1Prunus genetics and applications after de novo genome sequencing:achievements and prospects显示文摘Prior to the availability of whole-genome sequences,our understanding of the structural and functional aspects of Prunus tree genomes was limited mostly to molecular genetic mapping of important traits and development of EST resources.With public release of the peach genome and others that followed,significant advances in our knowledge of Prunus genomes and the genetic underpinnings of important traits ensued.In this review,we highlight key achievements in Prunus genetics and breeding driven by the availability of these whole-genome sequences.Within the structural and evolutionary contexts,we summarize:(1)the current status of Prunus whole-genome sequences;(2)preliminary and ongoing work on the sequence structure and diversity of the genomes;(3)the analyses of Prunus genome evolution driven by natural and man-made selection;and(4)provide insight into haploblocking genomes as a means to define genome-scale patterns of evolution that can be leveraged for trait selection in pedigree-based Prunus tree breeding programs worldwide.Functionally,we summarize recent and ongoing work that leverages whole-genome sequences to identify and characterize genes controlling 22 agronomically important Prunus traits.These include phenology,fruit quality,allergens,disease resistance,tree architecture,and self-incompatibility.Translationally,we explore the application of sequence-based marker-assisted breeding technologies and other sequence-guided biotechnological approaches for Prunus crop improvement.Finally,we present the current status of publically available Prunus genomics and genetics data housed mainly in the Genome Database for Rosaceae(GDR)and its updated functionalities for future bioinformatics-based Prunus genetics and genomics inquiry.Maria JoséAranzana Véronique Decroocq Elisabeth Dirlewanger Iban Eduardo Zhong Shan Gao Ksenija Gasic Amy Iezzoni Sook Jung Cameron Peace Humberto Prieto Ryutaro Tao Ignazio Verde Albert G.Abbott Pere Arús 2019Horticulture Research2019,6,1:8
2Vitamin B12 modulates Parkinson’s disease LRRK2 kinase activity through allosteric regulation and confers neuroprotection显示文摘Missense mutations in Leucine-Rich Repeat Kinase 2(LRRK2)cause the majority of familial and some sporadic forms of Parkinson’s disease(PD).The hyperactivity of LRRK2 kinase induced by the pathogenic mutations underlies neurotoxicity,promoting the development of LRRK2 kinase inhibitors as therapeutics.Many potent and specific small-molecule LRRK2 inhibitors have been reported with promise.However,nearly all inhibitors are ATP competitive—some with unwanted side effects and unclear clinical outcome—alternative types of LRRK2 inhibitors are lacking.Herein we identify 5′-deoxyadenosylcobalamin(AdoCbl),a physiological form of the essential micronutrient vitamin B12 as a mixed-type allosteric inhibitor of LRRK2 kinase activity.Multiple assays show that AdoCbl directly binds LRRK2,leading to the alterations of protein conformation and ATP binding in LRRK2.STD-NMR analysis of a LRRK2 homologous kinase reveals the contact sites in AdoCbl that interface with the kinase domain.Furthermore,we provide evidence that AdoCbl modulates LRRK2 activity through disrupting LRRK2 dimerization.Treatment with AdoCbl inhibits LRRK2 kinase activity in cultured cells and brain tissue,and prevents neurotoxicity in cultured primary rodent neurons as well as in transgenic C.elegans and D.melanogaster expressing LRRK2 disease variants.Finally,AdoCbl alleviates deficits in dopamine release sustainability caused by LRRK2 disease variants in mouse models.Our study uncovers vitamin B12 as a novel class of LRRK2 kinase modulator with a distinct mechanism,which can be harnessed to develop new LRRK2-based PD therapeutics in the future.Adam Schaffner Xianting Li Yacob Gomez-Llorente Emmanouela Leandrou Anna Memou Nicolina Clemente Chen Yao Farinaz Afsari Lianteng Zhi Nina Pan Keita Morohashi Xiaoluan Hua Ming-Ming Zhou Chunyu Wang Hui Zhang Shu G.Chen Christopher J.Elliott Hardy Rideout Iban Ubarretxena-Belandia Zhenyu Yue 2019Cell Research2019,29,4:7

4High-density mapping suggests cytoplasmic male sterility with two restorer genes in almond×peach progenies显示文摘Peach(Prunus persica)and almond(Prunus dulcis)are two sexually compatible species that produce fertile offspring.Almond,a highly polymorphic species,is a potential source of new genes for peach that has a strongly eroded gene pool.Here we describe the genetics of a male sterile phenotype that segregated in two almond(‘Texas’)×peach(‘Earlygold’)progenies:an F2(T×E)and a backcross one(T1E)to the‘Earlygold’parent.High-density maps were developed using a 9k peach SNP chip and 135 simple-sequence repeats.Three highly syntenic and collinear maps were obtained:one for the F2(T×E)and two for the backcross,T1E(for the hybrid)and E(for‘Earlygold’).A major reduction of recombination was observed in the interspecific maps(T×E and T1E)compared to the intraspecific parent(E).The E map also had extensive monomorphic genomic regions suggesting the presence of large DNA fragments identical by descent.Our data for the male sterility character were consistent with the existence of cytoplasmic male sterility,where individuals having the almond cytoplasm required the almond allele in at least one of two independent restorer genes,Rf1 and Rf2,to be fertile.The restorer genes were located in a 3.4 Mbp fragment of linkage group 2(Rf1)and 1.4 Mbp of linkage group 6(Rf2).Both fragments contained several genes coding for pentatricopeptide proteins,demonstrated to be responsible for restoring fertility in other species.The implications of these results for using almond as a source of novel variability in peach are discussed.JoséManuel Donoso Iban Eduardo Roger Picañol Ignasi Batlle Werner Howad María JoséAranzana Pere Arús 2015Horticulture Research2015,2,1:1

17Perspectives of nano-carrier drug delivery systems to overcome cancer drug resistance in the clinics显示文摘Advanced cancer is still considered an incurable disease because of its metastatic spread to distal organs and progressive gain of chemoresistance.Even though considerable treatment progress and more effective therapies have been achieved over the past years,recurrence in the long-term and undesired side effects are still the main drawbacks of current clinical protocols.Moreover,a majority of chemotherapeutic drugs are highly hydrophobic and need to be diluted in organic solvents,which cause high toxicity,in order to reach effective therapeutic dose.These limitations of conventional cancer therapies prompted the use of nanomedicine,the medical application of nanotechnology,to provide more effective and safer cancer treatment.Potential of nanomedicines to overcome resistance,ameliorate solubility,improve pharmacological profile,and reduce adverse effects of chemotherapeutical drugs is thus highly regarded.Their use in the clinical setting has increased over the last decade.Among the various existing nanosystems,nanoparticles have the ability to transform conventional medicine by reducing the adverse effects and providing a controlled release of therapeutic agents.Also,their small size facilitates the intracellular uptake.Here,we provide a closer review of clinical prospects and mechanisms of action of nanomedicines to overcome drug resistance.The significance of specific targeting towards cancer cells is debated as well.Anna Ulldemolins Joaquin Seras-Franzoso Fernanda Andrade Diana Rafael Ibane Abasolo Petra Gener Simo Schwartz Jr 2021Cancer Drug Resistance2021,4,1:1
