| 1 | Case of pediatric traditional serrated adenoma resected via endoscopic submucosal dissection显示文摘Traditional serrated adenoma(TSA)is a type of serrated polyp of the colorectum and is thought to be a precancerous lesion.There are three types of serrated polyps,namely,hyperplastic polyps,sessile serrated adenomas/polyps,and TSAs.TSA is the least common of the three types and accounts for about 5% of serrated polyps.Here we report a pediatric case of TSA that was successfully resected by endoscopic submucosal dissection(ESD).This rare case report describes a pediatric patient with no family history of colonic polyp who was admitted to our hospital with hematochezia.On colonoscopy,we found a polypoid lesion measuring 10 mm in diameter in the lower rectum.We selected ESD as a surgical option for en bloc resection,and histopathological examination revealed TSA.The findings in this case suggest that TSA with precancerous potential can occur in children,and that ESD is useful for treating this lesion. | Sonoko Kondo Hirohito Mori Noriko Nishiyama Takeo Kondo Ryuichi Shimono Hitoshi Okada Takashi Kusaka | 2017 | World Journal of Gastroenterology2017,23,24: | 4 |

| 5 | The KDM4B–CCAR1–MED1 axis is a critical regulator of osteoclast differentiation and bone homeostasis显示文摘Bone undergoes a constant and continuous remodeling process that is tightly regulated by the coordinated and sequential actions of bone-resorbing osteoclasts and bone-forming osteoblasts.Recent studies have shown that histone demethylases are implicated in osteoblastogenesis;however,little is known about the role of histone demethylases in osteoclast formation.Here,we identified KDM4B as an epigenetic regulator of osteoclast differentiation.Knockdown of KDM4B significantly blocked the formation of tartrate-resistant acid phosphatase-positive multinucleated cells.Mice with myeloid-specific conditional knockout of KDM4B showed an osteopetrotic phenotype due to osteoclast deficiency.Biochemical analysis revealed that KDM4B physically and functionally associates with CCAR1 and MED1 in a complex.Using genome-wide chromatin immunoprecipitation(ChIP)-sequencing,we revealed that the KDM4B–CCAR1–MED1 complex is localized to the promoters of several osteoclast-related genes upon receptor activator of NF-κB ligand stimulation.We demonstrated that the KDM4B–CCAR1–MED1 signaling axis induces changes in chromatin structure(euchromatinization)near the promoters of osteoclast-related genes through H3K9 demethylation,leading to NF-κB p65 recruitment via a direct interaction between KDM4B and p65.Finally,small molecule inhibition of KDM4B activity impeded bone loss in an ovariectomized mouse model.Taken together,our findings establish KDM4B as a critical regulator of osteoclastogenesis,providing a potential therapeutic target for osteoporosis. | Sun-Ju Yi You-Jee Jang Hye-Jung Kim Kyubin Lee Hyerim Lee Yeojin Kim Junil Kim Seon Young Hwang Jin Sook Song Hitoshi Okada Jae-Il Park Kyuho Kang Kyunghwan Kim | 2021 | Bone Research2021,9,3: | 1 |
| 6 | BCR–ABL-specific CD4^(+) T-helper cells promote the priming of antigen-specific cytotoxic T cells via dendritic cells显示文摘The advent of tyrosine kinase inhibitor(TKI)therapy markedly improved the outcome of patients with chronic-phase chronic myeloid leukemia(CML).However,the poor prognosis of patients with advanced-phase CML and the lifelong dependency on TKIs are remaining challenges;therefore,an effective therapeutic has been sought.The BCR–ABL p210 fusion protein’s junction region represents a leukemia-specific neoantigen and is thus an attractive target for antigen-specific T-cell immunotherapy.BCR–ABL p210 fusion-region-specific CD4+T-helper(Th)cells possess antileukemic potential,but their function remains unclear.In this study,we established a BCR–ABL p210 b3a2 fusion-region-specific CD4+Th-cell clone(b3a2-specific Th clone)and examined its dendritic cell(DC)-mediated antileukemic potential.The b3a2-specific Th clone recognized the b3a2 peptide in the context of HLA-DRB1*09:01 and exhibited a Th1 profile.Activation of this clone through T-cell antigen receptor stimulation triggered DC maturation,as indicated by upregulated production of CD86 and IL-12p70 by DCs,which depended on CD40 ligation by CD40L expressed on b3a2-specific Th cells.Moreover,in the presence of HLA-A*24:02-restricted Wilms tumor 1(WT1)235–243 peptide,DCs conditioned by b3a2-specific Th cells efficiently stimulated the primary expansion of WTI-specific cytotoxic T lymphocytes(CTLs).The expanded CTLs were cytotoxic toward WT1235–243-peptide-loaded HLA-A*24:02-positive cell lines and exerted a potent antileukemic effect in vivo.However,the b3a2-specific Th-clone-mediated antileukemic CTL responses were strongly inhibited by both TKIs and interferon-α.Our findings indicate a crucial role of b3a2-specific Th cells in leukemia antigen-specific CTL-mediated immunity and provide an experimental basis for establishing novel CML immunotherapies. | Norihiro Ueda Rong Zhang Minako Tatsumi Tian-YiLiu Shuichi Kitayama YutakaYasui Shiori Sugai Tatsuaki Iwama Satoru Senju Seiji Okada Tetsuya Nakatsura Kiyotaka Kuzushima Hitoshi Kiyoi Tomoki Naoe Shin Kaneko Yasushi Uemura | 2018 | Cellular & Molecular Immunology2018,15,1: | 1 |

| 17 | Operation and Consideration of a Pipe Corrosion Inspection System Based on Human-in-the-Loop Machine Learning显示文摘The aim of this study is to improve the efficiency of external corrosion inspection of pipes in chemical plants.Currently,the preferred method involves manual inspection of images of corroded pipes;however,this places significant workload on human experts owing to the large number of required images.Furthermore,visual assessment of corrosion levels is prone to subjective errors.To address these issues,we developed an AI(artificial intelligence)-based corrosion-diagnosis system(AI corrosion-diagnosis system)and implemented it in a factory.The proposed system architecture was based on HITL(human-in-the-loop)ML(machine learning)[1].To overcome the difficulty of developing a highly accurate ML model during the PoC(proof-of-concept)stage,the system relies on cooperation between humans and the ML model,utilizing human expertise during operation.For instance,if the accuracy of the ML model was initially 60%during the development stage,a cooperative approach would be adopted during the operational stage,with humans supplementing the remaining 40%accuracy.The implemented system’s ML model achieved a recall rate of approximately 70%.The system’s implementation not only contributed to the efficiency of operations by supporting diagnosis through the ML model but also facilitated the transition to systematic data management,resulting in an overall workload reduction of approximately 50%.The operation based on HITL was demonstrated to be a crucial element for achieving efficient system operation through the collaboration of humans and ML models,even when the initial accuracy of the ML model was low.Future efforts will focus on improving the detection of corrosion at elevated locations by considering using video cameras to capture pipe images.The goal is to reduce the workload for inspectors and enhance the quality of inspections by identifying corrosion locations using ML models. | Toshihiro Shimbo Yousuke Okada Hitoshi Matsubara | 2023 | Journal of Mechanics Engineering and Automation2023,13,5: | 0 |
