1Discovery and Analysis of Shale Gas in a Carboniferous Reservoir and its Enrichment Characteristics in the Northern Nanpanjiang Depression, Guizhou Province, China显示文摘Shale gas resources are considered to be extremely abundant in southern China,which has dedicated considerable attention to shale gas exploration in recent years.Exploration of shale gas has considerably progressed and several breakthroughs have been made in China.However,shale gas explorations are still scarce.Summary and detailed analysis studies on black shale reservoirs are still to be performed for many areas.This lack of information slows the progress of shale gas explorations and results in low quantities of stored black shale.The Carboniferous Dawuba Formation,which is widely distributed and considerably thick,is one of the black shale formations targeted for shale gas exploration in southern China in the recent years.The acquisition and analysis of total organic carbon,vitrinite reflectance,types of organic matter,mineral composition,porosity,and permeability are basic but important processes.In addition,we analyzed the microscopic pores present in the shale.This study also showesd the good gas content of the Dawuba Formation,as well as the geological factors affecting its gas content and other characteristics.To understand the prospect of exploration,we compared this with other shale reservoirs which have been already successfully explored for gas.Our comparison showesd that those shale reservoirs have similar but not identical geological characteristics.LU Shufan LUO Xiangjian DU Shengjiang HE Ben FU Hongbin LIU Kuiyong 2019Acta Geologica Sinica(English Edition)2019,93,3:5
2Anti-apoptotic effects of aspirin following cerebral ischemia-reperfusion injury in rats显示文摘BACKGROUND:The pharmacological effects of aspirin on apoptosis are complex.The underlying mechanisms have not been properly defined.OBJECTIVE:To observe the effect of different doses of aspirin on brain cell apoptosis following focal cerebral ischemia-reperfusion injury(CIRI) in rats.DESING,TIME AND SETTING:A randomized,controlled,animal experiment,performed at the School of Medicine and Pharmaceutics,Jiangnan University between June and October 2006.MATERIALS:Twenty-six male,adult,Sprague Dawley rats(grade II),weighing 240-290 g,were obtained from Shanghai Experimental Animal Center,Chinese Academy of Sciences.Aspirin was provided by Sigma(USA).METHODS:The rats were randomly divided into four groups:sham-operation(SO),CIRI + vehicle,CIRI + aspirin(6 mg/kg),and CIRI + aspirin(60 mg/kg).Rats in the lesion groups were intragastrically administrated saline,aspirin(6 mg/kg),or aspirin(60 mg/kg),respectively.MAIN OUTCOME MEASURES:The number of pyramidal neurons with normal appearance in the cerebral cortex at 2-4 mm from the midline;apoptotic cell death as measured by TUNEL;Bcl-2 and Bax protein localization was determined by immunohistochemistry;malondialdehyde(MDA) and super oxidation(SOD) content were determined by biochemistry method;adenosine triphosphate(ATP) content measured by capillary electrophoresis.RESULTS:Following CIRI,the following parameters were altered compared with sham-operated animals:the number of neurons with normal appearance was significantly reduced in the cerebral cortex;the number of apoptotic cells increased;Bax protein expression was enhanced;and the ratio between Bcl-2 and Bax decreased.In addition,MDA content increased significantly,whereas ATP content decreased(P < 0.01).Aspirin ameliorated the loss of healthy pyramidal neurons.Both 6 and 60 mg/kg aspirin increased the ratio between Bcl-2 and Bax,with no significant difference between the treatment groups.In addition,60 mg/kg aspirin decreased MDA content and increased ATP levels.However,6 mg/kg aspirin did not have the same effect.CONCLUSION:Aspirin reduced the number of apoptotic cells following CIRI.These results suggest that the neuroprotective mechanism of aspirin could be related to elevated Bcl-2 protein levels or decreased Bax protein expression.The increase in the ratio of Bcl-2 to Bax appears to be a common anti-apoptotic mechanism of aspirin.Liying Qiu Bin Du Ying Li Hongbin Fan Zhiyong Yang 2008Neural Regeneration Research2008,3,9:4
3Changes in neuronal apoptosis and apoptosis modulatory factors in cerebral ischemia/reperfusion显示文摘BACKGROUND: The high concentration of glutamate release is the main cause for neuronal cell death. The relationship between glutamate level and apoptosis during ischemia/reperfusion injury is still unclear. OBJECTIVE: To observe the neuronal apoptosis at 24 and 72 hours following cerebral ischemia/reperfusion in rats,and analyze the possible influencing factors. DESIGN: A randomized controlled animal experiment. SETTING: School of Medicine,Southern Yangtze University. MATERIALS: Totally 30 male adult Sprague Dawley (SD) rats of clean grade,weighing 240-290 g,were obtained from Shanghai Experimental Animal Center,Chinese Academy of Sciences. The rats were randomly divided into sham-operated group (n=10) and model group (n=20). Each group was observed at 24 and 72 hours after ischemia/reperfusion,5 rats at each time point in the sham-operated group,whereas 12 at 24 hours and 8 at 72 hours in the model group. Kits for determining apoptosis and Bcl-2 were bought from Wuhan Boster Biological Technology,Co.,Ltd.; Kit for calcineurin from Nanjing Jiancheng Bioengineering Institute. METHODS: The experiment was carried out in the Functional Scientific Research Room of Southern Yangtze University from June to October in 2006. ① Right middle cerebral artery was occluded by inserting a thread through internal carotid artery (ICA). The surgical process for the sham-operated rats was the same as that in the model group except a nylon suture inserted the ICA. According to Longa five-degree standard,the neurological deficit evaluation of rats was evaluated after surgery,and grades 1-3 were taken as successful model establishment. The blood was recirculated by withdrawing the nylon filament under anesthesia at 2 hours after ischemia in successful rat models. ②After reperfusion,the brain tissue was quickly removed at 24 or 72 hours and the slices were obtained from optic chiasma to funnel manubrium. The changes of the number of apoptotic cells were observed using the terminal deoxynucleotidyl transferase mediated dUTP-biotin nick-end labeling method. The expressions of Bcl-2 protein were determined with immunohistochemical staining. The activity of calcineurin was determined by the inorganic phosphorus method. The content of excitatory amino acid was detected by high performance liquid chromatography. MAIN OUTCOME MEASURES: ① Glutanate content in brain tissue; ② Conditions of apoptosis; ③ Calcineurin activity in brain tissue; ④ Bcl-2 expression in brain tissue. RESULTS: Totally 30 SD rats were used,5 died and the other 25 were involved in the analysis of results. ① Changes of apoptosis: There were 0-3 apoptotic cells in the sham-operated group. In the model group,the numbers of apoptotic cells were obviously increased at 24 and 72 hours of reperfusion (P < 0.01),and it was markedly reduced at 72 hours as compared with 24 hours (P < 0.01). ② Changes of glutanate content: The glutamate contents at 24 and 72 hours of reperfusion in the model group were obviously higher than those in the sham-operated group (P < 0.01); In the model group,it was obviously increased at 24 hours as compared with 72 hours (P < 0.01). ③ Changes of Bcl-2 protein: In the model group,the Bcl-2 protein expression had no obvious changes at 24 hours of reperfusion,and it was obviously enhanced at 72 hours,which was obviously different from that in the sham-operated group and that at 24 hours (P < 0.01). ④ Changes of calcinerin activity: In the model group,the activity of calcineurin in brain tissue had no obvious changes at 24 hours of reperfusion; The activity of calcineurin at 72 hours was obviously higher than that in the sham-operated group and that at 24 hours (P < 0.01). 5Early changes to the extracellular space in the hippocampus under simulated microgravity conditions显示文摘The smooth transportation of substances through the brain extracellular space(ECS)is crucial to maintaining brain function;however,the way this occurs under simulated microgravity remains unclear.In this study,tracer-based magnetic resonance imaging(MRI)and DECS-mapping techniques were used to image the drainage of brain interstitial fluid(ISF)from the ECS of the hippocampus in a tail-suspended hindlimb-unloading rat model at day 3(HU-3)and 7(HU-7).The results indicated that drainage of the ISF was accelerated in the HU-3 group but slowed markedly in the HU-7 group.The tortuosity of the ECS decreased in the HU-3 group but increased in the HU-7 group,while the volume fraction of the ECS increased in both groups.The diffusion rate within the ECS increased in the HU-3 group and decreased in the HU-7 group.The alterations to ISF drainage and diffusion in the ECS were recoverable in the HU-3 group,but neither parameter was restored in the HU-7 group.Our findings suggest that early changes to the hippocampal ECS and ISF drainage under simulated microgravity can be detected by tracer-based MRI,providing a new perspective for studying microgravity-induced nano-scale structure abnormities and developing neuroprotective approaches involving the brain ECS.Yajuan Gao Hongbin Han Jichen Du Qingyuan He Yanxing Jia Junhao Yan Hui Dai Bin Cui Jing Yang Xunbin Wei Liu Yang Rui Wang Ren Long Qiushi Ren Xing Yang Jiabin Lu 2022Science China(Life Sciences)2022,65,3:2
6Identification of microsatellites in cattle unigenes显示文摘To identify EST-SSR molecular markers, 41,986 cattle UniGene sequences from NCBI were mined for analyzing SSRs. A total of 1,831 SSRs were identified from 1,666 ESTs, which represented an average density of 19.88 kb per SSR. The frequency of EST-SSRs was 4.0%. The dinucleotide repeat motif was the most abundant SSR, accounting for 54%, followed by 22%, 13%, 7% and 4%, respec- tively, for tri-, hexa-, penta- and tetra-nucleotide repeats. Depending upon the length of the repeat unit, the length of microsatellites varied from 14 to 86 bp. Among the di- and tri-nucleotide repeats, AC/TG (57%) and AGC (12%) were the most abundant type. Annotation of EST-SSRs was also carried out.
