[Methods]The effects of 6-BA concentration,NAA concentration,potato amount and the amount of mashed banana on the growth of seedlings were determined by single factor experiment and were analyzed by Box-Behnken design and response surface methodology. [Results]The optimal culture conditions: rooting medium 1/2 MS + 6-BA 0. 24 mg/L + mashed banana 87. 63 g/L + potato 89. 30 g/L + NAA 0. 52 mg/L + sucrose 20. 0 g/L + activated carbon 4. 0 g/L + agar 7. 0 g/L,p H 5. 8,and light intensity 2 000 Lx. | 3 | Scalp acupuncture Yikang therapy on Baihui(GV20),Sishencong(EX-HN1),Zhisanzhen,Niesanzhen improves neurobehavior in young rats with cerebral palsy through Notch signaling pathway显示文摘OBJECTIVE:To investigate the efficacy of scalp acupuncture Yikang therapy on Baihui(GV20),Sishencong(EX-HN1),Zhisanzhen,Niesanzhen,on neurobehavior in young rats with cerebral palsy based on Notch signaling pathway.METHODS:Thirty 7-day-old rats were randomly divided into sham,model and acupuncture,10 rats in each group.The cerebral palsy model was established by the accepted modeling method,the acupuncture group selected'Baihui(GV20)','Sishencong(EX-HN1)','Zhisanzhen'and'Niesanzhen'for intervention 24 h after the model was made.The body masses were recorded before and after the treatment,respectively.After the intervention,the rats were subjected to suspension experiment,slope experiment,tactile stimulation experiment and Morris water maze experiment.After the end of the experiment,the morphological changes of hippocampal histology were observed by hematoxylineosin(HE)staining under light microscope,and the expression of Notch1,Notch3 and Hes5 were detected by Western blot and quantitative real-time polymerase chain reaction(PCR).RESULTS:The changes in body mass of the rats in each group were different;in behavioral experiments,compared with the sham,the suspension time of the model was shortened,the slope experiment,tactile stimulation experiment,and escape latency time were prolonged,and the number of platform crossing was reduced in the model,compared with the model,the suspension time of the acupuncture was prolonged,the slope experiment,tactile stimulation experiment,and escape latency time were shortened,and the number of platform crossing times was increased;HE staining showed severe hippocampal damage in the model and reduced hippocampal damage in the acupuncture.Western Blot and real-time fluorescence quantitative PCR showed that the expression of Notch1,Notch3 and Hes5 were increased in the model and the expression of Notch1,Notch3,Hes5 in acupuncture were decreased.CONCLUSIONS:Scalp acupuncture Yikang therapy may improve neurobehavior and reduce brain injury in rats with cerebral palsy by downregulating the expression of Notch1,Notch3,and Hes5. | XUE Jianyi XU Jinyan HUANG Mao YU Wentao YAN Yihui YAN Yuanjie YIN Zhenjin LU Qian PENG Wanying YAN Siyang | 2023 | Journal of Traditional Chinese Medicine2023,43,2: | 1 |
| 5 | RPL18aB helps maintain suspensor identity during early embryogenesis显示文摘During embryogenesis, plants are thought to use a mechanism that allows the suspensor to maintain its identity. Here, we reported that RPL_18aB is involved in this mechanism in Arabidopsis thaliana. The suspensor cells proliferated in rpl_18aB and formed a multicellular structure rather than undergo programmed cell death, as in wild type. Suspensors of rpl_18aB expressed the embryo proper marker, DRN::GFP, but not the suspensor marker, WOX8::GFP. In addition, auxin accumulated throughout the suspensors of rpl_18aB proembryos. Suspensor-specific expression of RPL_18aB could rescue the cell proliferation defects in rpl_18aB suspensors.
