| 1 | Spatial-temporal Evolution and Determinants of the Belt and Road Initiative: A Maximum Entropy Gravity Model Approach显示文摘The spatial interaction model is an effective way to explore the geographical disparities inherent in the Belt and Road Initiative(BRI) by simulating spatial flows. The traditional gravity model implies the hypothesis of equilibrium points without any reference to when or how to achieve it. In this paper, a dynamic gravity model was established based on the Maximum Entropy(MaxEnt) theory to estimate and monitor the interconnection intensity and dynamic characters of bilateral relations. In order to detect the determinants of interconnection intensity, a Geodetector method was applied to identify and evaluate the determinants of spatial networks in five dimensions. The empirical study clearly demonstrates a heterogeneous and non-circular spatial structure. The main driving forces of spatial-temporal evolution are foreign direct investment, tourism and railway infrastructure construction, while determinants in different sub-regions show obvious spatial differentiation. Southeast Asian countries are typically multi-island area where aviation infrastructure plays a more important role. North and Central Asian countries regard oil as a pillar industry where power and port facilities have a greater impact on the interconnection. | 2 | Human Recombinant B7-H3 Expressed in E.colt Enhances T Lymphocyte Proliferation and IL-10 Secretion in Vitro显示文摘o explore the biofunctions of human B7-H3 on activated T lymphocyte, the gene of humanB7-H3 encoding the extracellular region (IgV-like and IgC-like domains) was obtained by RT-PCR from human lung cells and subcloned into the prokaryotic expression vector pGEX-5X-3 to express glutathioneS-transferase (GST) fusion protein, A 49 kD fusion protein (named as GST/hB7-H3 hereafter) was induced by IPTG and purified by standard methods reported in prokaryotic system. In the presence of the first signal imitated by anti-CD3 monoclonal antibody, T lymphocyte proliferation was observed by incubating purified T cells with soluble GST/hB7-H3 fusion protein by MTT assay. The concentrations of JFN-y and LL-10 in the supernatants of T cells were determined by ELISA. | 3 | The Functional Roles of Lipid Rafts in T Cell Activation,Immune Diseases and HIV Infection and Prevention显示文摘The first appearance of lipid rafts,or lipid rafts-like structure,was occasionally observed by cryo-electronic microscopy in 1980s as cavity,such as caveolae. However,the fully understanding of lipid raft was attributed by the studies of T cell activation,virus entry/budding,and other membrane events. During the interaction of T cell and antigen presenting cell,a highly organized structure is formed at the interface of the two cells,where cholesterol and sphingolipids are enriched,and form a liquid ordered phase that facilitates the signaling proteins on and off. Lipid rafts are also involved in virus entry and assembly. In this review,we will discuss cholesterol-sphingolipid floating microdomain,the lipid raft as a unique compartment of the plasma membrane,with biological functions that ensure correct intracellular traffic of proteins and lipids,such as protein-protein interactions by concentrating certain proteins in these microdomains,while excluding others. | 9 | Vitamin D attenuates TGF-β1-induced lung fibroblast proliferation and migration through repression of RasGRP3显示文摘Background:Transforming growth factor-β1(TGF-β1)is a pleiotropic cytokine that plays a central role in the pathogenesis of idiopathic pulmonary fibrosis(IPF).While previous studies have revealed a cross-talk between vitamin D and TGF-β1 signaling,it is still unclear how they interact with each other to regulate the progression of IPF.Methods:In this work,we searched for a novel mediator of TGF-β1 activity in lung fibroblasts and examined its regulation by vitamin D.In addition,we investigated the mechanism underlying the interaction between vitamin D and TGF-β1 signaling in lung fibroblast activation.Bioinformatic analysis was performed to identify TGF-β1 downstream target genes.Knockdown and overexpression expression experiments were conducted to determine gene function in the regulation of lung fibroblast proliferation and migration.Results:Analysis of publicly available datasets revealed that RAS guanyl releasing protein 3(RasGRP3)was upregulated in TGF-β1-treated lung fibroblasts and lung tissues from IPF patients relative to healthy controls.Our data confirmed the upregulation of RasGRP3 by TGF-β1 in human MRC5 lung fibroblasts.Overexpression of RasGRP3 enhanced MRC5 cell proliferation and migration.Knockdown of RasGRP3 blocked TGF-β1-induced MRC5 proliferation and migration.Vitamin D abolished TGF-β1-induced RasGRP3 upregulation,which was reversed by inhibition of the vitamin D receptor(VDR).Mechanistically,vitamin D promoted VDR enrichment and prevented mothers against decapentaplegic homolog(SMAD)2 and 3 occupancy at the promoter of RasGRP3.Additionally,overexpression of RasGRP3 reversed the suppressive effect of vitamin D on MRC5 cell proliferation and migration.Conclusion:In conclusion,vitamin D antagonizes TGF-β1-induced lung fibroblast activation by repressing RasGRP3 transcription. | GUANQIONG HU RISHENG HUANG LUELUE LU QINSHI PAN XIAOXIN CHEN | 2023 | BIOCELL2023,47,6: | 0 |
