| 2 | Circular dichroism-like response of terahertz wave caused by phase manipulation via all-silicon metasurface显示文摘Chiral metasurfaces based on asymmetric meta-atoms have achieved artificial circular dichroism(CD),spin-dependent wavefront control,near-field imaging,and other spin-related electromagnetic control.In this paper,we propose and experimentally verify a scheme for achieving high-efficiency chiral response similar to CD of terahertz(THz)wave via phase manipulation.By introducing the geometric phase and dynamic phase in an all-silicon metasurface,the spin-decoupled terahertz transmission is obtained.The giant circular dichroism-like effect in the transmission spectrum is observed by using a random phase distribution for one of the circular polarization components.More importantly,the effect can be adjusted when we change the area of the metasurface illuminated by an incident terahertz beam.In addition,we also demonstrate the spin-dependent arbitrary wavefront control of the transmitted terahertz wave,in which one of the circularly polarized components is scattered,while the other forms a focused vortex beam.Simulated and experimental results show that this method provides a new idea for spin selective control of THz waves. | Jie Li Chenglong Zheng Guocui Wang Jitao Li Hongliang Zhao Yue Yang Zhang Zhang Maosheng Yang Liang Wu Jining Li Yating Zhang Yan Zhang Jianquan Yao | 2021 | Photonics Research2021,9,4: | 3 |
| 3 | Expression of P-gp and miRNA-21 in Colorectal Cancer Tissues and the Clinical Significance显示文摘[Objectives]To study the expression of P-gp and miRNA-21 in colorectal cancer tissues and the relationship between the expression of P-gp and clinicopathological features,and to analyze the correlation between the expression of miRNA-21 and P-gp.[Methods]Immunohistochemical SP method was used to detect the expression level of P-gp in the tissues of 40 cases of colorectal cancer and the normal mucosal tissues 5 cm away from the corresponding cancer tissues,and the expression level of miRNA-21 in the paraffin tissues of the 40 cases was detected by RT-qPCR.[Results]The positive expression of P-gp was localized in the cell membrane,and the expression in colorectal cancer tissues was significantly higher than that in the corresponding adjacent tissues(P<0.05);the positive expression of P-gp was closely related to the degree of tumor differentiation and the depth of invasion(P<0.05),and the comparison results of P-gp expression in the differentiation groups of different cancer tissues showed that the expression level of P-gp in the moderately and poorly differentiated groups was significantly higher than that in the highly differentiated group(P<0.05),and the expression level of the moderately and poorly differentiated groups was similar(P>0.05).The expression of miRNA-21 in all cancer tissues was higher than that in the corresponding adjacent tissues(P<0.05),and the expression levels of miRNA-21 in differentiation groups of different cancer tissues were 1.286,1.445 and 1.992 times of those in the corresponding adjacent tissues,respectively;the comparison results of miRNA-21 between different differentiation groups showed that the expression level of miRNA-21 in the poorly differentiated group was significantly higher than that in the highly differentiated group and the moderately differentiated group(P<0.05),while the expression level of miRNA-21 in the highly differentiated group and the moderately differentiated group was similar(P>0.05).Spearman correlation analysis showed that there was a positive correlation on the expression between the two in colorectal cancer tissues(r=0.423,P<0.05).[Conclusions]The expression levels of miRNA-21 and P-gp are closely related to the differentiation degree of clinical colorectal cancer,the lower the differentiation degree,the higher the expression level of the two. | Xiaoli YAN Peng YAN Yanfang FAN Jing XUE Mingjuan WANG Jining ZHENG | 2020 | Medicinal Plant2020,11,6: | 1 |
| 4 | Expression and Significance of MicroRNA-21 and P-gp in Colon Cancer Cells显示文摘[Objectives]To investigate the expression and significance of miR-21 and P-gp in colon cancer cell line HCT-15.[Methods]RT-qPCR was used to detect the miR-21 and ABCB1 mRNA expression level of P-gp coding genes in normal colon mucosal cell NCM460 and colon cancer cell HCT-15,and Western Blot was used to detect the expression of P-gp in these two cell lines.[Results]The results of RT-qPCR showed that the expression level of miR-21 and ABCB1 mRNA in colon cancer cell HCT-15 was significantly higher than that in normal colon cell NCM460,and the difference was statistically significant(P<0.05).Western Blot results showed that the expression level of P-gp in HCT-15 was significantly higher than that in NCM460,and the difference was statistically significant(P<0.05).[Conclusions]The expression level of miR-21 and ABCB1 mRNA/P-gp in colon cancer cell HCT-15 was higher than that in normal colonic mucosal cell NCM460,and the relationship between miR-21 and multidrug resistance in colon cancer might be related to P-gp. | Xiaoli YAN Yanfang FAN Peng YAN Jing XUE Zhenhong LIU Jining ZHENG | 2020 | Medicinal Plant2020,11,2: | 1 |

| 6 | Resistin protein expression in abdominal subcutaneous adipose tissue and placenta in pregnant women显示文摘Objective: To explore the expression of resistin protein in abdominal subcutaneous adipose tissue and placental tissue in pregnant women and the relationship between pregnant physiological insulin resistance(IR) and gestational diabetes mellitus(GDM). Methods: The expression of resistin protein in abdominal subcutaneous adipose tissue of pregnant and nonpregnant women and placental tissue was measured using western blotting method. Fasting serum glucose concentration was measured by glucose oxidase assay. Systolic blood pressure (SBP) and diastolic blood pressure (DBP) were measured. Results: Resistin protein expression in placental tissue(67905±8441)(arbitrary OD units)was much higher than that in abdominal subcutaneous fat tissue in pregnant women (40718±3818) (P<0.01)and nonpregnant women (38288±2084) (P<0.01) respectively, and there was no significant difference in abdominal subcutaneous fat tissue among pregnant and nonpregnant women. Conclusion: Resistin protein expression in placental tissue is much higher than that in abdominal subcutaneous fat tissue in pregnant and nonpregnant women. Resistin protein is secreted from human placental tissue. | 8 | miR-15a-5p Regulates Oxaliplatin Resistance in Colorectal Cancer through the Wnt/β-catenin Pathway显示文摘[Objectives]To explore the effects and mechanism of miR-15a-5p on oxaliplatin resistance in colorectal cancer HCT116/L cells.[Methods]The expression of miR-15a-5p in colorectal cancer sensitive cells HCT116 and resistant cells HCT116/L was detected by RT-qPCR method;the effect of oxaliplatin on the proliferation of HCT116 and HCT116/L cells was detected by MTT,and its IC_(50) and drug resistance fold of HCT116/L cells were calculated;the expressions of Wnt3a,β-Catenin and P-gp in HCT116 and HCT116/L cells were detected by Western Blot method.HCT-116/L cells were divided into 5 groups:blank control group HCT116/L,control group 1 transfected with miR-15a-5p mimics NC,experimental group 1 transfected with miR-15a-5p mimics,control group 2 transfected with miR-15a-5p inhibitor NC,and experimental group 2 transfected with miR-15a-5p inhibitor.The expression of miR-15a-5p in each group was detected by RT-qPCR method and the transfection efficiency was detected.The effect of different concentrations of oxaliplatin on the proliferation of cells in each group after transfection was detected by MTT and the half inhibitory concentration(50%inhibiting concentration,IC_(50))was calculated.The expressions of Wnt3a,β-catenin and P-gp in each group after transfection were detected by Western Blot method,and the expressions of Wnt3a,β-catenin and MDR1 mRNA in each group after transfection were detected by RT-qPCR method.[Results](i)RT-qPCR results showed that the expression of miR-15a-5p in HCT116/L cells was(0.16±0.05)significantly lower than that in HCT116 cells(P<0.05).(ii)The IC50 of oxaliplatin for HCT-116 cells and HCT116/L cells detected by MTT method were(13.51±2.62)and(103.08±12.29)μg/mL,respectively.The drug resistance index of HCT116/L was 7.63.(iii)Western Blot results showed that the expressions of Wnt3a,β-catenin and P-gp in HCT116/L cells were significantly higher than those in HCT-116 cells(P<0.05).(iv)After successful transfection,the IC50 of miR-15a-5p mimics group to oxaliplatin decreased to(40.78±2.47)μg/mL by MTT method,and its sensitivity to the drug was significantly improved compared with the control group.Western Blot results showed that the relative expressions of P-gp,Wnt3a andβ-catenin were significantly down-regulated(all P<0.05);RT-qPCR results showed that the relative expressions of MDR1,Wnt3a andβ-catenin mRNA were significantly down-regulated(all P<0.05).(v)After successful transfection,the expression of miR-15a-5p in the miR-15a-5p inhibitor transfection group was(0.38±0.04);MTT results showed that its IC50 for oxaliplatin was up-regulated to(132.77±7.97)μg/mL,and its sensitivity to chemotherapy drugs was significantly lower than that of the control group;Western Blot results showed that the relative expressions of P-gp,Wnt3a andβ-catenin were significantly up-regulated(all P<0.05);RT-qPCR results showed that the relative expressions of MDR1,Wnt3a andβ-catenin mRNA were significantly up-regulated(all P<0.05).[Conclusions]Up-regulation of miR-15a-5p expression can reverse the resistance of HCT116/L cell line to oxaliplatin.Up-regulation or down-regulation of miR-15a-5p will affect the expression of P-gp and Wnt/β-catenin signaling pathway-related proteins,suggesting that the mechanism of miR-15a-5p reversal of drug resistance may be related to the inhibition of Wnt/β-catenin pathway,thereby down-regulating the expression of P-gp. | Yanjie HUANG Tong XU Luwen WEN Qi FENG Jining ZHENG | 2022 | Medicinal Plant2022,13,3: | 0 |
