1Effects of dendritic cells from cord blood CD34^+ cells on human hepatocarcinoma cell line BEL-7402 in vitro and in SCID mice显示文摘AIM: To develop a cancer vaccine of dendritic cells derived from human cord blood CD34+ cells and to investigate its cytotoxicity on human hepatocarcinoma cells in vitro and in sever combined immunodeficiency (SCTD) mice.METHODS: Lymphocytes from cord blood or peripheral blood were primed by DCs, which were derived from cord blood and pulsed with whole tumor cell lysates. Nonradiative neutral red uptake assay was adopted to detect the cytotoxicity of primed lymphocytes on human hepatocarcinoma cell line BEL-7402 in vitro. The anti-tumor effect of primed lymphocytes in vivo was detected in SCID mice, including therapeutic effect and vaccination effect.RESULTS: The cytotoxicity of DC vaccine primed lymphocytes from cord blood or peripheral blood on human hepatocarcinoma cell line BEL-7402 was significantly higher than that of unprimed lymphocytes in vitro (44.09% vs 14.69%,47.92% vs 19.44%, P<0.01). There was no significant difference between the cytotoxicity of primed lymphocytes from cord blood and peripheral blood (P>0.05). The tumor growth rate and tumor size were smaller in SCID mice treated or vaccinated with primed lymphocytes than those with unprimed lymphocytes. SCID mice vaccinated with primed lymphocytes had a lower tumor incidence (80%vs 100%, P<0.05) and delayed tumor latent period compared with mice vaccinated with unprimed lymphocytes (11 d vs 7 d, P<0.01).CONCLUSION: Vaccine of cord blood derived-DCs has an inhibitory activity on growth of human hepatocarcinoma cells in vitro and in SCID mice. 2NF-κB activation and zinc finger protein A20 expression in mature dendritic cells derived from liver allografts undergoing acute rejection显示文摘AIM: To investigate the role of NF-κB activation and zinc finger protein A20 expression in the regulation of maturation of dendritic cells (DCs) derived from liver allografts undergoing acute rejection. METHODS: Sixty donor male SD rats and sixty recipient male LEW rats weighing 220-300 g were randomly divided into whole liver transplantation group and partial liver transplantation group. Allogeneic (SD rat to LEW rat) whole and 50 % partial liver transplantation were performed. DCs from liver grafts 0 hour and 4 days after transplantation were isolated and propagated in the presence of GM-CSF in vitro. Morphological characteristics and phenotypical features of DCs propagated for 10 days were analyzed by electron microscopy and flow cytometry, respectively. NF-κB binding activity, IL-12p70 protein and zinc finger protein A20expression in these DCs were measured by EMSA and Western blotting, respectively. Histological grading of rejection was determined. RESULTS: Allogeneic whole liver grafts showed no signs of rejection on day 4 after the transplantation. In contrast,allogeneic partial liver grafts demonstrated moderate to severe rejection on day 4 after the transplantation. After propagation for 10 days in the presence of GM-CSF in vitro,DCs from allogeneic whole liver grafts exhibited features of immature DC with absence of CD40 surface expression,these DCs were found to exhibit detectable but very low level of NF-κB activity, IL-12 p70 protein and zinc finger protein A20 expression. Whereas, DCs from allogeneic partial liver graft 4 days after transplantation displayed features of mature DC, with high level of CD40 surface expression, and as a consequence, higher expression of IL-12p70 protein, higher activities of NF-κB and higher expression of zinc finger protein A20 compared with those of DCs from whole liver grafts (P<0.001). CONCLUSION: These results suggest that A20expression is up-regulated in response to NF-κB activation in mature DCs derived from allogeneic liver grafts undergoing acute rejection. Given the NF-κB inhibition function of this gene, it is suggested that their expression survives to limit NF-κB activation and maturation of DCs,and consequently inhibits the acute rejection and induces acceptance of liver graft.Ming-QingXu WeiWang LanXue Lv-NanYan 2003World Journal of Gastroenterology2003,9,6:8
3Comparative study on efficacy of three sperm-separation techniques显示文摘Aim: To evaluate the comparative effectiveness of real-time sperm separation technique (Wang's tube method) and other two conventional methods in isolating high-quality sperm preparation, and to compare the spouse pregnancy rate in intrauterine insemination (IUI) with sperm preparations isolated by these methods. Methods: The effectiveness of the real-time sperm separation technique, the conventional swim-up and the Percoll discontinuous density gradient methods in isolating sperm preparations from 60 infertile patients (20 with apparently normal semen and 40, abnormal semen contaminated with microorganisms and other impurities) was evaluated and compared. The microorganisms to bere moved included bacteria, virus, Chlamydia trachomaticum, Ureaplsama urealyticum, etc. The spouse pregnancy rates in IUI with sperm preparations isolated by these three techniques from 80 oligoasthenoteratospermic patients werealso compared. Results: The quality (including the percentages of normal form, normal-chromatin and motilesperm, and the grade of motility) of sperm obtained by the real-time sperm separation technique was much higher ( P<0.01 ) as compared with those by the other two methods. The Wang's tube method was also more effective in removing microorganisms and other impurities. The method provided a higher IUI pregnancy rate than the other two sperm separation techniques ( P < 0.05). 6Immunotherapy for Alzheimer’s disease:targetingβ-amyloid and beyond显示文摘Alzheimer’s disease(AD)is the most common neurodegenerative disease in the elderly worldwide.However,the complexity of AD pathogenesis leads to discrepancies in the understanding of this disease,and may be the main reason for the failure of AD drug development.Fortunately,many ongoing preclinical and clinical studies will continually open up avenues to unravel disease mechanisms and guide strategies for AD diagnosis and drug development.For example,immunotherapeutic strategies targeting amyloid-β(Aβ)and tau proteins were once deemed almost certainly effective in clinical treatment due to the excellent preclinical results.However,the repeated failures of clinical trials on vaccines and humanized anti-Aβand anti-tau monoclonal antibodies have resulted in doubts on this strategy.Recently,a new anti-Aβmonoclonal antibody(Aducanumab)has been approved by the US Food and Drug Administration,which brings us back to the realization that immunotherapy strategies targeting Aβmay be still promising.Meanwhile,immunotherapies based on other targets such as tau,microglia and gut-brain axis are also under development.Further research is still needed to clarify the forms and epitopes of targeted proteins to improve the accuracy and effectiveness of immunotherapeutic drugs.In this review,we focus on the immunotherapies based on Aβ,tau and microglia and their mechanisms of action in AD.In addition,we present up-to-date advances and future perspectives on immunotherapeutic strategies for AD.Chenghuan Song Jiyun Shi Pingao Zhang Yongfang Zhang Jianrong Xu Lanxue Zhao Rui Zhang Hao Wang Hongzhuan Chen 2022Translational Neurodegeneration2022,11,1:1

8An effective global learning framework for hyperspectral image classification based on encoder-decoder architecture显示文摘Most deep learning methods in hyperspectral image(HSI)classification use local learning methods,where overlapping areas between pixels can lead to spatial redundancy and higher computational cost.This paper proposes an efficient global learning(EGL)framework for HSI classification.The EGL framework was composed of universal global random stratification(UGSS)sampling strategy and a classification model BrsNet.The UGSS sampling strategy was used to solve the problem of insufficient gradient variance resulted from limited training samples.To fully extract and explore the most distinguishing feature representation,we used the modified linear bottleneck structure with spectral attention as a part of the BrsNet network to extract spectral spatial information.As a type of spectral attention,the shuffle spectral attention module screened important spectral features from the rich spectral information of HSI to improve the classification accuracy of the model.Meanwhile,we also designed a double branch structure in BrsNet that extracted more abundant spatial information from local and global perspectives to increase the performance of our classification framework.Experiments were conducted on three famous datasets,IP,PU,and SA.Compared with other classification methods,our proposed method produced competitive results in training time,while having a greater advantage in test time.Lanxue Dang Chongyang Liu Weichuan Dong Yane Hou Qiang Ge Yang Liu 2022International Journal of Digital Earth2022,15,1:0
