| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 2 | The influence of DC stray current on pipeline corrosion显示文摘DC stray current can cause severe corrosion on buried pipelines.In this study,firstly,we deduced the equation of DC stray current interference on pipelines.Next,the cathode boundary condition was discretized with pipe elements,and corresponding experiments were designed to validate the mathematical model.Finally,the numerical simulation program BEASY was used to study the corrosion effect of DC stray current that an auxiliary anode bed generated in an impressed current cathodic protection system.The effects of crossing angle,crossing distance,distance of the two pipelines,anode output current,depth,and soil resistivity were investigated.Our results indicate that pipeline crossing substantially affects the corrosion potential of both protected and unprotected pipelines.Pipeline crossing angles,crossing distances,and anode depths,our results suggest,have no significant influence.Decreasing anode output current or soil resistivity reduces pipeline corrosion gradually.A reduction of corrosion also occurs when the distance between two parallel pipelines increases. | Gan Cui Zi-Li Li Chao Yang Meng Wang | 2016 | Petroleum Science2016,13,1: | 18 |

| 4 | Statistical scenarios forecasting method for wind power ramp events using modified neural networks显示文摘Wind power ramp events increasingly affect the integration of wind power and cause more and more problems to the safety of power grid operation in recent years.Several forecasting techniques for wind power ramp events have been reported.In this paper,the statistical scenarios forecasting method is proposed for wind power ramp event probabilistic forecasting based on the probability generating model.Multi-objective fitness functions are established considering cumulative density functions and higher order moment autocorrelation functions with respect to the consistency of distribution and timing characteristics,respectively.Parameters of probability generating model are calculated by the iterative optimization using the modified genetic algorithm with multi-objective fitness functions.A number of statistical scenarios captured bands are generated accordingly.Eventually,ramp event probability characteristics are detected from scenarios captured bands to evaluate the ramp event forecasting method.A wind plant of Bonneville Power Administration with actual wind power data is selected for calculation and statistical analysis.It is shown that statistical results with multi-objective functions are more accurate than the results with single objective functions.Moreover,the statistical scenarios forecasting method can accurately estimate the characteristics of wind power ramp events.The results verify that the proposed method can guide the generation method of statistical scenarios and forecasting models for ramp events. | Mingjian CUI Deping KE Di GAN Yuanzhang SUN | 2015 | Journal of Modern Power Systems and Clean Energy2015,3,3: | 13 |
| 5 | An efficient methodology for utilization of K-feldspar and phosphogypsum with reduced energy consumption and CO2 emissions显示文摘The issues of reducing CO_2 emissions, sustainably utilizing natural mineral resources, and dealing with industrial waste offer challenges for sustainable development in energy and the environment. We propose an efficient methodology via the co-reaction of K-feldspar and phosphogypsum for the extraction of soluble potassium salts and recovery of SO_2 with reduced CO_2 emission and energy consumption. The results of characterization and reactivity evaluation indicated that the partial melting of K-feldspar and phosphogypsum in the hightemperature co-reaction significantly facilitated the reduction of phosphogypsum to SO_2 and the exchange of K^+(K-feldspar) with Ca^(2+)(CaSO_4 in phosphogypsum). The reaction parameters were systematically investigated with the highest sulfur recovery ratio of ~ 60% and K extraction ratio of ~ 87.7%. | 6 | A de novo Genome of a Chinese Radish Cultivar显示文摘Here, we report a high-quality draft genome of a Chinese radish(Raphanus sativus) cultivar. This draft contains 387.73 Mb of assembled scaffolds, 83.93% of the scaffolds were anchored onto nine pseudochromosomes and 95.09% of 43 240 protein-coding genes were functionally annotated. 184.75 Mb(47.65%) of repeat sequences was identified in the assembled genome. By comparative analyses of the radish genome against 10 other plant genomes, 2 275 genes in 780 gene families were found unique to R. sativus. | 7 | Achieving and maintaining biological nitrogen removal via nitrite under normal conditions显示文摘The principal aim of this paper is to develop an approach to realize stable biological nitrogen removal via nitrite under normal conditions. Validation of the new method was established on laboratory-scale experiments applying the sequencing batch reactor(SBR) activated sludge process to domestic wastewater with low C/N ratio. The addition of sodium chloride(NaCl) to influent was established to achieve nitrite build-up. The high nitrite accumulation, depending on the salinity in influent and the application duration of salt, was obtained in SBRs treating saline wastewater. The maintenance results indicated that the real-time SBRs can maintain stable nitrite accumulation, but conversion from shorter nitrification-denitrification to full nitrification-denitrification was observed after some operation cycles in the other SBR with fixed-time control. | 10 | Involvement of the Mitochondrion-dependent and the Endoplasmic Reticulum Stress-signaling Pathways in Isoliquiritigenin-induced Apoptosis of HeLa Cell显示文摘Objective Isoliquiritigenin(ISL),a licorice chalconoid,is considered to be a bioactive agent with chemopreventive potential.This study investigates the mechanisms involved in ISL-induced apoptosis in human cervical carcinoma HeLa cells.Methods Cell viability was evaluated using a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide(MTT)assay.Apoptosis was determined by flow cytometry using an Annexin V-FITC Apoptosis Detection Kit.The intracellular ROS levels were assessed using a 2,7-dichlorofluorescein probe assay.The mitochondrial membrane potential was measured with the dual-emission potential-sensitive probe 5,5',6,6'-tetra-chloro-1,1',3,3'-tetraethyl-imidacarbocyanine iodide(JC-1).The degradation of poly-ADP-ribose polymerase(PARP)protein,the phosphorylation of PKR-like ER kinase(PERK),the phosphorylation of the α-subunit of eukaryotic initiation factor 2(eIF2α),the expression of the 78 kD glucose-regulated protein(GRP 78),and the activation of caspase-12 were analyzed via western blot analysis.Results ISL significantly inhibited the proliferation,the increase in ROS levels and apoptotic rates of HeLa cells in a concentration-dependent manner.Moreover,ISL induced mitochondrial dysfunction,caspase activation,and PARP cleavage,which displayed features of mitochondria dependent on apoptotic signals.Besides,exposure of HeLa cells to ISL triggered endoplasmic reticulum(ER)stress,as indicated by the increase in p-eIF2α and GRP78 expression,ER stress-dependent apoptosis is caused by the activation of ER-specific caspase-12.Conclusion The findings from our study suggest that ISL-induced oxidative stress causes HeLa cell apoptosis via the mitochondrion-dependent and the ER stress-triggered signaling pathways. | YUAN Xuan ZHANG Bo GAN Lu WANG Zhen Hua YU Ba Cui LIU Liang Liang ZHENG Qiu Sheng WANGZhi Ping | 2013 | Biomedical and Environmental Sciences2013,26,4: | 8 |

| 15 | Mesoporous Ag nanocubes synthesized via selectively oxidative etching at room temperature for surface- enhanced Raman spectroscopy显示文摘Silver nanocubes enriched with {100} facets have been extensively used for surface-enhanced Raman scattering. Herein, we report a new water-phase synthesis method for weU-defined Ag nanocubes with tunable sizes via a two-step procedure at room temperature. First, irregularly shaped Ag nanoparticles (INPs) were prepared by reducing silver ammonia solution using ethylal. Second, the agglomerated INPs were selectively etched with HNO3 and C1- to yield {100} facet-rich mesoporous Ag nanocubes. The mechanism of Ag-nanocube formation and growth was investigated in detail by elucidating the involved chemical reactions and physical changes at each step during the synthesis. The addition of C1- anions was responsible for facilitating Ag nanoparticle growth by removing surface-adsorbed Ag+ species, thereby eliminating inter-particle repulsive forces. This agglomeration was found crucial for the subsequent selective oxidation of Ag nanoparticles because the protective agent used, polyvinylpyrrolidone (PVP), was the most effective one for adsorption on the surfaces of Ag nanoparticles of size greater than approximately 50 nm. Importantly mesopores were found inside the Ag nanocubes; this can be attributed to the unavoidable imperfect packing during the agglomeration of INPs. | 17 | Spatial modulation of nanopattern dimensions by combining interference lithography and grayscale-patterned secondary exposure显示文摘Functional nanostructures are exploited for a variety of cutting-edge fields including plasmonics,metasurfaces,and biosensors,just to name a few.Some applications require nanostructures with uniform feature sizes while others rely on spatially varying morphologies.However,fine manipulation of the feature size over a large area remains a substantial challenge because mainstream approaches to precise nanopatterning are based on low-throughput pixel-by-pixel processing,such as those utilizing focused beams of photons,electrons,or ions.In this work,we provide a solution toward wafer-scale,arbitrary modulation of feature size distribution by introducing a lithographic portfolio combining interference lithography(IL)and grayscale-patterned secondary exposure(SE).Employed after the high-throughput IL,a SE with patterned intensity distribution spatially modulates the dimensions of photoresist nanostructures.Based on this approach,we successfully fabricated 4-inch wafer-scale nanogratings with uniform linewidths of<5%variation,using grayscale-patterned SE to compensate for the linewidth difference caused by the Gaussian distribution of the laser beams in the IL.Besides,we also demonstrated a wafer-scale structural color painting by spatially modulating the filling ratio to achieve gradient grayscale color using SE. | Zhuofei Gan Hongtao Feng Liyang Chen Siyi Min Chuwei Liang Menghong Xu Zijie Jiang Zhao Sun Chuying Sun Dehu Cui Wen-Di Li | 2022 | Light(Science & Applications)2022,11,5: | 3 |
| 18 | Coordinated transcription of ANRIL and P16 genes is silenced by P16 DNA methylation显示文摘Objective: To investigate the relationship between the transcription of ANRIL, P15, P14 and P16 at the same locus and the regulation mechanism of ANRIL.Methods: Publicly available database of Cancer Cell Line Encyclopedia(CCLE) was used in bioinformatic analyses. Methylation of Cp G islands was detected by denaturing high performance liquid chromatography(DHPLC). Gene transcript levels were determined using quantitative real-time polymerase chain reaction(q RTPCR) assays. An engineered P16-specific transcription factor and DNA methyltransferase were used to induce P16-specific DNA demethylation and methylation.Results: The expression level of ANRIL was positively and significantly correlated with that of P16 but not with that of P15 in the CCLE database. This was confirmed in human cell lines and patient colon tissue samples. In addition, ANRIL was significantly upregulated in colon cancer tissues. Transcription of ANRIL and P16 was observed only in cell lines in which the P16 alleles were unmethylated and not in cell lines with fully methylated P16 alleles.Notably, P16-specific methylation significantly decreased transcription of P16 and ANRIL in BGC823 and GES1 cells. | 19 | Chronic inflammation deteriorates structure and function of collagen fibril in rat temporomandibular joint disc显示文摘Collagen is the building component of temporomandibular joint(TMJ) discs and is often affected by inflammation in temporomandibular disorders. The macromechanical properties of collagen are deteriorated by chronic inflammation. However,the mechanism by which inflammation influences disc function remains unknown. The relationship between the ultrastructure and nanomechanical properties of collagen in inflamed discs should be clarified. Seven-week-old female Sprague–Dawley rats were randomly divided into two groups. Chronic TMJ inflammation was induced by intra-articular injection of complete Freund's adjuvant, and samples were harvested after 5 weeks. Picrosirius staining revealed multiple colours under polarized light, which represented alternative collagen bundles in inflamed discs. Using atomic force microscopy scanning, the magnitude of Young's modulus was reduced significantly accompanied with disordered collagen fibril arrangement with porous architecture of inflamed discs. Transmission electron microscopy scanning revealed a non-uniform distribution of collagen fibres, and oversized collagen fibrils were observed in inflamed discs. Fourier transform infrared microspectroscopy revealed a decrease in 1 338 cm^(-1)/amide II area ratio of collagen in different regions. The peak positions of amide I and amide II bands were altered in inflamed discs,indicating collagen unfolding. | 20 | Protecting lithium metal anode in all-solid-state batteries with a composite electrolyte显示文摘The volume of the metallic lithium anode in allsolid-state Li metal batteries increases significantly due to the lithium dendrite formation during the battery cycling,and the rough surface of lithium metal also reduces Li-ion transport in Li/electrolyte interface.In this work,we developed a solid polymer composite by adding the lowcost Si_(3)N_(4)particles to protect the lithium anode in allsolid-state batteries.The Fourier transform infrared spectroscopy(FTIR)data show that the surface of 10 wt%Si_(3)N_(4)particles interacts with the polyethylene oxide(PEO)and lithium bis(trifluoromethanesulfonyl)imide(LiTFSI)salt;the interaction restricts the anion mobility and improves the ionic conductivity(1×10^(-4)S·cm^(-1))and lithium-ion transference number(0.28)of the composite electrolyte.The lithium metal anode is well protected by the composite electrolyte in all-solid-state cells,including symmetric and Li/LiFePO_(4)cells.The lithium dendrite growth suppression by this composite electrolyte indicates the possible application of these low-cost composite electrolytes for lithium metal protection. | Wen-Qing Wei Bing-Qiang Liu Yi-Qiang Gan Hai-Jian Ma Da-Wei Cui | 2021 | Rare Metals2021,40,2: | 2 |
