| 1 | Biosynthesis and molecular engineering of templated natural products显示文摘Bioactive small molecules that are produced by living organisms, often referred to as natural products(NPs), historically play a critical role in the context of both medicinal chemistry and chemical biology. How nature creates these chemical entities with stunning structural complexity and diversity using a limited range of simple substrates has not been fully understood. Focusing on two types of NPs that share a highly evolvable ‘template'-biosynthetic logic, we here provide specific examples to highlight the conceptual and technological leaps in NP biosynthesis and witness the area of progress since the beginning of the twenty-first century. The biosynthesis of polyketides, non-ribosomal peptides and their hybrids that share an assembly-line enzymology of modular multifunctional proteins exemplifies an extended ‘central dogma'that correlates the genotype of catalysts with the chemotype of products; in parallel, post-translational modifications of ribosomally synthesized peptides involve a number of unusual biochemical mechanisms for molecular maturation. | 3 | Strong partitioning of soil bacterial community composition and co-occurrence networks along a small-scale elevational gradient on Zijin Mountain显示文摘The elevational distributions of bacterial communities in natural mountain forests,especially along large elevational gradients,have been studied for many years.However,the distributional patterns that underlie variations in soil bacterial communities along small-scale elevational gradients in urban ecosystems are not yet well understood.Using Illumina MiSeq DNA sequencing,we surveyed soil bacterial communities at three elevations on Zijin Mountain in Nanjing City:the hilltop(300 m a.s.l.),the hillside(150 m a.s.l.),and the foot of the hill(0 m a.s.l.).The results showed that edaphic properties differed significantly with elevation.Bacterial community composition,rather than alpha diversity,strongly differed among the three elevations(Adonis:R2=0.12,P<0.01).Adonis and DistLM analyses demonstrated that bacterial community composition was highly correlated with soil pH,elevation,total nitrogen(TN),and dissolved organic carbon(DOC).The degree scores,betweenness centralities,and composition of keystone species were distinct among the elevations.These results demonstrate strong elevational partitioning in the distributions of soil bacterial communities along the gradient on Zijin Mountain.Soil pH and elevation together drove the smallscale elevational distribution of soil bacterial communities.This study broadens our understanding of distribution patterns and biotic co-occurrence associations of soil bacterial communities from large elevational gradients to short elevational gradients. | Xu Liu Teng Yang Yu Shi Yichen Zhu Mulin He Yunke Zhao Jonathan MAdams Haiyan Chu | 2021 | Soil Ecology Letters2021,3,4: | 2 |
| 4 | Development of gradient microstructure in the lattice structure of AlSi10Mg alloy fabricated by selective laser melting显示文摘To identify the microstructural features of the lattice structures of Al alloys built via the selective laser melting(SLM)process,AlSil OMg alloy with a body-centered cubic(BCC)-type lattice structure was prepared.Characteristic microstructures comprising melt pools with several columnarα-Al phases with<001>orientations along the elongation direction and surrounded by eutectic Si particles were observed at all portions of the built lattice structure.In the node portions of the lattice structure,a gradient microstructure(continuous change in microstructure)was observed.The columnarα-Al phases were observed near the top surface of the node portion,whereas they became coarser and more equiaxed near the bottom surface,resulting in softening localized near the bottom surface.In the strut portions of the lattice structure,the columnarα-Al phases were elongated along the inclined direction of struts.This trend was more prevalent near the bottom surface.Theα-Al phases became coarser and more equiaxed near the bottom surface as well.The aforementioned results were the basis of a discussion of the development of the gradient microstructure in lattice-structured Al alloys during the SLM process in terms of thermal conductivities at the boundaries between the manufactured(locally melted and rapidly solidified)portions and adjacent(unmelted)alloy powder. | Mulin Liu Naoki Takata Asuka Suzuki Makoto Kobashi | 2020 | Journal of Materials Science & Technology2020,36,1: | 2 |

| 9 | O-GlcNAcylation regulates phagocytosis by promoting Ezrin localization at the cell cortex显示文摘O-GlcNAcylation is a post-translational modification that serves as a cellular nutrient sensor and participates in multiple physiological and pathological processes.However,it remains uncertain whether O-GlcNAcylation is involved in the regulation of phagocytosis.Here,we demonstrate a rapid increase in protein OGlcNAcylation in response to phagocytotic stimuli.Knockout of the O-GlcNAc transferase or pharmacological inhibition of O-GlcNAcylation dramatically blocks phagocytosis,resulting in the disruption of retinal structure and function.Mechanistic studies reveal that the O-GlcNAc transferase interacts with Ezrin,a membrane-cytoskeleton linker protein,to catalyze its O-GlcNAcylation.Our data further show that Ezrin OGlcNAcylation promotes its localization to the cell cortex,thereby stimulating the membrane-cytoskeleton interaction needed for efficient phagocytosis.These findings identify a previously unrecognized role for protein O-GlcNAcylation in phagocytosis with important implications in both health and diseases. | Song Yang Hanyu Liu Hua Ni Lingyu Jiang Mulin Yang Quan Chen Jun Zhou Fan Yu | 2023 | Journal of Genetics and Genomics2023,50,7: | 0 |
