| 2 | Complement C3 activation regulates the production of tRNA-derived fragments Gly-tRFs and promotes alcohol-induced liver injury and steatosis显示文摘Complement is known to play a role in alcoholic fatty liver disease (AFLD), but the underlying mechanisms are poorly understood, thereby constraining the development of a rational approach for therapeutic intervention in the complement system. C3 deficiency has been shown to impart protective effects against ethanol-induced hepatic steatosis and inflammation. Here we demonstrate a protection effect in wild-type mice by treatment with CR2-Crry, a specific inhibitor of C3 activation. The expression of glycine transfer (t) RNA-derived fragments (Gly-tRFs) is upregulated in ethanol-fed mice and inhibition of Gly-tRFs in vivo decreases chronic ethanol feeding-induced hepatosteatosis without affecting inflammation. The expression of Gly-tRF was downregulated in C3-deficient or CR2-Crry-treated mice, but not in C5-deficient mice;Gly-tRF expression was restored by the C3 activation products C3a or Asp (C3a-des-Arg) via the regulation of CYP2E1. Transcriptome profiling of hepatic tissues showed that Gly-tRF inhibitors upregulate the expression of sirtuin1 (Sirt1) and subsequently affect downstream lipogenesis and β-oxidation pathways. Mechanistically, Gly-tRF interacts with AGO3 to downregulate Sirt1 expression via sequence complementarity in the 3′ UTR. Notably, the expression levels of C3d, CYP2E1 and Gly-tRF are upregulated, whereas Sirt1 is decreased in AFLD patients compared to healthy controls. Collectively, our findings suggest that C3 activation products contribute to hepatosteatosis by regulating the expression of Gly-tRF. | 3 | Zircon U–Pb and Hf isotopic study of Neoproterozoic granitic gneisses from the Alatage area, Xinjiang:constraints on the Precambrian crustal evolution in the Central Tianshan Block显示文摘The architecture and growth history of Precambrian crustal basements in the Central Tianshan Block play a key role in understanding the tectonic evolution of the Chinese Tianshan Orogenic Belt.In this study,we present precise LA-ICP-MS zircon U–Pb dating and LAMC-ICPMS zircon Hf isotopic data for two granitic gneisses from Alatage area in the Central Tianshan Block.The magmatic zircons from both samples yield similar protolith ages of 945±6 and 942±6 Ma,indicating that the early Neoproterozoic magmatism is prevailed in the Alatage area.These zircons have crustal Hf model ages of1.82–2.22 and 1.70–2.03 Ga,respectively,which are significantly older than their crystallization ages.It indicates that their parental magmas were derived from the reworking of ancient crust.However,we suggest that these Paleoproterozoic Hf model ages might result from mixing of continental materials with different ages in the Neoproterozoic crust.The inherited(detrital)zircon cores not only yield a wide age range of ca.989–1617 Ma,but also exhibit large Hf-isotope variations with Hf model ages of1.54–2.30 Ga.In particular,some 1.4–1.6 Ga zircons show high initial176Hf/177Hf ratios,consistent with those of depleted mantle,which indicates that the Mesoproterozoic event involved both reworking of older crust and generation of juvenile crust.The Central Tianshan Block has different Precambrian crustal growth history from the Tarim Craton.Therefore,it would not be a fragment of the Precambrian basement of the Tarim Craton. | Botao Huang Zhenyu He Keqing Zong Zeming Zhang | 2014 | Chinese Science Bulletin2014,59,1: | 12 |
| 4 | Leveraging Auxiliary Knowledge for Web Service Clustering显示文摘By grouping Web services that share similar functionalities, Web service clustering can greatly enhance Web service discovery and selection. Most existing clustering techniques are designed to handle long text documents. However, the descriptions of most publicly available Web services are in the form of short text, which impairs the quality of service clustering due to the sparseness of useful information. Towards this issue, we propose a new service clustering approach based on transfer learning from auxiliary long text data obtained from Wikipedia.To handle the inconsistencies in semantics and topics between service descriptions and auxiliary data, we introduce a novel topic model – Tag aided dual Author topical model(TD-ATM), which jointly learns two sets of topics on the two data sets and automatically couples the topic parameters to avoid the potential inconsistencies between these two data sets. | 6 | Non-matrix-matched analysis of U-Th-Pb geochronology of bastn?site by laser ablation inductively coupled plasma mass spectrometry显示文摘Bastn?site is widespread in alkali granite, carbonatite and REE ore deposits. The U-Th-Pb ages of bastn?site can constrain the mineralization age. Currently, the lack of suitable in situ analysis reference material is the major obstacle of bastn?site U-Th-Pb geochronology. In this study, the matrix effects of U-Pb and Th-Pb age determination in bastn?site using glass NIST 610 as the external calibration reference were evaluated with the 193-nm excimer LA-ICP-MS analysis in normal and additional gases laser ablation modes. The obtained U-Th-Pb ages of bastn?site were approximately 7–11% lower than their reference values in the normal analytical mode(without additional gas). Fortunately, the determined systematic age biases in UTh-Pb dating were significantly reduced(less than 1–2%) with the addition of water vapor within the ablation chamber. This reduction may be attributed to the observed significant suppression of206 Pb/238 U and208 Pb/232 Th downhole fractionation in both NIST 610 glass and bastn?site by introducing a small quantity of water vapor in the ablation chamber. In addition, the obtained206 Pb/238 U and208 Pb/232 Th ages of bastn?site K-9, LZ1384, and MAD809 showed great consistency with their corresponding recommended values. The obtained results show that accurate and precise U-Th-Pb ages of bastn?site can be simultaneously obtained by using glass NIST 610 as the primary calibration standard in 193-nm LA-ICP-MS with the water vapor-assisted method.
