1The optimized preparation of HA/L-TiO2/D-TiO2 composite coating on porous titanium and its effect on the behavior osteoblasts显示文摘Various surface bioactivation technology has been confirmed to improve the osteogenic ability of porous titanium(pTi)implants effectively.In this study,a three-layered composite coating,i.e.outer layer of hydroxyapatite(HA),middle layer of loose titanium dioxide(L-TiO2)and inner layer of dense TiO2(D-TiO2),was fabricated on pTi by a combined processing procedure of pickling,alkali heat(AH),anodic oxidation(AO),electrochemical deposition(ED)and hydrothermal treatment(HT).After soaking in simulated body fluid for 48 h,the surface of the AHAOEDHT-treated pTi was completely covered by a homogeneous apatite layer.Using MC3T3-E1 pro-osteoblasts as cell model,the cell culture revealed that both the pTi without surface treatment and the AHAOEDHT sample could support the attachment,growth and proliferation of the cells.Compared to the pTi sample,the AHAOEDHT one induced higher expressions of osteogenesis-related genes in the cells,including alkaline phosphatase,Type I collagen,osteopontin,osteoclast inhibitor,osteocalcin and zinc finger structure transcription factor.As thus,besides the good corrosion resistance,the HA/L-TiO2/D-TiO2-coated pTi had good osteogenic activity,showing good potential in practical application for bone defect repair.Xi Fu Xingyu Zhou Pin Liu Hewei Chen Zhanwen Xiao Bo Yuan Xiao Yang Xiangdong Zhu Kai Zhang Xingdong Zhang 2020Regenerative Biomaterials2020,7,5:5

3Total body irradiation of donors can alter the course of tolerance and induce acute rejection in a spontaneous tolerance rat liver transplantation model显示文摘Liver transplantation is an established therapy for end-stage liver diseases. Graft rejection occurs unless the recipient receives immunosuppression after transplantation. This study aimed to explore the mechanism of acute rejection of liver allografts in rats pre-treated with total body irradiation to eliminate passenger lymphocytes and to define the role of CD4 + CD25 + regulatory T cells in the induction of immunotolerance in the recipient. Male Lewis rats were used as donors and male DA rats were recipients. Rats were randomly assigned to the following four groups: control group, homogeneity liver transplantation group, idio-immunotolerance group and acute rejection group. After transplantation, the survival time of each group, serum alanine aminotransferase, total bilirubin levels, number of Foxp3 + CD4 + CD25 + regulatory T cells, expression of glucocorticoid-induced tumor necrosis factor receptor on T cell subgroups, histopathology of the hepatic graft and spleen cytotoxic T lymphocyte lytic activity were measured. In the acute rejection group, where donors were preconditioned with total body irradiation before liver transplantation, all recipients died between day 17 and day 21. On day 14, serum alanine aminotransferase increased signifi- cantly to (459.2±76.9) U L -1 , total bilirubin increased to (124.1±33.7) μmol L -1 (P<0.05) and the ratio of Foxp3 + CD4 + CD25 + regulatory T cells decreased significantly to 1.50%±0.50% (P<0.05) compared with the other groups. Analysis of the T cell subpopulations in the acute rejection group varied from the other groups. Histological analysis showed typical changes of acute rejection in the acute rejection group only. 4Evaluation on the corrosion resistance, antibacterial property and osteogenic activity of biodegradable Mg-Ca and Mg-Ca-Zn-Ag alloys显示文摘The rapid degradation of magnesium(Mg)-based implants in physiological environment limits its clinical applications, and alloying treatment is an effective way to regulate the degradation rate of Mg-based materials. In the present study, three Mg alloys, including Mg-0.8Ca(denoted as ZQ), Mg-0.8Ca-5Zn-1.5Ag(denoted as ZQ71) and Mg-0.8Ca-5Zn-2.5Ag(denoted as ZQ63), were fabricated by alloying with calcium(Ca), zinc(Zn) and silver(Ag). The results obtained from electrochemical corrosion tests and in vitro degradation evaluation demonstrated that the three Mg alloys exhibited distinct corrosion resistance, and ZQ71 exhibited the lowest degradation rate in vitro among them. After addition of Zn and Ag, the antibacterial potential of Mg alloys was also enhanced. The in vitro cell experiments showed that all the three Mg alloys had good biocompatibility. After implantation in a rat femoral defect, ZQ71 showed significantly higher osteogenic activity and bone substitution rate than ZQ63 and ZQ, due to its higher corrosion resistance as well as the stimulatory effects of the released metallic ions. In addition, the average daily degradation rate of each Mg alloy in vivo was significantly higher than that in vitro, as could be due to the implantation site located in the highly vascularized trabecular region. Importantly, the correlations between the in vitro and in vivo degradation parameters of the Mg alloys were systematically analyzed to find out the potential predictors of the in vivo degradation performance of the materials. 6Construction of a magnesium hydroxide/graphene oxide/hydroxyapatite composite coating on Mg-Ca-Zn-Ag alloy to inhibit bacterial infection and promote bone regeneration显示文摘The improved corrosion resistance, osteogenic activity, and antibacterial ability are the key factors for promoting the large-scale clinical application of magnesium (Mg)-based implants. In the present study, a novel nanocomposite coating composed of inner magnesium hydroxide, middle graphene oxide, and outer hydroxyapatite (Mg(OH)_(2)/GO/HA) is constructed on the surface of Mg-0.8Ca-5Zn-1.5Ag by a combined strategy of hydrothermal treatment, electrophoretic deposition, and electrochemical deposition. The results of material characterization and electrochemical corrosion test showed that all the three coatings have high bonding strength, hydrophilicity and corrosion resistance. In vitro studies show that Mg(OH)2 indeed improves the antibacterial activity of the substrate. The next GO and GO/HA coating procedures both promote the osteogenic differentiation of MC3T3-E1 cells and show no harm to the antibacterial activity of Mg(OH)2 coating, but the latter exhibits the best promoting effect. In vivo studies demonstrate that the Mg alloy with the composite coating not only ameliorates osteolysis induced by bacterial invasion but also promotes bone regeneration under both normal and infected conditions. 9The binding epitope of sintilimab on PD-1 revealed by AbMap显示文摘PD-1 plays an important role as an immune checkpoint.Sintilimab is a newly approved PD-1 antibody for cancer immunotherapy with an unknown binding epitope on PD-1.In this study,to elucidate the molecular mechanism by which sintilimab blocks PD-1 activation,we applied Antibody binding epitope Mapping(AbMap)to identify the binding epitope of sintilimab.An epitope was successfully identified,i.e.SLAPKA,aa 127-132.By constructing a series of point mutations,the dominant residues S127,L128,A129,P130,and A132 of PD-1 were further validated by western blot analysis,biolayer interferometry,and flow cytometry.Structural analysis showed that the epitope is partially within the binding interface of PD-1 and PD-L1,and this epitope also partially overlaps with that of nivolumab and pembrolizumab.These results demonstrate that sintilimab can attenuate PD-1 activation by directly competing with the interaction between PD-1 and PD-L1 through binding with the key residues of the FG loop on PD-1.This study also demonstrates the high efficiency and accuracy of AbMap for determining the binding epitope of therapeutic antibodies.Mingliang Ma Huan Qi Chuansheng Hu Zhaowei Xu Fanlin Wu Nan Wang Danyun Lai Yang Li Hainan Zhang Hewei Jiang Qingfeng Meng Shujuan Guo Yani Kang Xiaodong Zhao Hua Li Sheng-ce Tao 2021Acta Biochimica et Biophysica Sinica2021,53,5:1

11Non-covalent interaction-based molecular electronics with graphene electrodes显示文摘Recent years have witnessed the fabrication of various non-covalent interaction-based molecular electronic devices.In the noncovalent interaction-based molecular devices,the strength of the interfacial coupling between molecule and electrode is weakened compared to that of the covalent interaction-based molecular devices,which provides wide applications in fabricating versatile molecular devices.In this review,we start with the methods capable of fabricating graphene-based nanogaps,and the following routes to construct non-covalent interaction-based molecular junctions with graphene electrodes.Then we give an introduction to the reported non-covalent interaction-based molecular devices with graphene electrodes equipped with different electrical functions.Moreover,we summarize the recent progress in the design and fabrication of new-type molecular devices based on graphene and graphene-like two-dimensional(2D)materials.The review ends with a prospect on the challenges and opportunities of non-covalent interaction-based molecular electronics in the near future.Shiqiang Zhao Hang Chen Qiaozan Qian Hewei Zhang Yang Yang Wenjing Hong 2023Nano Research2023,16,4:0

13SARS-CoV-2-specific antibody response characteristics in COVID-19 patients of different ages显示文摘Age has been found to be one of the main risk factors for the severity and outcome of COVID-19.However,differences in SARS-CoV-2 specific antibody responses among COVID-19 patients of different age groups remain largely unknown.In this study,we analyzed the IgG/IgM responses to 21 SARS-CoV-2 proteins and 197 peptides that fully cover the spike protein against 731 sera collected from 731 COVID-19 patients aged from 1 to 92 years.We show that there is no overall difference in SARS-CoV-2 antibody responses in COVID-19 patients in the 4 age groups.By antibody response landscape maps,we find that the IgG response profiles of SARS-CoV-2 proteins are positively correlated with age.The S protein linear epitope map shows that the immunogenicity of the S-protein peptides is related to peptide sequence,disease severity and age of the COVID-19 patients.Furthermore,the enrichment analysis indicates that low S1 IgG responses are enriched in patients aged<50 and high S1 IgG responses are enriched in mild COVID-19 patients aged>60.In addition,high responses of non-structural/accessory proteins are enriched in severe COVID-19 patients aged>70.These results suggest the distinct immune response of IgG/IgM to each SARS-CoV-2 protein in patients of different age,which may facilitate a deeper understanding of the immune responses in COVID-19 patients.Linfang Lu Siqi Yu Min Liu Yang Li Qing Lei Mingxi Lin Danyun Lai Shujuan Guo Hewei Jiang Hongyan Hou Yunxiao Zheng Xuening Wang Mingliang Ma Bo Zhang Hong Chen Junbiao Xue Hainan Zhang Huan Qi Ziyong Sun Feng Wang Xionglin Fan Zhaowei Xu 2022Acta Biochimica et Biophysica Sinica2022,54,4:0
