| 1 | The role of the HIF-1α/ALYREF/PKM2 axis in glycolysis and tumorigenesis of bladder cancer显示文摘Background:As a rate-limiting enzyme of glycolysis,pyruvate kinase muscle isozyme M2(PKM2)participates in tumor metabolism and growth.The regulatory network of PKM2 in cancer is complex and has not been fully studied in bladder cancer.The 5-methylcytidine(m5C)modification in PKM2 mRNA might participate in the pathogenesis of bladder cancer and need to be further clarified.This study aimed to investigate the biological function and regulatory mechanism of PKM2 in bladder cancer.Methods:The expression of PKM2 and Aly/REF export factor(ALYREF)was measured by Western blotting,qRT-PCR,and immunohistochemistry.The bioprocesses of bladder cancer cells were demonstrated by a series of experiments in vitro and in vivo.RNA immunoprecipitation,RNA-sequencing,and dualluciferase reporter assays were conducted to explore the potential regulatory mechanisms of PKM2 in bladder cancer.Results:In bladder cancer,we first demonstrated that ALYREF stabilized PKM2 mRNA and bound to its m5C sites in 3′-untranslated regions.Overexpression of ALYREF promoted bladder cancer cell proliferation by PKM2-mediated glycolysis.Furthermore,high expression of PKM2 and ALYREF predicted poor survival in bladder cancer patients.Finally,we found that hypoxia-inducible factor-1alpha(HIF-1α)indirectly up-regulated the expression of PKM2 by activating ALYREF in addition to activating its transcription directly.Conclusions:The m5C modification in PKM2 mRNA in the HIF-1α/ALYREF/PKM2 axis may promote the glucose metabolism of bladder cancer,providing a new promising therapeutic target for bladder cancer. | Jing-Zi Wang Wei Zhu Jie Han Xiao Yang Rui Zhou Hong-Cheng Lu Hao Yu Wen-Bo Yuan Peng-Chao Li Jun Tao Qiang Lu Ji-Fu Wei Haiwei Yang | 2021 | Cancer Communications2021,41,7: | 17 |

| 3 | Exosome-mediated metabolic reprogramming:the emerging role in tumor microenvironment remodeling and its influence on cancer progression显示文摘Metabolic reprogramming is reported to be one of the hallmarks of cancer,which is an adaptive mechanism by which fast-growing cancer cells adapt to their increasing energy demands.Recently,extracellular vesicles(EVs)known as exosomes have been recognized as crucial signaling mediators in regulating the tumor microenvironment(TME).Meanwhile,the TME is a highly heterogeneous ecosystem incorporating cancer cells,fibroblasts,adipocytes,endothelial cells,mesenchymal stem cells,and extracellular matrix.Accumulated evidence indicates that exosomes may transfer biologically functional molecules to the recipient cells,which facilitate cancer progression,angiogenesis,metastasis,drug resistance,and immunosuppression by reprogramming the metabolism of cancer cells and their surrounding stromal cells.In this review,we present the role of exosomes in the TME and the underlying mechanism of how exosomes exacerbate tumor development through metabolic reprogramming.In addition,we will also discuss the potential role of exosomes targeting metabolic process as biomarkers for tumor diagnosis and prognosis,and exosomes-mediated metabolic reprogramming as potential targets for cancer therapy.Furthermore,a better understanding of the link between exosomes and metabolic reprogramming,and their impact on cancer progression,would provide novel insights for cancer prevention and treatment in the future. | Enli Yang Xuan Wang Zhiyuan Gong Miao Yu Haiwei Wu Dongsheng Zhang | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 7 |
| 4 | Genotypic variation in phosphorus acquisition from sparingly soluble P sources is related to root morphology and root exudates in Brassica napus显示文摘Genotypic variations in the adaptive response to low-phosphorus(P) stress and P-uptake efficiency have been widely reported in many crops.We conducted a pot experiment to evaluate the P-acquisition ability of two rapeseed(Brassica napus) genotypes supplied with two sparingly soluble sources of P,Al-P and Fe-P.Then,the root morphology,proton concentrations,and carboxylate content were investigated in a solution experiment to examine the genotypic difference in P-acquisition efficiency.Both genotypes produced greater biomass and accumulated more P when supplied with Al-P than when supplied with Fe-P.The P-efficient genotype 102 showed a significantly greater ability to deplete sparingly soluble P from the rhizosphere soil because of its greater biomass and higher P uptake compared with those of the P-inefficient genotype 105.In the solution experiment,the P-efficient genotype under low-P conditions developed dominant root morphological traits,and it showed more intensive rhizosphere acidification because of greater H+ efflux,higher H+-ATPase activity,and greater exudation of carboxylates than the P-inefficient genotype.Thus,a combination of morphological and physiological mechanisms contributed to the genotypic variation in the utilization of different sparingly soluble P sources in B.napus. | ZHANG HaiWei HUANG Yu YE XiangSheng XU FangSen | 2011 | Science China(Life Sciences)2011,54,12: | 6 |

However, dissociation of the FMDV 146S particle in mildly acidic conditions renders inactivated foot-and-mouth disease(FMD) vaccines much less effective. Type Asia1 FMDV mutants with increased resistance to acid inactivation were selected to study the molecular basis of viral resistance to acid-induced disassembly and improve the acid stability of FMDV. Sequencing of capsid-coding regions revealed four amino acid replacements(VP1 N17D, VP2 H145Y, VP2 G192D, and VP3 K153E) in the viral population of the acid-selected 10th passage. | 15 | Multimodal 3D Convolutional Neural Networks for Classification of Brain Disease Using Structural MR and FDG-PET Images显示文摘The classification and identification of brain diseases with multimodal information have attracted increasing attention in the domain of computer-aided. Compared with traditional method which use single modal feature information, multiple modal information fusion can classify and diagnose brain diseases more comprehensively and accurately in patient subjects. Existing multimodal methods require manual extraction of features or additional personal information, which consumes a lot of manual work. Furthermore, the difference between different modal images along with different manual feature extraction make it difficult for models to learn the optimal solution. In this paper, we propose a multimodal 3D convolutional neural networks framework for classification of brain disease diagnosis using MR images data and PET images data of subjects. We demonstrate the performance of the proposed approach for classification of Alzheimer’s disease (AD) versus mild cognitive impairment (MCI) and normal controls (NC) on the Alzheimer’s Disease National Initiative (ADNI) data set of 3D structural MRI brain scans and FDG-PET images. Experimental results show that the performance of the proposed method for AD vs. NC, MCI vs. NC are 93.55% and 78.92% accuracy respectively.
