| 1 | A novel gene delivery system targeting cells expressing VEGF receptors显示文摘Two ligand oligopeptides GV1 and GV2 were designed according to the putative binding region of VEGF to its receptors. GV1, GV2 and endosome releasing oligopeptide HA20 were conjugated with poly-L-lysine or protamine and the resulting conjugates could interact with DNA in a noncovalent bond to form a complex. Using pSV2-β-galactosidase as a reporter gene, it has been demonstrated that exogenous gene was transferred into bovine aortic arch-derived endothelial cells (ABAE) andhuman malignant melanoma cell lines (A375) in vitro. In vivo experiments, exogenous gene was transferred into tumor vascular endothelial cells and tumor cells of subcutaneously transplanted human colon cancer LOVO, human malignant melanoma A375 and human hepatoma graft in nude mice. This system could also target gene to intrahepatically transplanted human hepatoma injected via portal vein in nude mice. | 2 | Determination of Seven Major Ginsenosides in Different Parts of Panax quinquefolius L.(American Ginseng) with Different Ages显示文摘Ginsenosides Rg1, Re, Rb1, Rc, Rb2, Rb3, and Rd in different parts of the American ginseng plant were investigated. The extraction process was a pressurized microwave-assisted extraction(PMAE). The seven ginsenosides were separated and determined by high-performance liquid chromatography(HPLC) with a ultraviolet(UV) detector, at 203 nm. The experiment results showed significant variations in the individual ginsenoside contents of the American ginseng in different parts and ages of the plant. The results demonstrated that the leaves, root hairs, and rhizomes of Panax quinquefolius L. contained higher ginsenoside contents, followed by the main roots and stems. The leaves contained dramatically higher levels of ginsenoside Rg1, Rb3, and Rd than the other four parts. Higher contents of Rb1 and Re were present in the main roots, root hairs, and rhizomes. The amount of ginsenoside content in the stems was the lowest. The total content of the seven ginsenosides in main roots, root hairs and rhizomes increased with the age of the plant. | 4 | Overexpression of Sweet Pepper Glycerol-3-Phosphate Acyltransferase Gene Enhanced Thermotolerance of Photosynthetic Apparatus in Transgenic Tobacco显示文摘In order to investigate the relationship between the lipid composition in thylakoid membrane and thermostability of pho-tosynthetic apparatus,tobacco transformed with sweet pepper sense glycerol-3-phosphate acyltransferase (GPAT) gene were used to analyze the lipid composition in thylakoid membrane,the net photosynthetic rate and chlorophyll fluorescence parameters under high temperature stress. The results showed that the saturated extent of monogalactosyldiacylglycerol (MGDG),sulfoquinovosyldiacylglycerol,digalactosyldiacylglycerol and phosphatidylglycerol in thylakoid membrane of transgenic tobacco T1 lines increased generally. Particularly,the saturated extent in MGDG increased obviously by 16.2% and 12.0% in T1-2 and T1-1,respectively. With stress temperature elevating,the maximum efficiency of photosystem II (PSII) photochemistry (Fv/Fm),actual photochemical efficiency of PSII in the light ( PSII) and net photosynthetic rate (Pn) of the two lines and wild type tobacco plants decreased gradually,but those parameters decreased much less in transgenic plants. Even though the recovery process appeared differently in the donor and acceptor side of PSII in transgenic tobacco compared with wild-type plants,the entire capability of PSII recovered faster in transgenic tobacco,which was shown in the parameters of PI,Fv/Fm and PSII,as a result,the recovery of Pn was accelerated. | 8 | Anatomical variation of infra-pyloric artery origination: A prospective multicenter observational study (IPA-Origin)显示文摘Objective: Infra-pyloric artery(IPA) is an important anatomical landmark in treatment of gastric cancer and is the key vessel for pylorus-preserving gastrectomy and subgroup of infra-pyloric lymph nodes. However, its anatomical variation is not thoroughly understood. Our study aimed to clarify the origination of the IPA.Methods: We did this prospective, multicenter, open-label, observational study at gastric surgery departments of34 hospitals in China. Gastric cancer patients aged 18 years or older and scheduled to undergo elective total or distal gastrectomy were assigned. During the surgery, IPA dissecting and exposing the origination point with photographs or video clips were required. The primary outcome was the origination of the IPA. Analysis of variance, χ~2 tests and Fisher's tests were used to analyze the differences between groups. The study is registered at Clinicaltrials.gov(No. NCT03071237).Results: Between May 8 and July 31, 2017, 429 patients were assigned for the study, and 419(97.7%) patients had the IPA dissected and recorded through photograph or video and were included in the primary outcome analysis. The median age was 62 years old, and 73.7% were male. Among the patients, 78.5% received laparoscopic surgery. Single IPA origination was identified in 398(95.0%) patients, including gastroduodenal artery(GDA) in154(36.8%) patients, anterior superior pancreaticoduodenal artery(ASPDA) in 130(31.0%) patients, and right gastroepiploic artery(RGEA) in 114(27.2%) patients. Fifteen(3.6%) patients were identified with multiple IPA and 6(1.4%) patients were identified as IPA absence. The differences in the distribution of surgical approach(P=0.003) and geographic area(P=0.030) were statistically significant. | 9 | 泌尿生殖系统小细胞癌:11年临床治疗经验显示文摘当前对于泌尿生殖系统的小细胞癌治疗及判断预后的影响因素尚存在较大争议。本研究回顾性分析了在2002年至2013年在复旦大学附属肿瘤医院诊治的18例泌尿生殖系统小细胞癌患者的临床资料,其中有10例来源于前列腺,7例来源于膀胱,1例来源于肾上腺。平均随访时间为15.5个月,平均无疾病进展时间为9.3个月。对其中的13例患者施行原发病灶切除术,在诊断为局限性泌尿生殖系统小细胞癌的患者中6例接受了根治性手术治疗,13例患者接受了以顺铂为基础的化疗。经统计学分析发现与乳酸脱氢酶较高的患者相比,乳酸脱氢酶处于正常范围的患者无疾病进展时间及总生存时间都明显延长(P=0.030,P=0.010),对于局限性小细胞癌患者,接受根治性手术治疗的患者与未接受根治手术的患者相比在无疾病进展时间上无统计学差异,但是在总生存时间上有明显差别。因此血液中乳酸脱氢酶高于正常值往往预示着患者的预后较差,对于诊断为局限性泌尿生殖系统小细胞肿瘤的患者,强烈推荐及早对患者施行根治性手术并联合以顺铂为基础的化疗。 | Kun Chang | 2014 | Asian Journal of Andrology2014,16,5: | 5 |
| 10 | 肥胖对临床局限性前列腺癌患者临床病理特征的影响显示文摘本研究旨在探讨不同肥胖测量指标对中国临床局限性前列腺癌患者临床病理特征的影响。本研究共入组734例在我院接受根治性前列腺切除术治疗的临床局限性前列腺癌患者,我们回顾性收集了全组患者的临床病理资料。BMI的计算即体重公斤数除以身高米数的平方。本组共413例患者术前在我院行盆腔增强核磁共振(magnetic resonance imaging,MRI)扫描。在MRJ扫描T2加权、矢状位图像上测量413例患者的腹部肥胖测量指标。利用方差分析或卡方检验比较不同BMI组患者的临床病理特征。利用Logistic回归模型分析术前血清睾酮水平和各肥胖测量指标对术后病理结果的影响。在多因素分析中,我们并未发现BMI与患者术后病理结果存在相关性。然而,多因素分析结果显示,内脏脂肪比例为术后病理Gleason评分≥8分(P〈O.001),包膜外侵犯(P=0.002)和精囊腺侵犯(P=0.007)的独立预后因素。此外,我们还发现术前血清睾酮水平与内脏脂肪比例呈负相关(R=-0.485,P〈0.001)而与皮下脂肪厚度成正相关(R=0.413,P〈0.001)。综上所述,本研究结果显示,腹部脂肪的分布,尤其是内脏脂肪比例,是进展性前列腺癌的危险因素。而且,内脏脂肪比例与血清睾酮水平呈负相关。内脏肥胖影响进展性前列腺癌的分子生物学机制仍需进一步研究。 | Yuan-Yuan Qu | 2013 | Asian Journal of Andrology2013,15,6: | 4 |

A small number(5%–10%) of these injuries fail to heal due to non-union and require surgical intervention. Currently, the principal treatment options for these defects are autografts, allografts, xenografts or synthetic grafts. The main problems associated with these therapies include pain,infection and donor site morbidity. Bone tissue engineering is a diverse field that focuses on the regeneration of bone by combining cells, scaffolds, growth factors and dynamic forces. There have been many recent studies utilizing biomineralized polymer matrix composites which mimic the natural structure of bone. The principal focus of this review is on recent advances in the synthesis of various types of biomineralized polymer matrix composite. | 12 | Host-Associated Quantitative Abundance Profiling Reveals the Microbial Load Variation of Root Microbiome显示文摘Plant-associated microbes are critical for plant growth and survival under natural environmental conditions.To date,most plant microbiome studies involving high-throughput amplicon sequencing have focused on the relative abundance of microbial taxa.However,this technique does not assess the total microbial load and the abundance of individual microbes relative to the amount of host plant tissues.Here,we report the development of a host-associated quantitative abundance profiling(HA-QAP)method that can accurately examine total microbial load and colonization of individual root microbiome members relative to host plants by the copy-number ratio of microbial marker gene to plant genome.We validate the HAQAP method using mock experiments,perturbation experiments,and metagenomic sequencing.The HA-QAP method eliminates the generation of spurious outputs in the classical method based on microbial relative abundance,and reveals the load of root microbiome to host plants.Using the HA-QAP method,we found that the copy-number ratios of microbial marker genes to plant genome range from 1.07 to 6.61 for bacterial 16S rRNA genes and from 0.40 to 2.26 for fungal internal transcribed spacers in the root microbiome samples from healthy rice and wheat.Furthermore,using HA-QAP we found that an increase in total microbial load represents a key feature of changes in root microbiome of rice plants exposed to drought stress and of wheat plants with root rot disease,which significantly influences patterns of differential taxa and species interaction networks.Given its accuracy and technical feasibility,HA-QAP would facilitate our understanding of genuine interactions between root microbiome and plants. | Xiaoxuan Guo Xiaoning Zhang Yuan Qin Yong-Xin Liu Jingying Zhang Na Zhang Kun Wu Baoyuan Qu Zishan He Xin Wang Xinjian Zhang Stephane Hacquard Xiangdong Fu Yang Bai | 2020 | Plant Communications2020,1,1: | 3 |

| 14 | Uptake of oxidative stress-mediated extracellular vesicles by vascular endothelial cells under low magnitude shear stress显示文摘Extracellular vesicles(EVs)are increasingly used as delivery vehicles for drugs and bioactive molecules,which usually require intravascular administration.The endothelial cells covering the inner surface of blood vessels are susceptible to the shear stress of blood flow.Few studies demonstrate the interplay of red blood cell-derived EVs(RBCEVs)and endothelial cells.Thus,the phagocytosis of EVs by vascular endothelial cells during blood flow needs to be elucidated.In this study,red blood cell-derived extracellular vesicles(RBCEVs)were constructed to investigate endothelial cell phagocytosis in vitro and animal models.Results showed that low magnitude shear stress including low shear stress(LSS)and oscillatory shear stress(OSS)could promote the uptake of RBCEVs by endothelial cells in vitro.In addition,in zebrafish and mouse models,RBCEVs tend to be internalized by endothelial cells under LSS or OSS.Moreover,RBCEVs are easily engulfed by endothelial cells in atherosclerotic plaques exposed to LSS or OSS.In terms of mechanism,oxidative stress induced by LSS is part of the reason for the increased uptake of endothelial cells.Overall,this study shows that vascular endothelial cells can easily engulf EVs in areas of low magnitude shear stress,which will provide a theoretical basis for the development and utilization of EVs-based nano-drug delivery systems in vivo. | Xian Qin Kun Zhang Juhui Qiu Nan Wang Kai Qu Yuliang Cui Junli Huang Li Luo Yuan Zhong Tian Tian Wei Wu Yi Wang Guixue Wang | 2022 | Bioactive Materials2022,7,3: | 3 |
| 15 | A male-ABCD algorithm for hepatocellular carcinoma risk prediction in HBs Ag carriers显示文摘Objective: Hepatocellular carcinoma(HCC) development among hepatitis B surface antigen(HBs Ag) carriers shows gender disparity, influenced by underlying liver diseases that display variations in laboratory tests. We aimed to construct a risk-stratified HCC prediction model for HBs Ag-positive male adults.Methods: HBs Ag-positive males of 35-69 years old(N=6,153) were included from a multi-center populationbased liver cancer screening study. Randomly, three centers were set as training, the other three centers as validation. Within 2 years since initiation, we administrated at least two rounds of HCC screening using Bultrasonography and α-fetoprotein(AFP). We used logistic regression models to determine potential risk factors,built and examined the operating characteristics of a point-based algorithm for HCC risk prediction.Results: With 2 years of follow-up, 302 HCC cases were diagnosed. A male-ABCD algorithm was constructed including participant's age, blood levels of GGT(γ-glutamyl-transpeptidase), counts of platelets, white cells,concentration of DCP(des-γ-carboxy-prothrombin) and AFP, with scores ranging from 0 to 18.3. The area under receiver operating characteristic was 0.91(0.90-0.93), larger than existing models. At 1.5 points of risk score,26.10% of the participants in training cohort and 14.94% in validation cohort were recognized at low risk, with sensitivity of identifying HCC remained 100%. At 2.5 points, 46.51% of the participants in training cohort and 33.68% in validation cohort were recognized at low risk with 99.06% and 97.78% of sensitivity, respectively. | 17 | CD4+Foxp3+CD25+/-Tregs characterize liver tissue specimens of patients suffering from drug-induced autoimmune hepatitis:A clinical-pathological study显示文摘Background: The diagnosis of drug-induced autoimmune hepatitis(DIAIH) and its differentiation from idiopathic autoimmune hepatitis(AIH) is challenging. This study aimed to differentiate DIAIH from AIH by comparing the biochemical changes, histological features, and frequencies of CD4^+Foxp3^+CD25^(+/-)regulatory T cells(Tregs) in liver tissues or peripheral blood lymphocytes.Methods: A total of 15 DIAIH patients and 24 AIH patients who underwent liver biopsies at initial presentation were enrolled in this study. The liver histological changes were assessed by HE staining. The phenotypic recognition and distribution of CD4^+Foxp3^+CD25^(+/-)Tregs in liver tissues were evaluated by single/double immunostains in serial sections. The CD4^+Foxp3^+CD25^(+/-)Tregs in peripheral blood were analyzed by flow cytometry.Results: The median values of ALT and AST were 404.50 U/L and 454.10 U/L in DIAIH patients and309.50 U/L and 315.00 U/L in AIH patients, respectively. More importantly, for the first time we found that patients with DIAIH had higher levels of serum ALT and AST, more severe degree of lobular inflammation,higher frequencies of zone 3 necrosis and higher number of lobular CD4^+Foxp3^+CD25^-Tregs compared with AIH(P < 0.05). Furthermore, there were positive correlations in DIAIH between the degree of lobular inflammation and either the AST/ALT level or the number of lobular CD4^+Foxp3^+CD25^-Tregs(P < 0.05).However, the frequency of peripheral blood CD4^+Foxp3^+CD25^(+/-)Tregs were not significantly different between DIAIH and AIH.Conclusions: The differences of ALT, AST and the number of lobular CD4^+Foxp3^+CD25^-Tregs between patients with DIAIH and those with AIH are clinically helpful in differentiating these two diseases in their early stage. | Li-Mei Qu Shu-Hua Wang Kun Yang David R.Brigstock Li Su Run-Ping Gao | 2018 | Hepatobiliary & Pancreatic Diseases International2018,17,2: | 3 |
| 18 | Ciliate Uronema marinum is the causative agent of scuticociliatosis in farm raised turbot Scophthalmus maximus显示文摘To identify the pathogen that causes scuticociliatosis in farmed-raised turbot Scophthalmus maximus , we isolated a ciliate from the brain tissue of an infected turbot and identifi ed it as Uronema marinum based on morphological and molecular evidence. We then infected the turbots in artifi cial laboratory settings with pure cultured U . marinum . The infected turbots showed syndromes similar to those observed in naturally infected ones. Furthermore, microscopic examination and PCR detection confi rmed the presence of the ciliate in the tissues of those laboratory-infected turbots.
