1Association of a genetic variant in AKT1 gene with features of the metabolic syndrome显示文摘Metabolic syndrome(MetS)is a clustering of metabolic abnormalities that is associated with increased risk of developing cardiovascular disease and type 2 diabetes.There is growing body of data showing the associations of genetic variants of the genes involved in the PI3K/AKT/mTOR pathway with diabetes and obesity.We aimed to investigate the association between MetS and its components with the genetic polymorphism in AKT1,rs1130233(T>C).Total of 618 participants,recruited from Mashhad stroke and heart atherosclerosis disorder cohort(MASHAD study).Patients with MetS were defined by using international diabetes federation(IDF)criteria(n Z 326)and those without MetS(n Z 261)were recruited.Anthropometric and biochemical parameters were measured in all subjects.Genetic analysis for the rs1130233 polymorphism was performed,using the ABI-StepOne instruments with SDS version-2.0 software.Individuals with MetS had a significantly higher levels of BMI,waistcircumference,total cholesterol,triglyceride,high sensitivity-c reactive protein(hs-CRP)and blood-pressure,and lower concentrations of high density lipoprotein(HDL-C),compared to non-MetS individuals(P<0.05).The association between the rs1130233 and MetS was not significant.Subjects with a CC or CT genotypes had a significantly higher serum hs-CRP-level(OR:1.5;95%CI(1.05e2.1),P Z 0.02).Additionally,subjects who carried the TC genotype had a higher BMI compared to the CC genotype(p value Z 0.045).Our findings demonstrated that AKT1,rs1130233(T>C)polymorphism was associated with major components of MetS such as hs-CRP,and BMI,indicating further investigation in a multi-center setting to explore its value as an emerging biomarker of risk stratification marker.Fateme Sadat Eshaghi Hamideh Ghazizadeh Sakine Kazami-Nooreini Ameneh Timar Habibollah Esmaeily Mehrane Mehramiz Amir Avan Majid Ghayour-Mobarhan 2019Genes & Diseases2019,6,3:6
2The Hippo Tumor Suppressor Pathway(YAP/TAZ/TEAD/MST/LATS)and EGFR-RAS-RAF-MEK in cancer metastasis显示文摘Hippo Tumor Suppressor Pathway is the main pathway for cell growth that regulates tissue enlargement and organ size by limiting cell growth.This pathway is activated in response to cell cycle arrest signals(cell polarity,transduction,and DNA damage)and limited by growth factors or mitogens associated with EGF and LPA.The major pathway consists of the central kinase of Ste20 MAPK(Saccharomyces cerevisiae),Hpo(Drosophila melanogaster)or MST kinases(mammalian)that activates the mammalian AGC kinase dmWts or LATS effector(MST and LATS).YAP in the nucleus work as a cofactor for a wide range of transcription factors involved in proliferation(TEA domain family,TEAD1-4),stem cells(Oct4 mononuclear factor and SMAD-related TGFb effector),differentiation(RUNX1),and Cell cycle/apoptosis control(p53,p63,and p73 family members).This is due to the diverse roles of YAP and may limit tumor progression and establishment.TEAD also coordinates various signal transduction pathways such as Hippo,WNT,TGFb and EGFR,and effects on lack of regulation of TEAD cancerous genes,such as KRAS,BRAF,LKB1,NF2 and MYC,which play essential roles in tumor progression,metastasis,cancer metabolism,immunity,and drug resistance.However,RAS signaling is a pivotal factor in the inactivation of Hippo,which controls EGFR-RAS-RAF-MEK-ERKmediated interaction of Hippo signaling.Thus,the loss of the Hippo pathway may have significant consequences on the targets of RAS-RAF mutations in cancer.Mohammad Reza Zinatizadeh Seyed Rouhollah Miri Peyman Kheirandish Zarandi Ghanbar Mahmoodi Chalbatani Catarina Raposo Hamid Reza Mirzaei Mohammad Esmaeil Akbari Habibollah Mahmoodzadeh 2021Genes & Diseases2021,8,1:5
3大豆提取物抗戊四唑所致卵巢切除大鼠痉挛的作用(英文)显示文摘目的:研究大豆提取物抗戊四唑所致卵巢切除大鼠痉挛的作用。方法:雌性Wistar大鼠随机分为4组(n=15),分别为假手术组、卵巢切除组、低剂量大豆提取物组和高剂量大豆提取物组。假手术组大鼠行假手术,其余3组大鼠行卵巢切除术。每组大鼠再随机分为2个亚组,其中一个亚组(n=7)每日予低剂量戊四唑(40mg/kg)腹腔内注射,连续14d;另一亚组(n=8)予一次高剂量戊四唑(90mg/kg)腹腔内注射。在戊四唑注射前30min,低剂量大豆提取物(20mg/kg)及高剂量大豆提取物(60mg/kg)组分别腹腔内注射大豆提取物,假手术组及卵巢切除组注射生理盐水。上述步骤完成后,将大鼠置于树脂玻璃箱中观察60min。结果:重复低剂量注射戊四唑14d,卵巢切除组大鼠的痉挛评分在第3、5、8、10、11、12及13天低于假手术组(P<0.05或P<0.01),而低剂量大豆提取物组和高剂量大豆提取物组大鼠的痉挛评分高于卵巢切除组(P<0.05或P<0.01)。一次性高剂量注射戊四唑后,与假手术组相比,卵巢切除组大鼠阵发性强直性抽搐潜伏期延长,而最小阵发性痉挛潜伏期无明显变化;与卵巢切除组相比,低剂量及高剂量的大豆提取物显著缩短了大鼠阵发性强直性抽搐及最小阵发性痉挛的潜伏期(P<0.01)。结论:雌激素能够影响戊四唑所致大鼠痉挛的严重程度,而大豆中所含的植物雌激素也具有类似作用,这种作用还有待更多的研究予以证实。Ali Reza Ebrahimzadeh Bideskan Mahmoud Hosseini Toktam Mohammadpour Reza Karami Mehdi Khodamoradi Habibollah Nemati Karimooy Hassan Alavi 2011中西医结合学报2011,9,6:4

5An Investigation of the Effects of Curcumin on Anxiety and Depression in Obese Individuals:A Randomized Controlled Trial显示文摘Objective:To investigate the effectiveness of curcumin,a natural polyphenoiic compound with antioxidant and anti-inflammatory activities,on the frequency of symptoms of anxiety and depression in obese individuals.Methods:In this double blind,cross-over trial,30 obese subjects were randomized to receive either curcumin(1 g/day) or placebo for a period of 30 days.Following a wash-out interval of 2 weeks,each subject was crossed over to the alternative regimen for a further 30 days.Severity of anxiety and depression was assessed at baseline and at weeks 4,6 and 10 of the trial using the Beck Anxiety Inventory(BAI) and Beck Depression Inventory(BDI) scales,respectively.Results:Mean BAI score was found to be significantly reduced following curcumin therapy(P=0.03).However,curcumin supplementation did not exert any significant impact on BDI scores(P=0.7).Conclusion:Curcumin has a potential anti-anxiety effect in individuals with obesity.Habibollah Esmaily Amirhossein Sahebkar Mehrdad Iranshahi Shiva Ganjali Akram Mohammadi Gordon Ferns Majid Ghayour-Mobarhan 2015Chinese Journal of Integrative Medicine2015,21,5:2
