| 2 | Phase I study of CAR-T cells with PD-1 and TCR disruption in mesothelin-positive solid tumors显示文摘Programmed cell death protein-1(PD-1)-mediated immunosuppression has been proposed to contribute to the limited clinical efficacy of chimeric antigen receptor T(CAR-T)cells in solid tumors.We generated PD-1 and T cell receptor(TCR)deficient mesothelin-specific CAR-T(MPTK-CAR-T)cells using CRISPR-Cas9 technology and evaluated them in a dose-escalation study.A total of 15 patients received one or more infusions of MPTK-CAR-T cells without prior lymphodepletion.No dose-limiting toxicity or unexpected adverse events were observed in any of the 15 patients.The best overall response was stable disease(2/15 patients).Circulating MPTK-CAR-T cells peaked at days 7–14 and became undetectable beyond 1 month.TCR-positive CAR-T cells rather than TCR-negative CAR-T cells were predominantly detected in effusion or peripheral blood from three patients after infusion.We further confirmed the reduced persistence of TCR-deficient CAR-T cells in animal models.Our results establish the preliminary feasibility and safety of CRISPR-engineered CAR-T cells with PD-1 disruption and suggest that the natural TCR plays an important role in the persistence of CAR-T cells when treating solid tumors. | Zhenguang Wang Na Li Kaichao Feng Meixia Chen Yan Zhang Yang Liu Qingming Yang Jing Nie Na Tang Xingying Zhang Chen Cheng Lianjun Shen Jiaping He Xun Ye Wei Cao Haoyi Wang Weidong Han | 2021 | Cellular & Molecular Immunology2021,18,9: | 7 |

| 6 | Self-cleaning property of AZ31 Mg alloy during plasma electrolytic oxidation process显示文摘The rolling of metal sheet often leads to strong contamination of the resulting surface. The present work reports the effects of the surface contamination remaining after rolling of AZ31 Mg alloy on plasma electrolytic oxidation(PEO) treatment of the resulting sheet material. Results indicate that most of the contaminants on the surface of as-rolled AZ31 Mg alloy are removed in the early stages of PEO process. The contaminations slightly influence the phase composition of PEO coating. Non-uniform distribution of contaminations results in aggregation of pores on the surface of PEO samples. On the bare substrate the contaminations are responsible for poor wear and corrosion resistance but final PEO coatings reveals similar performance on contaminated substrate compared to the abraded one. | 13 | Analysis of Compressive Strength Development of Ultra-high Performance Concrete显示文摘A numerical procedure was presented for evaluating the compressive strength development of ultra-high performance concrete(UHPC) with cement-silica fume-slag binder.This numerical procedure started with initial packing behavior of designed UHPC using a random sequential packing method.Furthermore,synergistic effect of combined mineral admixtures was addressed with respect to hydration heat.Accordingly,hydration degree of cement and reaction degrees of mineral admixtures were determined based on a blended cement hydration model.Finally,a compressive strength evolution model was proposed and the evolution of compressive strength of three mixes with different binder recipes was compared.The results showed that the both initial packing behavior of UHPC mixes and synergistic effect of mineral admixtures are critical for predicting the properties of UHPC.A remarkable void fraction of 0.2042 was observed for UHPC mix designed by optimization algorithms under random packing.Furthermore,a negative synergistic effect of the combination of silica fume and slag was obtained with regarding to compressive strength.In addition,for a given mineral admixture replacement of 20%,the formulation of mineral admixture should be carefully tailored where the UHPC incorporating 5%silica fume and 15%slag shows the highest compressive strength. | HAN Fangyu LIU Jianzhong ZHANG Qianqian LIU Jiaping SHI Liang | 2016 | Journal of the Chinese Ceramic Society2016,,3: | 0 |
| 14 | Organophosphine ligand derived sandwich-structural electrocatalyst for oxygen evolution reaction显示文摘The synchronous construction of metal phosphate and phosphorus-doped carbon structures is of great significance to innovate the design,synthesis,and application of catalysts,as these phosphoruscontaining composite materials have shown a remarkable contribution to electrocatalysts.However,their preparation procedure generally involves using large amounts of excess phosphorus sources for phosphorization,which inevitably release poisonous PH_(3) or dangerous phosphorus vapor.Here,a strategy for in-situ formation of FePO_(4) embedded in P-doped carbon 2D nano film(FePO_(4)/PdC)is developed using a highly integrated precursor,which is a small molecular organophosphine ligand,1,1’bis(diphenylphosphine)ferrocene(DPPF).The multi-source precursor DPPF that contains Fe,P,and C is molecular-vapor-deposited on the nickel foam(NF)supported ZIF-67 nanosheets to obtain the composite catalyst,namely DPPF-500/ZIF-67/NF.FePO_(4)/PdC encapsulated the ZIF-67 derived Co/N-doped carbon matrix(Co NC)to form a sandwich structure FePO_(4)/PdC@CoNC.The constructed catalyst shows good performance for OER,requiring an overpotential of only 297 m V to deliver 600 m A/cm^(2) with a Tafel slope of 42.7 m V dec^(-1).DFT calculations demonstrate that the synergistic effects between the metal active center and P-doped carbon film reduce the energy barriers and improve electron transport.This method of constructing P-containing catalysts overcomes the demand for additional P sources to realize eco-friendly fabrication and yields a unique structure with good catalytic activity. | Zejun Sun Wenjuan Xu Liutao Guo Qiang Han Jianyi Gao Jiaping Wang Yanru Feng Chengrui Li Qionglin Liang Hong-bin Sun Yang Yang | 2022 | Journal of Energy Chemistry2022,31,7: | 0 |
