| 1 | From Golden Rice to aSTARice:Bioengineering Astaxanthin Biosynthesis in Rice Endosperm显示文摘Carotenoids are important phytonutrients with antioxidant properties,and are widely used in foods and feedstuffs as Supplements.Astaxanthin,a red-colored ketocarotenoid,has strong antioxidant activity and thus can benefit human health.However,astaxanthin is not produced in most higher plants.Here we report the bioengineering of astaxanthin biosynthesis in rice endosperm by introducing four synthetic genes,sZmPSY1,sPaCrtl,sCrBKT,and sHpBHY,which encode the enzymes phytoene synthase,phytoene desaturase,β-carotene ketolase,and β-carotene hydroxylase,respectively.Transgneic overexpression of two (sZmPSY1 and sPaCrtl),three (sZmPSY1,sPaCrtl and sCrBKT),and all these four genes driven by rice endosperm-specific promoters established the Carotenoid/ketocarotenoid/astaxanthin biosynthetic pathways in the endosperm and thus resulted in various types of germplasm,from the yellow-grained β-caro- tene-enriched Golden Rice to orange-red-grained Canthaxanthin Rice and Astaxanthin Rice,respectively. | 2 | Vulnerability of Water Resources and Its Spatial Heterogeneity in Haihe River Basin, China显示文摘To manage water resources effectively, a multiscale assessment of the vulnerability of water resources on the basis of political boundaries and watersheds is necessary. This study addressed issues on the vulnerability of water resources and provided a multiscale comparison of spatial heterogeneity under a climate change background. Using improved quantitative evaluation methods of vulnerability, the Theil index and the Shannon-Weaver index, we evaluated the vulnerability of water resources and its spatial heterogeneity in the Haihe River Basin in four scales, namely, second-class water resource regions(Class ⅡWRRs), third-class water resource regions(Class Ⅲ WRRs), Province-Class ⅡWRRs, and Province-Class Ⅲ WRRs. Results show that vulnerability enhances from the north to south in the different scales, and shows obvious spatial heterogeneity instead of moving toward convergence in multiscale assessment results. Among the Class Ⅱ WRRs, the Tuhai-Majia River is the most vulnerable area, and the vulnerability of the Luanhe River is lower than that of the north of the Haihe River Basin, which in turn is lower than that of the south of the Haihe River Basin. In the scales of Class Ⅲ WRRs and Province-Class Ⅲ WRRs, the vulnerability shows obvious spatial heterogeneity and diversity measured by the Theil index and the Shannon-Weaver index. | 4 | Dissection of two quantitative trait loci for grain weight linked in repulsion on the long arm of chromosome 1 of rice(Oryza sativa L.)显示文摘Grain weight is a key determinant of grain yield in rice. Three sets of rice populations with overlapping segregating regions in isogenic backgrounds were established in the generations of BC2 F5, BC2 F6 and BC2 F7, derived from Zhenshan 97 and Milyang 46, and used for dissection of quantitative trait loci(QTL) for grain weight. Two QTL linked in repulsion phase on the long arm of chromosome 1 were separated. One was located between simple sequence repeat(SSR) markers RM11437 and RM11615, having a smaller additive effect with the enhancing allele from the maintainer line Zhenshan 97 and a partially dominant effect for increasing grain weight. The other was located between SSR markers RM11615 and RM11800, having a larger additive effect with the enhancing allele from the restorer line Milyang 46 and a partially dominant effect for increasing grain weight. When the two QTL segregated simultaneously, a residual additive effect with the enhancing allele from Milyang 46 and an over-dominance effect for increasing grain weight were detected. This suggests that dominant QTL linked in repulsion phase might play an important role in heterosis in rice. | 6 | Fermented mixed feed alters growth performance,carcass traits,meat quality and muscle fatty acid and amino acid profiles in finishing pigs显示文摘This study was conducted to investigate the effects of fermented mixed feed(FMF)on growth performance,carcass traits,meat quality,muscle amino acid and fatty acid composition and mRNA expression levels of genes related to lipid metabolism in finishing pigs.In the present study,144 finishing pigs(Duroc×Berkshire×Jiaxing Black)were randomly allocated to 3 dietary treatments with 4 replicate pens per group and 12 pigs per pen.The dietary treatments included a basal diet(CON),a basal diet+5%FMF and a basal diet+10%FMF.The experiment lasted 38 d after 4 d of acclimation.The results showed that 5%and 10%FMF significantly increased the average daily gain(ADG)of the females but not the males(P<0.05),but FMF supplementation showed no impact on carcass traits.Moreover,10%FMF supplementation increased the meat color45 min and meat color24 h values,while it decreased the shear force relative to CON(P<0.05).In addition,10%FMF significantly increased the contents of flavor amino acids(FAA),total essential AA(EAA),total non-EAA(NEAA)and total AA relative to CON(P<0.05).Furthermore,the diet supplemented with 10%FMF significantly increased the concentration of n-3 polyunsaturated fatty acids(PUFA),n-6 PUFA and total PUFA,and the PUFA to saturated fatty acids ratio(P<0.05),suggesting that FMF supplementation increased meat quality.Moreover,compared with the CON,10%FMF supplementation increased the mRNA expression of lipogenic genes,including CEBPα,PPARγ,SREBP1 and FABP4,and upregulated the expression of unsaturated fatty acid synthesis(ACAA1 and FADS2).Together,our results suggest that 10%FMF dietary supplementation improved the female pigs’growth performance,improved the meat quality and altered the profiles of muscle fatty acids and amino acids in finishing pigs.This study provides a reference for the production of high-quality pork. | Shiqi Liu Man Du Yuang Tu Wenjing You Wentao Chen Guoliang Liu Junyue Li Yizhen Wang Zeqing Lu Tenghao Wang Tizhong Shan | 2023 | Animal Nutrition2023,,1: | 4 |
| 7 | Lotus seedpod-inspired internal vascularized 3D printed scaffold for bone tissue repair显示文摘In the field of bone defect repair,3D printed scaffolds have the characteristics of personalized customization and accurate internal structure.However,how to construct a well-structured vascular network quickly and effectively inside the scaffold is essential for bone repair after transplantation.Herein,inspired by the unique biological structure of“lotus seedpod”,hydrogel microspheres encapsulating deferoxamine(DFO)liposomes were prepared through microfluidic technology as“lotus seeds”,and skillfully combined with a three-dimensional(3D)printed bioceramic scaffold with biomimetic“lotus”biological structure which can internally grow blood vessels.In this composite scaffold system,DFO was effectively released by 36%in the first 6 h,which was conducive to promote the growth of blood vessels inside the scaffold quickly.In the following 7 days,the release rate of DFO reached 69%,which was fundamental in the formation of blood vessels inside the scaffold as well as osteogenic differentiation of bone mesenchymal stem cells(BMSCs).It was confirmed that the composite scaffold could significantly promote the human umbilical vein endothelial cells(HUVECs)to form the vascular morphology within 6 h in vitro.In vivo,the composite scaffold increased the expression of vascularization and osteogenic related proteins Hif1-α,CD31,OPN,and OCN in the rat femoral defect model,significantly cutting down the time of bone repair.To sum up,this“lotus seedpod”inspired porous bioceramic 3D printed scaffold with internal vascularization functionality has broad application prospects in the future. | Xiaoyu Han Mingjie Sun Bo Chen Qimanguli Saiding Junyue Zhang Hongliang Song Lianfu Deng Peng Wang Weiming Gong Wenguo Cui | 2021 | Bioactive Materials2021,6,6: | 4 |

| 10 | Minor-effect QTL for heading date detected in crosses between indica rice cultivar Teqing and near isogenic lines of IR24显示文摘Identification of quantitative trait loci(QTL) having small effects on heading date(HD) is important for fine-tuning flowering time in rice(Oryza sativa L.). In this study, minor-effect QTL for HD were identified using five segregating rice populations, including a recombinant inbred line population derived from crosses between indica cultivar Teqing and near isogenic lines of IR24,and four populations derived from residual heterozygotes identified in the original population.HD data from these populations were obtained in multiple years or at two locations with different photoperiods. A total of 11 QTL were detected; they had small additive effects ranging from 0.21 to1.63 days. The QTL were all detected in different populations, locations and/or years, having consistent allelic effects across experiments and a stable magnitude across years at the same location. These QTL, and other minor-effect QTL that have been cloned or fine-mapped, generally do not have strong photoperiod sensitivity, and thus can be used in a wide range of ecogeographical conditions. Seven of the 11 QTL were different from those that have been cloned or fine-mapped, providing new candidates for gene cloning and marker-assisted breeding. Allelic effects of QTL corresponding to those that had been cloned or fine-mapped, were much smaller in this study than previously reported. | 11 | Numerical model to quantify biogenic volatile organic compound emissions:The Pearl River Delta region as a case study显示文摘This article compiles the actual knowledge of the biogenic volatile organic compound(BVOC) emissions estimated using model methods in the Pearl River Delta(PRD) region, one of the most developed regions in China. The developed history of BVOC emission models is presented briefly and three typical emission models are introduced and compared. The results from local studies related to BVOC emissions have been summarized. Based on this analysis, it is recommended that local researchers conduct BVOC emission studies systematically, from the assessment of model inputs, to compiling regional emission inventories to quantifying the uncertainties and evaluating the model results. Beyond that,more basic researches should be conducted in the future to close the gaps in knowledge on BVOC emission mechanisms, to develop the emission models and to refine the inventory results. | 14 | Spatiotemporal Mapping of Salt Marshes in the Intertidal Zone of China during 1985-2019显示文摘This study mapped the areal extent,identified the species composition,and analyzed the changes of salt marshes in the intertidal zone of China during the period 1985–2019.With the aid of the cloud platform of the Google Earth Engine,we selected Landsat 5/8 and Sentinel-2 images and used the support vector machine classification method to extract salt marsh information for the years of 1985,1990,1995,2000,2005,2010,2015,and 2019.Seven major species of salt marshes:Phragmites australis,Suaeda spp.,Spartina alterniflora,Scirpus mariqueter,Tamarix chinensis,Cyperus malaccensis,and Sesuvium portulacastrum were identified.Our results showed that salt marshes are mainly distributed in Liaoning,Shandong,Jiangsu,Shanghai,and Zhejiang,with varying patterns of shrinking,expansion,or wavering in different places.The distribution of salt marshes has declined considerably from 151,324 ha in 1985 to 115,397 ha in 2019,a drop of 23.7%.During the same period,the area of native species has dropped 95.4%from 77,741 ha to 3,563 ha for Suaeda spp.and 45.1%from 60,511 ha to 33,193 ha for P.australis;on the contrary,the area of exotic species,S.alterniflora,has exhibited a sharp rise from just 99 ha to 67,527 ha.For the past 35 years,the driving factors causing salt marsh changes are mainly land reclamation,variations in water and sand fluxes,and interspecific competition and succession of salt marsh vegetation.These results provide fundamental reference information and could form the scientific basis for formulating policies for the conservation and utilization of salt marsh resources in China. | Guangwei Chen Runjie Jin Zhanjiang Ye Qi Li Jiali Gu Min Luo Yongming Luo George Christakos James Morris Junyu He Dan Li Hengwei Wang Li Song Qiuxuan Wang Jiaping Wu | 2022 | Journal of Remote Sensing2022,,1: | 2 |

In the present study, we investigated the effects of ZIKV-encoded proteins in the activation of p53 signaling pathway and found that, among the ten viral proteins,the nonstructural protein 5(NS5) of ZIKV most significantly activated the transcription of p53 target genes. Using the immunoprecipitation-coupled mass spectrometry approach, we identified that ZIKV-NS5 interacted with p53 protein. The NS5-p53 interaction was further confirmed by co-immunoprecipitation and GST pull-down assays. In addition, the MTase domain of NS5 and the C-terminal domain of p53 were mapped to be responsible for the interaction between these two proteins. We further showed that ZIKV-NS5 was colocalized with p53 and increased its protein level in the nuclei and able to prolong the half-life of p53. Furthermore, lentivirus-mediated expression of ZIKV-NS5 in hNPCs led to an apparent cell death phenotype.
