| 2 | Formability,microstructure evolution and mechanical properties of wire arc additively manufactured AZ80M magnesium alloy using gas tungsten arc welding显示文摘Wire arc additive manufacturing(WAAM)technology has been used to fabricate the multi-layer single-pass deposited wall of AZ80M magnesium(Mg)alloy by gas tungsten arc welding.The formability,thermal cycles,microstructural evolution and mechanical properties of the WAAM AZ80M Mg alloy were investigated.The results show that there was significant difference in the temperature variation and the geometries between the original several layers and the subsequent deposited layers.Owing to the arc energy input,the interpass temperature rised rapidly and then stabilized at 150℃.As a result,the width of the deposited wall increased and then kept stable.There were obvious differences in the microstructure of the WAAM AZ80M Mg alloy among the top zone,intermediate zone and bottom zone of deposited wall.During the arc deposition process,theβphase of the WAAM AZ80M Mg alloy redissolved due to the cyclic heat accumulation,and then precipitated in the grain boundary.The cyclic heat accumulation also led to weakening of dendrite segregation.From the substrate to the top zone,the hardness of the deposited wall decreased gradually,and the intermediate zone which was the main body of deposited wall had relatively uniform hardness.The tensile properties of the WAAM AZ80M Mg alloy were different between the vertical direction and the horizontal direction.And the maximum ultimate tensile strength of the WAAM AZ80M Mg alloy was 308 MPa which was close to that of the as-extruded AZ80M Mg alloy. | Yangyang Guo Gaofeng Quan Yinglong Jiang Lingbao Ren Lingling Fan Houhong Pan | 2021 | Journal of Magnesium and Alloys2021,9,1: | 10 |
| 3 | WIP1 Phosphatase Plays a Critical Neuroprotective Role in Brain Injury Induced by High-Altitude Hypoxic Inflammation显示文摘The hypobaric hypoxic environment in highaltitude areas often aggravates the severity of inflammation and induces brain injury as a consequence. However, the critical genes regulating this process remain largely unknown. The phosphatase wild-type p53-induced phosphatase 1(WIP1) plays important roles in various physiological and pathological processes, including the regulation of inflammation in normoxia, but its functions in hypoxic inflammation-induced brain injury remain unclear.Here, we established a mouse model of this type of injury and found that WIP1 deficiency augmented the release of inflammatory cytokines in the peripheral circulation and brain tissue, increased the numbers of activated microglia/macrophages in the brain, aggravated cerebral histological lesions, and exacerbated the impairment of motor and cognitive abilities. | 5 | Sequential fate-switches in stem-like cells drive the tumorigenic trajectory from human neural stem cells to malignant glioma显示文摘Glioblastoma(GBM)is an incurable and highly heterogeneous brain tumor,originating from human neural stem/progenitor cells(hNSCs/hNPCs)years ahead of diagnosis.Despite extensive efforts to characterize hNSCs and end-stage GBM at bulk and single-cell levels,the de novo gliomagenic path from hNSCs is largely unknown due to technical difficulties in early-stage sampling and preclinical modeling.Here,we established two highly penetrant hNSC-derived malignant glioma models,which resemble the histopathology and transcriptional heterogeneity of human GBM.Integrating time-series analyses of whole-exome sequencing,bulk and single-cell RNA-seq,we reconstructed gliomagenic trajectories,and identified a persistent NSC-like population at all stages of tumorigenesis.Through trajectory analyses and lineage tracing,we showed that tumor progression is primarily driven by multi-step transcriptional reprogramming and fate-switches in the NSC-like cells,which sequentially generate malignant heterogeneity and induce tumor phenotype transitions.We further uncovered stage-specific oncogenic cascades,and among the candidate genes we functionally validated C1QL1 as a new glioma-promoting factor.Importantly,the neurogenic-to-gliogenic switch in NSC-like cells marks an early stage characterized by a burst of oncogenic alterations,during which transient AP-1 inhibition is sufficient to inhibit gliomagenesis.Together,our results reveal previously undercharacterized molecular dynamics and fate choices driving de novo gliomagenesis from hNSCs,and provide a blueprint for potential early-stage treatment/diagnosis for GBM. | Xiaofei Wang Ran Zhou Yanzhen Xiong Lingling Zhou Xiang Yan Manli Wang Fan Li Chuanxing Xie Yiming Zhang Zongyao Huang Chaoqiong Ding Kaidou Shi Weida Li Yu Liu Zhongwei Cao Zhen-Ning Zhang Shengtao Zhou Chong Chen Yan Zhang Lu Chen Yuan Wang | 2021 | Cell Research2021,31,6: | 8 |
| 6 | Evolution of the East China Sea sedimentary environment in the past 14 kyr: Insights from tetraethers-based proxies显示文摘We reconstruct the environmental evolution of the East China Sea in the past 14 kyr based on glycerol dialkyl glycerol tetraethers(GDGTs) in a sediment core from the subaqueous Yangtze River Delta. Two primary phases are recognized. Phase I(13.8–8 cal kyr BP) reflects a predominantly continental influence, showing distinctly higher concentrations of branched GDGTs(averaged 143 ng/g dry sediment weight, dsw) than isoprenoid GDGTs(averaged 36 ng/g dsw), high BIT index(branched vs. isoprenoid tetraethers) values(>0.78) and a fluctuating GDGT-0/crenarchaeol ratio(R_(0/5), varied from 0.52 to 3.81). Within this interval, temporal increases of terrestrial and marine influence are attributed to Younger Dryas(YD)(ca. 12.9–12.2 cal kyr BP) cold event and melt-water pulse(MWP)-1B(11.5–11.1 cal kyr BP), respectively. The prominent transition from 8 to 7.9 cal kyr BP shows a sharp decrease in BIT index value(<0.4) and increase in crenarchaeol, which marks the beginning of phase II. Afterwards, the proxies remain relatively constant, which indicates that phase II(7.9 cal kyr BP-present) is a shelf sedimentary environment with high stand of sea level. Overall, the BIT index in our record serves as a good marker for terrestrial influence at the site, and likely reflects the flooding history of the region. The TEX_(86)(Tetra Ether Index of tetraethers consisting of 86 carbons) proxy is not applicable in phase I because of an excess terrestrial influence; but it seems to be valid for revealing the annual SST in phase II(21.6±0.9°C, n=49). | 7 | The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. | 10 | Loss of Wip1 aggravates brain injury after ischaemia/reperfusion by overactivating microglia显示文摘Background and purpose The inflammatory response mediated by microglia/macrophages is closely related to cerebral ischaemia/reperfusion injury.Wild-type p53-induced protein phosphatase 1(Wip1),a serine/threonine phosphatase,is expressed in various tissues.A growing number of reports have suggested that Wip1 is a negative regulator of inflammation in peripheral tissue;however,its role in the central nervous system(CNS)remains unclear.This study aimed to clarify whether Wip1 can inhibit CNS inflammation by regulating microglia/macrophage functions after ischaemic injury.Methods A model of middle cerebral artery occlusion and reperfusion was established in mice.CNS inflammation was simulated by lipopolysaccharide treatment of primary microglia.Laser speckle imaging was used to monitor regional cerebral blood flow.Behavioural outcomes were assessed with a TreadScan gait analysis system.TTC staining was used to evaluate the infarct volume,and western blotting and immunofluorescence staining were applied to detect the phenotypical transformation of microglia.ELISA was performed to detect the levels of inflammatory factors.Results Wip1 expression was increased after ischaemia/reperfusion.Wip1-knockout(KO)mice displayed more severe brain injury than wild-type mice,as indicated by aggravated motor dysfunction,greater brain infarct volumes and higher expression of inflammatory cytokines(interleukin-6 and tumour necrosis factor alpha)in the brain.We also found that Wip1 depletion increased microglial/macrophage activation in both in vitro and in vivo models,which all showed activation of microglia/macrophages.Lentivirus-Ppm1d reversed the injury induced by Wip1-KO.Conclusions Our results suggest that Wip1 may inhibit neuroinflammation by inhibiting microglial/macrophage activation after brain ischaemia/reperfusion injury. | Feng Yan Xiang Cheng Ming Zhao Shenghui Gong Ying Han Liping Ding Di Wu Yumin Luo Wei Zuo Lingling Zhu Ming Fan Xunming Ji | 2021 | Stroke & Vascular Neurology2021,6,3: | 4 |
| 11 | Oceanic barrier layer variation induced by tropical cyclones in the Northwest Pacific显示文摘According to Argo profiles and one-dimensional Price-Weller-Pinkel models, the oceanic barrier layer variation induced by tropical cyclones is adequately analyzed in the Northwest Pacific. Results show that tropical cyclones mainly aff ect the oceanic barrier layer through intensifying and weakening pre-existed barrier layer. The former even may generate new one after tropical cyclones’ passage. The latter can make pre-existed one disappear. Local wind stress and precipitation, the dominant factors, primarily determine the variation of barrier layer. Negative eff ects of wind mainly focus on the north of 20°N. This phenomenon is more meaningful for slow tropical cyclones. Conversely, positive eff ects of wind and precipitation center on the south of 20°N in the Northwest Pacific. | 14 | Salivary protease spectrum biomarkers of oral cancer显示文摘Proteases are important molecules that are involved in many physiological and pathological processes of the human body,such as growth,apoptosis and metastasis cancer cells.They are potential targets in cancer diagnosis and biotherapy.In this study,we analyzed the salivary protease spectrum of patients with oral squamous cell carcinoma (OSCC),oral benign masses and chronic periodontitis,as well as that of health,using human protease array kits,enzyme-linked immunosorbent assay,western blot and immunofluorescence.The salivary protease spectrum was found to be associated with oral diseases.For example,the saliva of patients with OSCC contained increased numbers of proteases than those of other oral diseases and health.The levels of matrix metalloproteinase (MMP)-1,MMP-2,MMP-10,MMP-12,A disintegrin and metalloprotease (ADAM)9,A disintegrin and metalloprotease with thrombospondin type 13 motifs (ADAMST13),cathepsin V and kallikrein 5 in the saliva of patients with OSCC were significantly increased compared with those of other groups.Taking MMP-1,cathepsin V,kallikrein 5 and ADAM9 as biomarkers of OSCC,cutoff values were199,11.34,9.29 and 202.55 pg·mL?1,respectively.From the area under the curve,sensitivity and specificity,the combination of cathepsin V/kallikrein5/ADAM9 was an optimal biomarker for diagnosing OSCC.Thus,analysis of the salivary protease spectrum may be an innovative and cost-efficient approach to evaluating the health status of the oral cavity.Specifically,increases in cathepsin V,kallikrein 5 and ADAM9 may be useful biomarkers in the screening and diagnosis of OSCC. | Yun Feng Qian Li Jiao Chen Ping Yi Xin Xu Yaping Fan Bomiao Cui Yu Yu Xiaoying Li Yue Du Qianming Chen Lingling Zhang Jingjing Jiang Xuedong Zhou Ping Zhang | 2019 | International Journal of Oral Science2019,11,2: | 2 |

| 17 | RNA modifications:importance in immune cell biology and related diseases显示文摘RNA modifications have become hot topics recently.By influencing RNA processes,including generation,transportation,function,and metabolization,they act as critical regulators of cell biology.The immune cell abnormality in human diseases is also a research focus and progressing rapidly these years.Studies have demonstrated that RNA modifications participate in the multiple biological processes of immune cells,including development,differentiation,activation,migration,and polarization,thereby modulating the immune responses and are involved in some immune related diseases.In this review,we present existing knowledge of the biological functions and underlying mechanisms of RNA modifications,including N6-methyladenosine(m^(6)A),5-methylcytosine(m^(5)C),N1-methyladenosine(m1 A),N7-methylguanosine(m^(7)G),N4-acetylcytosine(ac^(4)C),pseudouridine(Ψ),uridylation,and adenosine-to-inosine(A-to-I)RNA editing,and summarize their critical roles in immune cell biology.Via regulating the biological processes of immune cells,RNA modifications can participate in the pathogenesis of immune related diseases,such as cancers,infection,inflammatory and autoimmune diseases.We further highlight the challenges and future directions based on the existing knowledge.All in all,this review will provide helpful knowledge as well as novel ideas for the researchers in this area. | Lian Cui Rui Ma Jiangluyi Cai Chunyuan Guo Zeyu Chen Lingling Yao Yuanyuan Wang Rui Fan Xin Wang Yuling Shi | 2022 | Signal Transduction and Targeted Therapy2022,7,10: | 2 |
| 18 | Reactive oxygen species in paraventricular nucleus involved in cardiac sympathetic afferent reflex in rats显示文摘Objective: The present study was designed to determine if reactive oxygen species (ROS) in the paraventricular nucleus (PVN) were involved in modulating cardiac sympathetic afferent reflex (CSAR) in anesthetized rats. Methods: Malondialdehyde (MDA), the end product of lipid peroxidation, in the PVN, was determined by thiobarbituric acid (TBA) spectrometric method. Renal sympathetic nerve activity (RSNA) and arterial pressure were recorded in sinoaortic-denervated and cervical-vagotomized rats. The CSAR was evaluated by the response of the RSNA evoked by epicardial application of bradykinin (BK, 0.4 μg). Results: The MDA in the PVN was significantly increased after epicardial application of BK compared with control (2.0±0.3 vs 0.8±0.1 nmol/mg protein, P<0.01). Microinjection of a superoxide anion scavenger, tiron (20 nmol) into the PVN significantly inhibited the CSAR evoked by BK (12.3±1.9 vs 4.2±1.2%, P<0.01) and decreased MDA level (1.9±0.3 vs 0.6±0.1 nmol/mg protein, P<0.01) compared with control. | 19 | Upregulation of GLT25D1 in Hepatic Stellate Cells Promotes Liver Fibrosis via the TGF-β1/SMAD3 Pathway In Vivo and In vitro显示文摘Background and Aims:Collagenβ(1-O)galactosyltransferase 25 domain 1(GLT25D1)is associated with collagen production and glycosylation,and its knockout in mice results in embryonic death.However,its role in liver fibrosis remains elusive,particularly in hepatic stellate cells(HSCs),the primary collagen-producing cells associated with liver fibrogenesis.Herein,we aimed to elucidate the role of GLT25D1 in HSCs.Methods:Bile duct ligation(BDL)-induced mouse liver fibrosis models,primary mouse HSCs(mHSCs),and transforming growth factor beta 1(TGF-β1)-stimulated LX-2 human hepatic stellate cells were used in in vivo and in vitro studies.Stable LX-2 cell lines with either GLT25D1 overexpression or knockdown were established using lentiviral transfection.RNA-seq was performed to investigate the genomic differences.HPLCMS/MS were used to identify glycosylation sites.Scanning electronic microscopy(SEM)and second-harmonic generation/two-photon excited fluorescence(SHG/TPEF)were used to image collagen fibril morphology.Results:GLT25D1 expression was upregulated in nonparenchymal cells in human cirrhotic liver tissues.Meanwhile,its knockdown attenuated collagen deposition in BDL-induced mouse liver fibrosis and inhibited mHSC activation.GLT25D1 was overexpressed in activated versus quiescence LX-2 cells and regulated in vitro LX-2 cell activation,including proliferation,contraction,and migration.GLT25D1 also significantly increased liver fibrogenic gene and protein expression.GLT25D1 upregulation promoted HSC activation and enhanced collagen expression through the TGF-β1/SMAD signaling pathway.Mass spectrometry showed that GLT25D1 regulated the glycosylation of collagen in HSCs,affecting the diameter of collagen fibers.Conclusions:Collectively,the upregulation of GLT25D1 in HSCs promoted the progression of liver fibrosis by affecting HSCs activation and collagen stability. | Shiwei Wang Lingling He Fan Xiao Meixin Gao Herui Wei Junru Yang Yang Shu Fuyang Zhang Xiaohui Ye Ping Li Xiaohua Hao Xingang Zhou Hongshan Wei | 2023 | Journal of Clinical and Translational Hepatology2023,11,1: | 1 |
