2Blood Lipid Profile and Prevalence of Dyslipidemia in Chinese Adults显示文摘Objective To investigate the plasma lipid levels in a national representative sample of subjects and to determine the prevalence of dyslipidemia in the Chinese population.Methods Plasma lipid profile was analyzed using the data obtained during the Chinese national nutrition and health survey(CNHS)in 2002 which involved 14 252 participants at the age of 18 years or older.Results The mean levels of total cholesterol(TC),triglyceride(TG)and high density lipoprotein cholesterol(HDL-C)in the participants were 3.81 mmol/L,1.10 mmol/L,and 1.30 mmol/L,respectively.In the groups of participants at the age of 18-44 years,45-59 years,and over 60 years the mean TC level was 3.70 mmol/L,4.09 mmol/L and 4.21 mmol/L,respectively,and the mean TG level was 07 mmol/L,1.21 mmol/L,1.20 mmol/L,1.29 mmol/L,1.33 mmol/L,and 1.33 mmol/L,respectively.The prevalence of dyslipidemia in Chinese adults was 18.6% and 22.2% in males and 15.9% in females.Dyslipidemia prevalence was higher in urban districts than in rural areas(21.0% vs.17.7%).The prevalence of hypercholesterolemia,hypertriglyceridemia,and low HDL cholesterol was 2.9%,11.9%,and 7.4% respectively among the participants.Conclusion Dyslipidemia has become one of the important health risk factors in the Chinese population.There is no significantly difference in the prevalence of dyslipidemia between the groups of participants at the age of 45-59 years and over 60 years.This study provides important lipid profile data for policy making and guideline development for the prevention of dyslipidemia in the Chinese population.WEN-HUA ZHAO JIAN ZHANG YI ZHAI YUE YOU QING-QING MAN CHUN-RONG WANG HONG LI YING LI XIAO-GUANG YANG 2007Biomedical and Environmental Sciences2007,20,4:22
3Phase 1 clinical trial demonstrated that MUC1 positive metastatic seminal vesicle cancer can be effectively eradicated by modified Anti-MUC1 chimeric antigen receptor transduced T cells显示文摘Recent progress in chimeric antigen receptor-modified T-cell(CAR-T cell) technology in cancer therapy is extremely promising, especially in the treatment of patients with B-cell acute lymphoblastic leukemia. In contrast, due to the hostile immunosuppressive microenvironment of a solid tumor, CAR T-cell accessibility and survival continue to pose a considerable challenge, which leads to their limited therapeutic efficacy. In this study, we constructed two anti-MUC1 CAR-T cell lines. One set of CAR-T cells contained SM3 single chain variable fragment(sc Fv) sequence specifically targeting the MUC1 antigen and co-expressing interleukin(IL) 12(named SM3-CAR). The other CAR-T cell line carried the SM3 sc Fv sequence modified to improve its binding to MUC1 antigen(named p SM3-CAR) but did not co-express IL-12. When those two types of CAR-T cells were injected intratumorally into two independent metastatic lesions of the same MUC1+ seminal vesicle cancer patient as part of an interventional treatment strategy, the initial results indicated no side-effects of the MUC1 targeting CAR-T cell approach, and patient serum cytokines responses were positive. 4Comparable effects of Jiedu Granule, a compound Chinese herbal medicine, and sorafenib for advanced hepatocellular carcinoma: A prospective multicenter cohort study显示文摘Objective:Sorafenib has been extensively used for the treatment of advanced hepatocellular carcinoma(HCC),and Chinese herbal medicine has also been used to manage advanced HCC.The present work evaluates the effectiveness and safety of Jiedu(JD)Granule,a compound of traditional Chinese herbal medicine,side-by-side with sorafenib for the treatment of advance HCC.Methods:Patients with advanced HCC receiving treatment with JD Granule or sorafenib were enrolled from December 2014 to March 2018.The primary endpoint was overall survival(OS).The secondary endpoints were progression-free survival(PFS)and safety.Propensity score matching(PSM)analysis was used to control for possible selection bias from the study group allocation process.Results:Of the 325 patients included,161 received JD Granule and 164 received sorafenib.No significant differences were found in OS or PFS among patients receiving JD Granule compared to sorafenib(P>0.05).Median OS of the two study groups was 6.83 months(95%confidence interval[CI]:5.83–9.47)in the group receiving JD Granule and 8 months(95%CI:6.67–9.80)in the group receiving sorafenib,with half-,1-and 2-year survival rates of 53.6%,31.2%and 13.2%vs 60.1%,35.5%and 14.2%,respectively.Even after PSM,the median survival time did not differ between the JD Granule group(9.03 months;95%CI:6.37–14.2)and the sorafenib group(7.93 months;95%CI:6.5–9.97),with comparable half-,1-and 2-year survival rates.The most common adverse events(AEs)were diarrhea(13.7%)and fatigue(5.6%)in the JD Granule group,and hand-foot skin reaction(46.3%)and diarrhea(36.6%)in the sorafenib group.The JD Granule was more cost-effective than sorafenib treatment for advanced HCC.Conclusion:Compared to sorafenib,JD Granule was more cost-effective and caused fewer AEs for the treatment of Chinese patients with advanced HCC.He-tong Zhao Yong-bin Meng Xiao-feng Zhai Bin-bin Cheng Sha-sha Yu Man Yao Hui-xia Yin Xu-ying Wan Yun-ke Yang Hui Liu Feng Shen Chang-quan Ling 2020Journal of Integrative Medicine2020,18,4:5
5Thiabendazole inhibits ubiquinone reduction activity of mitochondrial respiratory complex Ⅱ via a water molecule mediated binding feature显示文摘The mitochondrial respiratory complex Ⅱ or succinate:ubiquinone oxidoreductase(SQR)is a key membrane complex in both the tricarboxylic acid cycle and aerobic respiration.Five disinfectant compounds were investigated with their potent inhibition effects on the ubiquinone reduction activity of the porcine mitochondrial SQR by enzymatic assay and crystallography.Crystal structure of the SQR bound with thiabendazole(TBZ)reveals a different inhibitor-binding feature at the ubiquinone binding site where a water molecule plays an important role.The obvious inhibitory effect of TBZ based on the biochemical data(IC50~100μmol/L)and the significant structure-based binding affinity calculation(~94μmol/L)draw the suspicion of using TBZ as a good disinfectant compound for nematode infections treatment and fruit storage.Qiangjun Zhou Yujia Zhai Jizhong Lou Man Liu Xiaoyun Pang Fei Sun 2011Protein & Cell2011,2,7:2

8Highly efficient generation of blastocyst-like structures from spliceosomes-repressed mouse totipotent blastomere-like cells显示文摘Mammalian embryogenesis begins with a totipotent zygote.Blastocyst-like structures can be captured by aggregated cells with extended pluripotent properties in a three-dimensional(3D)culture system.However,the efficiency of generating blastoids is low,and it remains unclear whether other reported totipotent-like stem cells retain a similar capacity.In this study,we demonstrated that spliceosomal repression-induced totipotent blastomere-like cells(TBLCs)form blastocyst-like structures within around 80%of all microwells.In addition,we generated blastoids initiating from a single TBLC.TBLC-blastoids express specific markers of constituent cell lineages of a blastocyst and resemble blastocyst in cell-lineage allocation.Moreover,singlecell RNA sequencing revealed that TBLC-blastoids share a similar transcriptional profile to natural embryos,albeit composed of fewer primitive endoderm-like cells.Furthermore,TBLC-blastoids can develop beyond the implantation stage in vitro and induce decidualization in vivo.In summary,our findings provided an alternative cell type to efficiently generate blastoids for the study of early mouse embryogenesis.Pengfei Zhang Xuzhao Zhai Boyan Huang Shu Sun Wen Jing Wang Man Zhang 2023Science China(Life Sciences)2023,66,3:1

14High-throughput“read-on-ski”automated imaging and label-free detection system for toxicity screening of compounds using personalised human kidney organoids显示文摘Organoid models are used to study kidney physiology,such as the assessment of nephrotoxicity and underlying disease processes.Personalized human pluripotent stem cell-derived kidney organoids are ideal models for compound toxicity studies,but there is a need to accelerate basic and translational research in the field.Here,we developed an automated continuous imaging setup with the“read-on-ski”law of control to maximize temporal resolution with minimum culture plate vibration.High-accuracy performance was achieved:organoid screening and imaging were performed at a spatial resolution of 1.1µm for the entire multi-well plate under 3 min.We used the in-house developed multi-well spinning device and cisplatin-induced nephrotoxicity model to evaluate the toxicity in kidney organoids using this system.The acquired images were processed via machine learning-based classification and segmentation algorithms,and the toxicity in kidney organoids was determined with 95%accuracy.The results obtained by the automated“read-on-ski”imaging device,combined with label-free and non-invasive algorithms for detection,were verified using conventional biological procedures.Taking advantage of the close-to-in vivo-kidney organoid model,this new development opens the door for further application of scaled-up screening using organoids in basic research and drug discovery.Qizheng WANG Jun LU Ke FAN Yiwei XU Yucui XIONG Zhiyong SUN Man ZHAI Zhizhong ZHANG Sheng ZHANG Yan SONG Jianzhong LUO Mingliang YOU Meijin GUO Xiao ZHANG 2022Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2022,23,7:0

16Association between Baseline SBP/DBP and All-Cause Mortality in Residents of Shanxi,China:A Population-based Cohort Study from 2002 to 2015显示文摘Objective To investigate the association between blood pressure and all-cause mortality in Shanxi,China.Methods The‘2002 China Nutrition and Health Survey’baseline data in Shanxi province was used.A retrospective investigation was performed in 2015.The effects of SBP and DBP on the all-cause mortality were analyzed using the Cox regression model.The hazard ratio(HR)and 95%confidence interval(CI)were estimated by the sex and age groups.Results The follow-up rate was 76.52%over 13 years,while the cumulative mortality rate for all participants was 917.12/100,000 person-years.The mortality rose with an increasing SBP(χ_(trend)^(2)=270.537,P<0.001)or DBP level(χ_(trend)^(2)=57.240,P<0.001).After adjustment for the confounding factors,a significant association between mortality and high SBP(≥160 mm Hg)and high DBP(≥100 mm Hg),with adjusted HR ranging from 1.405-to 2.179-fold for SBP and 1.550-to 2.854-fold for DBP,was noted.Significant HRs for most DBP subgroups were found in>60-year-old participants.Males with DBP≥100 mm Hg had a significantly higher mortality,with an HR(95%CI)of 2.715(1.377–5.351).Conclusion Adults with SBP>160 mm Hg and DBP>100 mm Hg had a higher mortality risk.Sex and age difference was noted in both DBP and mortality risk.WANG Zhuo Qun ZHAI Yi LI Man YANG Xiu Feng ZHANG Jian REN Ze Ping ZHANG Mei SONG Peng Kun ZHAO Yan Fang MI Sheng Quan ZHANG Lu YANG Mao Xiang ZHAO Wen Hua 2021Biomedical and Environmental Sciences2021,34,1:0
17Image analysis and machine learning-based malaria assessment system显示文摘Malaria is an important and worldwide fatal disease that has been widely reported by the World Health Organization(WHO),and it has about 219 million cases worldwide,with 435,000 of those mortal.The common malaria diagnosis approach is heavily reliant on highly trained experts,who use a microscope to examine the samples.Therefore,there is a need to create an automated solution for the diagnosis of malaria.One of the main objectives of this work is to create a design tool that could be used to diagnose malaria from the image of a blood sample.In this paper,we firstly developed a graphical user interface that could be used to help segment red blood cells and infected cells and allow the users to analyze the blood samples.Secondly,a Feed-forward Neural Network(FNN)is designed to classify the cells into two classes.The achieved results show that the proposed techniques can be used to detect malaria,as it has achieved 92%accuracy with a database that contains 27,560 benchmark images.Kyle Manning Xiaojun Zhai Wangyang Yu 2022Digital Communications and Networks2022,8,2:0
