bge.f.alba or NBP (1 mL/100 g) was respectively perfused at 30 minutes following cerebral ischemia/reperfusion.MAIN OUTCOME MEASURES:Alterations in cerebral blood flow before and after ischemia/reperfusion, NF200-and bromodeoxyuridine-double positive cells in striatum of affected tissue as well as neuronal apoptosis rate at days 5 and 7 following cerebral ischemia/reperfusion.RESULTS:Subsequent to cerebral ischemia reperfusion, cerebral blood flow was reduced.Following treatment with S.miltiorrhiza Bge.f.alba, cerebral blood flow significantly increased (P<0.05).NBP treatment was inferior to S.miltiorrhiza Bge.f.alba with regard to stabilization of cerebra blood flow (P<0.05).S.miltiorrhiza Bge.f.alba significantly increased the number of newly formed neurons in rats following cerebral ischemia (P<0.05) and significantly reduced neuronal apoptosis (P<0.05), with no significant difference compared with NBP treatment (P>0.05).CONCLUSION:S.miltiorrhiza Bge.f.alba significantly increased cerebral blood flow, reduced neuronal apoptosis, and promoted neuronal regeneration in rats with cerebral ischemia/reperfusion impairment.Qiuling Zhang Yuanbiao Sun Bo Bai Hao Huang 2010Neural Regeneration Research2010,5,14:5

8PRMT5 determines the pattern of polyploidization and prevents liver from cirrhosis and carcinogenesis显示文摘Human hepatocellular carcinoma(HCC)occurs almost exclusively in cirrhotic livers.Here,we report that hepatic loss of protein arginine methyltransferase 5(PRMT5)in mice is sufficient to cause cirrhosis and HCC in a clinically relevant way.Furthermore,pathological polyploidization induced by hepatic loss of PRMT5 promotes liver cirrhosis and hepatic tumorigenesis in aged liver.The loss of PRMT5 leads to hyperaccumulation of P21 and endoreplication-dependent formation of pathological mono-nuclear polyploid hepatocytes.PRMT5 and symmetric dimethylation at histone H4 arginine 3(H4R3me2s)directly associate with chromatin of P21 to suppress its transcription.More importantly,loss of P21 rescues the pathological mono-nuclear polyploidy and prevents PRMT5-deficiency-induced liver cirrhosis and HCC.Thus,our results indicate that PRMT5-mediated symmetric dimethylation at histone H4 arginine 3(H4R3me2s)is crucial for preventing pathological polyploidization,liver cirrhosis and tumorigenesis in mouse liver.Jincheng Wang Xiang Huang Daoshan Zheng Qiuling Li Mei Mei Shilai Bao 2023Journal of Genetics and Genomics2023,50,2:1

11Experimental study on the regulation effect of Qiaoqin Qingfei agent on cough variant asthma based on airway neurogenic inflammation显示文摘Objective:To investigate the mechanism of regulation of airway neurogenic inflammation by Qiaoqin Qingfei agent in rats with cough variant asthma(CVA).Methods:48 SD rats were randomly divided into blank group,model group,montelukast sodium group(1.05 mg/kg)and high,medium and low dose groups(26,13,6.5 g/kg),with 8 rats in each group.The rat CVA model was established by the method of ovalbumin(OVA)combined with aluminum hydroxide(Al(OH)3)sensitization and repeated stimulation.From the second day of sensitization,the rat CVA model was given by gavage for 28 days.The pathological changes of lung tissue were observed under microscope by HE staining.The content changes of nerve growth factor(NGF)and substance P(SP)in alveolar lavage fluid(BALF)were determined by double-antibody sandwich ABC-ELISA,and the protein expression levels of NGF and SP in lung tissue were detected by immunohistochemistry.Results:Pathological findings showed significant inflammatory manifestations in the model group,and the inflammatory infiltration in the high-dose,medium-dose and low-dose groups of Qiaoqin Qingfei agent and montelukast sodium groups were alleviated to varying degrees.Compared with blank group,the protein expression levels of NGF and SP in lung tissue of model group were significantly increased(P<0.01).Compared with model group,the protein expression levels of NGF and SP in lung tissue and the contents of NGF and SP in alveolar lavage fluid in high-dose,medium-dose and low-dose groups and montelukast sodium group were significantly decreased(P<0.05).Conclusion:Qiaoqin Qingfei agent may reduce airway inflammation and relieve cough variant asthma by regulating the protein expression levels of NGF and SP in airway neurogenic inflammation.HUANG Hui-si HAN Chao SHI Yan-hong QI Chun-li XIONG Ai-hua ZHANG Qiuling LIANG Hui-ling 2023Journal of Hainan Medical University2023,29,9:0

13A New Edge Perturbation Mechanism for Privacy-Preserving Data Collection in IOT显示文摘A growing amount of data containing the sensitive information of users is being collected by emerging smart connected devices to the center server in Internet of things(IoT)era,which raises serious privacy concerns for millions of users.However,existing perturbation methods are not effective because of increased disclosure risk and reduced data utility,especially for small data sets.To overcome this issue,we propose a new edge perturbation mechanism based on the concept of global sensitivity to protect the sensitive information in IoT data collection.The edge server is used to mask users’sensitive data,which can not only avoid the data leakage caused by centralized perturbation,but also achieve better data utility than local perturbation.In addition,we present a global noise generation algorithm based on edge perturbation.Each edge server utilizes the global noise generated by the center server to perturb users’sensitive data.It can minimize the disclosure risk while ensuring that the results of commonly performed statistical analyses are identical and equal for both the raw and the perturbed data.Finally,theoretical and experimental evaluations indicate that the proposed mechanism is private and accurate for small data sets.CHEN Qiuling YE Ayong ZHANG Qiang HUANG Chuan 2023Chinese Journal of Electronics2023,32,3:0
