1Pathological features of COVID-19-associated lung injury:a preliminary proteomics report based on clinical samples显示文摘The COVID-19 pandemic has emerged as a global health emergency due to its association with severe pneumonia and relative high mortality.However,the molecular characteristics and pathological features underlying COVID-19 pneumonia remain largely unknown.To characterize molecular mechanisms underlying COVID-19 pathogenesis in the lung tissue using a proteomic approach,fresh lung tissues were obtained from newly deceased patients with COVID-19 pneumonia.After virus inactivation,a quantitative proteomic approach combined with bioinformatics analysis was used to detect proteomic changes in the SARS-CoV-2-infected lung tissues.We identified significant differentially expressed proteins involved in a variety of fundamental biological processes including cellular metabolism,blood coagulation,immune response,angiogenesis,and cell microenvironment regulation.Several inflammatory factors were upregulated,which was possibly caused by the activation of NF-κB signaling.Extensive dysregulation of the lung proteome in response to SARS-CoV-2 infection was discovered.Our results systematically outlined the molecular pathological features in terms of the lung response to SARS-CoV-2 infection,and provided the scientific basis for the therapeutic target that is urgently needed to control the COVID-19 pandemic.Ling Leng Ruiyuan Cao Jie Ma Danlei Mou Yunping Zhu Wei Li Luye Lv Dunqin Gao Shikun Zhang Feng Gong Lei Zhao Bintao Qiu Haiping Xiang Zhongjie Hu Yingmei Feng Yan Dai Jiang Zhao Zhihong Wu Hongjun Li Wu Zhong 2020Signal Transduction and Targeted Therapy2020,5,1:4
2Ribonuclease H-like gene SMALL GRAIN2 regulates grain size in rice through brassinosteroid signaling pathway显示文摘Grain size is a key agronomic trait that determines the yield in plants.Regulation of grain size by brassinosteroids(BRs)in rice has been widely reported.However,the relationship between the BR signaling pathway and grain size still requires further study.Here,we isolated a rice mutant,named small grain2(sg2),which displayed smaller grain and a semi-dwarf phenotype.The decreased grain size was caused by repressed cell expansion in spikelet hulls of the sg2 mutant.Using map-based cloning combined with a Mut Map approach,we cloned SG2,which encodes a plant-specific protein with a ribonuclease H-like domain.SG2 is a positive regulator downstream of GLYCOGEN SYNTHASE KINASE2(GSK2)in response to BR signaling,and its mutation causes insensitivity to exogenous BR treatment.Genetical and biochemical analysis showed that GSK2 interacts with and phosphorylates SG2.We further found that BRs enhance the accumulation of SG2 in the nucleus,and subcellular distribution of SG2 is regulated by GSK2 kinase activity.In addition,Oryza sativa OVATE family protein 19(OsOFP19),a negative regulator of grain shape,interacts with SG2 and plays an antagonistic role with SG2 in controlling gene expression and grain size.Our results indicated that SG2 is a new component of GSK2-related BR signaling response and regulates grain size by interacting with Os OFP19.Yunshuai Huang Hui Dong Changling Mou Ping Wang Qixian Hao Min Zhang Hongmin Wu Fulin Zhang Tengfei Ma Rong Miao Kai Fu Yaping Chen Ziyan Zhu Cheng Chen Qikai Tong Zhuoran Wang Shirong Zhou Xi Liu Shijia Liu Yunlu Tian Ling Jiang Jianmin Wan 2022Journal of Integrative Plant Biology2022,64,10:1

5Rapid Detection of Rifampin-resistant Clinical Isolates of Mycobacterium tuberculosis by Reverse Dot Blot Hybridization显示文摘Objective A PCR-reverse dot blot hybridization(RDBH) assay was developed for rapid detection of rpo B gene mutations in ‘hot mutation region' of Mycobacterium tuberculosis(M. tuberculosis). Methods 12 oligonucleotide probes based on the wild-type and mutant genotype rpoB sequences of M. tuberculosis were designed to screen the most frequent wild-type and mutant genotypes for diagnosing RIF resistance. 300 M. tuberculosis clinical isolates were detected by RDBH, conventional drug-susceptibility testing(DST) and DNA sequencing to evaluate the RDBH assay. Results The sensitivity and specificity of the RDBH assay were 91.2%(165/181) and 98.3%(117/119), respectively, as compared to DST. When compared with DNA sequencing, the accuracy, positive predictive value(PPV) and negative predictive value(NPV) of the RDBH assay were 97.7%(293/300), 98.2%(164/167), and 97.0%(129/133), respectively. Furthermore, the results indicated that the most common mutations were in codons 531(48.6%), 526(25.4%), 516(8.8%), and 511(6.6%), and the combinative mutation rate was 15(8.3%). One and two strains of insertion and deletion were found among all strains, respectively. 6Characterization of Chlamydia trachomatis omp1 gene among sexually transmitted disease patients in south China显示文摘To investigate the DNA sequence polymorphism of Chlamydia trachomatis omp1 gene, urogenital samples were collected from 4 different cities in South China, DNA was extracted, and an approximately 980-bp-long fragment of the omp1 gene was amplified by nested polymerase chain reaction (nPCR). DNA sequence was determined, genotyping was performed by BLAST similarity search, and multiple alignment was performed with CLUSTAL X. Then a phylogenetic tree was constructed by Mega 3 software to illustrate the evolutionary relationships between clinical isolates and reference strains. Ninety-six specimens were sequenced, and 28 genetic variants were detected, among which E was the most prevalent genotype. The omp1 gene was highly conserved for genotypes E and F, but appeared slightly less conserved for other genotypes, where the sequences displayed one to several nucleotide substitutions relative to the reference sequence. Phylogenetic tree showed that C.trachomatis serotypes were mainly divided into three clusters, according to previous grouping in the B, F-G, and C complexes, and the clinical isolates were highly related to the corresponding reference strains. 7Denoising enabled channel estimation for underwater acoustic communications:A sparsity-aware model-driven learning approach显示文摘It has always been difficult to achieve accurate information of the channel for underwater acoustic communications because of the severe underwater propagation conditions,including frequency-selective property,high relative mobility,long propagation latency,and intensive ambient noise,etc.To this end,a deep unfolding neural network based approach is proposed,in which multiple layers of the network mimic the iterations of the classical iterative sparse approximation algorithm to extract the inherent sparse features of the channel by exploiting deep learning,and a scheme based on the Sparsity-Aware DNN(SA-DNN)for UAC estimation is proposed to improve the estimation accuracy.Moreover,we propose a Denoising Sparsity-Aware DNN(DeSA-DNN)based enhanced method that integrates a denoising CNN module in the sparsity-aware deep network,so that the degradation brought by intensive ambient noise could be eliminated and the estimation accuracy can be further improved.Simulation results demonstrate that the performance of the proposed schemes is superior to the state-of-the-art compressed sensing based and iterative sparse recovery schems in the aspects of channel recovery precision,pilot overhead,and robustness,particularly under unideal circumstances of intensive ambient noise or inadequate measurement pilots.Sicong Liu Younan Mou Xianyao Wang Danping Su Ling Cheng 2023Intelligent and Converged Networks2023,4,1:0
8Artemin对胰腺癌细胞系MIA PaCa-2浸润能力的研究(英文)显示文摘Objective:The aim of this study was to analyze the capacity of Artemin promoting the motility and invasiveness of MIA PaCa-2 pancreatic cancer (PAC) cells.Methods:The PAC cell line MIA PaCa-2 was cultured in vitro and studied using Transwell chamber analysis.The motility and invasiveness ability affected by different concentrations of Artemin and its receptor GFRα3 were determined.Expression level of matrix metalloproteinase-2 (MMP-2),epithelial cadherin (E-cadherin) were quantitative analysis using RT-PCR and Western blot in MIA PaCa-2 cells stimulated with Artemin and receptor GFRα3.Results:MIA PaCa-2 PAC cell motility and invasiveness was significantly increased with Artemin and its receptor GFRα3 increasing concentrations than control (P<0.01).150 ng/mL was the best of both the role of concentration.MMP-2 was increased significantly (t=6.35,t=7.32),while E-cadherin was significantly lower (t=4.27,t=5.61),after affected by the 150 ng/mL Artemin and GFRα3,respectively.The difference was statistically significant compared with the control group (P<0.01).Conclusion:Artemin and its receptor GFRα3 can promote PAC cell motility and invasiveness ability and contribute to the aggressive behavior.The mechanism may be related to increased expression of MMP-2 molecule and E-cadherin down-regulation expression.Lingxin Meng Yuhua Chi Xiangxu Wang Zhaojun Ding Ling Mou Wen Cui Yingjie Xue 2012The Chinese-German Journal of Clinical Oncology2012,11,4:0

12Evaluation of the efficacy and safety of endoscopic band ligation in the treatment of bleeding from mild to moderate gastric varices type 1显示文摘BACKGROUND According to practice guidelines,endoscopic band ligation(EBL)and endoscopic tissue adhesive injection(TAI)are recommended for treating bleeding from esophagogastric varices.However,EBL and TAI are known to cause serious complications,such as hemorrhage from dislodged ligature rings caused by EBL and hemorrhage from operation-related ulcers resulting from TAI.However,the optimal therapy for mild to moderate type 1 gastric variceal hemorrhage(GOV1)has not been determined.Therefore,the aim of this study was to discover an individualized treatment for mild to moderate GOV1.AIM To compare the efficacy,safety and costs of EBL and TAI for the treatment of mild and moderate GOV1.METHODS A clinical analysis of the data retrieved from patients with mild or moderate GOV1 gastric varices who were treated under endoscopy was also conducted.Patients were allocated to an EBL group or an endoscopic TAI group.The differences in the incidence of varicose relief,operative time,operation success rate,mortality rate within 6 wk,rebleeding rate,6-wk operation-related ulcer healing rate,complication rate and average operation cost were compared between the two groups of patients.RESULTS The total effective rate of the two treatments was similar,but the efficacy of EBL(66.7%)was markedly better than that of TAI(39.2%)(P<0.05).The operation success rate in both groups was 100%,and the 6-wk mortality rate in both groups was 0%.The average operative time(26 min)in the EBL group was significantly shorter than that in the TAI group(46 min)(P<0.01).The rate of delayed postoperative rebleeding in the EBL group was significantly lower than that in the TAI group(11.8%vs 45.1%)(P<0.01).At 6 wk after the operation,the healing rate of operation-related ulcers in the EBL group was 80.4%,which was significantly greater than that in the TAI group(35.3%)(P<0.01).The incidence of postoperative complications in the two groups was similar.The average cost and other related economic factors were greater for the EBL than for the TAI(P<0.01).CONCLUSION For mild to moderate GOV1,patients with EBL had a greater one-time varix eradication rate,a greater 6-wk operation-related ulcer healing rate,a lower delayed rebleeding rate and a lower cost than patients with TAI.Yue Deng Ya Jiang Tong Jiang Ling Chen Hai-Jun Mou Bi-Guang Tuo Guo-Qing Shi 2024World Journal of Gastroenterology2024,30,5:0
13The Analysis of Clinical Features of Systemic Lupus Erythematosus (SLE) in Children显示文摘Objective:Observe the clinical characteristics of children with SLE,namely,to observe the symptoms and laboratory examinations,such as blood routine,blood lipid,immunoglobulin,complement,autoantibodies,serum 25(OH)D and other indicators,and to explore the clinical characteristics,the difference and the significance of vitamin D supplements between male and female SLE patients in children respectively.Methods:We enrolled 64 cases of SLE patients in children who were admitted into the department of pediatrics and rheumatology of the third affiliated hospital of sun yat-sen university in guangzhou from May 1,2011 to February 1,2019,They were analyzed retrospectively,adoptingΧ²test for statistical analysis.Results:64 cases of SLE in children,which included 10 cases of male and 54 cases of female.Clinical manifestations:facial skin rash in 48 patients(75%),fever in 38 cases(59.4%),arthritis in 28 cases(43.8%),oral ulcer in 18 cases(28.1%),serositis in13 cases(20.3%),and the sun allergy in 9 cases(14.1%),the damage of central nervous system in 7 cases(10.9%).Laboratory examination:30 cases of leukopenia(46.9%),anemia in 30 cases(46.9%),thrombocytopenia in 12 cases(18.8%),hematuria in 18 cases(28.1%),proteinuria in 33 cases(51.2%),6 patients with renal impairment(9.4%),antinuclear antibody positive in 63 cases(98.4%),anti-double-stranded DNA(dsDNA)antibody positive in 48 cases(75%),anti SSA antibody positive in 44 cases(68.7%),SSB antibody positive in 33 cases(51.6%),Sm antibody positive in 40 cases(62.5%),nucleosome antibody positive in 28 cases(43.8%).Among these children,male SLE patients were higher than female children with SLE in the damage of kidney,Sm antibodies and resisting nucleosome antibody positive rates(Χ²=4.451,8.336,6.803,P<0.05),the female children with SLE was higher than male SLE Children in the anti-SSB antibody positive rate(Χ²=4.945,P<0.05).In 64 cases of SLE children,which included 52 cases were lower than the normal level of serum 25(OH)D measurements,12 cases were in the normal lower limit of serum 25(OH)D measurements,at the same time,the female SLE.Patients was higher than male children with SLE in the reduce rate of serum 25(OH)D(Χ²=8.351,P<0.05).Conclusion:Male SLE patients which appeared damage of kidney easier than female patients,the proteinuria was the most common in the damage of kidney.Resistance to Sm antibodies which was the risk factor of renal injury with higher incidence in male children with SLE;Anti nucleosome antibody which was the risk factor for the disease activity in male children with SLE were higher than female children with SLE.It was estimated that the risk of Sjogren’s syndrome appeared in female with SLE were higher than that in male SLE children.In this retrospective study,the serum 25(OH)D levels were significantly lower in children with SLE,and vitamin D supplementation was required.Ying Liang Pingping Zhang Huiqin Chen Xiangqin Luo Yesheng Ling Yating Li Ou Jin Qian Kong Fenhua Chen Yikun Mou 2020Journal of Clinical and Nursing Research2020,4,5:0
14Young Leaf White Stripe encodes a P-type PPR protein required for chloroplast development显示文摘Pentatricopeptide repeat(PPR) proteins function in post-transcriptional regulation of organellar gene expression. Although several PPR proteins are known to function in chloroplast development in rice(Oryza sativa), the detailed molecular functions of many PPR proteins remain unclear.Here, we characterized a rice young leaf white stripe(ylws) mutant, which has defective chloroplast development during early seedling growth.Map-based cloning revealed that YLWS encodes a novel P-type chloroplast-targeted PPR protein with 11 PPR motifs. Further expression analyses showed that many nuclear-and plastid-encoded genes in the ylws mutant were significantly changed at the RNA and protein levels. The ylws mutant was impaired in chloroplast ribosome biogenesis and chloroplast development under low-temperature conditions. The ylws mutation causes defects in the splicing of atpF, ndhA, rpl2,and rps12, and editing of ndhA, ndhB, and rps14transcripts. YLWS directly binds to specific sites in the atpF, ndhA, and rpl2 pre-mRNAs.
