| 1 | Auxin response factor 6A regulates photosynthesis,sugar accumulation,and fruit development in tomato显示文摘Auxin response factors(ARFs)are involved in auxin-mediated transcriptional regulation in plants.In this study,we performed functional characterization of SlARF6A in tomato.SlARF6A is located in the nucleus and exhibits transcriptional activator activity.Overexpression of SlARF6A increased chlorophyll contents in the fruits and leaves of tomato plants,whereas downregulation of SlARF6A decreased chlorophyll contents compared with those of wild-type(WT)plants.Analysis of chloroplasts using transmission electron microscopy indicated increased sizes of chloroplasts in SlARF6A-overexpressing plants and decreased numbers of chloroplasts in SlARF6A-downregulated plants.Overexpression of SlARF6A increased the photosynthesis rate and accumulation of starch and soluble sugars,whereas knockdown of SlARF6A resulted in opposite phenotypes in tomato leaves and fruits.RNA-sequence analysis showed that regulation of SlARF6A expression altered the expression of genes involved in chlorophyll metabolism,photosynthesis and sugar metabolism.SlARF6A directly bound to the promoters of SlGLK1,CAB,and RbcS genes and positively regulated the expression of these genes.Overexpression of SlARF6A also inhibited fruit ripening and ethylene production,whereas downregulation of SlARF6A increased fruit ripening and ethylene production.SlARF6A directly bound to the SAMS1 promoter and negatively regulated SAMS1 expression.Taken together,these results expand our understanding of ARFs with regard to photosynthesis,sugar accumulation and fruit development and provide a potential target for genetic engineering to improve fruit nutrition in horticulture crops. | Yujin Yuan Xin Xu Zehao Gong Yuwei Tang Mengbo Wu Fang Yan Xiaolan Zhang Qian Zhang Fengqing Yang Xiaowei Hu Qichen Yang Yingqing Luo Lihua Mei Wenfa Zhang Cai-Zhong Jiang Wangjin Lu Zhengguo Li Wei Deng | 2019 | Horticulture Research2019,6,1: | 3 |
| 2 | RING finger 138 deregulation distorts NF-κB signaling and facilities colitis switch to aggressive malignancy显示文摘Prolonged activation of nuclear factor(NF)-кB signaling significantly contributes to the development of colorectal cancer(CRC).New therapeutic opportunities are emerging from targeting this distorted cell signaling transduction.Here,we discovered the critical role of RING finger 138(RNF138)in CRC tumorigenesis through regulating the NF-кB signaling,which is independent of its Ubiquitin-E3 ligase activity involved in DNA damage response.RNF138^(−/−) mice were hyper-susceptible to the switch from colitis to aggressive malignancy,which coincided with sustained aberrant NF-кB signaling in the colonic cells.Furthermore,RNF138 suppresses the activation of NF-кB signaling pathway through preventing the translocation of NIK and IKK-Beta Binding Protein(NIBP)to the cytoplasm,which requires the ubiquitin interaction motif(UIM)domain.More importantly,we uncovered a significant correlation between poor prognosis and the downregulation of RNF138 associated with reinforced NF-кB signaling in clinical settings,raising the possibility of RNF138 dysregulation as an indicator for the therapeutic intervention targeting NF-кB signaling.Using the xenograft models built upon either RNF138-dificient CRC cells or the cells derived from the RNF138-dysregulated CRC patients,we demonstrated that the inhibition of NF-кB signaling effectively hampered tumor growth.Overall,our work defined the pathogenic role of aberrant NF-кB signaling due to RNF138 downregulation in the cascade events from the colitis switch to colonic neoplastic transformation and progression,and also highlights the possibility of targeting the NF-кB signaling in treating specific subtypes of CRC indicated by RNF138-ablation. | Yalan Lu Rong Huang Jianming Ying Xingchen Li Tao Jiao Lei Guo Haitao Zhou Han Wang Amannisa Tuersuntuoheti Jianmei Liu Qichen Chen Yanhong Wang Luying Su Changyuan Guo Fu Xu Ziyi Wang Yan Lu Kai Li Junbo Liang Zhen Huang Xiao Chen Jinjie Yao Hanjie Hu Xiaowen Cheng Yufeng Wan Xinyan Chen Ning Zhang Shiying Miao Jianqiang Cai Linfang Wang Changzheng Liu Wei Song Hong Zhao | 2022 | Signal Transduction and Targeted Therapy2022,7,7: | 0 |
| 3 | Smart industrial IoT empowered crowd sensing for safety monitoring in coal mine显示文摘The crowd sensing technology can realize the sensing and computing of people,machines,and environment in smart industrial IoT-based coal mine,which provides a solution for safety monitoring through distributed intelligence optimization.However,due to the difficulty of neural network training to achieve global optimality and the fact that traditional LSTM methods do not consider the relationship between adjacent machines,the accuracy of human body position prediction and pressure value prediction is not high.To solve these problems,this paper proposes a smart industrial IoT empowered crowd sensing for safety monitoring in coal mine.First,we propose a Particle Swarm Optimization-Elman Neural Network(PE)algorithm for the mobile human position prediction.Second,we propose an ADI-LSTM neural network prediction algorithm for pressure values of machines supports in underground mines.Among them,our proposed PE algorithm has the lowest average cumulative prediction error,and the trajectory fit rate is improved by 24.1%,13.9%and 8.7%compared with Kalman filtering,Elman and Kalman plus Elman algorithms,respectively.Meanwhile,compared with single-input ARIMA,RNN,LSTM,and GRU,the RMSE values of our proposed ADI-LSTM are reduced by 36.6%,52%,32%,and 13.7%,respectively;and the MAPE values are reduced by 0.0003%,0.9482%,1.1844%,and 0.3620%,respectively. | Jing Zhang Qichen Yan Xiaogang Zhu Keping Yu | 2023 | Digital Communications and Networks2023,9,2: | 0 |
| 4 | A novel method for identification and relative quantification of N-terminal peptides using metal-element-chelated tags coupled with mass spectrometry显示文摘By means of the reaction between a DOTA-NHS-ester bifunctional reagent and N-terminal peptides of proteins,and then chelation of lanthanide metal ions as tags,we established a novel method for the identification of N-terminal peptides of proteins and their relative quantification using metal-element-chelated tags coupled with mass spectrometry.The experimental results indicate that metal elements are able to completely label N-terminal peptides at the protein level.The N-terminal peptides are enriched as the peptides digested with trypsin are selectively eliminated by isothiocyanate-coupled silica beads.We successfully identified the N-terminal peptides of 158 proteins of Thermoanaerobacter tengcongensis incubated at 55 and 75°C,among which N-terminal peptides of 24 proteins are partially acetylated.Moreover,metal-element tags with high molecule weights make it convenient for N-terminal peptides consisting of less than 6 amino acids to be identified;these make up 55percent of the identified proteins.Finally,we developed a general approach for the relative quantification of proteins based on N-terminal peptides.We adopted lysozyme and ribonuclease B as model proteins;the correlation coefficients(R2)of the standard curves for the quantitative method were 0.9994 and 0.9997,respectively,with each concentration ratio ranging from0.1 to 10 and both relative standard derivations(RSD)measured at less than 5%.In T.tengcongensis at two incubation temperatures,80 proteins possess quantitative information.In addition,compared with the proteins of T.tengcongensis incubated at 55°C,in T.tengcongensis incubated at 75°C,7 proteins upregulate whereas 16 proteins downregulate,and most differential proteins are related to protein synthesis. | YAN Hui HAO FeiRan CAO QiChen LI JiaBin LI NanNan TIAN Fang BAI HaiHong REN XiaoJun LI XianYu ZHANG YangJun QIAN XiaoHong | 2014 | Science China Chemistry2014,57,5: | 0 |
