A reactive oxygen species burst causes haploid induction in maize显示文摘Haploid induction (HI) is an important tool in crop breeding. Phospholipase A1 (ZmPLA1)/NOT LIKE DAD (NLD)/MATRILINEAL (MTL) is a key gene controlling HI in maize;however, the underlying molecular mechanism remains unclear. In this study, to dissect why loss of ZmPLA1 function could mediate HI we performed a comprehensive multiple omics analysis of zmpla1 mutant anthers by integrating transcriptome, metabolome, quantitative proteome, and protein modification data. Functional classes of significantly enriched or differentially abundant molecular entities were found to be associated with the oxidative stress response, suggesting that a reactive oxygen species (ROS) burst plays a critical role in HI. In support of this, we further discovered that a simple chemical treatment of pollen with ROS reagents could lead to HI. Moreover, we identified ZmPOD65, which encodes a sperm-specific peroxidase, as a new gene controlling HI. Taken together, our study revealed a likely mechanism of HI, discovered a new gene controlling HI, and created a new method for HI in maize, indicating the importance of ROS balance in maintaining normal reproduction and providing a potential route to accelerate crop breeding.Chenglin Jiang Ju Sun Rui Li Shijuan Yan Wei Chen Liang Guo Guochen Qin Pengcheng Wang Cheng Luo Wenjie Huang Qinghua Zhang Alisdair R.Fernie David Jackson Xiang Li Jianbing Yan 2022Molecular Plant2022,15,6:2
Localized in‑situ deposition:a new dimension to control in fabricating surface micro/nano structures via ultrafast laser ablation显示文摘Controllable fabrication of surface micro/nano structures is the key to realizing surface functionalization for various applications. As a versatile approach, ultrafast laser ablation has been widely studied for surface micro/nano structuring.Increasing research eforts in this feld have been devoted to gaining more control over the fabrication processes to meet the increasing need for creation of complex structures. In this paper,we focus on the in-situ deposition process following the plasma formation under ultrafast laser ablation. From an overview perspective,we frstly summarize the diferent roles that plasma plumes, from pulsed laser ablation of solids, play in diferent laser processing approaches. Then, the distinctive in-situ deposition process within surface micro/nano structuring is highlighted. Our experimental work demonstrated that the in-situ deposition during ultrafast laser surface structuring can be controlled as a localized micro-additive process to pile up secondary ordered structures, through which a unique kind of hierarchical structure with fort-like bodies sitting on top of micro cone arrays were fabricated as a showcase. The revealed laser-matter interaction mechanism can be inspiring for the development of new ultrafast laser fabrication approaches, adding a new dimension and more fexibility in controlling the fabrication of functional surface micro/nano structures.Peixun Fan Guochen Jiang Xinyu Hu Lizhong Wang Hongjun Zhang Minlin Zhong 2023Frontiers of Optoelectronics2023,16,4:0
A novel protein complex that regulates active DNA demethylation in Arabidopsis显示文摘Active DNA demethylation is critical for altering DNA methylation patterns and regulating gene expression. The 5-methylcytosine DNA glycosylase/lyase ROS1 initiates a base-excision repair pathway for active DNA demethylation and is required for the prevention of DNA hypermethylation at 1000 s of genomic regions in Arabidopsis. How ROS1 is regulated and targeted to specific genomic regions is not well understood. Here,we report the discovery of an Arabidopsis protein complex that contains ROS1,regulates ROS1 gene expression, and likely targets the ROS1 protein to specific genomic regions.ROS1 physically interacts with a WD40 domain protein(RWD40),which in turn interacts with a methyl-DNA binding protein(RMB1)as well as with a zinc finger and homeobox domain protein(RHD1).RMB1 binds to DNA that is methylated in any sequence context, and this binding is necessary for its function in vivo.Loss-of-function mutations in RWD40,RMB1,or RHD1 cause DNA hypermethylation at several tested genomic regions independently of the known ROS1 regulator IDM1.Because the hypermethylated genomic regions include the DNA methylation monitoring sequence in the ROS1 promoter, plants mutated in RWD40,RMB1,or RHD1 show increased ROS1 expression.Importantly,ROS1 binding to the ROS1 promoter requires RWD40,RMB1,and RHD1,suggesting that this complex dictates ROS1 targeting to this locus. Our results demonstrate that ROS1 forms a protein complex with RWD40,RMB1,and RHD1,and that this novel complex regulates active DNA demethylation at several endogenous loci in Arabidopsis.Pan Liu Wen-Feng Nie Xiansong Xiong Yuhua Wang Yuwei Jiang Pei Huang Xueqiang Lin Guochen Qin Huan Huang Qingfeng Niu Jiamu Du Zhaobo Lang Rosa Lozano-Duran Jian-Kang Zhu 2021Journal of Integrative Plant Biology2021,63,4:0
Porous reduced graphene oxide for ultrasensitive detection of nitrogen dioxide显示文摘The defect engineering in graphene plays a significant role for the application of gas sensors. In this work, we proposed an efficient method to prepare ultrasensitive gas sensors based on the porous reduced graphene oxide(PRGO). Photo-Fenton etching was carried out on GO nanosheets in a controlled manner to enrich their vacancy defects. The resulting porous graphene oxide(PGO) was then drop-coated on interdigital electrodes and hydrothermal reduced at 180 °C. Controllable reduction was achieved by varying the water amount. The gas sensor based on PRGO-5 min-6h exhibited superior sensing and selective performance toward nitrogen dioxide(NO2), with an exceptional high sensitivity up to 12 ppm-1.The theoretical limit of detection is down to 0.66 ppb. The excellent performance could be mainly attributed to the typical vacancy defects of PRGO. Some residue carboxylic groups on the edges could also facilitate the adsorption of polar molecules. The process has a great potential for scalable fabrication of high-performance NO2gas sensors.Zengyong Chu Min Xiao Qichao Dong Guochen Li Tianjiao Hu Ye Zhang Zhenhua Jiang 2023Chinese Chemical Letters2023,34,1:0
Superomniphobic surfaces for easy-removals of environmentalrelated liquids after icing and melting显示文摘Superhydrophobic surfaces often lose the easy-removal ability of liquids during icing&melting cycles due to the impalement phenomena of air pockets.Especially for the most common mixed liquids in normal life, their difficult-removals after icing and melting have brought colossal troubles in the fields of aviation, energy, biomedicine, etc. Here we adopt the ultrafast laser to fabricate the optimal micro-nanostructured surfaces, realizing excellent superomniphobicity for seven environmental-related liquids. It is demonstrated that different droplets on the surfaces recover well to the original Cassie-Baxter state after melting, and can be removed easily at low tilted angles. The ice adhesion strengths of the seven liquids as low as 5 kPa and the micro-nanostructure durability ensure the long-term easy-removal after icing.Compared with the ice adhesion strength of untreated surfaces(264.4±17.6 kPa),those of our designed surfaces have decreased by over 50 times.Icing and melting processes are investigated to reveal the easy-removal mechanisms that specifically distributed solutes and bubbles after icing impact downwards significantly to accelerate the recovery of the Cassie–Baxter state during melting.A series of environmental-related durability experiments including continuous icing&melting cycles,long-term salt spray, and high-pressure water jet impact further demonstrate the surfaces promising for real applications.Lizhong Wang Ze Tian Xiao Luo Changhao Chen Guochen Jiang Xinyu Hu Rui Peng Hongjun Zhang Minlin Zhong 2023Nano Research2023,16,2:0
