2Sequencing of Cultivated Peanut, Arachis hypogaea, Yields Insights into Genome Evolution and Oil Improvement显示文摘Cultivated peanut (Arachis hypogaea) is an allotetraploid crop planted in Asia, Africa, and America for edible oil and protein. To explore the origins and consequences of tetraploidy, we sequenced the allotetraploid A. hypogaea genome and compared it with the related diploid Arachis duranensis and Arachis ipaensis genomes. We annotated 39 888 A-subgenome genes and 41 526 B-subgenome genes in allotetraploid peanut. The A. hypogaea subgenomes have evolved asymmetrically, with the B subgenome resembling the ancestral state and the A subgenome undergoing more gene disruption, loss, conversion, and transposable element proliferation, and having reduced gene expression during seed development despite lacking genome-wide expression dominance. Genomic and transcriptomic analyses identified more than 2 500 oil metabolism-related genes and revealed that most of them show altered expression early in seed development while their expression ceases during desiccation, presenting a comprehensive map of peanut lipid biosynthesis. 3Cancer incidence and mortality in Shandong province,2012显示文摘Objective: Population-based cancer registration data in 2012 from all available cancer registries in Shandong province were collected by Shandong Center for Disease Control and Prevention(SDCDC). SDCDC estimated the numbers of new cancer cases and cancer deaths in Shandong province with compiled cancer incidence and mortality rates.Methods: In 2015, there were 21 cancer registries submitted data of cancer incidence and deaths occurred in 2012. All the data were checked and evaluated based on the National Central Cancer Registry(NCCR) criteria of data quality. Qualified data from 15 registries were used for cancer statistics analysis as provincial estimation. The pooled data were stratified by area(urban/rural), gender, age group(0, 1-4, 5-9, 10-14, …, 85+ years) and cancer type. New cancer cases and deaths were estimated using age-specific rates and corresponding provincial population in 2012. The Chinese census data in 2000 and Segi's population were applied for age-standardized rates. All the rates were expressed per 100,000 person-year.Results: Qualified 15 cancer registries(4 urban and 11 rural registries) covered 17,189,988 populations(7,486,039 in urban and 9,703,949 in rural areas). The percentage of cases morphologically verified(MV%) and death certificate-only cases(DCO%) were 66.12% and 2.93%, respectively, and the mortality to incidence rate ratio(M/I) was 0.60. A total of 253,060 new cancer cases and 157,750 cancer deaths were estimated in Shandong province in 2012. The incidence rate was 263.86/100,000(303.29/100,000 in males, 223.23/100,000 in females), the age-standardized incidence rates by Chinese standard population(ASIRC) and by world standard population(ASIRW) were 192.42/100,000 and 189.50/100,000 with the cumulative incidence rate(0-74 years old) of 22.07%. The cancer incidence, ASIRC and ASIRW in urban areas were 267.64/100,000, 195.27/100,000 and 192.02/100,000 compared to 262.32/100,000, 191.26/100,000 and 188.48/100,000 in rural areas, respectively. The cancer mortality was 164.47/100,000(207.42/100,000 in males, 120.23/100,000 in females), the age-standardized incidence rates by Chinese standard population(ASMRC) and by world standard population(ASMRW) were 117.54/100,000 and 116.90/100,000, and the cumulative mortality rate(0-74 years old) was 13.53%. The cancer mortality, ASMRC and ASMRW were 141.59/100,000, 101.17/100,000 and 100.33/100,000 in urban areas, and 173.79/100,000, 124.20/100,000 and 123.64/100,000 in rural areas, respectively. Cancers of the lung, stomach, liver, esophagus, colorectum, female breast, brain, leukemia, bladder and pancreas were the most common cancers, accounting for about 82.12% of all cancer new cases. Lung cancer, stomach cancer, liver cancer, esophageal cancer, colorectal cancer, female breast cancer, pancreatic cancer, brain tumor, leukemia and lymphoma were the leading causes of cancer death, accounting for about 89.01% of all cancer deaths. The cancer spectrum showed difference between urban and rural, males and females both in incidence and mortality rates.Conclusions: Cancer surveillance information in Shandong province is making great progress with the increasing number of cancer registries, population coverage and the improving data quality. Cancer registration plays a fundamental role in cancer control by providing basic information on population-based cancer incidence, mortality, survival and time trend. 4Tiller Bud Formation Regulators MOC1 and MOC3 Cooperatively Promote Tiller Bud Outgrowth by Activating F0N1 Expression in Rice显示文摘Tillering in rice is one of the most important agronomic traits.Rice tiller development can be divided into two main processes: the formation of the axillary bud and its subsequent outgrowth.Several genes critical for bud formation in rice have been identified by genetic studies;however,their molecular functions and relationships are still largely unknown.Here,we report that MONOCULM 1 (MOC1) and MONOCULM 3/ TILLERS ABSENT 1/STERILE AND REDUCED TILLERING 1 (MOC3/TAB1/SRT1),two vital regulators for tiller formation in rice,physically interact to regulate tiller bud outgrowth through upregulating the expression of FLORAL ORGAN NUMBER 1 (FON1),the homolog of CLAVATA1 in rice.We found that M0C3 is able to directly bind the promoter ofFONI and subsequently activate FON1 expression.MOC1 functions as a coactivator of MOC3,whereas it could not directly bind the FON1 promoter,and further activated FON1 expression in the presence of MOC3.Accordingly,FON1 is highly expressed at axillary meristems and shows remarkably decreased expression levels in mod and moc3 mutants.Loss-of-function mutants of FON1 exhibit normal bud formation but defective bud outgrowth and reduced tiller number.Collectively,these results shed light on a joint transcriptional regulatory mechanim by MOC1 and MOC3,and establish a new framework for the control of tiller bud formation and outgrowth.Gaoneng Shao Zefu Lu Jinsong Xiong Bing Wang Yanhui Jing Xiangbing Meng Guifu Liu Haiyan Ma Yan Liang Fan Chen Yonghong Wang Jiayang Li Hong Yu 2019Molecular Plant2019,12,8:15

7Environmental factors influencing cyanobacteria community structure in Dongping Lake, China显示文摘The present study was conducted to provide a detailed understanding of the variation in cyanobacterial communities of Dongping Lake, which is the final water volume adjusting and storing lake in the east route of the South-to-North Water Diversion Project in China. The spatial and temporal distribution of cyanobacteria was assessed from May 2010 to October 2012 based on monthly samples collected from three stations. Over the 30-month survey, 15 genera and 25 species of cyanobacteria were identified, with cyanobacterial abundance at each monitoring station ranging from undetected to 3.04×107 cells/L, average of 4.27×106 cells/L. The dominant cyanobacterial species were Pseudanabaena limnetica and Aphanizomenon issatschenkoi and not the usual bloom-forming genera such as Microcystis and Anabaena. Cyanobacterial community structure and water quality variables exhibited substantial changes over the period of survey. Redundancy analysis, Pearson correlations, and regression analysis were applied to analyze the relationships among the variables. The results suggested that temperature and chemical oxygen demand were key drivers of the cyanobacterial community composition in Dongping Lake. 12Phytoplankton composition and its ecological effect in subsurface cold pool of the northern Bering Sea in summer as revealed by HPLC derived pigment signatures显示文摘CHEMTAX analysis of high-performance liquid chromatography(HPLC) pigment was conducted to study phytoplankton community structure in the northern Bering Sea shelf, where a seasonal subsurface cold pool emerges. The results showed that fucoxanthin(Fuco) and chlorophyll a(Chl a) were the most abundant diagnostic pigments, with the integrated water column values ranging from 141 to 2 160 μg/m2 and 477 to 5 535 μg/m2, respectively. Moreover, a diatom bloom was identified at Sta. BB06 with the standing stock of Fuco up to 9 214 μg/m3. The results of CHEMTAX suggested that the phytoplankton community in the northern Bering Sea shelf was dominated by diatoms and chrysophytes with an average relative contribution to Chl a of 80% and 12%, respectively, followed by chlorophytes, dinoflagellates, and cryptophytes. Diatoms were the absolutely dominant algae in the subsurface cold pool with a relative contribution exceeding 90%, while the contribution of chrysophytes was generally higher in oligotrophic upper water. 13Chemical genomics reveals inhibition of breast cancer lung metastasis by Ponatinib via c-Jun显示文摘Metastasis is the leading cause of human cancer deaths.Unfortunately,no approved drugs are available for antimetastatic treatment.In our study,high-throughput sequencing-based high-throughput screening(HTS^2)and a breast cancer lung metastasis(BCLM)-associated gene signature were combined to discover anti-metastatic drugs.After screening of thousands of compounds,we identified Ponatinib as a BCLM inhibitor.Ponatinib significantly inhibited the migration and mammosphere formation of breast cancer cells in vitro and blocked BCLM in multiple mouse models.Mechanistically,Ponatinib represses the expression of BCLM-associated genes mainly through the ERK/c-Jun signaling pathway by inhibiting the transcription of JUN and accelerating the degradation of c-Jun protein.Notably,JUN expression levels were positively correlated with BCLM-associated gene expression and lung metastases in breast cancer patients.Collectively,we established a novel approach for the discovery of anti-metastatic drugs,identified Ponatinib as a new drug to inhibit BCLM and revealed c-Jun as a crucial factor and potential drug target for BCLM.Our study may facilitate the therapeutic treatment of BCLM as well as other metastases.Wei Shao Shasha Li Lu Li Kequan Lin Xinhong Liu Haiyan Wang Huili Wang Dong Wang 2019Protein & Cell2019,10,3:7
14Symptoms of long-term exposure to moxa smoke in acupuncturists: a correlation analysis显示文摘OBJECTIVE: To investigate the effects of long-term exposure to moxa smoke on acupuncturists and to consider the association between physical symptoms and gender, age, secondhand smoke(SHS) exposure, and the duration of exposure.METHODS: A self-report, web-based questionnaire was used to evaluate the effects of moxa smoke onacupuncturists. Logistic regression was used to analyze the relationship between physical symptoms and possible correlative factors.RESULTS: A total of 858 questionnaires were analyzed. The data showed that 42.42% of acupuncturists had been exposed to moxa smoke for 5-10 years. The most frequent symptom was tearing(32.98%) and the least frequent symptom was asthma(5.24%). Logistic regression analysis showed that female acupuncturists were slightly more susceptible to cough and tearing than males(cough:OR: 1.583, 95% CI: 1.079, 2.321, P = 0.019; tearing:OR: 1.519, 95% CI: 1.094, 2.108, P = 0.013). Acupuncturists aged over 25 years reported a slightly lower incidence of eye itchiness than those aged under25 years(26-40 years: OR: 0.604, 95% CI: 0.379,0.965, P = 0.035; over 40 years: OR: 0.330, 95% CI:0.114, 0.958, P = 0.042). Ophthalmodynia occurred less in acupuncturists aged 26-40 years(OR: 0.591,95% CI: 0.359, 0.970, P = 0.038) than in those younger than 25 years. The only association between prevalence of symptoms and tobacco use and SHS exposure was that smokers had a lower occurrence of phlegm in the throat(OR: 0.579, 95% CI: 0.392,0.856, P = 0.006). Shortness of breath was less frequent in participants exposed to moxa smoke for more than 5 years(5-10 years: OR: 0.400, 95% CI:0.204, 0.785, P = 0.008; 11-20 years: OR: 0.392, 95%CI: 0.178, 0.864, P = 0.02), but a slightly higher incidence of eye itchiness was found in those with over20 years of moxa exposure(OR: 4.200, 95% CI:1.344, 3.128, P = 0.014).CONCLUSION: The most frequent symptom of moxa smoke exposure in acupuncturists was tear-ing. 15First observation of Karlodinium venef icum from the East China Sea and the coastal waters of Germany显示文摘Harmful algal blooms often occurred in the East China Sea (ECS) and the German coastal waters of the North Sea but Karlodinium veneficum had not been taxonomically reported. Two strains of Karlodinium (LAMB090611 and LAMB010601) were isolated from the two areas in 2009. The morphological characteristics and molecular phylogeny of two strains are compared on the basis of observation of a light microscope, a scanning electron microscope (SEM), a laser scanning micro-scope (LSM) and an internal transcribed spacer (ITS) sequence data. The mean cell length of strain LAMB090611 is (14.2 ± 1.8) μm (range 11.1-18.7 μm) and mean width is (10.8 ± 1.5) μm (range 8.2- 14.7 μm). The mean cell length of strain LAMB010601 is (15.1 ± 1.2) μm (range 12.7-17.9 μm) and the mean width is (11.4 ± 1.1) μm (range 9.1-14.7 μm), respectively. The two strains are similar in morphological characteristics, including a straight apical groove, distinct ventral pore, sulcal extension, cingulum displacement, two or four irregular shaped chloroplasts within the cell and almost equal sized epicone and hypocone. The large and round nucleus is located at the center or at the hypocone of the cell. The sequence length of the ECS strain LAMB090611 and the German strain LAMB010601 is 640 and 646 bp, respectively. The GC content is 49%. The nucleotide similarity of the two strains is 98.1%. The sequence divergence is 0.003. Both strains are confirmed as Karlo-dinium veneficum (D. Ballantine) J. Larsen and this is the first taxonomic report from China and Germany coastal waters. 16Composition of algal pigments in surface freshen layer after ice melt in the central Arctic显示文摘Seasonal meltwater input creates a thin freshen layer in surface seawater under ice,which largely shifts the algae assemblages.Our recent observation of photosynthetic pigments in the high Arctic showed that ice bottom and 5m of seawater under ice contained relatively high concentration of fucoxanthin,while chlorophyll b and lutein were the major diagnostic pigments in ice-water interface and 0 m of seawater under ice.Additionally,a notable change of dominant phytoplankton occurred in the top 5 m of seawater under ice,from chlorophytes-dominated at surface to diatoms-dominated at 5 m depth,which might attribute to the sharp salinity gradient(salinity from12.5 to 28.1) in the surface seawater under ice.Our results imply that phytoplankton community in surface layer under ice would become more chlorophytes in the future warming Arctic Ocean.ZHUANG Yanpei JIN Haiyan GU Fan ZHANG Yang BAI Youcheng JI Zhongqiang LU Yong CHEN lianfang 2017Acta Oceanologica Sinica2017,36,8:6
17Sequential therapy according to distinct disease progression patterns in advanced ALK-positive non-small-cell lung cancer after crizotinib treatment显示文摘Objective: Crizotinib is recommended as the first-line therapy for advanced anaplastic lymphoma kinase(ALK)-positive non-small-cell lung cancer(NSCLC). Despite its initial efficacy, patients ultimately acquire resistance to crizotinib within 1 year. In such patients, the optimal sequential therapy after crizotinib treatment remains unknown. This study explored which sequential therapy option confers the greatest benefit.Methods: A total of 138 patients with advanced ALK-positive NSCLC resistant to crizotinib were studied. Based on patterns of disease progression of metastases, patients were divided into 3 groups: brain progression, non-liver progression, and liver progression. Sequential therapies included crizotinib continuation plus local therapy, nextgeneration ALK inhibitors(ALKi's), and chemotherapy. The primary endpoint was overall survival(OS) from the time of crizotinib resistance to death or last follow-up.Results: The 138 patients included 64 cases with progression in brain, 57 cases in non-liver sites and 17 cases in liver. A significant difference in OS was observed among the distinct progression pattern(median OS, 25.4 months in brain, 15.8 months in non-liver, and 10.8 months in liver, respectively, P=0.020). The difference in OS among sequential therapies was statistically significant in the non-liver progression group(median OS, 27.6 months with next-generation ALKi's, 13.3 months with crizotinib continuation, and 10.8 months with chemotherapy,respectively, P=0.019). However, crizotinib continuation plus local therapy seems to provide non-inferior median OS compared with next-generation ALKi's for patients with brain progression(median OS, 28.9 months vs.32.8 months, P=0.204). And no significant differences in OS were found in patients with progression in liver(P=0.061).Conclusions: Crizotinib continuation together with local therapy might be a feasible strategy for patients with progression in brain beyond crizotinib resistance, as well as next-generation ALKi's. 18Characterization of a Putative New Semi-Dominant Reduced Height Gene, Rht_NM9, in Wheat(Triticum aestivum L.)显示文摘Plant height is an important agronomic trait in cereal crops, and can affect both plant architecture and grain yield. New dwarfing genes are required for improving the genetic diversity of wheat. In this study, a novel dwarf mutant, NM9, was created by treating seeds of the wheat variety NAU9918 with ethyl methanesulfonate(EMS). NM9 showed obvious phenotypic changes, which were distinct from those caused by other dwarfing genes, especially the reduced plant height, increased effective tiller number, and elongated spike and grain length. The reduced plant height in NM9 was attributable to a semi-dominant dwarfing gene Rht_NM9, which was flanked by two closely linked SNP markers, SNP34 and SNP41, covering an 8.86-Mb region on the chromosome arm 2AS. The results of gibberellic acid(GA) sensitivity evaluation, comparative genomics analysis and allelism test indicated that Rht_NM9 was neither allelic to Rht7 and Rht21 nor homoeoallelic to Rht8, so Rht_NM9 was proposed to be a new dwarfing locus on the homoeologous group 2 chromosomes of wheat. 20FABP4 secreted by M1-polarized macrophages promotes synovitis and angiogenesis to exacerbate rheumatoid arthritis显示文摘Increasing evidence shows that adipokines play a vital role in the development of rheumatoid arthritis(RA).Fatty acid-binding protein 4(FABP4),a novel adipokine that regulates inflammation and angiogenesis,has been extensively studied in a variety of organs and diseases.However,the effect of FABP4 on RA remains unclear.Here,we found that FABP4 expression was upregulated in synovial M1-polarized macrophages in RA.The increase in FABP4 promoted synovitis,angiogenesis,and cartilage degradation to exacerbate RA progression in vivo and in vitro,whereas BMS309403(a FABP4 inhibitor)and anagliptin(dipeptidyl peptidase 4 inhibitor)inhibited FABP4 expression in serum and synovial M1-polarized macrophages in mice to alleviate RA progression.Further studies showed that constitutive activation of mammalian target of rapamycin complex 1(mTORC1)by TSC1 deletion specifically in the myeloid lineage regulated FABP4 expression in macrophages to exacerbate RA progression in mice.In contrast,inhibition of mTORC1 by ras homolog enriched in brain(Rheb1)disruption specifically in the myeloid lineage reduced FABP4 expression in macrophages to attenuate RA development in mice.Our findings established an essential role of FABP4 that is secreted by M1-polarized macrophages in synovitis,angiogenesis,and cartilage degradation in RA.BMS309403 and anagliptin inhibited FABP4 expression in synovial M1-polarized macrophages to alleviate RA development.Hence,FABP4 may represent a potential target for RA therapy.Dong Guo Chuangxin Lin Yuheng Lu Hong Guan Weizhong Qi Hongbo Zhang Yan Shao Chun Zeng Rongkai Zhang Haiyan Zhang Xiaochun Bai Daozhang Cai 2022Bone Research2022,10,4:5
