1GS6, A Member of the GRAS Gene Family,Negatively Regulates Grain Size in Rice显示文摘Grain size is an important yield related trait in rice.Intensive arti cial selection for grain size during domestication is evidenced by the larger grains of most of today’s cultivars compared with their wild relatives.However,the molecular genetic control of rice grain size is still not well characterized.Here,we report the identi cation and cloning of Grain Size 6(GS6),which plays an important role in reducing grain size in rice.A premature stop at the t348 position in the coding sequence(CDS)of GS6 increased grain width and weight signi cantly.Alignment of the CDS regions of GS6 in 90 rice materials revealed three GS6alleles.Most japonica varieties(95%)harbor the Type I haplotype,and 62.9%of indica varieties harbor the Type II haplotype.Association analysis revealed that the Type I haplotype tends to increase the width and weight of grains more than either of the Type II or Type III haplotypes.Further investigation of genetic diversity and the evolutionary mechanisms of GS6showed that the GS6gene was strongly selected in japonica cultivars.In addition,a'ggc'repeat region identi ed in the region that encodes the GRAS domain of GS6 played an important historic role in the domestication of grain size in rice.Knowledge of the function of GS6might aid efforts to elucidate the molecular mechanisms that control grain development and evolution in rice plants,and could facilitate the genetic improvement of rice yield.Lianjun Sun Xiaojiao Li Yongcai Fu Zuofeng Zhu Lubin Tan Fengxia Liu Xianyou Sun Xuewen Sun Chuanqing Sun 2013Journal of Integrative Plant Biology2013,55,10:44
2An Effective Method for Total RNA Isolation from Bamboo显示文摘Trizol reagent was used for RNA isolation from fresh leaves of Dendrocalamopsis oldhami, Bambusa ventricosa and Phyllostachys aureosulcata cv. Pekinensis. The extracted RNA from leaves had the normal ultraviolet absorption, the value of OD260/OD280 varied between 1.8-2.0. The 28s rRNA was more than two times brighter than 18s rRNA in electrophoresis. These results indicated that the total RNA was complete and not degraded. 3Identification and mapping of quantitative trait loci controlling cold-tolerance of Chinese common wild rice (O. rufipogon Griff.) at booting to flowering stages显示文摘An advanced backcross population of rice was used to identify the quantitative trait locus (QTL) controlling the cold-tolerance at booting to flowering stages. The recipient, Guichao 2 (GC2), was a commercial Indica rice; the donor Dongxiang common wild rice, was an accession of common wild rice (DXCWR, Oryza rufipogon Griff.). Three QTLs for cold-tolerance were detected on chromosomes 1, 6 and 11. Two of them coming from DXCWR could enhance the cold-tolerance of the backcross progenies. 4Zircon U-Pb geochronological framework of Qitianling granite batholith, middle part of Nanling Range, South China显示文摘The Qitianling granite batholith (QGB) is located in the southern Hunan Province, middle part of the Nanling Range, South China. Its total exposure area is about 520 km2. Based on our 25 single grain zircon U-Pb age data and 7 published data as well as the geological, petrological, and space distribution characteristics, we conclude that QGB is an Early Yanshanian (Jurassic) multi-staged composite pluton. Its formation process can be subdivided into three major stages. The first stage, emplaced at 163―160 Ma with a peak at about 161 Ma, is mainly composed of hornblende-biotite monzonitic granites and locally biotite granites, and distributed in the eastern, northern, and western peripheral parts of the pluton. The second stage, emplaced at 157―153 Ma with a peak at 157―156 Ma, is mainly composed of biotite granites and locally containing hornblende, and distributed in the middle and southeastern parts of the pluton. The third stage, emplaced at 150―146 Ma with a peak at about 149 Ma, is mainly composed of fine-grained (locally porphyritic) biotite granites, and distributed in the middle-southern part of the pluton. Each stage can be further disintegrated into several granite bodies. The first two intrusive stages comprise the major phase of QGB, and the third intrusive stage comprises the additional phase. Many second stage fine-grained granite bosses and dykes intruded into the first stage host granites with clear chilling margin-baking phenomena at their intrusive contacts. They were emplaced in the open fracture space of the earlier stage consolidated rocks. Their isotopic ages are mostly 2―6 Ma younger than their hosts. Conceivably, the time interval from magma emplacement, through cooling, crystallization, solidification, up to fracturing of the earlier stage granites cannot exceed 2―6 Ma. During the Middle-Late Jurassic in the Qitianling area and neighboring Nanling Range, the coeval granitic and basic-intermediate magmatic activities were widely developed. It indicates that the Early Yanshanian period was the culmination time of magmatic activities in this region. The Nanling Range was under a post-orogenic, intracontinental geotectonic environment with an obvious lithospheric extension and thinning. 8LHD1,an Allele of DTH8/Ghd8,Controls Late Heading Date in Common Wild Rice (Oryza rufipogon)显示文摘Flowering at suitable time is very important for plants to adapt to complicated environments and produce their seeds successfully for reproduction.In rice (Oryza rufipogon Griff.) photoperiod regulation is one of the important factors for controlling heading date.Common wild rice,the ancestor of cultivated rice,exhibits a late heading date and a more sensitive photoperiodic response than cultivated rice.Here,through map-based cloning,we identified a major quantitative trait loci (QTL) LHD1 (Late Heading Date 1),an allele of DTH8/Ghd8,which controls the late heading date of wild rice and encodes a putative HAP3/NF-YB/CBF-A subunit of the CCAAT-box-binding transcription factor.Sequence analysis revealed that several variants in the coding region of LHD1 were correlated with a late heading date,and a further complementary study successfully rescued the phenotype.These results suggest that a functional site for LHD1 could be among those variants present in the coding region.We also found that LHD1 could down-regulate the expression of several floral transition activators such as Ehd1,Hd3a and RFT1 under long-day conditions,but not under short-day conditions.This indicates that LHD1 may delay flowering by repressing the expression of Ehd1,Hd3a and RFT1 under long-day conditions.Xiaodong Dai Younian Ding Lubin Tan Yongcai Fu Fengxia Liu Zuofeng Zhu Xianyou Sun Xuewen Sun Ping Gu Hongwei Cai Chuanqing Sun 2012Journal of Integrative Plant Biology2012,54,10:23
9NARROW AND ROLLED LEAF 2 regulates leaf shape,male fertility, and seed size in rice显示文摘Grain yield in rice(Oryza sativa L.) is closely related to leaf and flower development. Coordinative regulation of leaf, pollen, and seed development in rice as a critical biological and agricultural question should be addressed. Here we identified two allelic rice mutants with narrow and semirolled leaves, named narrow and rolled leaf 2-1(nrl2-1) and nrl2-2. Map-based molecular cloning revealed that NRL2 encodes a novel protein with unknown biochemical function. The mutation of NRL2 caused pleiotropic effects, including a reduction in the number of longitudinal veins, defective abaxial sclerenchymatous cell differentiation, abnormal tapetum degeneration and microspore development, and the formation of more slender seeds compared with the wild type(WT). The NRL2 protein interacted with Rolling-leaf(RL14),causing the leaves of the nrl2 mutants to have a highercellulose content and lower lignin content than the WT, which may have been related to sclerenchymatous cell differentiation and tapetum degeneration. 13Particle size distribution in CPFD modeling of gas-solid flows in a CFB riser显示文摘A computational particle fluid dynamics(CPFD) numerical method to model gas-solid flows in a circulating fluidized bed(CFB) riser was used to assess the effects of particle size distribution(PSD) on solids distribution and flow.We investigated a binary PSD and a polydisperse PSD case.Our simulations were compared with measured solids concentrations and velocity profiles from experiments,as well as with a published Eulerian-Eulerian simulation.Overall flow patterns were similar for both simulation cases,as confirmed by experimental measurements.However,our fine-mesh CPFD simulations failed to predict a dense bottom region in the riser,as seen in other numerical studies.Above this bottom region,distributions of particle volume fraction and particle vertical velocity were consistent with our experiments,and the simulated average particle diameter decreased as a power function with riser height.Interactions between particles and walls also were successfully modeled,with accurate predictions for the lateral profiles of particle vertical velocity.It was easy to implement PSD into the CPFD numerical model,and it required fewer computational resources compared with other models,especially when particles with a polydisperse PSD were present in the heterogeneous flow.Qinggong Wang Timo Niemi Juho Peltola Sirpa Kallio Hairui Yang Junfu Lu Lubin Weic 2015Particuology2015,13,4:8
14Characterization of a novel high-tillering dwarf 3 mutant in rice显示文摘Tiller number and culm length are important components of plant architecture and determinate grain production in rice.A line SIL046, derived from an introgression lines population developed by an accession of common wild rice(Oryza rufipogon Griff.) and a high-yielding indica cultivar Guichao 2(Oryza sativa L.),exhibits a higher tiller number and shorter culm length phenotype than the recipient parent Guichao 2(GC2).Genetic analysis showed that the high-tillering dwarf phenotype was controlled by a novel single recessive gene,referred to as the high-tillering dwarf 3(htd3),which located within the genetic distance of 13.4 cM between SSR makers RM7003 and RM277 on chromosome 12.By means of fine-mapping strategy,we mapped HTD3 gene within the genetic distance of 2.5 cM and the physical distance of 3100 kb in the centromere of chromosome 12.Further identification of HTD3 gene would provide a new opportunity to uncover the molecular mechanism of the development of culm and tiller,two important components of yields in rice.Bosen Zhang Feng Tian Lubin Tan Daoxin Xie Chuanqing Sun 2011Journal of Genetics and Genomics2011,38,9:7

16RLS3, a protein with AAA+ domain localized in chloroplast, sustains leaf longevity in rice显示文摘Leaf senescence plays an important role in crop developmental processes that dramatically affect crop yield and grain quality. The genetic regulation of leaf senescence is complex, involving many metabolic and signaling pathways.Here, we identified a rapid leaf senescence 3(rls3) mutant that displayed accelerated leaf senescence, shorter plant height and panicle length, and lower seed set rate than the wild type. Map-based cloning revealed that RLS3 encodes a protein with AAA+ domain, localizing it to chloroplasts. Sequence analysis found that the rls3 gene had a single-nucleotide substitution(G→A) at the splice site of the 10^(th)intron/11^(th) exon, resulting in the cleavage of the first nucleotide in 11 ^(th) exon and premature termination of RLS3 protein translation.Using transmission electron microscope, the chloroplasts of the rls3 mutant were observed to degrade much faster than those of the wild type. The investigation of the leaf senescence process under dark incubation conditions furtherrevealed that the rls3 mutant displayed rapid leaf senescence.Thus, the RLS3 gene plays key roles in sustaining the normal growth of rice, while loss of function in RLS3 leads to rapid leaf senescence. 17Analysis of QTLs for yield-related traits in Yuanjiang common wild rice (Oryza rufipogon Griff.)显示文摘Using an accession of common wild rice (Oryza rufipogon Griff.) collected from Yuanjiang County, Yunnan Province, China, as the donor and an elite cultivar 93-11, widely used in two-line indica hybrid rice production in China, as the recurrent parent, an advanced backcross populations were developed. Through genotyping of 187 SSR markers and investigation of six yield-related traits of two gen- erations (BC4F2 and BC4F4), a total of 26 QTLs were detected by employing single point analysis and interval mapping in both genera-tions. Of the 26 QTLs, the alleles of 10 (38.5%) QTLs originating from O. rufipogon had shown a beneficial effect for yield-related traits in the 93-11 genetic background. In addition, five QTLs controlling yield and its components were newly identified, indicating that there are potentially novel alleles in Yuanjiang common wild rice. Three regions underling significant QTLs for several yield-related traits were detected on chromosome 1, 7 and 12. The QTL clusters were founded and corresponding agronomic traits of those QTLs showed highly significant correlation, suggesting the pleiotropism or tight linkage. 18Ciprofloxacin Loaded Bovine Serum Albumin Microspheres: Prepa-ration and Drug Release Characterization In Vitro显示文摘Ciprofloxacin loaded microspheres were prepared by spray drying technique, with bovine serum albumin as the natural biodegradable wall materials. The obtained microspheres, using aqueous system, were organic solvent-free. The diameters of the spherical microspheres were in the range of 1-5 1:4. The drug entrapment of microspheres, formulated with different ciprofloxacin/albumin ratios as 1:1, 1:2 and 1:4, were 46.93%, 32.96% and 20.56% (n=3). And the encapsulation efflciencies for ciprofloxacin during spray drying were higher than 90%. Thermal denaturation programs at different temperatures (100-120℃) for different time intervals (3-6-12 h) were further processed to stabilize the spray-dried microspheres. The higher the extent for thermal denaturation, the slower the rate of ciprofloxacin released from microspheres in vitro. 19Preparation,characterization and performance of an electrospun carbon nanofiber mat applied in hexavalent chromium removal from aqueous solution显示文摘Hexavalent chromium, Cr(VI), a highly toxic oxyanion known as a carcinogen and mutagen,is an issue of concern due to its adverse impact on human health. Therefore, development of effective technologies and/or materials for Cr(VI) removal from water has been of great interest for researchers. In this study, an electrospun carbon nanofiber(CNF) mat was prepared via electrospinning polyacrylonitrile(PAN), followed by thermal pre-oxidation and carbonization. Scanning electron microscopy(SEM) observation showed that the fiber diameter of the CNF with carbonization temperature of 950°C(CNF_(950)) was about 266 nm.Potentiometric titration analysis demonstrated that the point of zero charge p H(pHpzc) of CNF_(950) was around 7.93. CNF_(950) demonstrated high adsorption capacity and fast adsorption kinetics for Cr(VI) at pH < 3. Langmuir isotherm calculations showed that the maximum adsorption capacity of Cr(VI) on CNF_(950) was 118.8 mg/g at pH 2. The adsorption isotherm of Cr(VI) on CNF_(950) was well described by the Redlich–Peterson model, revealing that Cr(VI)adsorption was the result of a combination of monolayer and multilayer adsorption,depending on the initial Cr(VI) concentration. Solution pH greatly affected Cr(VI) adsorption onto CNF_(950) due to the electrostatic interaction, and the adsorption capacity was relatively high when pH was below 3. X-ray photoelectron spectroscopy(XPS) analysis revealed that the removal of Cr(VI) might be the result of a combination of redox reaction and electrostatic adsorption. The adsorption-saturated CNF_(950) could be regenerated by NaOH solution. 20Removal of tetracycline from aqueous solution by a Fe_3O_4 incorporated PAN electrospun nanofiber mat显示文摘Pollution of antibiotics, a type of emerging contaminant, has become an issue of concern,due to their overuse in human and veterinary application, persistence in environment and great potential risk to human and animal health even at trace level. In this work, a novel adsorbent, Fe3O4 incorporated polyacrylonitrile nanofiber mat(Fe-NFM), was successfully fabricated via electrospinning and solvothermal method, targeting to remove tetracycline(TC), a typical class of antibiotics, from aqueous solution. Field emission scanning electron microscopy and X-ray diffraction spectroscopy were used to characterize the surface morphology and crystal structure of the Fe-NFM, and demonstrated that Fe-NFM was composed of continuous, randomly distributed uniform nanofibers with surface coating of Fe3O4 nanoparticles. A series of adsorption experiments were carried out to evaluate the removal efficiency of TC by the Fe-NFM. The pseudo-second-order kinetics model fitted better with the experimental data. The highest adsorption capacity was observed at initial solution p H 4 while relative high adsorption performance was obtained from initial solution p H 4 to 10. The adsorption of TC on Fe-NFM was a combination effect of both electrostatic interaction and complexation between TC and Fe-NFM. Freundlich isotherm model could better describe the adsorption isotherm. The maximum adsorption capacity calculated from Langmuir isotherm model was 315.31 mg/g.
