| 2 | Tom20 senses iron-activated ROS signaling to promote melanoma cell pyroptosis显示文摘Iron has been shown to trigger oxidative stress by elevating reactive oxygen species (ROS) and to participate in different modes of cell death,such as ferroptosis,apoptosis and necroptosis. However,whether iron-elevated ROS is also linked to pyroptosis has not been reported. Here,we demonstrate that iron-activated ROS can induce pyroptosis via a Tom20-Bax-caspase-GSDME pathway. In melanoma cells,iron enhanced ROS signaling initiated by CCCP,causing the oxidation and oligomerization of the mitochondrial outer membrane protein Tom20. Bax is recruited to mitochondria by oxidized Tom20,which facilitates cytochrome c release to cytosol to activate caspase-3;eventually triggering pyroptotic death by inducing GSDME cleavage. Therefore,ROS acts as a causative factor and Tom20 senses ROS signaling for iron-driven pyroptotic death of melanoma cells. Since iron activates ROS for GSDME-dependent pyroptosis induction and melanoma cells specifically express a high level of GSDME,iron may be a potential candidate for melanoma therapy. Based on the functional mechanism of iron shown above,we further demonstrate that iron supplementation at a dosage used in iron-deficient patients is sufficient to maximize the anti-tumor effect of clinical ROS-inducing drugs to inhibit xenograft tumor growth and metastasis of melanoma cells through GSDME-dependent pyroptosis. Moreover,no obvious side effects are observed in the normal tissues and organs of mice during the combined treatment of clinical drugs and iron. | 3 | Simulation of energy and water balance in Soil-Vegetation-Atmosphere Transfer system in the mountain area of Heihe River Basin at Hexi Corridor of northwest China显示文摘In the mountain area of inland Heihe River Basin at Hexi Corridor of northwest China during the vegetation growing season from May to September, the Simultaneous Heat and Water (SHAW) model of Soil-Vegetation-Atmosphere Transfer (SVAT) system is applied to simulating and studying energy and water balance of the soil-residue-plant canopy layers in the Picea crassifolia forest and the grassland by the forest at the shaded slope and the grassland at the sun-facing slope. The simulation of energy balance indicates that net radiation of the grass- land at the sun-facing slope is more than that of the Picea crassifolia forest and the grassland by the forest at the shaded slope. The energy outgoing components are the first latent heat and next sensible heat from the grassland both at the shaded slope and the sun-facing slope, but those at the former are less. The energy outgoing components are the first sensible heat and next latent heat from the Picea crassifolia forest. The composition and distribution of energy in the soil-residue-plant canopy layers in the Picea crassifolia forest and the grassland by the forest at the shaded slope make the soil layer receive less energy, which therefore, especially the forest possesses the energy conditions for soil water conservation. The simulation of water balance indicates that the water loss of the grassland at the sun-facing slope is mainly caused by soil evaporation, while evapotranspiration of the Picea crassifolia forest and the grassland by the forest at the shaded slope is less than that of the grassland at the sun-facing slope. Half of the evapotranspiration of the Picea crassifolia forest and the grassland by the forest at the shaded slope is consumed by transpiration. After precipitation, the soil water storage is increased much more for the Picea crassifolia forest and also more for the grassland by the forest at the shaded slope. | 4 | A multi-center study of hemodynamic characteristics exhibited by children with unexplained syncope显示文摘Background Syncope is common in children and adolescents,with 15% estimated to have had at least one syncopal episode by age 18.In recent years,an increasing number of children,especially girls at their school age,have developed unexplained syncope.The mechanism of an unexplained syncope exhibited by children is incompletely studied;the association between different hemodynamic patterns and clinical features is also not clear.The aim of the study was to investigate the hemodynamic patterns of children with unexplained syncope and to examine the clinical relevance.Methods Two hundred and eight children [87 boys,121 girls,aged 3-19 years,mean(11.66±2.72) years] were selectively recruited from May 2000 to April 2006 when they presented syncope as their main complaint at the Multi-center Network for Childhood Syncope in Beijing,Hunan Province,Hubei Province,and Shanghai of China.All of the patients underwent head-up tilt tests;data were analyzed using SPSS version 10.0 for Windows.Continuous variables were expressed as the mean±standard deviation.Dichotomous variables were compared through a χ2 test.A value of P<0.05(two sided) was regarded as statistically significant.Results The age distribution of children with syncope was approximately normal.Head-up tilt tests was positive in 155 children,and the incidence of positive response of the baseline head-up tilt test for diagnosing unexplained syncope was 50.48%.The sensitivity value and diagnostic value of sublingual nitroglycerin head-up tilt test were both 74.52%.The hemodynamic pattern was normal in 53 children.The 155 children,who were positive in head-up tilt tests,showed signs of postural orthostatic tachycardia syndrome(60,28.8%),the vasoinhibitory pattern(72,34.6%),the cardioinhibitory pattern(5,2.4%),and the mixed pattern(18,8.7%).The gender distribution between the two age groups(age < 12 years vs age ≥ 12 years) was not different(P>0.05).The distribution of hemodynamic patterns between the children of the two age groups(age < 12 years vs age ≥ 12 years),and the children with different complaints(dizziness vs syncope) was significantly different(P<0.05),while the distribution between the children of different sexes and different lasting time of syncope(≤ 5 minutes vs> 5 minutes) was not significantly different(P>0.05).Different hemodynamic patterns were differentiated by differing syncope inducements,presymptoms,and complicated symptoms during and after syncope.Conclusion The tested girls were more prone when compared with the boys to have unexplained syncope,and the peak age was around twelve years old.The incidence of positive response of head-up tilt tests was also relatively higher for the girls.The distribution of hemodynamic patterns for different ages was different.For children with unexplained syncope,we should use head-up tilttests to distinguish the hemodynamic patterns in order to adopt rational therapeutic measures. | CHEN Li YANG Yuan-yuan WANG Cheng WANG Hong-wei TIAN Hong ZHANG Qing-you CHEN Jian-jun WANG Yu-li KANG Yi-long TANG Chao-shu DU Jun-bao | 2006 | Chinese Medical Journal2006,,24: | 23 |

| 7 | Corrosion Behavior of Mg-Zn-Y Alloy with Long-period Stacking Ordered Structures显示文摘Mg-Zn-Y alloys with long-period stacking ordered structures were prepared by an ingot casting method. The corrosion performance of Mg-Zn-Y alloys was studied by combining gas-collecting test, immersion test and electrochemical measurements in order to determine the corrosion rate and mechanism of the alloys. The results showed that the volume fraction of Mg(12)YZn phase increased and the shape of the Mg(12)YZn phase changed from discontinuous to continuous net-like with increasing Zn and Y content. The corrosion rate of the alloys greatly depended on the distribution and volume fraction of the Mg(12)YZn phase. Corrosion products appeared at the junction of Mg phase and Mg(12)YZn phase, indicating that the Mg(12)YZn phase accelerated galvanic corrosion of Mg matrix. During their preparation, some inhomogeneous residual stress is induced, resulting in competition between interface mismatching and surface energy and even leading to structure failure. This work presents a methodological study on the measurement of residual stress in a multi-layer semiconductor heterostructure. Scanning electron microscopy(SEM), micro-Raman spectroscopy(MRS), and transmission electron microscopy(TEM) were applied to measure the geometric parameters of the multilayer structure. The relationship between the Raman spectrum and the stress/strain on the [100] and [110] crystal orientations was determined to enable surface and crosssection residual stress analyses, respectively. | 13 | Plant Polyploidy: Origin, Evolution, and Its Influence on Crop Domestication显示文摘The prevalence and recurrence of polyploidization in plant species make it one of the most important evolutionary events in plants, and as a result, polyploidization is an extensively investigated research field. Due to the rapid development of sequencing technologies, there is increased evidence to support that polyploidization plays an important role in the diversification of plant species, evolution of genes, and the domestication of crops. Here, we reviewed the influence of polyploidization on various aspects of plant evolution, mainly focused on polyploid origin, characteristics, subsequent genome divergence, and its impact on gene function innovation and crop domestication. The occurrence of many independent polyploidization events in plants was found to be tightly associated with the timing of extreme climate events or natural disasters on earth, leading to mass extinction while possibly facilitating increased polyploidization. Following allo-polyploidization, a distinct phenomenon known as sub-genome dominance occurred during sub-genome evolution, which was found to be associated with the methylation of transposons. Extensive gene fractionations(lost) following polyploidization were reported in almost all paleo-polyploids, and the evolutionary fates of multi-copy genes, such as sub-/neo-functionalization, were further proposed to illustrate their underlying mechanisms. Moreover,polyploidization was found to significantly impact species diversification, with subsequent effects on crop domestication and the development of traits with agronomic importance. | 15 | Emerging role of Hippo pathway in gastric and othergastrointestinal cancers显示文摘More evidence has underscored the importance of Hippo signaling pathway in gastrointestinal tissue homeostasis, whereas its deregulation induces tumorigenesis. Yes-associated protein 1(YAP1) and its close paralog TAZ, transcriptional co-activator with a PDZbinding motif, function as key effectors negatively controlled by the Hippo pathway. YAP1/TAZ exerts oncogenic activities by transcriptional regulation via physical interaction with TEAD transcription factors. In various cancers, Hippo pathway cross-talks with pro- or anti-tumorigenic pathways such as GPCR, Wnt/β-catenin, Notch and TGF-β signaling and is deregulated by multiple factors including cell density/junction and micro RNAs. As YAP1 expression is significantly associated with poor prognosis of gastric and other gastrointestinal cancers, detailed delineation of Hippo regulation in tumorigenesis provides novel insight for therapeutic intervention. | 18 | Comparison of different methods of intestinal obstruction in a rat model显示文摘AIM:To investigate different methods of creating incomplete intestinal obstruction in a rat model and to compare their electrophysiologic,morphologic and histologic characteristics. METHODS:Rat ileum was partially obstructed by the respective application of:braided silk(penetrated the mesentery and surrounded intestine);half ligation (penetrated directly and ligated 1/2 cross-section of the intestine);wide pipe(6 mm in width,surrounded the intestine);narrow pipe(2 mm in width,surrounded the intestine).A control was also included(no obstruction). Various behavioral and electrophysiologic variables, as well as morphologic and immunohistochemical observations were recorded by blinded investigators at different time points(12,24,48,72 h),including daily general condition,ileal wet weight and circumference, macromorphous and micromorphous intestine,bowel movement capability in vivo and in vitro,slow wave and neural electrical activity,and the number of c-Kit positive interstitial cells of Cajal(ICC). RESULTS:Despite being of a similar general condition, these methods resulted in different levels of obstruction in each group compared with the control at different time points(12,24,48,72 h).However,these fields of the wide pipe rat showed significantly differences when compared with the other three obstructed groups at 12 to 72 h,including macroscopic and histological presentation,intestinal transit ratio and contractility,circumference and wet weight,amplitude and frequency of nerve electrical discharge and slow wave,and ICC numbers(all P<0.01).
