| 1 | Advances in the development of molecular genetic tools for Mycobacterium tuberculosis显示文摘Mycobacterium tuberculosis, a Gram-positive bacterium of great clinical relevance, is a lethal pathogen owing to its complex physiological characteristics and development of drug resistance. Several molecular genetic tools have been developed in the past few decades to study this microorganism. These tools have been instrumental in understanding how M. tuberculosis became a successful pathogen. Advanced molecular genetic tools have played a significant role in exploring the complex pathways involved in M. tuberculosis pathogenesis. Here, we review various molecular genetic tools used in the study of M. tuberculosis. Further, we discuss the applications of clustered regularly interspaced short palindromic repeat interference(CRISPRi), a novel technology recently applied in M. tuberculosis research to study target gene functions. | 2 | Effect of CO_2 on the structural variation of Na_2WO_4/Mn/SiO_2 catalyst for oxidative coupling of methane to ethylene显示文摘In this work,the influence of CO2 on the structural variation and catalytic performance of Na2WO4/Mn/Si O2 for oxidative coupling of methane to ethylene was investigated. The catalyst was prepared by impregnation method and characterized by XRD,Raman and XPS techniques. Appropriate amount of CO2 in the reactant gases enhanced the formation of surface tetrahedral Na2WO4 species and promoted the migration of O in MOx,Na,W from the catalyst bulk to surface,which were favorable for oxidative coupling of methane. When the molar ratio of CH4/O2/CO2 was 3/1/2,enriched surface tetrahedral Na2WO4 species and high surface concentration of O in MOx,Na,W were detected,and then high CH4 conversion of 33.1% and high C2H4 selectivity of 56.2% were obtained. With further increase of CO2 in the reagent gases,the content of active surface tetrahedral Na2WO4 species and surface concentration of O in MOx,Na,W decreased,while that of inactive species(Mn WO4 and Mn2O3) increased dramatically,leading to low CH4 conversion and low C2H4 selectivity. It could be speculated that Na2WO4 crystal was transformed into Mn WO4 crystal with excessive CO2 added under the reaction conditions. | 4 | Genome-Wide Linkage and Positional Association Analyses Identify Associations of Novel AFF3 and NTM Genes with Triglycerides:The GenSalt Study显示文摘We conducted a genome-wide linkage scan and positional association study to identify genes and variants influencing blood lipid levels among participants of the Genetic Epidemiology Network of Salt-Sensitivity(Gen Salt) study. The Gen Salt study was conducted among1906 participants from 633 Han Chinese families. Lipids were measured from overnight fasting blood samples using standard methods.Multipoint quantitative trait genome-wide linkage scans were performed on the high-density lipoprotein, low-density lipoprotein, and logtransformed triglyceride phenotypes. Using dense panels of single nucleotide polymorphisms(SNPs), single-marker and gene-based association analyses were conducted to follow-up on promising linkage signals. Additive associations between each SNP and lipid phenotypes were tested using mixed linear regression models. Gene-based analyses were performed by combining P-values from singlemarker analyses within each gene using the truncated product method(TPM). Significant associations were assessed for replication among777 Asian participants of the Multi-ethnic Study of Atherosclerosis(MESA). Bonferroni correction was used to adjust for multiple testing.In the Gen Salt study, suggestive linkage signals were identified at 2p11.2-2q12.1 [maximum multipoint LOD score(MML)=2.18 at2q11.2] and 11q24.3-11q25(MML=2.29 at 11q25) for the log-transformed triglyceride phenotype. Follow-up analyses of these two regions revealed gene-based associations of charged multivesicular body protein 3(CHMP3), ring finger protein 103(RNF103),AF4/FMR2 family, member 3(AFF3), and neurotrimin(NTM) with triglycerides(P=4 10 4, 1.00 10 5, 2.00 10 5, and1.00 10 7, respectively). Both the AFF3 and NTM triglyceride associations were replicated among MESA study participants(P=1.00 10 7and 8.00 10 5, respectively). Furthermore, NTM explained the linkage signal on chromosome 11. | 7 | Effect of methane co-feeding on the selectivity of ethylene produced from oxidative dehydrogenation of ethane with CO_2 over a Ni-La/SiO_2 catalyst显示文摘A Ni-La/SiO2 catalyst was prepared through the incipient wetness impregnation method and tested in the oxidative dehydrogenation of ethane (ODHE) with CO2 . The fresh and used catalysts were characterized by XRD and SEM techniques. The Ni-La/SiO2 catalyst exhibited catalytic activity for the oxidative dehydrogenation of ethane, but with low ethylene selectivity in the absence of methane. The selectivity to ethylene increased with increasing molar ratio of methane in the feed. The carbon deposited on the catalyst surface in the sole ODHE with CO2 was mainly inert carbon, while much more filamentous carbon was formed in the presence of methane. The filamentous carbon was easy to be removed by CO2 , which might play a role in improving the conversion of ethane to ethylene. The introduction of methane might affect the equilibrium of the CO2 reforming of ethane and the ODHE with CO2 .
