| 1 | Chondroitinase ABC plus bone marrow mesenchymal stem cells for repair of spinal cord injury显示文摘As chondroitinase ABC can improve the hostile microenvironment and cell transplantation is proven to be effective after spinal cord injury, we hypothesized that their combination would be a more effective treatment option. At 5 days after T8 spinal cord crush injury, rats were injected with bone marrow mesenchymal stem cell suspension or chondroitinase ABC 1 mm from the edge of spinal cord damage zone. Chondroitinase ABC was first injected, and bone marrow mesenchymal stem cell suspension was injected on the next day in the combination group. At 14 days, the mean Basso, Beattie and Bresnahan score of the rats in the combination group was higher than other groups. Hematoxylin-eosin staining showed that the necrotic area was significantly reduced in the combination group compared with other groups. Glial fibrillary acidic protein-chondroitin sulfate proteoglycan double staining showed that the damage zone of astrocytic scars was significantly reduced without the cavity in the combination group. Glial fibrillary acidic protein/growth associated protein-43 double immunostaining revealed that positive fibers traversed the damage zone in the combination group. | 3 | Tuning charge density of chimeric antigen receptor optimizes tonic signaling and CAR-T cell fitness显示文摘Tonic signaling of chimeric antigen receptor(CAR),i.e.,the spontaneous CAR activation in the absence of tumor antigen stimulation,is considered to be a pivotal event controlling CAR-T efficacy.However,the molecular mechanism underlying the spontaneous CAR signals remains elusive.Here,we unveil that positively charged patches(PCPs)on the surface of the CAR antigenbinding domain mediate CAR clustering and result in CAR tonic signaling.For CARs with high tonic signaling(e.g.,GD2.CAR and CSPG4.CAR),reducing PCPs on CARs or boosting ionic strength in the culture medium during ex vivo CAR-T cell expansion minimizes spontaneous CAR activation and alleviates CAR-T cell exhaustion.In contrast,introducing PCPs into the CAR with weak tonic signaling,such as CD19.CAR,results in improved in vivo persistence and superior antitumor function.These results demonstrate that CAR tonic signaling is induced and maintained by PCP-mediated CAR clustering.Notably,the mutations we generated to alter the PCPs maintain the antigen-binding affinity and specificity of the CAR.Therefore,our findings suggest that the rational tuning of PCPs to optimize tonic signaling and in vivo fitness of CAR-T cells is a promising design strategy for the nextgeneration CAR. | Jian Chen Shizhen Qiu Wentao Li Kun Wang Yu Zhang Han Yang Baichuan Liu Guangfei Li Li Li Min Chen Junjie Lan Jiahua Niu Peijie He Lei Cheng Gaofeng Fan Xin Liu Xianmin Song Chenqi Xu Haitao Wu Haopeng Wang | 2023 | Cell Research2023,33,5: | 2 |
| 4 | Olfactory ensheathing cell transplantation for spinal cord injury An 18-year bibliometric analysis based on the Web of Science显示文摘OBJECTIVE:Olfactory ensheathing cell (OEC) transplantation is a promising new approach for the treatment of spinal cord injury (SCI),and an increasing number of scientific publications are devoted to this treatment strategy.This bibliometric analysis was conducted to assess global research trends in OEC transplantation for SCI.DATA SOURCE:All of the data in this study originate from the Web of Science maintained by the Institute for Scientific Information,USA,and includes SCI-EXPANDED,SSCI,A&HCI,CPCI-S,CPCI-SSH,BKCI-S,BKCI-SSH,CCR-EXPANDED and IC.The Institute for Scientific Information's Web of Science was searched using the keywords 'olfactory ensheathing cells' or 'OECs' or 'olfactory ensheathing glia' or 'OEG' or 'olfactory ensheathing glial cells' or 'OEGs' and 'spinal cord injury' or 'SCI' or 'spinal injury' or 'spinal transection' for literature published from January 1898 to May 2012.DATA SELECTION:Original articles,reviews,proceedings papers and meeting abstracts,book chapters and editorial materials on OEC transplantation for SCI were included.Simultaneously,unpublished literature and literature for which manual information retrieval was required were excluded.MAIN OUTCOME MEASURES:All selected literatures addressing OEC transplantation for SCI were evaluated in the following aspects:publication year,document type,language,author,institution,times cited,Web of Science category,core source title,countries/territories and funding agency.RESULTS:In the Web of Science published by the Institute for Scientific Information,the earliest literature record was in April,1995.Four hundred and fourteen publications addressing OEC transplantation for SCI were added to the data library in the past 18 years,with an annually increasing trend.Of 415 records,405 publications were in English.Two hundred and fifty-nine articles ranked first in the distribution of document type,followed by 141 reviews.Thirty articles and 20 reviews,cited more than 55 times by the date the publication data were downloaded by us,can be regarded as the most classical references.The journal Experimental Neurology published the most literature (32 records),followed by Glia.The United States had the most literature,followed by China.In addition,Yale University was the most productive institution in the world,while The Second Military Medical University contributed the most in China.The journal Experimental Neurology published the most OEC transplantation literature in the United States,while Neural Regeneration Research published the most in China.CONCLUSION:This analysis provides insight into the current state and trends in OEC transplantation for SCI research.Furthermore,we anticipate that this analysis will help encourage international cooperation and teamwork on OEC transplantation for SCI to facilitate the development of more effective treatments for SCI. | Zikuan Leng Xijing He Haopeng Li Dong Wang Kai Cao | 2013 | Neural Regeneration Research2013,8,14: | 2 |

| 7 | Proliferation and differentiation of reactive nestin^+/GFAP^+ cells in an adult rat model of compression-induced spinal cord injury显示文摘BACKGROUND:Studies have demonstrated that astrocytes may possess similar properties to neural stem cells/neural precursor cells and have the potential to differentiate into neurons. OBJECTIVE:To observe neuroepithelial stem cell protein(nestin) and glial fibrillary acidic protein (GFAP) expression following spinal cord injury,and to explore whether nestin^+/GFAP^+ cells,which are detected at peak levels in gray and white matter around the ependymal region of the central canal in injured spinal cord,possess similar properties of neural stem cells. DESIGN,TIME AND SETTING:A randomized,controlled experiment.The study was performed at the Key Laboratory of Environment and Genes Related to Diseases(Xi'an Jiaotong University), Ministry of Education between January 2004 and December 2006. MATERIALS:Rabbit anti-rat nestin,β-tubulinⅢ,mouse anti-rat GFAP,galactocerebroside(GaLC) antibodies were utilized,as well as flow cytometry. METHODS:A total of 60 male,Sprague Dawley rats,aged 8 weeks,were randomly assigned to control(n = 12) and model(n = 48) groups.The spinal cord injury model was established in the model group by aneurysm clip compression,while the control animals were not treated.The gray and white matter around the ependymal region of the central canal exhibited peak expression of nestin^+/GFAP^+ cells.These cells were harvested and prepared into single cell suspension,followed by primary and passage cultures.The cells were incubated with serum-containing neural stem cell complete medium. MAIN OUTCOME MEASURES:Nestin and GFAP expression in injured spinal cord was determined using immunohistochemistry and double-labeled immunofluorescence at 1,3,5,7,14,28,and 56 days post-injury.In addition,cell proliferation and differentiation were detected using immunofluorescence cytochemistry and flow cytometry. RESULTS:Compared with the control group,the model group exhibited significantly increased nestin and GFAP expression(P<0.05),which reached peak levels between 3 and 7 days.The majority of cells in the ependymal region around the central canal were nestin^+/GFAP^- cells,while the gray and white matter around the ependymal region were full of nestin^+/GFAP^+ cells,with an astrocytic-like appearance.A large number of nestin^+/GFAP^+ cells were observed in the model group cell culture,and the cells formed clonal spheres and displayed strong nestin-positive immunofluorescence staining.Following induced differentiation,a large number of GaLC-nestin,β-tubulinⅢ-nestin,and GFAP-nestin positive cells were observed.However,no obvious changes were seen in the control group.Cells in S stage,as well as the percentage of proliferating cells,in the model group were significantly greater than in the control group(P<0.01). The combination of p75 neurotrophin receptor (p75NTR) andprecursor of nerve growth factor (pro-NGF) expression initiates the apoptotic pathway and inducesneuronal apoptosis. However, few reports have focused on the p75-mediated mechanism ofneuronal apoptosis following cauda equine compression injuryOBJECTIVE: To determine apoptosis of spinal cord neurons and activation of thepro-NGF-p75NTR-JNK(c-Jun N-terminal kinase) signal pathway in rats following cauda equinacompression, and to verify experimental outcomes.DESIGN, TIME AND SETTING: A randomized, controlled, in vivo experiment was performed at theMedical Experimental Center of Xi'an Jiaotong University between April and November in 2008.MATERIALS: Streptavidin-perosidase kit was purchased from Wuhan Boster, China; in situ endlabeling detection kit was provided by Promega, USA; type AEG-220G electron microscope waspurchased from Hitachi, Japan.METHODS: A total of 48 healthy, adult, female, Sprague Dawley rats were randomly assigned tothree groups: normal (n = 6), sham-surgery (n = 6), and compression (n = 36). The compressiongroup was randomly assigned to six subsets at 1,3, 5, 7, 14, and 28 days, respectively, with 6 rats ineach subset. A cylindrical silica gel stick was implanted into the rats to compress 75% of thevertebral canal in the compression group; in the sham-surgery group, only vertebral resection wasperformed; and no procedures were performed in the normal group.MAIN OUTCOME MEASURES: At 1,3, 5, 7, 14, and 28 days following compression, L_(2-3) spinalcord segments were processed for immunohistochemistry, in situ cell apoptosis detection, andtransmission electron microscopy observation. Nissl staining was used to observe neuronal survivalin the L_2 spinal cord segment. Immunohistochemistry was applied to detect expressions of pro-NGF,p75NTR, and JNK in the L_2 segment. TUNEL fluorometric method was used to observe apoptosis ofneurons in the L_2 segment.RESULTS: In the normal and sham-surgery groups, little neuronal apoptosis was observed in theL_(2-3) spinal cord segment. At 3 days after compression injury, pro-NGF, p75NTR and JNK expressionwas observed in the spinal cord. Expression levels reached a peak at 7 days, and then graduallydecreased. In the compression and sham-surgery groups, neurons primarily expressed pro-NGFand p75NTR. The number of JNK-positive neurons in the compression group was dramaticallyincreased compared with the sham-surgery group (P < 0.05). A few neurons were apoptotic in thespinal cord 1 day after compression injury. The number of apoptotic neurons gradually increasedand reached a peak at 7 days, and subsequently decreased. Apoptosis was still detectable at 28days. There was a positive correlation between p75NTR expression and neuronal apoptosis(r= 0.75, P< 0.05).CONCLUSION: Following cauda equina compression injury, apoptosis of spinal cord neurons wasobserved. | 10 | Population genomic analysis reveals distinct demographics and recent adaptation in the black flying fox(Pteropus alecto)显示文摘As the only mammalian group capable of powered flight,bats have many unique biological traits.Previous comparative genomic studies in bats have focused on long-term evolution.However,the microevolutionary processes driving recent evolution are largely under-explored.Using resequencing data from 50 black flying foxes(Pteropus alecto),one of the model species for bats,we find that black flying fox has much higher genetic diversity and lower levels of linkage disequilibrium than most of the mammalian species.Demographic inference reveals strong population fluctuations(>100 fold)coinciding with multiple historical events including the last glacial change and Toba super eruption,suggesting that the black flying fox is a very resilient species with strong recovery abilities.While long-term adaptation in the black flying fox is enriched in metabolic genes,recent adaptation in the black flying fox has a unique landscape where recently selected genes are not strongly enriched in any functional category.The demographic history and mode of adaptation suggest that black flying fox might be a well-adapted species with strong evolutionary resilience.Taken together,this study unravels a vibrant landscape of recent evolution for the black flying fox and sheds light on several unique evolutionary processes for bats comparing to other mammalian groups. | Haopeng He Hechuan Yang Randy Foo Wharton Chan Feng Zhu Yunsong Liu Xuming Zhou Liang Ma Lin-Fa Wang Weiwei Zhai | 2023 | Journal of Genetics and Genomics2023,50,8: | 0 |
