1mTOR/miR-145-regulated exosomal GOLM1 promotes hepatocellular carcinoma through augmented GSK-3b/MMPs显示文摘Golgi membrane protein 1(GOLM1/GP73)is a serum marker of hepatocellular carcinoma(HCC).We have previously shown that mTOR promoted tumorigenesis of HCC through stimulating GOLM1 expression.In this study,we demonstrated that the mammalian target of rapamycin(mTOR)was a negative regulator of microRNA-145(miR-145)expression.miR-145 inhibited GOLM1 expression by targeting a coding sequence of GOLM1 gene.GOLM1 and miR-145 were inversely correlated in human HCC tissues.GOLM1-enriched exosomes activated the glycogen synthase kinase-3β/matrix metalloproteinases(GSK-3β/MMPs)signaling axis of recipient cells and accelerated cell proliferation and migration.In contrast,miR-145 suppressed tumorigenesis and metastasis.We suggest that mTOR/miR-145/GOLM1 signaling pathway should be targeted for HCC treatment.Xiaochen Gai Bufu Tang Fangming Liu Yuting Wu Fang Wang Yanling Jing Fuqiang Huang Di Jin Ling Wang Hongbing Zhang 2019Journal of Genetics and Genomics2019,46,5:13
2Organic carbon accumulation capability of two typical tidal wetland soils in Chongming Dongtan,China显示文摘We measured organic carbon input and content of soil in two wetland areas of Chongming Dongtan(Yangtze River Estuary) to evaluate variability in organic carbon accumulation capability in different wetland soils.Observed differences were investigated based on the microbial activity and environmental factors of the soil at the two sites.Results showed that the organic carbon content of wetland soil vegetated with Phragmites australis(site A) was markedly lower than that with P.australis and Spartina alterniflora(site B).Sites differences were due to higher microbial activity at site A,which led to higher soil respiration intensity and greater carbon outputs.This indicated that the capability of organic carbon accumulation of the site B soils was greater than at site A.In addition,petroleum pollution and soil salinity were different in the two wetland soils.After bio-remediation,the soil petroleum pollution at site B was reduced to a similar level of site A.However,the culturable microbial biomass and enzyme activity in the remediated soils were also lower than at site A.These results indicated that greater petroleum pollution at site B did not markedly inhibit soil microbial activity.Therefore,differences in vegetation type and soil salinity were the primary factors responsible for the variation in microbial activity,organic carbon output and organic carbon accumulation capability between site A and site B.Shiping Zhang Lei Wang Jiajun Hu Wenquan Zhang Xiaohua Fu Yiquan Le Fangming Jin 2011Journal of Environmental Sciences2011,23,1:11
3Internet of Things to network smart devices for ecosystem monitoring显示文摘Smart, real-time, low-cost, and distributed ecosystem monitoring is essential for understanding and managing rapidly changing ecosystems. However, new techniques in the big data era have rarely been introduced into operational ecosystem monitoring, particularly for fragile ecosystems in remote areas.We introduce the Internet of Things(IoT) techniques to establish a prototype ecosystem monitoring system by developing innovative smart devices and using IoT technologies for ecosystem monitoring in isolated environments. The developed smart devices include four categories: large-scale and nonintrusive instruments to measure evapotranspiration and soil moisture, in situ observing systems for CO2 and d13 C associated with soil respiration, portable and distributed devices for monitoring vegetation variables, and Bi-CMOS cameras and pressure trigger sensors for terrestrial vertebrate monitoring. These new devices outperform conventional devices and are connected to each other via wireless communication networks. The breakthroughs in the ecosystem monitoring IoT include new data loggers and longdistance wireless sensor network technology that supports the rapid transmission of data from devices to wireless networks. The applicability of this ecosystem monitoring IoT is verified in three fragile ecosystems, including a karst rocky desertification area, the National Park for Amur Tigers, and the oasis-desert ecotone in China. By integrating these devices and technologies with an ecosystem monitoring information system, a seamless data acquisition, transmission, processing, and application IoT is created. 4Deficient Rnf43 potentiates hyperactive Kras-mediated pancreatic preneoplasia initiation and malignant transformation显示文摘Background:Largely due to incidental detection,asymptomatic pancreatic cystic le-sions(PCLs)have become prevalent in recent years.Among them,intraductal papillary mucinous neoplasm(IPMN)infrequently advances to pancreatic ductal adenocarcinoma(PDAC).Conservative surveillance versus surgical intervention is a difficult clinical decision for both caregivers and PCL patients.Because RNF43 loss-of-function mutations and KRAS gain-of-function mutations concur in a subset of IPMN and PDAC,their biological significance and therapeutic potential should be elucidated.Methods:Pancreatic Rnf43 knockout and Kras activated mice(Rnf43^(−/−);Kras^(G12D))were generated to evaluate their clinical significance in pancreatic pre-neoplastic initiation and malignant transformation.Results:Loss of Rnf43 potentiated the occurrence and severity of IPMN and PDAC in oncogenic Kras mice.The Wnt/β-catenin signaling pathway was activated in pan-creatic Kras^(G12D)and Rnf43 knockout mice and the PORCN inhibitor LGK974 blocked pancreatic IPMN initiation and progression to PDAC accordingly.Conclusions:Rnf43 is a tumor suppressor in the prevention of pancreatic malignant transformation.This genetically reconstituted autochthonous pancreatic Rnf43^(−/−);Kras^(G12D)preclinical cancer model recapitulates the pathological process from pancreatic cyst to cancer in humans and can be treated with inhibitors of Wnt/β-catenin signaling.Since the presence of RNF43 and KRAS mutations in IPMNs predicts future development of advanced neoplasia from PCLs,patients with these genetic anomalies warrant surveillance,surgery,and/or targeted therapeutics such as Wnt/β-catenin inhibitors.Xian Zhou Zhichao Sun Mengdi Zhang Xiaoyu Qu Shuhui Yang Lianmei Wang Yanling Jing Li Li Weiwei Deng Fangming Liu Jin Di Jie Chen Jian Wu Hongbing Zhang 2022Animal Models and Experimental Medicine2022,5,1:3
5A numerical study of non-Darcy flow in EGS heat reservoirs during heat extraction显示文摘Underground non-Darcy fluid flow has been observed and investigated for decades in the petroleum industry. It is deduced by analogy that the fluid flow in enhanced geothermal system (EGS) heat reservoirs may also be in the non-Darcy regime under some conditions. In this paper, a transient 3D model was presented, taking into consideration the non-Darcy fluid flow in EGS heat reservoirs, to simulate the EGS long-term heat extraction process. Then, the non-Darcy flow behavior in water- and supercritical CO2 (SCCO2)-based EGSs was simulated and discussed. It is found that non-Darcy effects decrease the mass flow rate of the fluid injected and reduce the heat extraction rate of EGS as a flow resistance in addition to the Darcy resistance which is imposed to the seepage flow in EGS heat reservoirs. Compared with the water-EGS, the SCCO2-EGS are more prone to experiencing much stronger non-Darcy flow due to the much larger mobility of the SCCO2. The non-Darcy flow in SCCO2- EGSs may thus greatly reduce their heat extraction performance. Further, a criterion was analyzed and proposed to judge the onset of the non-Darcy flow in EGS heat reservoirs. The fluid flow rate and the initial thermal state of the reservoir were taken and the characteristic Forchheimer number of an EGS was calculated. 7Metal sulfide quantum dots-aggregated PAMAM dendrimer for cadmium ion-selective electrode-based immunoassay of alpha-fetoprotein显示文摘An ion-selective electrode(ISE)-based immunoassay has been innovatively designed for the sensitive detection of liver cancer biomarker(alpha-fetoprotein,AFP),using metal sulfide quantum dot(QD)-based nano labels.Cd S QDs-aggregated PAMAM dendrimer(QD-DE)was first synthesized and functionalized with polyclonal rabbit anti-human AFP antibodies.Thereafter,a sandwich immunoreaction was implemented on monoclonal mouse anti-human AFP antibody-coated microplate by using antibody-functionalized QD-DE as the secondary antibody.Accompanying the immunocomplex,subsequent potentiometric detection of cadmium ion dissolved from the QD-DE under acidic condition was conducted on a portable cadmium ion-selective electrode(Cd-ISE).Results revealed that the electrode potential of the Cd-ISE increased with the increment of AFP concentration from 0.1 to 100 ng m L^(-1)at a detection limit(LOD)of 68 pg m L^(-1).The relative standard deviations(RSD)were below9.09%and 10.54%for the intra-and inter-assay,respectively.Additionally,six human serum specimens were determined on CdISE-based immunosensor by using commercial human AFP ELISA kit as the reference,and gave good relationship between two methods.Importantly,Cd-ISE-based immunoassay offers the promise for simple and cost-effective screening of disease-related biomarkers.Qunfang Li Jing Jin Fangming Lou Dianping Tang 2018Science China Chemistry2018,61,6:2

9Universally improving effect of mixed electron donors on the CO_2 fixing efficiency of non-photosynthetic microbial communities from marine environments显示文摘The universality of improved CO2 fixing upon the addition of mixed electron donors(MEDs)composed of Na2 S,NO2-,and S2O32-to non-photosynthetic microbial communities(NPMCs)obtained from 12 locations in four oceans of the world was validated. The CO2 fixing efficiencies of NPMCs were universally enhanced by MED compared with those obtained using H2 alone as electron donor,with average increase of about 276%. An increase in microbial inoculation concentration could increase the net amount of CO2 fixing to853.34 mg/L in the presence of MED. NO2-and S2O32-may play the roles of both electron acceptor and electron donor under aerobic conditions,which may improve the energy utilization efficiency of NPMC and enhance the CO2 fixation efficiency. The sequence determination of 16 S ribosomal deoxyribonucleic acid(rDNA) from 150 bacteria of NPMC showed that more than 50% of the bacteria were symbiotic and there were many heterotrophic bacteria such as Vibrio natriegens. These results indicate that NPMC acts as a symbiotic CO2 fixing system. 10Catalytic transfer hydrogenation of levulinate ester intoγ-valerolactone over ternary Cu/ZnO/Al2O3 catalyst显示文摘An effective catalytic transfer hydrogenation (CTH) process of bio-based levulinate esters into γ-valerolactone (GVL) was explored over ternary Cu/ZnO/Al2O3 catalyst which was prepared by coprecipitation method and could be sustainably used. As a result, quantitative conversion of ethyl levulinate (EL) and 99.0% yield of GVL were obtained in the CTH process using i-PrOH as hydrogen donor. The Cu/ZnO/Al2O3 catalyst with high-surface-area could be reused at least four times without the loss of catalytic activity. Furthermore, the structure and properties of Cu/ZnO/Al2O3 catalyst was characterized through XRD, BET, SEM, TEM and H2-TPR. 11Photocatalytic reduction of carbon dioxide by titanium oxide- based semiconductors to produce fuels显示文摘To tackle the crisis of global warming, it is imperative to control and mitigate the atmospheric carbon dioxide level. Photocatalytic reduction of carbon dioxide into solar fuels furnishes a gratifying solution to utilize and reduce carbon dioxide emission and simultaneously generate renewable energy to sustain the societies. So far, titanium oxide-based semiconductors have been the most prevalently adopted catalysts in carbon dioxide photoreduction. This mini-review provides a general summary of the recent progresses in titanium oxide- catalyzed photocatalytic reduction of carbon dioxide. It first illustrates the use of structural engineering as a strategy to adjust and improve the catalytic performances. Then, it describes the introduction of one/two exogenous elements to modify the photocatalytic activity and/or selectivity. 12Improved two-step hydrothermal process for acetic acid production from carbohydrate biomass显示文摘An improved two-step process for converting carbohydrate biomass to acetic acid under hydrothermal conditions is proposed. The first step consists of the production of lactic acid from carbohydrate biomass, and the second step consists of conversion of the lactic acid obtained in the first step to acetic acid using CuO as an oxidant. The results indicated that CuO as an oxidant in the second step can significantly improve the production of high-purity acetic acid from lactic acid, and the maximum yield of acetic acid was 61%, with a purity of 90%. The yield of acetic acid obtained using the improved two-step hydrothermal process from carbohydrate biomass, such as glucose, cellulose and starch, was greater than that obtained using traditional two-step process with H2O2 orO2. In addition, a proposed pathway for the production of acetic acid from lactic acid in the second step with CuO was also discussed. 16Enhanced photocatalytic CO_(2) hydrogenation with wide-spectrum utilization over black TiO_(2) supported catalyst显示文摘Light-driven conversion of CO_(2)into chemicals/fuels is a desirable approach for achieving carbon neutral-ity using clean and sustainable energy.However,its scale-up application is restricted due to insufficient efficiency.Herein,we present a photothermal catalytic hydrogenation of CO_(2)into CH_(4)over Ru/black TiO_(2) catalysts,aiming to achieve the synergistic use of light and heat in solar energy during CO_(2)conversion.Owing to the desirable spectral response ability and photothermal conversion performance of black TiO_(2),an efficient combination of photocatalysis and thermocatalysis has been established.The CO_(2)hydrogena-tion was significantly accelerated because of the increased catalyst surface temperature enabled by the photothermal effect of black TiO_(2).Simultaneously,through the in situ X-ray photoelectron spectroscopy(XPS)observation,electron-rich Ru nanoparticles was achieved based on the photo-induced excitation,thereby providing more negative hydride to improve nucleophilic attack to the CO_(2),obtaining the CH_(4) yield of 93.8%.Binbin Jin Xin Ye Heng Zhong Fangming Jin Yun Hang Hu 2022Chinese Chemical Letters2022,33,2:1

20Catalytic conversion of ethyl lactate to 1,2-propanediol over CuO显示文摘An efficient conversion of biomass-derived ethyl lactate to 1,2-propanediol(1,2-PDO) over CuO was investigated.Among the catalysts we tested, CuO, Cu2 O and Co showed excellent catalytic activity for the conversion of ethyl lactate to 1,2-PDO in water, and CuO was more active and gave the best result. The 1,2-PDO yield of 93.6% was achieved when Zn acted as a reductant.
