| 1 | Mercury remediation potential of a mercury resistant strain Sphingopyxis sp.SE2 isolated from contaminated soil显示文摘A mercury resistant bacterial strain SE2 was isolated from contaminated soil.The 16 s rRNA gene sequencing confirms the strain as Sphingopyxis belongs to the Sphingomonadaceae family of the α-Proteobacteria group.The isolate showed high resistance to mercury with estimated concentrations of Hg that caused 50%reduction in growth(EC_(50)) of 5.97 and 6.22 mg/L and minimum inhibitory concentrations(MICs) of 32.19 and 34.95 mg/L in minimal and rich media,respectively.The qualitative detection of volatilized mercury and the presence of mercuric reductase enzyme proved that the strain SE2 can potentially remediate mercury.ICP-QQQ-MS analysis of the remaining mercury in experimental broths indicated that a maximum of 44%mercury was volatilized within 6 hr by live SE2 culture.Furthermore a small quantity(23%) of mercury was accumulated in live cell pellets.While no volatilization was caused by dead cells,sorption of mercury was confirmed.The mercuric reductase gene merA was amplified and sequenced.Homology was observed among the amino acid sequences of mercuric reductase enzyme of different organisms from a-Proteobacteria and ascomycota groups, | Khandaker Rayhan Mahbub Kannan Krishnan Ravi Naidu Mallavarapu Megharaj | 2017 | Journal of Environmental Sciences2017,29,1: | 10 |
| 2 | Nutrient dynamics in the Sundarbans mangrove estuarine system of Bangladesh under different weather and tidal cycles显示文摘Introduction:Seasonal and tidal variations in nutrient concentration and water quality were investigated in the western Sundarbans of Bangladesh during the post-monsoon,winter and monsoon seasons during 2010–2011.Methods:Water collected from the surface,middle and bottom layers at six locations of the Kholpetua-Arpangashia river system during high and low tides was analyzed for temperature,salinity,pH,dissolved oxygen(DO),total dissolved solids(TDS),electrical conductivity(EC)and dissolved nutrients(NO3–N,PO4–P,SO4 and NH3–N).Results:The study revealed that nutrient concentrations were influenced by seasonal changes.Mean nutrient levels during post-monsoon,winter and monsoon seasons,respectively,were in the following ranges:nitrate(0.06–0.40,0.06–0.46 and 0.08–0.46 mg/L);phosphate(0.09–0.18,0.05–0.42 and 0.10–0.16 mg/L);sulphate(58.71–86.14,68.68–119.01 and 78.15–136.47 mg/L)and ammonia(0.02–0.08,0.02–0.04 and 0.26–0.38 mg/L).Increased levels of PO4–P,SO4 and NH3–N and lower DO and salinity were recorded during the monsoon period.Most of the experimental sites showed higher NO3–N content during monsoon,whereas few elevated concentrations were observed during post-monsoon and winter periods.High and low tidal waters contained mean nutrient levels in the following ranges:nitrate(0.05–0.46 and 0.04–0.40 mg/L);phosphate(0.05–0.42 and 0.07–0.18 mg/L);sulphate(63.63–125.36 and 58.71–136.47 mg/L)and ammonia(0.02–0.38 and 0.02–0.37 mg/L)without following any distinct fluctuation patterns.The western part of the Sundarbans receives less freshwater input during the monsoon season than other areas of the ecosystem,which reduces the variability of nutrient levels and water quality components.Conclusions:This study provides considerable advances in understanding the seasonality of nutrient distribution with possible tidal influence.The data generated from this study will guide continuing efforts to support a sound management for coastal mangrove ecosystems. | Shak Md Bazlur Rahaman Lipton Sarder Md Sayadur Rahaman Alokesh Kumar Ghosh Sudhin Kumar Biswas SM Shahjahan Siraj Khandaker Anisul Huq Abul Farah Md Hasanuzzaman Shikder Saiful Islam | 2013 | Ecological Processes2013,2,1: | 2 |

| 19 | The anti-scarring effect of corneal stromal stem cell therapy is mediated by transforming growth factorβ3显示文摘Background:Corneal stromal stem cells(CSSC)reduce corneal inflammation,prevent fibrotic scarring,and regenerate transparent stromal tissue in injured corneas.These effects rely on factors produced by CSSC to block the fibrotic gene expression.This study investigated the mechanism of the scar-free regeneration effect.Methods:Primary human CSSC(hCSSC)from donor corneal rims were cultivated to passage 3 and co-cultured with mouse macrophage RAW264.7 cells induced to M1 pro-inflammatory phenotype by treatment with interferonγand lipopolysaccharides,or to M2 anti-inflammatory phenotype by interleukin-4,in a Transwell system.The timecourse expression of human transforming growth factorβ3(hTGFβ3)and hTGFβ1 were examined by immunofluorescence and qPCR.TGFβ3 knockdown for>70%in hCSSC[hCSSC-TGFβ3(si)]was achieved by small interfering RNA transfection.Naïve CSSC and hCSSC-TGFβ3(si)were transplanted in a fibrin gel to mouse corneas,respectively,after wounding by stromal ablation.Corneal clarity and the expression of mouse inflammatory and fibrosis genes were examined.Results:hTGFβ3 was upregulated by hCSSC when co-cultured with RAW cells under M1 condition.Transplantation of hCSSC to wounded mouse corneas showed significant upregulation of hTGFβ3 at days 1 and 3 post-injury,along with the reduced expression of mouse inflammatory genes(CD80,C-X-C motif chemokine ligand 5,lipocalin 2,plasminogen activator urokinase receptor,pro-platelet basic protein,and secreted phosphoprotein 1).By day 14,hCSSC treatment significantly reduced the expression of fibrotic and scar tissue genes(fibronectin,hyaluronan synthase 2,Secreted protein acidic and cysteine rich,tenascin C,collagen 3a1 andα-smooth muscle actin),and the injured corneas remained clear.However,hCSSC-TGFβ3(si)lost these anti-inflammatory and anti-scarring functions,and the wounded corneas showed intense scarring.Conclusion:This study has demonstrated that the corneal regenerative effect of hCSSC is mediated by TGFβ3,inducing a scar-free tissue response. | Lin Weng James L.Funderburgh Irona Khandaker Moira L.Geary Tianbing Yang Rohan Basu Martha L.Funderburgh Yiqin Du Gary Hin-Fai Yam | 2020 | Eye and Vision2020,7,1: | 1 |
