| 2 | Soya saponin improves egg-laying performance and immune function of laying hens显示文摘Background:Soya saponin(SS),an active compound in soybean meals,has been widely studied in the medical field.However,it was considered as an anti-nutritional factor in poultry diets.The objective of this experiment was to measure the effects of dietary SS using three dietary treatments on egg-laying performance and immune function of laying hens.Birds were fed a low soybean meal basal diet(CON),a low-SS diet(50 SS)containing 50mg/kg SS,or a high-SS diet(500 SS)containing 500 mg/kg SS for 10 weeks.At the end of the 5th and 10th week of the trial,samples were collected for analysis.Results:Results showed that with 50 mg/kg SS supplementation,the egg production rate,feed conversion ratio(FCR),and eggshell quality tended to be improved.Serum follicle stimulating hormone(FSH)and Interleukin-4(IL-4)levels were also elevated as well as the peripheral blood LPS stimulation index,the proportion of B lymphocytes,and antibody titer of bovine serum albumin(BSA).We also found that mRNA levels of follicle stimulating hormone receptor(FSHR)in ovarian,nuclear transcription factor kappa B(NF-κB),Transforming growth factor(TGF-β)and interferonγ(IFN-γ)in spleen were up-regulated at the end of the trial.Additionally,dietary 50 mg/kg SS improved the ileal flora via up-regulating the relative abundance of Lactobacillus,Romboutsia and Lactobacillus delbrueckii.Although the immune related indicators were improved with 500 mg/kg SS supplemented,it seemed to have a negative influence on the laying-performance.Specifically,serum alanine aminotransferase(ALT),alkaline phosphatase(ALP),and the ratio of IFN-γto IL-4 were increased in the 500 SS group at the end of the trial.The m RNA levels of gonadotropin releasing hormone 1(GnRH1)in Hypothalamus,the estrogen related receptor(ERR)in ovaries were downregulated as well as the egg production rate during the trial with 500 mg/kg SS supplemented.Conclusions:The egg production performance was improved by dietary supplemented with 50 mg/kg SS via increasing ovarian FSHR transcription level and serum estrogen level.A beneficial shift in intestinal microflora was recorded,and the immune function of laying hens was also improved with 50 mg/kg SS supplementation.Surprisingly,the long-term supplementation of 500 mg/kg SS exerted a negative impact on the laying performance and physiological functions of the liver of laying hens. | Peng Li Yizhu Zhao Shaojia Yan Bocheng Song Yongfa Liu Mingkun Gao Dazhi Tang Yuming Guo | 2022 | Journal of Animal Science and Biotechnology2022,13,3: | 5 |

| 5 | Application of next generation sequencing technology on contamination monitoring in microbiology laboratory显示文摘The surveillance and prevention of pathogenic microbiological contamination are the most important tasks of biosafety management in the lab.There is an urgent need to establish an effective and unbiased method to evaluate and monitor such contamination.This study aims to investigate the utility of next generation sequencing(NGS)method to detect possible contamination in the microbiology laboratory.Environmental samples were taken at multiple sites at the lab including the inner site of centrifuge rotor,the bench used for molecular biological tests,the benches of biosafety cabinets used for viral culture,clinical sample pre-treatment and nucleic acids extraction,by scrubbing the sites using sterile flocked swabs.The extracted total nucleic acids were used to construct the libraries for deep sequencing according to the protocol of Ion Torrent platform.At least 1G raw data was obtained for each sample.The reads of viruses and bacteria accounted for 0.01±0.02%,and 77.76±12.53%of total reads respectively.The viral sequences were likely to be derived from gene amplification products,the nucleic acids contaminated in fetal bovine serum.Reads from environmental microorganisms were also identified.Our results suggested that NGS method was capable of monitoring the nucleic acids contaminations from different sources in the lab,demonstrating its promising utility in monitoring and assessing the risk of potential laboratory contamination.The risk of contamination from reagents,remnant DNA and environment should be considered in data analysis and results interpretation. | Yan Xiao Li Zhang Bin Yang Mingkun Li Lili Ren Jianwei Wang | 2019 | Biosafety and Health2019,1,1: | 1 |
| 6 | Orlistat induces ferroptosis-like cell death of lung cancer cells显示文摘Aberrant de novo lipid synthesis is involved in the progression and treatment resistance of many types of cancers,including lung cancer;however,targeting the lipogenetic pathways for cancer therapy remains an unmet clinical need.In this study,we tested the anticancer activity of orlistat,an FDA-approved anti-obesity drug,in human and mouse cancer cells in vitro and in vivo,and we found that orlistat,as a single agent,inhibited the proliferation and viabilities of lung cancer cells and induced ferroptosis-like cell death in vitro.Mechanistically,we found that orlistat reduced the expression of GPX4,a central ferroptosis regulator,and induced lipid peroxidation.In addition,we systemically analyzed the genome-wide gene expression changes affected by orlistat treatment using RNA-seq and identified FAF2,a molecule regulating the lipid droplet homeostasis,as a novel target of orlistat.Moreover,in a mouse xenograft model,orlistat significantly inhibited tumor growth and reduced the tumor volumes compared with vehicle control(P<0.05).Our study showed a novel mechanism of the anticancer activity of orlistat and provided the rationale for repurposing this drug for the treatment of lung cancer and other types of cancer. | Wenjing Zhou Jing Zhang Mingkun Yan Jin Wu Shuo Lian Kang Sun Baiqing Li Jia Ma Jun Xia Chaoqun Lian | 2021 | Frontiers of Medicine2021,15,6: | 1 |
| 7 | Magnetic field induced synthesis of (Ni, Zn)Fe_(2)O_(4) spinel nanorod for enhanced alkaline hydrogen evolution reaction显示文摘Recently, the introduction of external fields (light, thermal, magnetism, etc.) during electrocatalysis reactions gradually becomes a new strategy to modulate the catalytic activities. In this work, an external magnetic field was innovatively employed for the synthesis progress of (Ni, Zn)Fe_(2)O_(4) spinel oxide (M-(Ni, Zn)Fe_(2)O_(4)). Results indicated the magnetic field (≤250 mT) would affect the morphology of catalyst due to the existing Fe ions, inducing the M-(Ni, Zn)Fe_(2)O_(4) nanosphere particles to be uniform and coral-like nanorods. In addition, the electronic structure of the catalyst was regulated by the valence state of Fe, changing the bonding of metal to oxygen atoms in different spinel sites. The results manifested that the M-(Ni, Zn)Fe_(2)O_(4) requires a lower overpotential of only 67 mV to deliver 10 mA cm−2 for hydrogen evolution reaction (HER) in alkaline electrolyte. Moreover, M-(Ni, Zn)Fe_(2)O_(4) respectively as the anode and cathode electrode for the overall water splitting, the catalysis system requires a cell voltage of only 1.76 V to gain a current density of 50 mA cm−2, combining with an excellent discharging stability after 10 h.
