| 1 | Bone morphogenetic protein-2 gene controls tooth root development in coordination with formation of the periodontium显示文摘Formation of the periodontium begins following onset of tooth-root formation in a coordinated manner after birth. Dental follicle progenitor cells are thought to form the cementum, alveolar bone and Sharpey's fibers of the periodontal ligament (PDL). However, little is known about the regulatory morphogens that control differentiation and function of these progenitor cells, as well as the progenitor cells involved in crown and root formation. We investigated the role of bone morphogenetic protein-2 (Bmp2) in these processes by the conditional removal of the Bmp2 gene using the Sp7-Cre-EGFP mouse model. Sp7-Cre-EGFP first becomes active at E18 in the first molar, with robust Cre activity at postnatal day 0 (P0), followed by Cre activity in the second molar, which occurs after P0. There is robust Cre activity in the periodontium and third molars by 2 weeks of age. When the Bmp2 gene is removed from Sp7+ (Osterix+) cells, major defects are noted in root, cellular cementum and periodontium formation. First, there are major cell autonomous defects in root-odontoblast terminal differentiation. Second, there are major alterations in formation of the PDLs and cellular cementum, correlated with decreased nuclear factor IC (Nfic), periostin and α-SMA+ cells. Third, there is a failure to produce vascular endothelial growth factor A (VEGF-A) in the periodontium and the pulp leading to decreased formation of the microvascular and associated candidate stem cells in the Bmp2-cKOSp7-Cre-EGFP . Fourth, ameloblast function and enamel formation are indirectly altered in the Bmp2-cKOSp7-Cre-EGFP . | 5 | Less common etiologies of exocrine pancreatic insufficiency显示文摘Exocrine pancreatic insufficiency(EPI), an important cause of maldigestion and malabsorption, results from primary pancreatic diseases or secondarily impaired exocrine pancreatic function. Besides cystic fibrosis and chronic pancreatitis, the most common etiologies of EPI, other causes of EPI include unresectable pancreatic cancer, metabolic diseases(diabetes); impaired hormonal stimulation of exocrine pancreatic secretion by cholecystokinin(CCK); celiac or inflammatory bowel disease(IBD) due to loss of intestinal brush border proteins; and gastrointestinal surgery(asynchrony between motor and secretory functions, impaired enteropancreatic feedback, and inadequate mixing of pancreatic secretions with food). This paper reviews such conditions that have less straightforward associations with EPI and examines the role of pancreatic enzyme replacement therapy(PERT). Relevant literature was identified by database searches. Most patients with inoperable pancreatic cancer develop EPI(66%-92%). EPI occurs in patients with type 1(26%-57%) or type 2 diabetes(20%-36%) and is typically mild to moderate; by definition, all patients with type 3 c(pancreatogenic) diabetes have EPI. EPI occurs in untreated celiac disease(4%-80%), but typically resolves on a gluten-free diet. EPI manifests in patients with IBD(14%-74%) and up to 100% of gastrointestinal surgery patients(47%-100%; dependent on surgical site). With the paucity of published studies on PERT use for these conditions, recommendations for or against PERT use remain ambiguous. | 6 | The role of landscape pattern analysis in understanding concepts of land cover change显示文摘Landscape ecology and landscape pattern analysis are important components of national-scale programs to identify trends in land cover change because: 1) Statistics on changes in land cover proportions are not spatial. A change matrix derived from GIS provides useful information, but it does not show the spatial form of change in the landscape. Landscape pattern metrics reveal spatial pattern. 2) A growing body of literature has shown that a change in landscape pattern might indicate important changes in ecological functions: forest connectivity and species movements, number and size of farm patches, effects on water quality. Spatial pattern is important in structuring ecological communities and in maintaining existence of competitors. Spatial pattern may be determined by disturbance and may in turn, determine how disturbances propagate through the system. 3) Sometimes landscape pattern may not significantly change, even though land cover proportions do change. Or, vice-versa, sometimes landscape pattern can significantly change, even though land cover proportions don’t significantly change. 4) Landscape pattern is an inherent and important part of describing landscapes: based on the literature, one of the most important descriptive characteristics of a landscape is its texture. | 8 | 局部缓释IUdR治疗恶性星形细胞瘤实验研究(英文)显示文摘目的研究脑局部缓释125碘标记的5-碘脱氧尿苷(IUdR)的作用。方法为了控制释放125碘-IUdR,合成了25μCi的羧基苯氧丙烷:癸二酸(PCPPSA)(2080)125碘-IUdR聚合体。体外将合成的三聚体缓释剂(10mgPCPPSA)置于37℃磷酸盐缓冲溶液(PBS)中孵育不同时间后移出溶剂并置换为新的PBS,将移出液于放射计数器上测量数值。在体研究采用雄性裸鼠(6周)颅内接种U251人恶性星形细胞瘤及皮下接种对比研究。将颅内荷瘤鼠瘤腔内及皮下接种肿瘤组织内分别应用三聚体缓释剂及空白缓释剂,于不同时间分别测量动物头颅及皮下结节的放射活性,间接定量研究缓释状况;采血了解全身代谢情况。对于多聚磷酸酯(PPE)缓释剂研究采用皮下肿瘤结节内局部应用并测量其缓释的方法,放射自显影研究及全身不同器官缓释剂的吸收代谢研究采用接种后2、4和8d收集标本,分别行切片放射自显影和器官放射活性测定。结果体内和体外研究均显示PCPP:SA缓释剂对125碘-IUdR的控制性释放作用,尤其在体研究分别显示颅内和皮下接种的肿瘤组织对标记的IUdR的释放程度存有极大的差异,提示颅内局部应用的优越性。放射自显影定性研究显示了随距离的增加被标记的IUdR的递减趋势,为确定缓释剂施放部位和间距提供了参考数据。各主要脏器放射性测定的结果显示早期小肠内放射性较高,其次为脾脏,提示IUdR主要的代谢渠道,为处理应用其而可能引发的其他脏器损害或作用提供了线索。结论放射标记的IUdR缓释剂主要集中在接种部位且缓释作用确切。 | 马晓东 Jerry R.Williams Larry E.Dillehay Jeffery A Williams | 2005 | 中华神经外科疾病研究杂志2005，4，4： | 3 |
