| 1 | The PI3K/Akt/GSK-3β/ROS/eIF2B pathway promotes breast cancer growth and metastasis via suppression of NK cell cytotoxicity and tumor cell susceptibility显示文摘Objective: To examine the effect of pSer9-GSK-3β on breast cancer and to determine whether the underlying metabolic and immunological mechanism is associated with ROS/eIF2B and natural killer(NK) cells.Methods: We employed TWS119 to inactivate GSK-3β by phosphorylating Ser9 and explored its effect on breast cancer and NK cells. The expression of GSK-3β, natural killer group 2 member D(NKG2D) ligands, eIF2B was quantified by PCR and Western blot. We measured intracellular reactive oxygen species(ROS) and mitochondrial ROS using DCFH-DA and MitoSOX^(TM) probe,respectively, and conducted quantitative analysis of cellular respiration on 4T1 cells with mitochondrial respiratory chain complex Ⅰ/Ⅲ kits.Results: Our investigation revealed that TWS119 downregulated NKG2D ligands(H60 a and Rae1), suppressed the cytotoxicity of NK cells, and promoted the migration of 4T1 murine breast cancer cells. Nevertheless, LY290042, which attenuates p-GSK-3β formation by inhibiting the PI3K/Akt pathway, reversed these effects. We also found that higher expression of p Ser9-GSK-3β induced higher levels of ROS, and observed that abnormality of mitochondrial respiratory chain complex Ⅰ/Ⅲ function induced the dysfunction of GSK-3β-induced electron transport chain, naturally disturbing the ROS level. In addition, the expression of NOX3 and NOX4 was significantly up-regulated, which affected the generation of ROS and associated with the metastasis of breast cancer. Furthermore, we found that the expression of pSer535-eIF2B promoted the expression of NKG2D ligands(Mult-1 and Rae1) following by expression of pSer9-GSK-3β and generation of ROS.Conclusions: The PI3K/Akt/GSK-3β/ROS/eIF2B pathway could regulate NK cell activity and sensitivity of tumor cells to NK cells,which resulted in breast cancer growth and lung metastasis. | 4 | Two-photon-excited near-infrared emissive carbon dotsas multifunctional agents for fluorescence imaging andphotothermal therapy显示文摘C dots (CDs) have shown great potential in bioimaging and phototherapy.However, it is challenging to manipulate their fluorescent properties andtherapeutic efficacy to satisfy the requirements for clinic applications. In thisstudy, we prepared S, Se-codoped CDs via a hydrothermal method anddemonstrated that the doping resulted in excitation wavelength-independentnear-infrared (NIR) emissions of the CDs, with peaks at 731 and 820 nm.Significantly, the CDs exhibited a photothermal conversion efficiency of -58.2%,which is the highest reported value for C nanostructures and is comparable tothat of Au nanostructures. Moreover, the CDs had a large two-photon absorptioncross section (-30,045 GM), which allowed NIR emissions and the photothermalconversion of the CDs through the two-photon excitation (TPE) mechanism. | 5 | Heavy metal(loid) pollution in mine wastes of a Carlin-type gold mine in southwestern Guizhou, China and its environmental impacts显示文摘Mercury pollution resulting from artisanal gold mining is a serious environmental problem in many developing countries. In this study, we analyzed heavy metal(loid) contamination in mine wastes from a Carlin-type gold mine in southwestern Guizhou, China. Highly elevated levels of As, Hg, Tl, Sb, and Cd—up to 5844, 28,29, 581 and 3.0 mg/kg, respectively—were observed in the mine wastes, but no significant accumulation of Cr, Ni, Cu,Zn, and Pb was found. The smelting process of gold ores had significant impacts on the enrichment of As, Tl, and Hg in the mine wastes. Due to the significant metal(loid)pollution in this gold mine, the ecological risks are classified as ‘‘very high.'' Hg and As are the major pollutants with a relative contribution of 55.9 % and 24.2 % to the risk index, respectively. Phytoremediation could be used to reduce heavy metal contamination and recycle the gold simultaneously. | 11 | Farmland mercury contamination in the vicinity of an organic chemical factory in Guizhou,China显示文摘This study assesses the level of contamination of Hg in farmland soils along the irrigating channel down-stream from Guizhou Organic Chemical Factory (GOCF),where metallic mercury is used as a catalyst to produce acetic acid. The total input of mercury into the environment,as announced by GOCF,is 140 t in the past 30 years (1971-2000). Sampling sites were chosen based on the distance from the source of pollution—the chemical factory. A total of 39 samples were collected from the study area and analyzed for total mercury concentrations and methyl mercury concentrations. The characteristics of vertical and horizontal distributions of total mercury and methyl mer-cury in the study area (farmland) are described in this paper. Much attention was paid to the transformation of inor-ganic Hg into organic mercury species in soils as well. The results showed that the farmland has been heavily con-taminated by Hg. Land cultivation activity,land utilization style and distance from the pollution source could be the dominant factors controlling the distribution of THg and MeHg. One problem identified is the asymmetric city connectivity, partially due to data availability. We present a data-driven approach based on location and toponym(place name) extracted from social media data, to assess the asymmetric connectivity between cities. The assumption is that a higher frequency of occurrences of the name of city i in posts located in city j would imply that the city i is more influential than other cities upon city j. In addition, we’ve developed a group of measurements such as the relatedness index, impact index, link strength index, dependence index, and structure similar index to characterize such interactions. This framework of connectivity measurements can also be used to support smart planning taking into account the evolving interplay among cities. The steady-state benefits of static instability are illustrated, while the corresponding control challenge is described both by the characteristic lag of airframe and the increment of necessary control usage. Control limitation led by unstable zero-pole pair is analyzed for preliminary design and evaluation. Linear control strategy is examined wherein two and three loop acceleration autopilots with different control usages are developed using an optimal control approach combined with frequency domain constraint. The weights selection and relation with system performance are detailed. Then the nonlinear backstepping recursive method is detailed to determine how well it is able to follow command and its engineering feasibility. The results show that a static unstable missile is controllable, while the actuator bandwidth is the crucial limited factor. | 17 | Wire-in-Wire TiO_(2)/C Nanofibers Free-Standing Anodes for Li-Ion and K-Ion Batteries with Long Cycling Stability and High Capacity显示文摘Wearable and portable mobile phones play a critical role in the market, and one of the key technologies is the flexible electrode with high specific capacity and excellent mechanical flexibility. Herein, a wire-in-wire TiO_(2)/C nanofibers (TiO_(2) ww/CN) film is synthesized via electrospinning with selenium as a structural inducer. The interconnected carbon network and unique wire- in-wire nanostructure cannot only improve electronic conductivity and induce effective charge transports, but also bring a superior mechanic flexibility. Ulti-mately, TiO_(2) ww/CN film shows outstanding electrochemical performance as free-standing electrodes in Li/K ion batteries. It shows a discharge capacity as high as 303 mAh g^(−1) at 5 A g^(−1) after 6000 cycles in Li half-cells, and the unique structure is well-reserved after long-term cycling. Moreover, even TiO_(2) has a large diffusion barrier of K^(+), TiO_(2) ww/CN film demonstrates excellent perfor-mance (259 mAh g^(−1) at 0.05 A g^(−1) after 1000 cycles) in K half-cells owing to extraordinary pseudocapacitive contribution. The Li/K full cells consisted of TiO_(2) ww/CN film anode and LiFePO_(4)/Perylene-3,4,9,10-tetracarboxylic dianhydride cathode possess outstanding cycling stability and demonstrate practical application from lighting at least 19 LEDs.
