3Variation of Cd concentration in various rice cultivars and derivation of cadmium toxicity thresholds for paddy soil by species-sensitivity distribution显示文摘It is imperative to derive an appropriate cadmium(Cd) health risk toxicity threshold for paddy soils to ensure the Cd concentration of rice grains meet the food safety standard. In this study, 20 rice cultivars from the main rice producing areas in China were selected, and a pot-experiment was conducted to investigate transformation of Cd in paddy soil-rice system with 0(CK), 0.3 mg kg–1(T1) and 0.6 mg kg–1(T2) Cd treatments in greenhouse. The results showed that Cd concentrations of rice grains existed significant difference(P<0.05) in 20 rice cultivars under the same Cd level in soil. The Cd concentrations of rice grains of the CK, T1 and T2 treatments were in the range of 0.143–0.202, 0.128–0.458 and 0.332–0.806 mg kg–1, respectively. Marked differences of the ratios of Cd concentration for soil to rice grain(BCFs) and transfer factors(TFs, root to grain and straw to grain) among the tested cultivars were observed in this study. The bioconcentration factors(BCFgrain) and TFs of the 20 rice cultivars were 0.300–1.112 and 0.342–0.817, respectively. The TFs of Cd from straw to grain ranged from 0.366 to 1.71, with significant differences among these 20 rice cultivars. The bioconcentration factors(BCFgrain) and TFs among the 20 rice cultivars ranged from 0.300–1.112 and 0.342–0.817, respectively. The species-sensitivity distribution(SSD) of Cd sensitivity of the rice species could be fitted well with Burr-III(R2=0.987) based on the data of BCFs. 5Sleep and Cognitive Abnormalities in Acute Minor Thalamic Infarction显示文摘In order to characterize sleep and the cognitive patterns in patients with acute minor thalamic infarction(AMTI), we enrolled 27 patients with AMTI and 12 matched healthy individuals. Questionnaires about sleep and cognition as well as polysomnography(PSG) were performed on days 14 and 90 post-stroke. Compared to healthy controls, in patients with AMTI, hyposomnia was more prevalent; sleep architecture was disrupted as indicated by decreased sleep efficiency, increased sleep latency, and decreased non-rapid eye movement sleep stages 2 and 3;more sleep-related breathing disorders occurred; and cognitive functions were worse, especially memory. While sleep apnea and long-delay memory recovered to a large extent in the patients, other sleep and cognitive function deficit often persisted. Patients with AMTI are at an increased risk for hyposomnia, sleep structure disturbance,sleep apnea, and memory deficits. 10Hot deformation behavior and constitutive model of TC18 alloy during compression显示文摘The hot deformation behavior of TC18 alloy at strain rates ranging from 1 9 10-4to 1 9 10-2s-1and temperatures ranging from 25 to 800 °C was studied using a WDW-300 electronic universal testing machine. The relationships between true stress and true strain show that flow stress decreases with the increase of temperature and increases as strain rate increases. The effect of strain rate on the flow stress becomes pronounced at higher temperatures. At room temperature, the river pattern characteristic of brittle fracture and the dimple pattern typical of ductile fracture are found to exist in different regions of fracture surfaces of the samples. An improved constitutive relationship is proposed to accurately describe the flow stress of TC18 by considering the effect of strain. And a microscopic model is also deduced which can link the physical mechanisms to the macroscopic experimental results. 12Petroleum hydrocarbons and their effects on fishery species in the Bohai Sea, North China显示文摘Systematic studies on the changes in concentrations of petroleum hydrocarbons (PHs) and their effects on fishery species in the Bohai Sea during 1974-2004 are presented. Changes in PHs concentrations were closely related to Yellow River runoff. Concentrations of PHs accumulated in fish and shrimp increased by about 0.712 mg/kg dry weight when trophic level of fish and shrimps increased by 1. Attention should also be paid to the high PHs concentrations in mollusks along the coastal waters of the Bohai Sea. Average concentration of PHs in the adjacent coastal waters of Tianjin City during 1996-2005 decreased the population growth rates of fish, crustaceans and mollusks in the Bohai Sea by 2.58%, 6.59% and 2.33%, respectively. 15Efficacy of totally laparoscopic compared with laparoscopic-assisted total gastrectomy for gastric cancer: A meta-analysis显示文摘BACKGROUND Laparoscopic radical gastrectomy is currently the most common surgical approach for gastric cancer.The main difference between totally laparoscopic total gastrectomy(TLTG)and laparoscopic-assisted total gastrectomy(LATG)is the route of digestive tract reconstruction.However,TLTG is currently not widespread as the safety and feasibility of intracorporeal esophagojejunostomy is uncertain.AIM To compare the short-term efficacy of TLTG and LATG for radical gastrectomy of gastric cancer,and to determine the safety and feasibility of intracorporeal esophagojejunostomy.METHODS PubMed,EMBASE,and Web of Science databases were searched for all relevant articles regarding TLTG vs LATG for gastric cancer published up to October 1,2019.Inclusion and exclusion criteria were established.All the basic conditions of patients and important clinical data related to surgery were extracted,and a meta-analysis was performed with RevMan 5.3 software.RESULTS Eight studies involving a total of 1883 cases(869 cases in the TLTG group and 1014 cases in the LATG group)were included.Compared with the LATG group,reduced intraoperative blood loss(weighted mean difference=-35.37,95%CI:-61.69--9.06,P=0.008)and a larger number of retrieved lymph nodes(weighted mean difference=3.11,95%CI:-2.60-12.00,P=0.01)were found in the TLTG group.There were no significant differences in operating time,anastomotic time,tumor size,proximal resection margin length,postoperative pain score,time to first flatus,time to first oral intake,postoperative hospital stay,postoperative anastomosis-related complication rate and overall complication rate between the two groups(P>0.05).CONCLUSION Intracorporeal esophagojejunostomy is safe and feasible.TLTG has the advantages of being minimally invasive,reduced intraoperative blood loss and easier access to lymph nodes compared with LATG.Totally laparoscopic gastrectomy is likely to be the surgical trend for gastric cancer in the future.Song Wang Mei-Lan Su Yang Liu Zhi-Ping Huang Ning Guo Tian-Jin Chen Zhong-Hui Zou 2020World Journal of Clinical Cases2020,8,5:14
16Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 19Efficiency and Inheritance of Targeted Mutagenesis in Maize Using CRISPR-Cas9显示文摘CRISPR/Cas(clustered regularly interspaced short palindromic repeats/CRISPR-associated proteins) is an adaptive immune system in bacteria and archaea to defend against invasion from foreign DNA fragments. Recently, it has been developed as a powerful targeted genome editing tool for a wide variety of species. However, its application in maize has only been tested with transiently expressed somatic cells or with a limited number of stable transgenic T0 plants. The exact efficiency and specificity of the CRISPR/Cas system in the highly complex maize genome has not been documented yet. Here we report an extensive study of the well-studied type II CRISPR-Cas9 system for targeted genome editing in maize, with the codon-optimized Cas9 protein and the short non-coding guide RNA generated through a functional maize U6 sn RNA promoter. Targeted gene mutagenesis was detected for 90 loci by maize protoplast assay, with an average cleavage efficiency of 10.67%. Stable knockout transformants for maize phytoene synthase gene(PSY1) were obtained. Mutations occurred in germ cells can be stably inherited to the next generation. Moreover, no off-target effect was detected at the computationally predicted putative off-target loci. No significant difference between the transcriptomes of the Cas9 expressed and non-expressed lines was detected. 20Low-dose metformin treatment in the subacute phase improves the locomotor function of a mouse model of spinal cord injury显示文摘Metformin,a first-line drug for type-2 diabetes,has been shown to improve locomotor recovery after spinal cord injury.However,there are studies reporting no beneficial effect.Recently,we found that high dose of metformin(200 mg/kg,intraperitoneal)and acute phase administration(immediately after injury)led to increased mortality and limited locomotor function recovery.Consequently,we used a lower dose(100 mg/kg,i.p.)metformin in mice,and compared the effect of immediate administration after spinal cord injury(acute phase)with that of administration at 3 days post-injury(subacute phase).Our data showed that metformin treatment starting at the subacute phase significantly improved mouse locomotor function evaluated by Basso Mouse Scale(BMS)scoring.Immunohistochemical studies also revealed significant inhibitions of microglia/macrophage activation and astrogliosis at the lesion site.Furthermore,metformin treatment at the subacute phase reduced neutrophil infiltration.These changes were in parallel with the increased survival rate of spinal neurons in animals treated with metformin.These findings suggest that low-dose metformin treatment for subacute spinal cord injury can effectively improve the functional recovery possibly through anti-inflammation and neuroprotection.This study was approved by the Institute Animal Care and Use Committee at the University of Texas Medical Branch(approval No.1008041C)in 2010.Wen-Ye Song Han Ding Tiffany Dunn Jun-Ling Gao Javier Allende Labastida Caitlin Schlagal Guang-Zhi Ning Shi-Qing Feng Ping Wu 2021Neural Regeneration Research2021,16,11:12
