| 1 | Phase 1 clinical trial demonstrated that MUC1 positive metastatic seminal vesicle cancer can be effectively eradicated by modified Anti-MUC1 chimeric antigen receptor transduced T cells显示文摘Recent progress in chimeric antigen receptor-modified T-cell(CAR-T cell) technology in cancer therapy is extremely promising, especially in the treatment of patients with B-cell acute lymphoblastic leukemia. In contrast, due to the hostile immunosuppressive microenvironment of a solid tumor, CAR T-cell accessibility and survival continue to pose a considerable challenge, which leads to their limited therapeutic efficacy. In this study, we constructed two anti-MUC1 CAR-T cell lines. One set of CAR-T cells contained SM3 single chain variable fragment(sc Fv) sequence specifically targeting the MUC1 antigen and co-expressing interleukin(IL) 12(named SM3-CAR). The other CAR-T cell line carried the SM3 sc Fv sequence modified to improve its binding to MUC1 antigen(named p SM3-CAR) but did not co-express IL-12. When those two types of CAR-T cells were injected intratumorally into two independent metastatic lesions of the same MUC1+ seminal vesicle cancer patient as part of an interventional treatment strategy, the initial results indicated no side-effects of the MUC1 targeting CAR-T cell approach, and patient serum cytokines responses were positive. | 2 | Effect of Metal Additives on Performance of Low-Carbon Magnesia-Carbon Materials显示文摘In this paper,both oxidation and corrosion resistance of low-carbon magnesia-carbon materials containing 4.0wt% graphite with metallic Al and Mg-Al alloy powders as antioxidants were investigated.Meanwhile,the microstructures of samples corroded by slag were observed with optical microscope as well.The test results revealed the properties of oxidation and corrosion resistance of low-carbon magnesia-carbon materials could be improved obviously by adding metal Al powder and Mg-Al alloy powder.The rule of improving oxidation resistance was illegibility when metal Al powder and Mg-Al alloy powder were added together.It was harmful to corrosion resistance by mixed adding metal Al powder and Mg-Al alloy powder into the materials,at the same time,the corrosion resistance would decreased with the increasing of Mg-Al alloy content.The corrosion resistance of samples with 0.5wt% or 3.0wt% Mg-Al alloy was better. The oxidation resistance and corrosion resistance of materials with metal Al or Mg-Al alloy respectively were better than that with mixed metal Al and Mg-Al alloy. The results show adding 0.3wt% B4C can get the best oxidation resistance and adding 0.2wt% B4C can get the highest hot modulus of rupture. | 5 | Effects of genetic polymorphisms on methotrexate levels and toxicity in Chinese patients with acute lymphoblastic leukemia显示文摘Methotrexate(MTX)has an antitumor effect when used for the treatment of acute lymphoblastic leukemia(ALL).This study aims at evaluating the associations between 14 polymorphisms of six genes involved in MTX metabolism with serum MTX concentration and toxicity accompanying high-dose MTX.Polymorphisms in 183 Chinese patients with ALL were analyzed using TaqMan single nucleotide polymorphism genotyping assay.The serum MTX concentration was determined using homogeneous enzyme immunoassay.MTX-related toxicities were also evaluated.Renal toxicity was significantly associated with higher serum MTX concentrations at 24,48,and 72 hours,and MTX elimination delay(P=0.001,P<0.001,P<0.001,and P<0.001,respectively),whereas SLCO1B1 rs4149056 was associated with serum MTX concentrations at 48 and 72 hours,and MTX elimination delay in candidate polymorphisms(P=0.014,P=0.019,and P=0.007,respectively).SLC19A1 rs2838958 and rs3788200 were associated with serum MTX concentrations at 24 hours(P=0.016,P=0.043,respectively).MTRR rs1801394 was associated with serum MTX concentrations at 72 hours(P=0.045).Neutropenia was related to SLC19A1 rs4149056(odds ratio[OR]:3.172,95%confidence interval[CI]:1.310–7.681,P=0.011).Hepatotoxicity was associated with ABCC2 rs2273697(OR:3.494,95%CI:1.236–9.873,P=0.018)and MTRR rs1801394(OR:0.231,95%CI:0.084–0.632,P=0.004).Polymorphisms of SLCO1B1,SLC19A1,ABCC2,and MTRR genes help predict higher risk of increased MTX levels or MTX-related toxicities in adult ALL patients. | Qishan Hao Yang Song Qiuyun Fang Yani Lin Long Chen Xiaodan Wang Ping Zhang Zhe Wang Xiaoyuan Gong Kaiqi Liu Qinghua Li Zheng Tian Min Wang Jianxiang Wang Yingchang Mi | 2023 | Blood Science2023,5,1: | 0 |
| 6 | Gas hydrate accumulation associated with fluid escape structure in the western margin of South China Sea显示文摘Gas hydrates have been found in the western continental margin of South China Sea,which are revealed by widespread bottom simulating reflectors(BSRs)imaged from a three-dimensional(3D)seismic volume near the Guangle carbonate platform in the western South China Sea.Fluid-escape structures(faults and gas chimneys)are originated below BSR were distinguished.A comprehensive model in three-level structure was proposed to depict the gas hydrate accumulation in the study area.In Level 1,regional major faults and gas chimneys provide the first pathways of upward migration of gas near basement.In Level 2,pervasive polygonal faults in carbonate layer promote the migration of gas.In Level 3,gases sourced from near-basement accumulate within shallow sediment layers and form gas hydrate above the unit with faults once appropriate temperature and pressure occur.The gas hydrates in the study area are mainly in microbial origin,and their accumulation occurs only when fluid-escape structures align in all the three levels.The proposed model of the gas hydrate accumulation in western SCS margin provides new insights for further studies in this poorly studied area. | Taotao YANG Lin LI Yintao LU Kaiqi YU Zhili YANG Xuefeng WANG Wei LI | 2023 | Journal of Oceanology and Limnology2023,41,3: | 0 |
