| 4 | Construction of recombinant plasmid pEGFP-C2-L539fs/47 and its expression in HEK293 cells显示文摘Objective:To reconstruct pEGFP-C2-L539fs/47,a HERG nonsense mutant in eukaryotic expression plasmid,and observe the fusion protein expressed in HEK293 cells(human embryo kidney cells).Methods:After double digestion of pcDNA3-L539fs/47 and pEGFP-C2-HERG with sbf I and Eco91 I,the small product fragment,from pcDNA3-L539fs/47,was subcloned into the big fragment of pEGFP-C2-HERG under T4 ligase.pEGFP-C2-L539fs/47 was identified by agarose gel electrophoresis and sequencing.pcDNA3-L539fs/47 and pEGFP-C2-L539fs/47 were transiently transfected into HEK293 cells by Lipofect,respectively.The expression of fusion protein in HEK293 cells was detected through immunofluorescence,laser confocal imaging scanning in vivo,Western blot and PCR.Results:Mutation region cDNA fragment(about 1 kb) and target vector fragment(about 7.2 kb) were ligated after purification and gel recovery.Agarose gel electrophoresis and sequencing successfully demonstrated eukaryotic expression plasmid pEGFP-C2-L539fs/47,constructed approximately 8.2 kb,sequencing consistent with template gene.The transfection efficiency of recombinant plasmid by fluorescence microscopy was more than60%.Western blot analysis detected pcDNA3-L539fs/47 expression of the protein size 60 KD,the expression of pEGFP-C2 fusion protein size of approximately 90 KD.The L539fs/47 gene expression in HEK293 cells was significant by PCR analysis.Confocal laser imaging showed that pEGFP-C2-L539fs/47 protein was successfully expressed in cytoplasm and cytomembrane of HEK293 cells.Conclusion:pEGFP-C2-L539fs/47 containing the HERG gene mutant was successfully constructed by double digestion method and expressed fusion protein in HEK293 cells,which laid a foundation for the further study on L539fs/47. | Lue Ying Zhang Aifeng Han Wenqi Li Guoliang Zhang Junbo Gao Jie Pan Junqiang Zhang Yong Sun Chaofeng | 2012 | Journal of Medical Colleges of PLA(China)2012,27,3: | 2 |
| 5 | Experimental realization to efficiently sort vector beams by polarization topological charge via Pancharatnam–Berry phase modulation显示文摘This paper reports the experimental realization of efficiently sorting vector beams by polarization topological charge(PTC). The PTC of a vector beam can be defined as the repetition number of polarization state change along the azimuthal axis, while its sign stands for the rotating direction of the polarization. Here, a couple of liquid crystal Pancharatnam–Berry optical elements(PBOEs) have been used to introduce conjugated spatial phase modulations for two orthogonal circular polarization states. Applying these PBOEs in a 4-f optical system,our experiments show the setup can work for PTC sorting with a separation efficiency of more than 58%. | 6 | Generation and characterisation of rabbit monoclonal antibodies against the native cell surface antigens of embryonic stem cells显示文摘Embryonic stem (ES) cells are potent resources for cell therapy,and monoclonal antibodies (mAbs) against native cell surface markers of ES cells could be useful tools for therapeutic applications.Here,we report the development of a feasible approach,which could be used in mass production,for experimentally producing rabbit mAbs against native cell surface antigens on the cell surface.Two of the 14 mAbs,which were selected at random,could be bound to the cell surface antigens of mES cells.The immunocytochemistry (ICC) and Western blot results showed that mAb 39 recognises conformational epitopes.The target antigen of mAb 39 was then successfully purified using an improved immunoprecipitation approach in which mAb was bounded to intact mES cells before the cells were lysed.The LC-LTQ mass spectrum analysis showed that the target antigen of mAb 39 was Glut3.This result was further confirmed by Western blot using commercially available antibodies against Glut3.Further experiments showed that mAb 39 exhibited an antiproliferative effect on mES cells.We also found that Glut3 was differentially expressed among the mES cell population as detected by flow cytometry. | Yongliang Lu Zhongyuan Su Ying Li Jingfeng Luo Zhou Tan Huijiao Ji Weimin Zhu Guoliang Yu Liangbiao Chen Ming Zhang | 2010 | Journal of Genetics and Genomics2010,37,7: | 1 |
| 7 | The development of Bi Fe O_3-based ceramics显示文摘The multiferroic properties of Bi Fe O3-based ceramics were improved through optimizing their sintering method and doping with certain rare earth elements in pure Bi Fe O3. Some methods, especially liquid-phase sintering method has largely decreased the densities of oxygen vacancies and Fe2in Bi Fe O3-based ceramics, and thus their resistivity became high enough to measure the saturated polarization and the large piezoelectric d33 coefficient under the high electric field of [150 k V/cm. Besides,multiferroic properties were improved through the rare earth elements' doping in pure Bi Fe O3. Magnetization commonly increases with the proportional increase of Nd,La, Sm and Dy contents up to *30 %, while ferroelectric phase can transform to paraelectric phase at a certain proportion. An improved magnetoelectric coupling was often observed at ferroelectric phase with a relatively large proportion. | 12 | Serum carbohydrate antigen-125 is elevated in patients with chronic atrial fibrillation显示文摘Objective: To determine whether CA-125 is elevated in medically stable patients with chronic atrial fibrillation (AF) compared with patients without AF and to examine whether levels of CA-125 are associated with demographic and clinical variables in a sample of patients under study. Methods: 55 patients with chronic AF and 58 control patients in sinus rhythm were included into the study. Patients with acute heart failure (HF), chronic inflammatory or neoplastic disease were excluded from the study. Circulating levels of CA-125 were assessed; all patients underwent clinical examination, assessment and medical records including demographic data, history of comorbid conditions, current use of cardiac medications, and the results of cardiac tests including electrocardiograms. Results: The mean age of the study sample was 53.2±6.5 years and 48% were men. Patients with sinus rhythm were significantly more likely to have lower heart rates, smaller dimensions of left atrium, and to be treated with aspirin. Coumadin, coumadin and digoxin were more often prescribed in patients with chronic AF. The CA-125 levels were significantly higher in patients with chronic AF than in patients in sinus rhythm (48.5±7.65 U/ml and 28.43±5.48 U/ml, P<0.005). An inverse relation was found between CA-125 levels and left ventricular ejection fraction (LVEF) (r=-0.48, P<0.001). CA-125 was significantly related to the left atrium (LA) diameter, left ventricular end-diastolic diameter (LVEDD), left ventricular end-systolic diameter (LVESD) and brain natriuretic peptide (BNP). There was no significant correlation between CA-125 and age.
