Synthesis of novel nanomaterials and their application in efficient removal of radionuclides显示文摘With the development of nuclear energy, large amounts of radionuclides are inevitably released into the natural environment. It is necessary to eliminate radionuclides from wastewater for the protection of environment. Nanomaterials have been considered as the potential candidates for the effective and selective removal of radionuclides from aqueous solutions under complicated conditions because of their high specific surface area, large amounts of binding sites, abundant functional groups, pore-size controllable and easily surface modification. This review mainly summarized the recent studies for the synthesis, fabrication and surface modification of novel nanomaterials and their applications in the efficient elimination and solidification of radionuclides, and discussed the interaction mechanisms from batch experiments, spectroscopy analysis and theoretical calculations. The sorption capacities with other materials, advantages and disadvantages of different nanomaterials are compared, and at last the perspective of the novel nanomaterials is summarized. | Xiangxue Wang Long Chen Lin Wang Qiaohui Fan Duoqiang Pan Jiaxing Li Fangting Chi Yi Xie Shujun Yu Chengliang Xiao Feng Luo Jun Wang Xiaolin Wang Changlun Chen Wangsuo Wu Weiqun Shi Shuao Wang Xiangke Wang | 2019 | Science China Chemistry2019,62,8: | 20
Chondroitin sulfate-mediated albumin corona nanoparticles for the treatment of breast cancer显示文摘Chondroitin sulfate-mediated albumin corona nanoparticles were readily prepared without any chemical reaction, and their active tumor targeting and therapeutic effects were examined.Negatively charged chondroitin sulfate(CS)and positively charged doxorubicin(DOX)self-assembled into nanoparticles(CS-DOX-NPs)via electrostatic interactions.Bovine serum albumin(BSA)was then adsorbed on the surface of CS-DOX-NPs to form albumin corona nanoparticles(BC-DOX-NPs)protected from endogenous proteins.Due to the dual effect of BSA and CS,BC-DOX-NPs interacted with the gp60,SPARC and CD44 receptors on tumor cells, facilitating their rapid and efficient transcytosis and improving their accumulation and uptake within tumor tissues. The simultaneous presence of BSA and CS also allowed BC-DOX-NPs to target CD44 efficiently, leading to greater cellular uptake and cytotoxicity against 4 T1 cells than CS-DOX-NPs or free DOX.Intravenous injection of BCDOX-NPs into orthotopic 4 T1 tumor-bearing mice led to greater drug accumulation at the tumor site than with CS-DOX-NPs or free DOX,resulting in significant inhibition of tumor growth and lower exposure of major organs to the drug. | Tiantian Tan Qin Yang Dan Chen Juan Zhao Ling Xiang Jiaxing Feng Xu Song Yao Fu Tao Gong | 2021 | Asian Journal of Pharmaceutical Sciences2021,16,4: | 3
Development of silicon-based microelectrode array显示文摘This paper introduces in details a kind of silicon-based microelectrode array. MEMS (micro-electromechanical system) technology is used in the fabrication of the microelectrode array, which is designed to perform signal recording and electrical stimulation for nerves in neural engineering. A simple fabrication process is developed. An improved model of microelectrodes is brought forward and successfully validated by the excellent match between circuit simulations and electrical measurements, including both magnitude and phase of microelectrode impedance. Compared with the simple one that is usually used, the improved model is believed to be an advance and more accurate. This modeling helps to improve the design of microelectrodes and understand the behavior of interface between electrode and cell. Furthermore, the microelectrode is proved to be a feasible tool for researches in neural engineering by successfully recording neural activities of sciatic nerve of a bullfrog. | LIN JianHui1,2,3, WU XiaoMing1,2,3,4, HUANG PengSheng1,2, FENG Lei5, REN TianLing1,2 & LIU LiTian1,2 1 Institute of Microelectronics, Tsinghua University, Beijing 100084, China 2 Tsinghua National Laboratory for Information Science and Technology, Beijing 100084, China 3 Yangtze Delta Region Institute of Tsinghua University, Jiaxing 314050, China 4 State Key Laboratory of Transducer Technology, Chinese Academy of Sciences, Shanghai 200050 China 5 Department of Biomedical Engineering, Tsinghua University, Beijing 100084, China | 2009 | Science China(Technological Sciences)2009,52,8: | 3
Arctic sea ice thickness variations from CryoSat-2 satellite altimetry data显示文摘Arctic sea ice plays an important role in Earth's climate and environmental system.Sea ice thickness is one of the most important sea ice parameters.Accurately obtaining the sea ice thickness and its changes has great significance to Arctic and global change research.Satellite altimeters can be used to derive long-term and large-scale changes in sea ice thickness. The leads detection is vital in sea ice thickness estimation by using satellite altimetry.Different leads detection methods are compared with remote sensing images, and results show that the detection method that uses waveform parameters can obtain improved results. The model for the conversion of freeboard to thickness is optimized by considering the incomplete penetration of snow for radar altimeters. We derive the estimates of the Arctic sea ice thickness for November 2010 to December 2019 by using the CryoSat-2 altimetry data. The sea ice thickness from the IceBridge and draft data from the upward-looking sonar are used to validate our thickness results.Validations show that the accuracy of our thickness estimates is within 0.2 m.Variations in the Arctic sea ice thickness are analyzed using the PIOMAS model and air and sea surface temperatures.A sharp increase in sea ice thickness is found in 2014. | Feng XIAO Shengkai ZHANG Jiaxing LI Tong GENG Yue XUAN Fei LI | 2021 | Science China Earth Sciences2021,64,7: | 2
Targeted delivery of celastrol to glomerular endothelium and podocytes for chronic kidney disease treatment显示文摘The etiology of chronic kidney disease(CKD)is complex and diverse, which could be briefly categorized to glomerular-or tubularoriginated. However, the final outcomes of CKD are mainly glomerular sclerosis, endothelial dysfunction and injury, and chronic inflammation. Thus, targeted delivery of drugs to the glomeruli in order to ameliorate glomerular endothelial damage may help alleviate CKD and help enrich our knowledge. The herb tripterygium wilfordii shows therapeutic effect on kidney disease, and celastrol(CLT)is one of its active ingredients but with strong toxicity. Therefore, based on the unique structure and pathological characteristics of the glomerulus,we designed a targeted delivery system named peptides coupled CLT-phospholipid lipid nanoparticles(PC-PLNs)to efficiently deliver CLT to damaged endothelial cells and podocytes in the glomerulus for CKD treatment and research.PC-PLNs could effectively inhibit inflammation, reduce endothelial damage, alleviate CKD severity, and reduce the toxicity of CLT.We also studied the mechanism of CLT in the treatment of nephropathy and found that CLT can increase the level of NO by increasing eNOS while inhibiting the expression of VCAM-1,thus provides an anti-inflammatory effect. Therefore, our study not only offered an efficient CKD drug formulation for further development, but also provided new medical knowledge about CKD. | Qingsi Wu Jiading Wang Yuanfang Wang Ling Xiang Yulu Tan Jiaxing Feng Zhirong Zhang Ling Zhang | 2022 | Nano Research2022,15,4: | 1

NudCL2 is an autophagy receptor that mediates selective autophagic degradation of CP110 at mother centrioles to promote ciliogenesis显示文摘Primary cilia extending from mother centrioles are essential for vertebrate development and homeostasis maintenance.Centriolar coiled-coil protein 110(CP110)has been reported to suppress ciliogenesis Initiation by capping the distal ends of mother centrioles. However, the mechanism underlying the specific degradation of mother centriole-capping CP110 to promote cilia initiation remains unknown. Here,we find that autophagy is crucial for CP110 degradation at mother centrioles after serum starvation in MEF cells. We further identify NudC-like protein 2(NudCL2)as a novel selective autophagy receptor at mother centrioles, which contains an LC3-interacting region(UR)motif mediating the association of CP110 and the autophagosome marker LC3.Knockout of NudCL2 induces defects in the removal of CP110 from mother centrioles and ciliogenesis, which are rescued by wild-type NudCL2 but not its UR motif mutant.Knockdown of CP110 significantly attenuates ciliogenesis defects in NudCL2-deficient cells. In addition,NudCL2 morphants exhibit ciliation-related phenotypes in zebrafish, which are reversed by wild-type NudCL2,but not its UR motif mutant.Importantly,CP110 depletion significantly reverses these ciliary phenotypes in NudCL2 morphants.Taken together, our data suggest that NudCL2 functions as an autophagy receptor mediating the selective degradation of mother centriole-capping CP110 to promote ciliogenesis, which is indispensable for embryo development in vertebrates. | Min Liu Wen Zhang Min Li Jiaxing Feng Wenjun Kuang Xiying Chen Feng Yang Qiang Sun Zhangqi Xu Jianfeng Hua Chunxia Yang Wei Liu Qiang Shu Yuehong Yang Tianhua Zhou Shanshan Xie | 2021 | Cell Research2021,31,11: | 1

Albumin-based multidrug delivery system enriched in Golgi apparatus against metastatic breast cancer显示文摘Breast cancer and metastasis remain great challenges in clinical therapy.Compared with monotherapy, combination therapy, especially mediated by nanomedicine delivery strategy, significantly improves the therapeutic efficacy and reduces undesired toxicity.Cyclooxygenase-2(COX-2)inhibitors are widely used for adjuvant chemotherapy because COX-2 is overexpressed in virtually all cancer cell lines to regulate tumor progression by catalyzing prostaglandin E2(PGE2)synthesis.This drug combination strategy is still required to be improved due to some unsatisfactory clinical trial results.Intricate processes of tumor growth and metastasis are orchestrated by multiple proteins in addition to COX-2,which are modified and transported by Golgi apparatus.Hence, disrupting the structure and function of Golgi apparatus can inhibit the secretion of tumor-related proteins and further suppress carcinoma progression and metastasis.Since COX-2 is also enriched within Golgi apparatus in tumor cells,COX-2 inhibitors and Golgi disrupting agents can be co-delivered to Golgi apparatus to maximize the synergy. In this work,we developed a human serum albumin(HSA)nanoparticle encapsulating pirarubicin(THP),retinoic acid(RA),and indomethacin(IMC),called TIR-HSA,which was observed to be localized in Golgi complex of 4T1 cells.Owing to the synergistic effect of these three drugs,TIR-HSA inhibited the proliferation, migration, and invasion of tumor cells, enhanced the apoptotic rate, and improved the immunosuppressive tumor microenvironment, which remarkably regressed the tumor growth and metastasis and prolonged the survival period of 4T1-bearing mice. | Chenqi Guo Xiong Peng Ting Zhao Jiaxing Feng Zhaofei Guo Mengying Wu Rongping Zhang Xun Sun Yuan Huang Zhirong Zhang Tao Gong | 2023 | Nano Research2023,16,4: | 0
AN ADVANCED DUAL-FREQUENCY POSITIONING ALGORITHM FOR SATELLITE NAVIGATION RECEIVERS显示文摘With the development of the Ground Positioning System (GPS) modernization and the expectable implementation of Galileo, people pay more and more attention to civil applications on multi-frequency signals. This paper proposes a new and advanced positioning algorithm for the dual-frequency satellite navigation receivers, concerning the various influences of all the ranging error sources and taking advantage of the Klobuchar single-frequency ionospheric model. The paper also presents positioning precision provided by the new algorithm. Theoretical analysis and experimental results show that, the new dual-frequency positioning algorithm can achieve higher positioning accu- racy than the single-frequency positioning algorithm and the traditional dual-frequency positioning algorithm. | Liu Jiaxing Lu Mingquan Feng Zhenming | 2009 | Journal of Electronics(China)2009,26,4: | 0
RUNX1-205, a novel splice variant of the human RUNX1 gene, has blockage effect on mesoderm-hemogenesis transition and promotion effect during the late stage of hematopoiesis显示文摘Runt-related transcription factor 1(RUNX1)is required for definitive hematopoiesis;however, the functions of most human RUNX1 isoforms are unclear. In particular, the effects of RUNX1-205(a novel splice variant that lacks exon 6 in comparison with RUNX1b)on human hematopoiesis are not clear. In this study,a human embryonic stem cell(hESC)line with inducible RUNX1-205 overexpression was established.Analyses of these cells revealed that induction of RUNX1-205 overexpression at early stage did not influence the induction of mesoderm but blocked the emergence of CD34+cells,and the production of hematopoietic stem/progenitor cells was significantly reduced. In addition, the expression of hematopoiesis-related factors was downregulated. However, these effects were abolished when RUNX1-205 overexpression was induced after Day 6 in co-cultures of hESCs and AGM-S3 cells, indicating that the inhibitory effect occurred prior to generation of hemogenic endothelial cells, while the promotive effect could be observed during the late stage of hematopoiesis. This is very similar to that of RUNX1b.Interestingly,the mRNA expression profile of RUNX1-205 during hematopoiesis was distinct from that of RUNX1b,and the protein stability of RUNX1-205 was much higher than that of RUNX1b.Thus,the function of RUNX1-205 in normal and diseased models should be further explored. | Wencui Sun Jiahui Zeng Jing Chang Yuan Xue Yonggang Zhang Xu Pan Ya Zhou Mowen Lai Guohui Bian Qiongxiu Zhou Jiaxing Liu Bo Chen Feng Ma | 2020 | Journal of Molecular Cell Biology2020,12,5: | 0

Structural repurposing of SGLT2 inhibitor empagliflozin for strengthening anti-heart failure activity with lower glycosuria显示文摘Sodium-glucose cotransporter 2(SGLT2)inhibitors have been reapproved for heart failure(HF)therapy in patients with and without diabetes. However, the initial glucose-lowering indication of SGLT2i has impeded their uses in cardiovascular clinical practice.A challenge of SGLT2i then becomes how to separate their anti-HF activity from glucose-lowering side-effect.To address this issue,we conducted structural repurposing of EMPA,a representative SGLT2 inhibitor,to strengthen anti-HF activity and reduce the SGLT2-inhibitory activity according to structural basis of inhibition of SGLT2.Compared to EMPA,the optimal derivative JX01,which was produced by methylation of C2—OH of the glucose ring, exhibited weaker SGLT2-inhibitory activity(IC_(50)>100 nmol/L),and lower glycosuria and glucose-lowering side-effect,better NHE1-inhibitory activity and cardioprotective effect in HF mice.Furthermore,JX01 showed good safety profiles in respect of single-dose/repeat-dose toxicity and hERG activity, and good pharmacokinetic properties in both mouse and rat species.Collectively, the present study provided a paradigm of drug repurposing to discover novel anti-HF drugs, and indirectly demonstrated that SGLT2-independent molecular mechanisms play an important role in cardioprotective effects of SGLT2 inhibitors. | Yixiang Xu Chao Zhang Kai Jiang Xinchun Yang Feng Chen Zhiyang Cheng Jinlong Zhao Jiaxing Cheng Xiaokang Li Xin Chen Luoyifan Zhou Hao Duan Yunyuan Huang Yaozu Xiang Jian Li | 2023 | Acta Pharmaceutica Sinica B2023,13,4: | 0
