1Artificial intelligence in healthcare:past,present and future显示文摘Artificial intelligence(AI)aims to mimic human cognitive functions.It is bringing a paradigm shift to healthcare,powered by increasing availability of healthcare data and rapid progress of analytics techniques.We survey the current status of AI applications in healthcare and discuss its future.AI can be applied to various types of healthcare data(structured and unstructured).Popular AI techniques include machine learning methods for structured data,such as the classical support vector machine and neural network,and the modern deep learning,as well as natural language processing for unstructured data.Major disease areas that use AI tools include cancer,neurology and cardiology.We then review in more details the AI applications in stroke,in the three major areas of early detection and diagnosis,treatment,as well as outcome prediction and prognosis evaluation.We conclude with discussion about pioneer AI systems,such as IBM Watson,and hurdles for real-life deployment of AI.Fei Jiang Yong Jiang Hui Zhi Yi Dong Hao Li Sufeng Ma Yilong Wang Qiang Dong Haipeng Shen Yongjun Wang 2017Stroke & Vascular Neurology2017,2,4:76

3Expression of miR-125b in the new,highly invasive glioma stem cell and progenitor cell line SU3显示文摘MicroRNA (miR)-125b has been shown to play a potential role in the development of glioma stem cells.However,the relationship between miRNA and glioma stem cells is still elusive.This study was designed to elucidate this potential relationship.We established a highly invasive glioma stem cell and progenitor (GSCP) cell line SU3.SU3 cell suspensions were injected into nude mice brains in situ,and the invasiveness of graft tumors was analyzed using hematoxylin and eosin staining as well as immunohistochemistry.Real-time polymerase chain reaction (PCR) was used to measure the expression levels of miR-125b in SU3 and other cells.In vitro,SU3 cells expressed CD133 and nestin as well as differentiation markers glial fibrillary acidic protein (GFAP) and β-tubulin III,which were consistent with the characteristics of glioma stem cells.Scratch assays indicated that the migration ability of SU3 cells was stronger than that of U251 stem cells (U251s).In vivo,SU3 cells invaded into each part of the mouse brain from the caudate nucleus in a diffuse pattern and highly expressed invasive and proliferative cell markers matrix metalloprotease 2 (MMP2),MMP9,and Ki-67.Real-time PCR results revealed that the levels of miR-125b and MMP9 were significantly higher in SU3 and SU2,also a highly invasive GSCP cell line we established before,than in U251s.High expression of miR-125b both in newly established GSCPs,SU3,and long-term cultured GSCPs,SU2 suggests that miR-125b exhibits oncogene-like behavior.This behavior should be considered in further studies of miR-125b in cancer stem cells.Furthermore,MMP9,which plays a role in cancer stem cell invasion,may be a target gene of miR-125b.Yi Wan Xi-Feng Fei Zhi-Min Wang Dong-Yi Jiang Han-Chun Chen Jian Yang Lei Shi Qiang Huang 2012Chinese Journal of Cancer2012,31,4:25
4Nomogram to predict overall survival after gallbladder cancer resection in China显示文摘AIM To integrate clinically significant variables related to prognosis after curative resection for gallbladder carcinoma(GBC) into a predictive nomogram.METHODS One hundred and forty-two GBC patients who underwent curative intent surgical resection at Peking Union Medical College Hospital(PUMCH) were included. This retrospective case study was conducted at PUMCH of the Chinese Academy of Medical Sciences and Peking Union Medical College(CAMS & PUMC) in China from January 1, 2003 to January 1, 2018. The continuous variable carbohydrate antigen 19-9(CA19-9) was converted into a categorical variable(cCA19-9) based on the normal reference range. Stages 0 to IIIA were merged into one category, while the remaining stages were grouped into another category. Pathological grade X(GX) was treated as a missing value. A multivariate Cox proportional hazards model was used to select variables to construct a nomogram. Discrimination and calibration of the nomogram were performed via the concordance index(C-index) and calibration plots. The performance of the nomogram was estimated using the calibration curve. Receiver operating characteristic(ROC) curve analysis and decision curve analysis(DCA) were performed to evaluate the predictive accuracy and net benefit of the nomogram, respectively.RESULTS Of these 142 GBC patients, 55(38.7%) were male, and the median and mean age were 64 and 63.9 years, respectively. Forty-eight(33.8%) patients in this cohort were censored in the survival analysis. The median survival time was 20 months. A series of methods, including the likelihood ratio test and Akaike information criterion(AIC) as well as stepwise, forward, and backward analyses, were used to select the model, and all yielded identical results. Jaundice [hazard ratio(HR) = 2.9; 95% confidence interval(CI): 1.60-5.27], cCA19-9(HR = 3.2; 95%CI: 1.91-5.39), stage(HR = 1.89; 95%CI: 1.16-3.09), and resection(R)(HR = 2.82; 95%CI: 1.54-5.16) were selected as significant predictors and combined into a survival time predictive nomogram(C-index = 0.803; 95%CI: 0.766-0.839). High prediction accuracy(adjusted C-index = 0.797) was further verified via bootstrap validation. The calibration plot demonstrated good performance of the nomogram. ROC curve analysis revealed a high sensitivity and specificity. 5Size distribution of chemical elements and their source apportionment in ambient coarse, fine, and ultrafine particles in Shanghai urban summer atmosphere显示文摘Ambient coarse particles (diameter 1.8-10 μm), fine particles (diameter 0.1-1.8 μm), and ultrafine particles (diameter < 0.1 μm) in the atmosphere of the city of Shanghai were sampled during the summer of 2008 (from Aug 27 to Sep 08). Microscopic characterization of the particles was investigated by scanning electron microscopy coupled with energy dispersive X-ray spectroscopy (SEM/EDX). Mass concentrations of Si, P, S, Cl, K, Ca, Ti, V, Cr, Mn, Fe, Ni, Cu, Zn, As, Se, Br, Rb, Sr, and Pb in the size-resolved particles were quantified by using synchrotron radiation X-ray fluorescence (SRXRF). Source apportionment of the chemical elements was analyzed by means of an enrichment factor method. Our results showed that the average mass concentrations of coarse particles, fine particles and ultrafine particles in the summer air were 9.38 ± 2.18, 8.82 ± 3.52, and 2.02 ± 0.41 μg/m3, respectively. The mass percentage of the fine particles accounted for 51.47% in the total mass of PM10, indicating that fine particles are the major component in the Shanghai ambient particles. SEM/EDX results showed that the coarse particles were dominated by minerals, fine particles by soot aggregates and fly ashes, and ultrafine particles by soot particles and unidentified particles. SRXRF results demonstrated that crustal elements were mainly distributed in the coarse particles, while heavy metals were in higher proportions in the fine particles. Source apportionment revealed that Si, K, Ca, Fe, Mn, Rb, and Sr were from crustal sources, and S, Cl, Cu, Zn, As, Se, Br, and Pb from anthropogenic sources. 6Mechanisms of aid-leaching reagent calcium fluoride in the extracting vanadium processes from stone coal显示文摘The mechanisms of aid-leaching reagent calcium fluoride in the extracting vanadium processes were researched by comparing the blank extraction with the extraction with calcium fluoride as aid-leaching reagent. The leaching experiments were carried out under the conditions of 95 ℃, 6h, 1.5 mlg-1 , 15 vol % H2SO4 , and 5 % calcium fluoride (mass ratio of CaF2 to stone coal). The concentrations of hydrogen ion were adjusted from initial 1.82 and 4.79 mol L -1 to the pH of 1.80 ± 0.05 by 200 g L-1 lime milk, respectively. The solvent extraction experiments were carried out under the organic phase using 15 vol% D2EHPA, 5 vol% TBP, and 80 vol% sulfonated kerosene and O/A phase ratio of 1/4. The leaching rate of vanadium and six-stage countercurrent solvent extraction rate of vanadium can be remarkably improved from 66.71 to 92.97 % and 81.15 to 98.81 %, respectively, when calcium fluoride was added as aid-leaching reagent. In the leaching process, fluorine can enhance the breakage of the structure of vanadium-bearing minerals. Meanwhile, fluoride ion can combine with iron ion to generate acid-insoluble iron fluoride hydrate (Fe2F57H2O) and consume all ferric ions and a portion of ferrous ions. In adjusting pH value process, the massive impurities are decreased to form KAlSO4 and FeAlF5 . 9Fast magnetic reconstruction of the portal vein with allogeneic blood vessels in canines显示文摘BACKGROUND:The resection and reconstruction of large vessels,including the portal vein,are frequently needed in tumor resection.Warm ischemia before reconstruction might have deleterious effects on the function of some vital organs and therefore,how to reconstruct the vessels quickly after resection is extremely important.The present study was to introduce a new type of magnetic compression anastomosis(MCA)device to establish a quick non-suture anastomosis of the portal vein after resection in canines.METHODS:The new MCA device consists of a pair of titanium alloy and neodymium-ferrum-boron magnet(Ti-Nd Fe B)composite rings.The Nd Fe B magnetic ring as a core of the device was hermetically sealed inside the biomedical titanium alloy case.Twelve canines were divided into two groups:a MCA group in which the end-to-end anastomoses was made with a new device after resection in the portal vein and a traditional manual suture(TMS)group consisted of 6 canines.The anastomosis time,anastomotic patency and quality were investigated at week 24 postoperatively.RESULTS:The portal vein was reconstructed successfully in all of the animals and they all survived.The duration of portal vein anastomosis was significantly shorter in the MCA group than in the TMS group(8.16±1.25 vs 36.24±2.17 min,P<0.05).Portography and ultrasound showed that the blood flow was normal without angiostenosis or thrombosis in all of the canines.Hematoxylin-eosin staining and electron microscope scanning showed in contrast to the TMS group,MCA anastomotic intimal was much smoother with more regularly arranged endothelial cells at week 24 postoperatively.Shan-Pei Wang Xiao-Peng Yan Fei Xue Ding-Hui Dong Xu-Feng Zhang Feng Ma Hao-Hua Wang Yi Lv 2015Hepatobiliary & Pancreatic Diseases International2015,14,3:20

13Higher CO_2-insufflation pressure inhibits the expression of adhesion molecules and the invasion potential of colon cancer cells显示文摘AIM:To investigate the influence of CO2-insufflation pressure on adhesion,invasion and metastatic potential of colon cancer cells based on adhesion molecules expression.METHODS:With an in vitro artificial pneumoperitoneum model,SW1116 human colon carcinoma cells were exposed to CO2-insufflation in 5 different pressure groups:6 mmHg,9 mmHg,12 mmHg,15 mmHg and control group,respectively for 1 h.Expression of E-cadherin,ICAM-1,CD44 and E-selectin was measured at 0,12,24,48 and 72 h after CO2-insufflation using flow cytometry.The adhesion and invasion capacity of SW1116 cells before and after exposure to CO2-insufflation was detected by cell adhesion/invasion assay in vitro.Each group of cells was injected intraperitoneally into 16 BALB/C mice.The number of visible abdominal cavity tumor nodules,visceral metastases and survival of the mice were recorded in each group.RESULTS:The expression of E-cadherin,ICAM-1,CD44 and E-selectin in SW1116 cells were changed significantly following exposure to CO2 insufflation at different pressures(P<0.05).The expression of E-cadherin,CD44 and ICAM-1 decreased with increasing CO2-insufflation pressure.The adhesive/invasive cells also decreased gradually with increasing pressure as determined by the adhesion/invasion assay.In animal experiments,the number of abdominal cavity tumor nodules in the 15 mmHg group was also signif icantly lower than that in the 6 mmHg group(29.7±9.91 vs 41.7±14.90,P=0.046).However,the survival in each group was not statistically different.CONCLUSION:CO2-insufflation induced a temporary change in the adhesion and invasion capacity of cancer cells in vitro.Higher CO2-insufflation pressure inhibited adhesion,invasion and metastatic potential in vitro and in vivo,which was associated with reduced expression of adhesion molecules.Jun-Jun Ma Bo Feng Yi Zhang Jian-Wen Li Ai-Guo Lu Ming-Liang Wang Yuan-Fei Peng Wei-Guo Hu Fei Yue Min-Hua Zheng 2009World Journal of Gastroenterology2009,15,22:16
14Evaluation of spatial-temporal dynamics in surface water temperature of Qinghai Lake from 2001 to 2010 by using MODIS data显示文摘Lake surface water temperature(SWT) is an important indicator of lake state relative to its water chemistry and aquatic ecosystem,in addition to being an important regional climate indicator.However,few literatures involving spatial-temporal changes of lake SWT in the Qinghai-Tibet Plateau,including Qinghai Lake,are available.Our objective is to study the spatial-temporal changes in SWT of Qinghai Lake from 2001 to 2010,using Moderate-resolution Imaging Spectroradiometer(MODIS) data.Based on each pixel,we calculated the temporal SWT variations and long-term trends,compared the spatial patterns of annual average SWT in different years,and mapped and analyzed the seasonal cycles of the spatial patterns of SWT.The results revealed that the differences between the average daily SWT and air temperature during the temperature decreasing phase were relatively larger than those during the temperature increasing phase.The increasing rate of the annual average SWT during the study period was about 0.01℃/a,followed by an increasing rate of about 0.05℃/a in annual average air temperature.The annual average SWT from 2001 to 2010 showed similar spatial patterns,while the SWT spatial changes from January to December demonstrated an interesting seasonal reversion pattern.The high-temperature area transformed stepwise from the south to the north regions and then back to the south region from January to December,whereas the low-temperature area demonstrated a reversed annual cyclical trace.The spatial-temporal patterns of SWTs were shaped by the topography of the lake basin and the distribution of drainages.Fei XIAO Feng LING Yun DU Qi FENG Yi YAN Hui CHEN 2013Journal of Arid Land2013,5,4:16
15Effects of Honghua preserved amniotic membrane on scar healing in experimental glaucoma surgery显示文摘AIM:To investigate the efficacy and safety of Honghua preserved amniotic membrane(AM)for preventing scar formation of the filtering bleb in a rabbit model of glaucoma trabeculectomy surgery.METHODS:Totally 36 rabbits(36 eyes)were randomly divided into 3 groups:the experimental group(ocular trabeculectomy in combination with Honghua preserved AM transplantation),the control group(ocular trabeculectomy surgery in combination with AM implantation),and the blank group(single trabeculectomy).Clinical observations[including intraocular pressure(IOP),filtering blebs and complications],MassonTrichrome staining,real-time quantitative reverse transcription-polymerase chain reaction(real-time PCR),Western blot were performed on different time points(D1,D7,D14,D21 and D56)after the surgery.RESULTS:After operated for 14d,there were statistically significant differences in the filtering blebs compared to the situation before operation(P<0.05),whereas no statistically difference on that among three groups(P>0.05).After 21d,the IOP of experimental group was lowest(P<0.05).There was significant difference between control group and blank group(P<0.05).On postoperative D14,the mean number of fibroblasts in the experimental group was significantlylower(40.6±10.2)compared to those in the control group(54.4±10.8)and blank group(68.2±11.6)(P<0.05,respectively).The mean numbers of the macrophage in the experimental and control groups were respcitively significantly lower versus the blank group(P<0.05,P<0.05,respectively).Compared to that in blank group,the level of transforming growth factor-β(TGF-β1)expression in sclera and conjunctival areas was reduced in the experimental and control groups on protein and mRNA level(P<0.05),but not significant difference between these two groups(P>0.05).CONCLUSION:The trabeculectory surgery with Honghua preserved AM can control IOP,sustain the functional filtration bleb,inhibit the proliferation of fibroblasts and open the filtrating pathway on the rabbit glaucoma models.Yi Shao Yao Yu Qiu-Ping Liu Jing-Ming Li Fei Dong Xin Huang Chong-Gang Pei Ping Tu Hen-Hui Li Gui-Ping Gao 2014International Journal of Ophthalmology(English edition)2014,7,2:16
16Experimental exploration and research prospect of physical bases and functional characteristics of meridians显示文摘The physical basis of meridians and acupoints (acupuncture points) is in a complex system mainly of connective tissue and interwoven with the blood capillaries, nerves, lymph vessels,etc. Elements of Ca, P, K, Fe, Zn, Mn, etc are found concentrated in the deep connective tissue structures in locations corresponding to acupoints. On this physical basis, the specific waveband of high efficiency for transmission of infrared rays exists in the collagenous fibre in a liquid crystal state. 17Choledochojejunostomy with an innovative magneticcompressive anastomosis:How to determine optimalpressure?显示文摘AIM: To investigate the optimal magnetic pressure and provide a theoretical basis for choledochojejunostomy magnetic compressive anastomosis(magnamosis).METHODS: Four groups of neodymium-iron-boron magnets with different magnetic pressures of 0.1, 0.2, 0.3 and 0.4 MPa were used to complete the choledochojejunostomy magnamosis. Twenty-six young mongrel dogs were randomly divided into five groups: four groups with different magnetic pressures and 1 group with a hand-suture anastomosis. Serum bilirubin levels were measured in all groups before and 1 wk, 2 wk, 3 wk, 1 mo and 3 mo after surgery. Daily abdominal X-ray fluoroscopy was carried out postoperatively to detect the path and the excretion of the magnet. The animals were euthanized at 1 or 3 mo after the operation, the burst pressure was detected in each anastomosis, and the gross appearance and histology were compared according to the observation.RESULTS: The surgical procedures were all successfully performed in animals. However, animals of group D(magnetic pressure of 0.4 MPa) all experienced complications with bile leakage(4/4), whereas half of animals in group A(magnetic pressure of 0.1 MPa) experienced complications(3/6), 1 animal in the manual group E developed anastomotic stenosis, and animals in group B and group C(magnetic pressure of 0.2 MPa and 0.3 MPa, respectively) all healed well without complications. These results also suggested that the time required to form the stoma was inversely proportional to the magnetic pressure; however, the burst pressure of group A was smaller than those of the other groups at 1 mo(187.5 ± 17.7 vs 290 ± 10/296.7 ± 5.7/287.5 ± 3.5, P < 0.05); the remaining groups did not differ significantly. 18Effects of Environmental Factors on Tree Seedling Regeneration in a Pine-oak Mixed Forest in the Qinling Mountains, China显示文摘Natural seedling regeneration and tree establishment are affected by various environmental factors.In this study,we established eight,eight,six,and four independent forest stands(each stand was further divided into five subplots) respectively based on the altitudinal gradient,stand density,slope location,and slope aspect to investigate the effects of environmental factors on tree seedling regeneration in a pine-oak mixed forest.The results indicated that the seedling density was significantly higher at altitudes of 1,283 m to 1,665 m,whereas the sapling density did not differ with altitudes.The seedling and sapling density decreased significantly at 1,835 m.The seedling densities on the upper slopes were much higher than those on the middle and lower slopes,whereas the sapling density had no difference.The sapling density decreased gradually from the southwest(20°-75°) whereas it increased on the shady slopes to the northeast(40°).The seedling density increased from southwest(20°) to northeast(40°).The seedling and sapling densities increased with the stand density(850 trees ha-1to 1,525 trees ha-1) whereas the sapling density was significantly lower in stands(1,900 trees ha-1).Principal components analysis showed that the slope aspect and stand density had more important roles in tree regeneration in this study region compared with the other two factors.Therefore,our findings suggest that it will be beneficial to keep stands at a moderate density on shady slopes.Appropriate thinning of higher density stands is also expected to promote the natural regeneration of pine-oak mixed forest.YU Fei WANG De-xiang SHI Xiao-xiao YI Xian-feng HUANG Qing-ping HU You-ning 2013Journal of Mountain Science2013,10,5:13
19Toripalimab combined with lenvatinib and GEMOX is a promising regimen as first-line treatment for advanced intrahepatic cholangiocarcinoma:a single-center,single-arm,phase 2 study显示文摘Advanced intrahepatic cholangiocarcinoma(ICC)has a dismal prognosis.Here,we report the efficacy and safety of combining toripalimab,lenvatinib,and gemcitabine plus oxaliplatin(GEMOX)as first-line therapy for advanced ICC.Thirty patients with pathologically confirmed advanced ICC received intravenous gemcitabine(1 g/m2)on Days 1 and 8 and oxaliplatin(85 mg/m2)Q3W for six cycles along with intravenous toripalimab(240 mg)Q3W and oral lenvatinib(8 mg)once daily for one year.The expression of programmed death-ligand 1(PD-L1)and genetic status was investigated in paraffin-embedded tissues using immunohistochemistry and whole-exome sequencing(WES)analysis.The primary endpoint was the objective response rate(ORR).Secondary outcomes included safety,overall survival(OS),progression-free survival(PFS),disease control rate(DCR)and duration of response(DoR).As of July 1,2022,the median follow-up time was 23.5 months,and the ORR was 80%.Twenty-three patients achieved partial response,and one achieved complete response.Patients(21/30)with DNA damage response(DDR)-related gene mutations showed a higher ORR,while patients(14/30)with tumor area positivity≥1(PD-L1 staining)showed a trend of high ORR,but without significant difference.The median OS,PFS,and DoR were 22.5,10.2,and 11.0 months,respectively.The DCR was 93.3%.Further,56.7%of patients experienced manageable grade≥3 adverse events(AEs),commonly neutropenia(40.0%)and leukocytopenia(23.3%).In conclusion,toripalimab plus lenvatinib and GEMOX are promising first-line regimens for the treatment of advanced ICC.A phase-III,multicenter,double-blinded,randomized study to validate our findings was approved by the National Medical Products Administration(NMPA,No.2021LP01825).Guo-Ming Shi Xiao-Yong Huang Dong Wu Hui-Chuan Sun Fei Liang Yuan Ji Yi Chen Guo-Huan Yang Jia-Cheng Lu Xian-Long Meng Xin-Ying Wang Lei Sun Ning-Ling Ge Xiao-Wu Huang Shuang-Jian Qiu Xin-Rong Yang Qiang Gao Yi-Feng He Yang Xu Jian Sun Zheng-Gang Ren Jia Fan Jian Zhou 2023Signal Transduction and Targeted Therapy2023,8,4:12
20Synthesis and curing of a novel amino-containing phthalonitrile derivative显示文摘A novel phthalonitrile derivative containing an amino group, 3,5-bis(3,4-dicyanophenoxy)aniline (CPA), was synthesized via a nucleophilic displacement of 4-nitrophthalonitrile and 5-aminoresorcinol hydrochloride. The structure of CPA was confirmed by Fourier transform infrared spectra (FT-IR) and nuclear magnetic resonance (1H NMR).
