2Integration of Metabolomics and Subcellular Organelle Expression Microarray to Increase Understanding the Organic Acid Changes in Post-harvest Citrus Fruit显示文摘Citric acid plays an important role in fresh fruit favor and its adaptability to post-harvest storage conditions.In order to explore organic acid regulatory mechanisms in post-harvest citrus fruit,systematic biological analyses were conducted on stored Hirado Buntan Pummelo(HBP;Citrus grandis)fruits.High-performance capillary electrophoresis,subcellular organelle expression microarray,real-time quantitative reverse transcription polymerase chain reaction,gas chromatography mass spectrometry(GC-MS),and conventional physiological and biochemical analyses were undertaken.The results showed that the concentration of organic acids in HBP underwent a regular fuctuation.GC-MS-based metabolic profling indicated that succinic acid,g-aminobutyric acid(GABA),and glutamine contents increased,but 2-oxoglutaric acid content declined,which further confrmed that the GABA shunt may have some regulatory roles in organic acid catabolism processes.In addition,the concentration of organic acids was signifcantly correlated with senescence-related physiological processes,such as hydrogen peroxide content as well as superoxide dismutase and peroxidase activities,which showed that organic acids could be regarded as important parameters for measuring citrus fruit post-harvest senescence processes.Xiaohua Sun Andan Zhu Shuzhen Liu Ling Sheng Qiaoli Ma Li Zhang Elsayed Mohamed Elsayed Nishawy Yunliu Zeng Juan Xu Zhaocheng Ma Yunjiang Cheng Xiuxin Deng 2013Journal of Integrative Plant Biology2013,55,11:14
3Allogeneic Vγ9Vδ2 T-cell immunotherapy exhibits promising clinical safety and prolongs the survival of patients with late-stage lung or liver cancer显示文摘Vγ9Vδ2 T cells are promising candidates for cellular tumor immunotherapy.Due to their HLA-independent mode of action,allogeneic Vγ9Vδ2 T cells can be considered for clinical application.To apply allogeneic Vγ9Vδ2 T cells in adoptive immunotherapy,the methodology used to obtain adequate cell numbers with optimal effector function in vitro needs to be optimized,and clinical safety and efficacy also need to be proven.Therefore,we developed a novel formula to improve the expansion of peripheralγδT cells from healthy donors.Then,we used a humanized mouse model to validate the therapeutic efficacy of expandedγδT cells in vivo;furthermore,the expandedγδT cells were adoptively transferred into late-stage liver and lung cancer patients.We found that the expanded cells possessed significantly improved immune effector functions,including proliferation,differentiation,and cancer cell killing,both in vitro and in the humanized mouse model.Furthermore,a phase I clinical trial in 132 late-stage cancer patients with a total of 414 cell infusions unequivocally validated the clinical safety of allogeneic Vγ9Vδ2 T cells.Among these 132 patients,8 liver cancer patients and 10 lung cancer patients who received≥5 cell infusions showed greatly prolonged survival,which preliminarily verified the efficacy of allogeneic Vγ9Vδ2 T-cell therapy.Our clinical studies underscore the safety and efficacy of allogeneic Vγ9Vδ2 T-cell immunotherapy,which will inspire further clinical investigations and eventually benefit cancer patients.Yan Xu Zheng Xiang Mohammed Alnaggar Léonce Kouakanou Jiawei Li Junyi He Jiashuang Yang Yi Hu Yan Chen Li Lin Jianlei Hao Jingxia Li Jibing Chen Man Li Qingling Wu Christian Peters Qinghua Zhou Jianshuang Li Yingqing Liang Xiaohua Wang Baohui Han Meili Ma Dieter Kabelitz Kecheng Xu Wenwei Tu Yangzhe Wu Zhinan Yin 2021Cellular & Molecular Immunology2021,18,2:8

5Growth and wood properties of a 38-year-old Populus simonii 3 P.nigra plantation established with different densities in semi-arid areas of northeastern China显示文摘To characterize the effects of plantation densities on the growth characteristics(diameter at breast level,tree height and volume)and the common wood properties,38-year-old Populus simonii 9 P.nigra clones planted with four levels of spacing(2 m 92 m,3 m 93 m,4 m 94 m,and 5 m 95 m)in a semi-arid area in northeastern China were examined.The results of ANOVA showed significant differences(P\0.01)for all the investigated growth traits and wood properties under different plantation densities,except for the chemical composition of wood.The repeatability and phenotypic variations of all the traits varied from 0.34 to 0.99 and from 13.45 to 59.65%,respectively.Except for wood density,which was significantly negatively correlated with the growth traits,a positive correlation was observed between the growth traits and all the other wood mechanical properties.However,most of the correlations between the growth traits and the chemical composition of the wood were not significant.The path analysis for the wood mechanical characteristics and the growth in the prediction of volume were significant and ranged from 0.18 to 0.72 for wood density and diameter at breast height,respectively,while those for the chemical composition of wood ranged from 0.001 to 0.336,which showed a low impact on the volume.The highest stand volume(610 m3)per hectare was observed with the 2 m 92 m spacing,which consequently provided a high total price and income,while a high individual volume growth per tree was observed with the 5 m 95 m spacing.The results suggested that for the poplar trees younger than 40 years in a semi-arid area in China,2 m 92 m spacing is suitable for obtaining a high volume per hectare,while 5 m 95 m spacing is best for obtaining a high individual volume per tree.Ammar Khalil Mohamed Ahmed Zhixiang Fu Changjun Ding Luping Jiang Xidong Han Aiguo Yang Yinghui Ma Xiyang Zhao 2020Journal of Forestry Research2020,31,2:6
6Phase Ⅱ study of apatinib in combination with oral vinorelbine in heavily pretreated HER2-negative metastatic breast cancer and clinical implications of monitoring ctDNA显示文摘Objective:Apatinib is an oral TKI targeting VEGFR-2.Single-agent apatinib treatment has been shown to produce an objective response in patients with pretreated m BC.Oral vinorelbine also holds promise as a treatment of choice in patients with m BC.This study aimed to investigate the efficacy and safety of the oral vinorelbine-apatinib combination in patients with pretreated m BC.In addition,we detected gene variants in ct DNA to explore the therapeutic implications.Methods:This study enrolled patients with HER2-negative m BC who were pretreated with anthracycline/taxanes.Patients were treated with apatinib at 500 mg/425 mg daily plus oral vinorelbine 60 mg/m2 on days 1,8,and 15 of every cycle(3 weeks).The primary endpoint was PFS.The secondary endpoints were ORR,CBR,OS,and safety.Patients eligible for ct DNA detection were evaluated before and during treatment.Results:Forty patients were enrolled.The median PFS was 5.2 months(95%CI,3.4–7.0 months),and the median OS was 17.4 months(95%CI,8.0–27.0 months).The ORR was 17.1%(6/35),and the CBR was 45.7%(16/35).The most common AEs included gastrointestinal reaction,myelosuppression,and hypertension.In 20 patients,ct DNA was detected at baseline and during treatment.A significant difference was found in PFS for undetected vs.detected baseline ct DNA(13.9 months vs.3.6 months,P=0.018).Conclusions:All-oral therapy with apatinib plus vinorelbine displayed objective efficacy in patients with heavily pretreated HER2-negative m BC,with acceptable and manageable toxicity profiles.Patients with no gene variant detected and lower variant allele frequencies in ct DNA at baseline showed longer PFS.Anjie Zhu Peng Yuan Nanlin Hu Mingzhou Li Wenmiao Wang Xue Wang Jian Yue Jiayu Wang Yang Luo Fei Ma Pin Zhang Qing Li Binghe Xu Shanbo Cao Giuseppe Lippi Yoichi Naito Mohammed A.Osman Gustavo N.Marta Gianluca Franceschini Armando Orlandi 2021Cancer Biology & Medicine2021,18,3:4

8Effect of oxygen plasma treatment on the nanofiltration performance of reduced graphene oxide/cellulose nanofiber composite membranes显示文摘Graphene based nanosheets have been widely used as building blocks for fabrication of superior separation membrane for water processing.In particular,membranes made of reduced graphene oxide(rGO)show better stability compared with graphene oxide(GO).However,densely stacked of rGO often results in low water flux.In this study,cellulose nanofibers(CNFs)were incorporated into the rGO laminates by vacuum filtration of dilute GO/CNF solution and thermal reduction at 150C for 1.5 h.The resulting rGO/CNF membrane was treated with oxygen plasma for 1–4 min to create nanopores on the membrane surface for the purpose of enhancing nano-filtration performance.The results showed that the optimum membrane performance was obtained by using the equal amount of GO(31.83 mg m^(-2))and CNFs accompanied by 3 min of plasma treatment,exhibiting a pure water permeance of 37.23.9 L m^(-2)h^(-1)bar^(-1)maintaining a rejection above 90%for Acid Fuchsin(1.2×1.1 nm),Rose Bengal(1.5×1.2 nm)and Brilliant Blue(2.2×1.7 nm).Shabin Mohammed Hanaa MHegab Ranwen Ou Shasha Liu Hongyu Ma Xiaofang Chen Tam Sridhar Huanting Wang 2021Green Chemical Engineering2021,2,1:3
9High-efficiency electrocatalytic NO reduction to NH_(3) by nanoporous VN显示文摘Electrocatalytic NO reduction reaction to generate NH_(3)under ambient conditions offers an attractive alternative to the energy-extensive Haber-Bosch route;however,the challenge still lies in the development of cost-effective and high-performance electrocatalysts.Herein,nanoporous VN film is first designed as a highly selective and stable electrocatalyst for catalyzing reduction of NO to NH_(3)with a maximal Faradaic efficiency of 85%and a peak yield rate of 1.05×10^(-7)mol·cm^(-2)·s^(-1)(corresponding to 5,140.8mg·h^(-1)·mg_(cat).^(-1))at-0.6 V vs.reversible hydrogen electrode in acid medium.Meanwhile,this catalyst maintains an excellent activity with negligible current density and NH_(3)yield rate decays over 40 h.Moreover,as a proof-of-concept of Zn-NO battery,it delivers a high power density of 2.0 mW·cm^(-2)and a large NH_(3)yield rate of 0.22×10^(-7)mol·cm^(-2)·s^(-1)(corresponding to 1,077.1mg·h^(-1)·mg_(cat).^(-1)),both of which are comparable to the best-reported results.Theoretical analyses confirm that the VN surface favors the activation and hydrogenation of NO by suppressing the hydrogen evolution.This work highlights that the electrochemical NO reduction is an eco-friendly and energy-efficient strategy to produce NH_(3).Defeng Qi Fang Lv Tianran Wei Mengmeng Jin Ge Meng Shusheng Zhang Qian Liu Wenxian Liu Dui Ma Mohamed SHamdy Jun Luo Xijun Liu 2022Nano Research Energy2022,1,2:2
10Identification of sequence-specific interactions of the CD44-intracellular domain with RUNX2 in the transcription of matrix metalloprotease-9 in human prostate cancer cells显示文摘Aim:The Cluster of differentiation 44(CD44)transmembrane protein is cleaved byγ-secretase,the inhibition of which blocks CD44 cleavage.This study aimed to determine the biological consequence of CD44 cleavage and its potential interaction with Runt-related transcription factor(RUNX2)in a sequence-specific manner in PC3 prostate cancer cells.Methods:Using full-length and C-terminal deletion constructs of CD44-ICD(D1-D5)expressed as stable green fluorescent protein-fusion proteins in PC3 cells,we located possible RUNX2-binding sequences.Results:Chromatin immunoprecipitation assays demonstrated that the C-terminal amino acid residues between amino acids 671 and 706 in D1 to D3 constructs were indispensable for sequence-specific binding of RUNX2.This binding was minimal for sequences in the D4 and D5 constructs.Correspondingly,an increase in matrix metalloprotease-9(MMP-9)expression was observed at the mRNA and protein levels in PC3 cells stably expressing D1–D3 constructs.Conclusion:These results provide biochemical evidence for the possible sequence-specific CD44-ICD/RUNX2 interaction and its functional relationship to MMP-9 transcription in the promoter region.Linda T.Senbanjo Hanan AlJohani Mohammed AlQranei Sunipa Majumdar Tao Ma Meenakshi A.Chellaiah 2020Cancer Drug Resistance2020,3,3:2
11Exposure to herbicides reduces larval sensitivity to insecticides in Spodoptera litura (Lepidoptera: Noctuidae)显示文摘Herbicides and insecticides are widely used in modern agriculture. It has been reported in various studies that application of insecticides can increase tolerance of herbivorous insects to insecticides. However, limited information exists on susceptibility to insecticides when insects are exposed to herbicides. This study was conducted to investigate the potential impact of the herbicides trifluralin and 2-methyl-4-chlorophenoxyacetic acid sodium salt (MCPA-Na) on the susceptibility of the nocturnal moth Spodoptera litura to the insecticides X-cyhalothrin, phoxim and bifenthrin. We found that larvae exposed to trifluralin or MCPA-Na became significantly less susceptible to both insecticides than nonexposed control larvae. Herbicide-treated larvae did not show altered growth under the used test conditions. However, heads of herbicide-treated larvae showed increased expression of the acetylcholinesterase genes SI Ace I and SI Ace 2. Moreover, the fat body and midgut of herbicide-treated larvae displayed elevated expression of detoxification genes (the carboxylesterase gene SI CarE;the glutathione S-transferase genes SlGSTe2 and SlGSTe3\ the cytochrome P450 monooxygenase genes CYP6B48, CYP9A40 and CYP321B1). The CYP6B48 gene exhibited highest inducibility. 12Insights into the sandwich-like ultrathin Ni-doped MoS_(2)/rGO hybrid as effective sulfur hosts with excellent adsorption and electrocatalysis effects for lithium-sulfur batteries显示文摘The design of sulfur hosts with high conductivity,large specific surface area,strong adsorption and electrocatalytic ability is crucial to advance high performance lithium-sulfur batteries.Herein,a novel ultrathin sandwich-type Ni-doped MoS_(2)/reduced graphene oxide(denote as Ni-doped MoS_(2)/rGO) hybrid is developed as a sulfur host through a simple one-step hydrothermal route.The two-dimensional layered structure Ni-doped MoS_(2)/rGO hybrid with heterostructure and heteroatom architecture defects not only plays a key role in adsorption of lithium polysulfide but also catalyzes on redox kinetics of sulfur and polysulfide species.Meanwhile,it can contribute to the large specific surface area for Li_(2) S/S_8 deposition,fast Li-ion and electron transportation,thus enhancing the electrocatalytic properties,as confirmed firstly by cyclic voltammetry(CV) results.Due to the adsorption-catalytic synergistic effect,the Ni-doped MoS_(2)/rGO cathode exhibits high specific capacity(1343.6 mA h g^(-1) at 0.2 C,921.6 mA h g^(-1) at 1 C),high coulombic efficiency and an outstanding cycle stability(with the low attenuation rate of 0.077% per cycle over 140 cycles at 0.5 C and 0.11% per cycle over 400 cycles at 1 C,respectively).This work proposes some inspiration for exploring the construction of advanced lithium-sulfur batteries through the rational design defects of atomic structure and electronic states of MoS_(2) as sulfur host.Ran Zhang Yutao Dong Mohammed AAl-Tahan Yingying Zhang Ruipeng Wei Yuhang Ma Changchun Yang Jianmin Zhang 2021Journal of Energy Chemistry2021,30,9:2
