| 2 | Zinc oxide nano-enabled microfluidic reactor for water purification and its applicability to volatile organic compounds显示文摘This paper reports fast and efficient chemical decontamination of water within a tree-branched centimeter-scale microfluidic reactor.The microreactor integrates Zinc oxide nanowires(ZnO NWs)in situ grown acting as an efficient photocatalytic nanomaterial layer.Direct growth of ZnO NWs within the microfluidic chamber brings this photocatalytic medium at the very close vicinity of the water flow path,hence minimizing the required interaction time to produce efficient purification performance.We demonstrate a degradation efficiency of 95%in o5 s of residence time in one-pass only.According to our estimates,it becomes attainable using microfluidic reactors to produce decontamination of merely 1 l of water per day,typical of the human daily drinking water needs.To conduct our experiments,we have chosen a laboratory-scale case study as a seed for addressing the health concern of water contamination by volatile organic compounds(VOCs),which remain difficult to remove using alternative decontamination techniques,especially those involving water evaporation.The contaminated water sample contains mixture of five pollutants:Benzene;Toluene;Ethylbenzene;m–p Xylenes;and o-Xylene(BTEX)diluted in water at 10 p.p.m.concentration of each.Degradation was analytically monitored in a selective manner until it falls below 1 p.p.m.for each of the five pollutants,corresponding to the maximum contaminant level(MCL)established by the US Environmental Protection Agency(EPA).We also report on a preliminary study,investigating the nature of the chemical by-products after the photocatalytic VOCs degradation process. | Imadeddine Azzouz Yamina Ghozlane Habba Martine Capochichi-Gnambodoe Frédéric Marty Jérôme Vial Yamin Leprince-Wang Tarik Bourouina | 2018 | Microsystems & Nanoengineering2018,4,1: | 4 |

| 4 | Targeting γ-secretase triggers the selective enrichment of oligomeric APP-CTFs in brain extracellular vesicles from Alzheimer cell and mouse models显示文摘Background:We recently demonstrated an endolysosomal accumulation of theβ-secretase-derived APP C-terminal fragment(CTF)C99 in brains of Alzheimer disease(AD)mouse models.Moreover,we showed that the treatment with theγ-secretase inhibitor(D6)led to further increased endolysosomal APP-CTF levels,but also revealed extracellular APP-CTF-associated immunostaining.We here hypothesized that this latter staining could reflect extracellular vesicle(EV)-associated APP-CTFs and aimed to characterize theseγ-secretase inhibitor-induced APPCTFs.Methods:EVs were purified from cell media or mouse brains from vehicle-or D6-treated C99 or APPswedish expressing cells/mice and analyzed for APP-CTFs by immunoblot.Combined pharmacological,immunological and genetic approaches(presenilin invalidation and C99 dimerization mutants(GXXXG))were used to characterize vesicle-containing APP-CTFs.Subcellular APP-CTF localization was determined by immunocytochemistry.Results:Purified EVs from both AD cell or mouse models were enriched in APP-CTFs as compared to EVs from control cells/brains.Surprisingly,EVs from D6-treated cells not only displayed increased C99 and C99-derived C83 levels but also higher molecular weight(HMW)APP-CTF-immunoreactivities that were hardly detectable in whole cell extracts.Accordingly,the intracellular levels of HMW APP-CTFs were amplified by the exosomal inhibitor GW4869.By combined pharmacological,immunological and genetic approaches,we established that these HMW APP-CTFs correspond to oligomeric APP-CTFs composed of C99 and/or C83.Immunocytochemical analysis showed that monomers were localized mainly to the trans-Golgi network,whereas oligomers were confined to endosomes and lysosomes,thus providing an anatomical support for the selective recovery of HMW APP-CTFs in EVs.The D6-induced APP-CTF oligomerization and subcellular mislocalization was indeed due toγ-secretase blockade,since it similarly occurred in presenilin-deficient fibroblasts.Further,our data proposed that besides favoring APP-CTF oligomerization by preventing C99 proteolysis,γ-secretase inhibiton also led to a defective SorLA-mediated retrograde transport of HMW APP-CTFs from endosomal compartments to the TGN.Conclusions:This is the first study to demonstrate the presence of oligomeric APP-CTFs in AD mouse models,the levels of which are selectively enriched in endolysosomal compartments including exosomes and amplified byγ-secretase inhibition.Future studies should evaluate the putative contribution of these exosome-associated APP-CTFs in AD onset,progression and spreading. | Inger Lauritzen Anaïs Bécot Alexandre Bourgeois Raphaëlle Pardossi-Piquard Maria-Grazia Biferi Martine Barkats Fréderic Checler | 2019 | Translational Neurodegeneration2019,8,1: | 1 |

| 8 | Genome-wide differences in hepatitis C-vs alcoholism-associated hepatocellular carcinoma显示文摘AIM:To look at a comprehensive picture of etiology- dependent gene abnormalities in hepatocellular carcinoma in Western Europe. METHODS:With a liver-oriented microarray,transcript levels were compared in nodules and cirrhosis from a training set of patients with hepatocellular carcinoma (alcoholism,12;hepatitis C,10)and 5 controls.Loose or tight selection of informative transcripts with an abnormal abundance was statistically valid and the tightly selected transcripts were next quantified by qRTPCR in the nodules from our training set(12+10) and a test set(6+7). RESULTS:A selection of 475 transcripts pointed to significant gene over-representation on chromosome 8 (alcoholism)or-2(hepatitis C)and ontology indicated a predominant inflammatory response(alcoholism)or changes in cell cycle regulation,transcription factors and interferon responsiveness(hepatitis C).A stringent selection of 23 transcripts whose differences betweenetiologies were significant in nodules but not in cirrhotic tissue indicated that the above dysregulations take place in tumor but not in the surrounding cirrhosis.These 23 transcripts separated our test set according to etiologies. The inflammation-associated transcripts pointed to limited alterations of free iron metabolism in alcoholic vs hepatitis C tumors.
