| 1 | Efficient Targeted Genome Modification in Maize Using CRISPR/Cas9 System显示文摘CRISPR(clustered regularly interspaced short palindromic repeats)/Cas9 system, which is a newly developed technology for targeted genome modification, has been successfully used in a number of species. In this study, we applied this technology to carry out targeted genome modification in maize. A marker gene Zmzb7 was chosen for targeting. The sg RNA-Cas9 construct was transformed into maize protoplasts, and indel(insertion and deletion) mutations could be detected. A mutant seedling with an expected albino phenotype was obtained from screening 120 seedlings generated from 10 callus events. Mutation efficiency in maize heterochromatic regions was also investigated. Twelve sites with different expression levels in maize centromeres or pericentromere regions were selected. The sg RNACas9 constructs were transformed into protoplasts followed by sequencing the transformed protoplast genomic DNA. The results show that the genes in heterochromatic regions could be targeted by the CRISPR/Cas9 system efficiently, no matter whether they are expressed or not. Meanwhile, off-target mutations were not found in the similar sites having no PAM(protospacer adjacent motif) or having more than two mismatches. | 3 | Difficult biliary cannulation: Historical perspective, practical updates, and guide for the endoscopist显示文摘Despite improvements in endoscopic technologies and accessories, development of advanced endoscopy fellowship programs, and advances in ancillary imaging techniques, biliary cannulation in endoscopic retrograde cholangiopancreatography(ERCP) can still be unsuccessful in up to 20% of patients, even in referral centers. Once cannulation has been deemed to be difficult, the risk of post-ERCP pancreatitis and technical failure inherently increases. A number of factors, including endoscopist experience and patient anatomy, have been associated with difficult biliary cannulation, but predicting a case of difficult cannulation a priori is often not possible. Numerous techniques such as pancreatic guidewire and stenting, early pre-cut, and rendezvous may be employed when standard approaches fail. Data regarding the rate of success and adverse events of these techniques have been variable, though most studies suggest that pancreatic duct stenting generally reduces the rate of post-ERCP pancreatitis in instances of difficult biliary cannulation. | 4 | Extreme Cold Events from East Asia to North America in Winter 2020/21:Comparisons,Causes,and Future Implications显示文摘Three striking and impactful extreme cold weather events successively occurred across East Asia and North America during the mid-winter of 2020/21.These events open a new window to detect possible underlying physical processes.The analysis here indicates that the occurrences of the three events resulted from integrated effects of a concurrence of anomalous thermal conditions in three oceans and interactive Arctic-lower latitude atmospheric circulation processes,which were linked and influenced by one major sudden stratospheric warming(SSW).The North Atlantic warm blob initiated an increased poleward transient eddy heat flux,reducing the Barents-Kara seas sea ice over a warmed ocean and disrupting the stratospheric polar vortex(SPV)to induce the major SSW.The Rossby wave trains excited by the North Atlantic warm blob and the tropical Pacific La Nina interacted with the Arctic tropospheric circulation anomalies or the tropospheric polar vortex to provide dynamic settings,steering cold polar air outbreaks.The long memory of the retreated sea ice with the underlying warm ocean and the amplified tropospheric blocking highs from the midlatitudes to the Arctic intermittently fueled the increased transient eddy heat flux to sustain the SSW over a long time period.The displaced or split SPV centers associated with the SSW played crucial roles in substantially intensifying the tropospheric circulation anomalies and moving the jet stream to the far south to cause cold air outbreaks to a rarely observed extreme state.The results have significant implications for increasing prediction skill and improving policy decision making to enhance resilience in“One Health,One Future”. | Xiangdong ZHANG Yunfei FU Zhe HAN James E.OVERLAND Annette RINKE Han TANG Timo VIHMA Muyin WANG | 2022 | Advances in Atmospheric Sciences2022,39,4: | 7 |
| 5 | Dicentric Chromosome Formation and Epigenetics of Centromere Formation in Plants显示文摘Plant centromeres are generally composed of tandem arrays of simple repeats that form a complex chromosome locus where the kinetochore forms and microtubules attach during mitosis and meiosis. Each chromosome has one centromere region, which is essential for accurate division of the genetic material. Recently, chromosomes containing two centromere regions (called dicentric chromosomes) have been found in maize and wheat. Interestingly, some dicentric chromosomes are stable because only one centromere is active and the other one is inactivated. Because such arrays maintain their typical structure for both active and inactive centromeres, the specification of centromere activity has an epigenetic component independent of the DNA sequence. Under some circumstances, the inactive centromeres may recover centromere function, which is called centromere reactivation. | 7 | Consensus on the digestive endoscopic tunnel technique显示文摘With the digestive endoscopic tunnel technique(DETT), many diseases that previously would have been treated by surgery are now endoscopically curable by establishing a submucosal tunnel between the mucosa and muscularis propria(MP). Through the tunnel, endoscopic diagnosis or treatment is performed for lesions in the mucosa, in the MP, and even outside the gastrointestinal(GI) tract.At present, the tunnel technique application range covers the following:(1)Treatment of lesions originating from the mucosal layer, e.g., endoscopic submucosal tunnel dissection for oesophageal large or circular early-stage cancer or precancerosis;(2) treatment of lesions from the MP layer, per-oral endoscopic myotomy, submucosal tunnelling endoscopic resection, etc.; and(3) diagnosis and treatment of lesions outside the GI tract, such as resection of lymph nodes and benign tumour excision in the mediastinum or abdominal cavity. With the increasing number of DETTs performed worldwide, endoscopic tunnel therapeutics, which is based on DETT, has been gradually developed and optimized. However, there is not yet an expert consensus on DETT to regulate its indications, contraindications, surgical procedure, and postoperative treatment.The International DETT Alliance signed up this consensus to standardize the procedures of DETT. | 8 | Antimicrobial peptide LL-37 forms complex with bacterial DNA to facilitate blood translocation of bacterial DNA and aggravate ulcerative colitis显示文摘Bacterial DNA(bacDNA) is frequently found in serum of patient with ulcerative colitis(UC) and Crohn's disease, even blood bacterial culture is negative. How bacDNA evades immune elimination and is translocated into blood remain unclear. Here, we showed that bacDNA avoids elimination and disables bacteriakilling function of antimicrobial peptide LL-37(Cramp in mice) by forming complex with LL-37, which is inducible after culture with bacteria or bacterial products. Elevated LL-37-bacDNA complex was found in plasma and lesions of patients with UC. LL-37-bacDNA promoted inflammation by inducing Th1, Th2 and Th17 differentiation and activating toll-like receptor-9(TLR9). The complex also increased paracellular permeability, which possibly combines its inflammatory effects to promote local damage and bacDNA translocation into blood. Cramp-bacDNA aggravated mouse colitis severity while interference with the complex ameliorated the disease. The study identifies that inflammatogenic bacDNA utilizes LL-37 as a vehicle for blood translocation and to evade immune elimination. | 10 | Non-invasive detection of pancreatic cancer by measuring DNA methylation of Basonuclin 1 and Septin 9 in plasma显示文摘Pancreatic cancer (PC), a deadly malignancy with an overall 5-year survival rate of 5% to 15%, is ranked as the seventh leading cause of cancer death in the world in spite of its low occurrence rate.[1,2] Early detection appears to be the most effective approach to improve the overall survival of patients with PC. However, the difficulty in early detection of PC is a lack of specific symptoms and reliable biomarkers. Currently, carbohydrate antigen 19-9 (CA 19-9) is a serum biomarker that is widely used in PC detection. However, 10% of patients with PC cannot produce CA 19-9 and serum CA 19-9 is frequently absent in patients with early-stage cancer. Furthermore, CA 19-9 is often found to be elevated in benign conditions or in other cancers, making its utility limited. | 15 | Time-dependent viscoelastic properties along rat small intestine显示文摘AIM: To measure the time-dependent (viscoelastic)behavior in the change of the small intestinal opening angle and to test how well the behavior could be described by the Kelvin model for a standard linear solid.METHODS: Segments from the duodenum, jejunum, and ileum were harvested from 10 female Wistar rats and the luminal diameter, wall thickness, and opening angleover time (θ(t)) were measured from rings cut from thesesegments.RESULTS: Morphometric variations were found along thesmall intestine with an increase in luminal area and adecrease in wall thickness from the duodenum to theileum. The opening angle obtained after 60 min washighest in the duodenum (220.8±12.9°) and decreasedalong the length of the intestine to 143.9±8.9° in the jejunum and 151.4±9.4° in the ileum. The change ofopening angle as a function of time, fitted well to theKelvin model using the equation θ(t)/θo = [1-ηexp (-λt)]after the ring was cut. The computed creep rate λ did notdiffer between the segments. Compared to constantcalculated from pig aorta and coronary artery, it showedthat α agreed well (within 5%), η was three times largerthan that for vascular tissue, and λ ranged ±40% from the value of the pig coronary artery and was a third of the value of pig aorta.CONCLUSION: The change of opening angle over timefor all the small intestine segments fits well to the standardlinear spring-dashpot model. This viscoelastic constantof the rat small intestine is fairly homogenous along itslength.
