1Whole-genome sequencing of 508 patients identifies key molecular features associated with poor prognosis in esophageal squamous cell carcinoma显示文摘Esophageal squamous cell carcinoma(ESCC)is a poor-prognosis cancer type with limited understanding of its molecular etiology.Using 508 ESCC genomes,we identified five novel significantly mutated genes and uncovered mutational signature clusters associated with metastasis and patients’outcomes.Several functional assays implicated that NFE2L2 may act as a tumor suppressor in ESCC and that mutations in NFE2L2 probably impaired its tumor-suppressive function,or even conferred oncogenic activities.Additionally,we found that the NFE2L2 mutations were significantly associated with worse prognosis of ESCC.We also identified potential noncoding driver mutations including hotspot mutations in the promoter region of SLC35E2 that were correlated with worse survival.Approximately 5.9%and 15.2%of patients had high tumor mutation burden or actionable mutations,respectively,and may benefit from immunotherapy or targeted therapies.We found clinically relevant coding and noncoding genomic alterations and revealed three major subtypes that robustly predicted patients’outcomes.Collectively,we report the largest dataset of genomic profiling of ESCC useful for developing ESCC-specific biomarkers for diagnosis and treatment.Yongping Cui Hongyan Chen Ruibin Xi Heyang Cui Yahui Zhao Enwei Xu Ting Yan Xiaomei Lu Furong Huang Pengzhou Kong Yang Li Xiaolin Zhu Jiawei Wang Wenjie Zhu Jie Wang Yanchun Ma Yong Zhou Shiping Guo Ling Zhang Yiqian Liu Bin Wang Yanfeng Xi Ruifang Sun Xiao Yu Yuanfang Zhai Fang Wang Jian Yang Bin Yang Caixia Cheng Jing Liu Bin Song Hongyi Li Yi Wang Yingchun Zhang Xiaolong Cheng Qimin Zhan Yanhong Li Zhihua Liu-Show 2020Cell Research2020,30,10:19

4Pharmacodynamics of frigid zone plant Taxus cuspidata S.et Z.against skin melanin deposition,oxidation,inflammation and allergy显示文摘Background:Taxus cuspidata S.et Z.is a precious species of frigid zone plant belonging to the Taxaceae family,which possesses anticancer,anti-inflammatory,hypoglycemic,and antibacterial pharmacological properties.While taxane extracted from Taxus chinensis has been reported to elicit antioxidant activities,whether Taxus cuspidata S.et Z.has skin-protective actions against injuries remained unknown.This study aims to explore the pharmacological effects of three Taxus extracts on skin melanin deposition,oxidation,inflammation,and allergy so as to provide new ideas for the prevention and treatment of various diseases related to skin damage.Methods:Skin melanin deposition was evaluated by measuring melanin content in the skin of guinea pigs by alkali lysis method.Antioxidant capacity was evaluated by measuring superoxide dismutase(SOD)concentration and glutathione(GSH)content in skin tissue homogenates of Kunming mice by SOD assay kit and micro reduced GSH assay kit.The quantitative real-time polymerase chain reaction(qRT-PCR)and western blotting were used to examine the levels of both SOD and recombinant glutathione peroxidase 4(GPX4).Skin inflammation was evaluated by xylene-induced ear swelling test and egg-white-induced paw swelling test in mice.In a mouse model of skin allergy induced by 4-aminopyridine(4-AP),allergy was determined by licking body counts and histamine concentrations in tissue homogenates using enzyme-linked immunosorbent assay(ELISA)kits.Two proinflammatory factors tumor necrosis factor(TNF)-αand interleukin(IL)-1βwere measured by qRT-PCR.Hematoxylin and eosin(HE)staining was conducted to assess the degree of skin lesion.Results:All three Taxus extracts including Taxus chinensis essential oil,Taxus chinensis extract and Taxus chinensis extract compound reduced the melanin deposits in the back skin relative to the non-treated control animals,of which Taxus chinensis essential oil produced the greatest effect.In contrast,the three Taxus extracts elevated SOD and GSH levels in the skin tissues,and the highest increase was seen with Taxus chinensis essential oil.Three Taxus extracts,especially Taxus chinensis essential oil,effectively reduce the rate of ear and paw swelling.All three Taxus extracts reduced the number of body licks,the levels of TNF-αand IL-1β,and the histamine content in tissue homogenates of mice and alleviated skin damage.Consistently,Taxus chinensis essential oil yielded the greatest magnitude of decreases.Conclusion:While all three Taxus extracts possessed the anti-skin melanin deposition,oxidation,and allergy properties,Taxus chinensis essential oil produced the superior effects.Yang Shi Ling Liu Heyang Sun Chen Chen Jing Feng Yongchao Chen Yuan Lin Philipp Kopylov Qi Wang Yong Zhang 2023Frigid Zone Medicine2023,3,1:0
5Spatial changes and driving factors of lake water quality in Inner Mongolia, China显示文摘Lakes play important roles in sustaining the ecosystem and economic development in Inner Mongolia Autonomous Region of China,but the spatial patterns and driving mechanisms of water quality in lakes so far remain unclear.This study aimed to identify the spatial changes in water quality and the driving factors of seven lakes(Juyanhai Lake,Ulansuhai Lake,Hongjiannao Lake,Daihai Lake,Chagannaoer Lake,Hulun Lake,and Wulannuoer Lake)across the longitudinal axis(from the west to the east)of Inner Mongolia.Large-scale research was conducted using the comprehensive trophic level index(TLI(Σ)),multivariate statistics,and spatial analysis methods.The results showed that most lakes in Inner Mongolia were weakly alkaline.Total dissolved solids and salinity of lake water showed obvious zonation characteristics.Nitrogen and phosphorus were identified as the main pollutants in lakes,with high average concentrations of total nitrogen and total phosphorus being of 4.05 and 0.21 mg/L,respectively.The values of TLI(Σ)ranged from 49.14 to 71.77,indicating varying degrees of lake eutrophication,and phosphorus was the main driver of lake eutrophication.The lakes of Inner Mongolia could be categorized into lakes to the west of Daihai Lake and lakes to the east of Daihai Lake in terms of salinity and TLI(Σ).The salinity levels of lakes to the west of Daihai Lake exceeded those of lakes to the east of Daihai Lake,whereas the opposite trend was observed for lake trophic level.The intensity and mode of anthropogenic activities were the driving factors of the spatial patterns of lake water quality.It is recommended to control the impact of anthropogenic activities on the water quality of lakes in Inner Mongolia to improve lake ecological environment.These findings provide a more thorough understanding of the driving mechanism of the spatial patterns of water quality in lakes of Inner Mongolia,which can be used to develop strategies for lake ecosystem protection and water resources management in this region.REN Xiaohui YU Ruihong LIU Xinyu SUN Heyang GENG Yue QI Zhen ZHANG Zhuangzhuang LI Xiangwei WANG Jun ZHU Penghang GUO Zhiwei WANG Lixin XU Jifei 2023Journal of Arid Land2023,15,2:0
6Greenhouse gas emissions from Daihai Lake,China:Should eutrophication and salinity promote carbon emission dynamics?显示文摘Greenhouse gases(GHGs)emitted or absorbed by lakes are an important component of the global carbon cycle.However,few studies have focused on the GHG dynamics of eutrophic saline lakes,thus preventing a comprehensive understanding of the carbon cycle.Here,we conducted four sampling analyses using a floating chamber in Daihai Lake,a eutrophication saline lake in Inner Mongolia Autonomous Region,China,to explore its carbon dioxide(CO_(2))and methane(CH_(4))emissions.The mean CO_(2)emission flux(FCO_(2))and CH_(4)emission flux(FCH_(4))were 17.54±14.54 mmol/m^(2)/day and 0.50±0.50 mmol/m^(2)/day,respectively.The results indicated that Daihai Lake was a source of CO_(2)and CH_(4),and GHG emissions exhibited temporal variability.The mean CO_(2)partial pressure(p CO_(2))and CH_(4)partial pressure(p CH_(4))were 561.35±109.59μatm and 17.02±13.45μatm,which were supersaturated relative to the atmosphere.The regression and correlation analysis showed that the main influencing factors of p CO_(2)were wind speed,dissolved oxygen(DO),total nitrogen(TN)and Chlorophyll a(Chl.a),whereas the main influencing factors of p CH_(4)were water temperature(WT),Chl.a,nitrate nitrogen(NO_(3)^(-)-N),TN,dissolved organic carbon(DOC)and water depth.Salinity regulated carbon mineralization and organic matter decomposition,and it was an important influencing factor of p CO_(2)and p CH_(4).Additionally,the trophic level index(TLI)significantly increased p CH_(4).Our study elucidated that salinity and eutrophication play an important role in the dynamic changes of GHG emissions.However,research on eutrophic saline lakes needs to be strengthened.Xiangwei Li Ruihong Yu Jun Wang Heyang Sun Xinyu Liu Xiaohui Ren Shuai Zhuang Zhiwei Guo Xixi Lu 2024Journal of Environmental Sciences2024,,1:0
7Self-adaptive label filtering learning for unsupervised domain adaptation显示文摘1 Introduction As an emerging machine learning paradigm,unsupervised domain adaptation(UDA)aims to train an effective model for unlabeled target domain by leveraging knowledge from related but distribution-inconsistent source domain.Most of the existing UDA methods[2]align class-wise distributions resorting to target domain pseudo-labels,for which hard labels may be misguided by misclassifications while soft labels are confusing with trivial noises so that both of them tend to cause frustrating performance.To overcome such drawbacks,as shown in Fig.1,we propose to achieve UDA by performing self-adaptive label filtering learning(SALFL)from both the statistical and the geometrical perspectives,which filters out the misclassified pseudo-labels to reduce negative transfer.Specifically,the proposed SALFL firstly predicts labels for the target domain instances by graph-based random walking and then filters out those noise labels by self-adaptive learning strategy.Qing TIAN Heyang SUN Shun PENG Tinghuai MA 2023Frontiers of Computer Science2023,17,1:0
